This study was conducted to evaluate the hair-growth promotion effect in C57BL/6 mice of a new substance mixed with Allium cepa (red)-bioconversion extract and Angelica gigas Nakai. The ethanol extract of Allium cepa (red) was bioconverted through the use of the Bacillus subtilis KJ-3 (BS3) strain, which was named Red-BCQ. The quercetin content of Red-BCQ increased by about 7.4-fold after bioconversion. Angelica gigas Nakai extract (Agnex) contains a large amount of coumarins such as decursin (D) and decursinol angelate (DA). A 1 mg portion of Agnex contained 0.4146 mg of D and 0.3659 mg of DA. Minoxidil has been known to promote hair growth. In this study, the hair-growth promotion effects of Red-BCQ, Agnex, and a mixture of both Red-BCQ and Agnex were compared with 5% minoxidil. Twenty-five mice were divided into five experimental groups including saline (CON), 5% minoxidil (PCON), Red-BCQ (RA), Agnex (AG), and a Red-BCQ-Agnex mixture (RAG)-treated group. Samples were administered orally once a day at a fixed time for 4 weeks. Hair growth was monitored by photograph at 7, 14, 21, and 28 days. We also observed 5α-reductase, alkaline phosphatase (ALP), γ-glutamyl transpeptidase (γ-GT), insulin-like growth factor (IGF)-1, transforming growth factor (TGF)-β1, antioxidant enzyme, and the hair follicles of the skin tissue. In all the results, the RAG-administered group showed greater antioxidative and hair-growth promotion effects than the other groups. These data suggest that RAG has potent stimulating activity on hair growth in C57BL/6 mice.
Guibi-tang is a traditional medicine used for the treatment of colds. We investigated the levels of several compounds in Guibi-tang before and after fermentation with Lactobacillus and tested their ability to inhibit angiotensin-converting enzyme. Six known compounds (decursin, decursinol angelate, nodakenin, liquiritin, formononetin, and 6-gingerol) and 2 unidentified compounds were detected in Guibi-tang (GB) and fermented Guibi-tang (FGB) by an established HPLC-DAD method. The levels of the 6 known compounds were decreased after fermentation. FGB showed more potent inhibition of angiotensin-converting enzyme activity than GB. In conclusion, fermentation with Lactobacillus affects the content of several compounds in GB and improves its angiotensin-converting enzyme inhibitory activity.

In this study, Gami-Sumiwon (GS; Cynanchi wilfordii R., Angelica gigantis R., Lycium chinense M., Betula platyphylla S., Cryptotympana atrata F. and Carthamus tinctorius L.) and/or Sinomenium acutum R. (SC) was extracted with 70% ethanol or water. And We investigated the antioxidant activity effect of GS±SC. The following experimental techniques were used to evaluate the antioxidant efficacy of GS±SC. HPLC chromatogram, heavy metal content, ABTS/DPPH radical scavenging analysis, SOD-like activity assay, FACS, and NO assay. As a result of the experiment, the sinomenine content was found to be higher in DW extracts, and decursin was found to be higher in 80% ethanol extracts. And, the amount of heavy metals in all extracts was below the standard value. ABTS, and DPPH radical scavenging activity was identified that GS±SC(EtOH) was found to have a higher scavenging activity than GS±SC(DW). But, SOD showed the opposite result. No cytotoxicity of GS was observed on Raw 264.7 cells at concentration of 1~100 ㎍/㎖. The ROS production was significantly decreased that GS±SC(DW) was found to have a higher scavenging activity than GS±SC(EtOH). However, NO production showed the opposite result. Looking at the results of SOD and ROS analysis, SC does not seem to have a function of prevention. SC is thought to have an effect on the removal of free radicals generated after oxidative stress. This result objectively confirmed the antioxidant effect of GS±SCs. We will continue to conduct in-depth research. Therefore, it is believed that the possibility of using GS±SCs as a functional material can be established. The more diverse the objectives, the higher the value of GS utilization is thought to be.

Angelica tenuissima, also known as Ligusticum tenuissimum, is classified as a food-related plant and has been used as traditional medicines treating headache and anemia in Asia. However, its anti-melanogenic effect has not been reported in detail. When the extract of Angelica tenuissima (ATE) was prepared by the extraction with 70% EtOH at $80^{\circ}C$ (final yield = 22%), the contents of decursin and Z-ligustilide in ATE were determined 0.06% and 8.43%, respectively. Total flavonoid and phenolic content in mg ATE were $5.52{\pm}0.07{\mu}g$ quercetin equivalents and $237.27{\pm}13.24{\mu}g$ gallic acid equivalents, respectively. Antioxidant capacity of ATE determined by DPPH and ABTS assay was increased with a dose dependent manner up to $1000{\mu}g/m{\ell}$. The amount of melanin synthesis followed by ${\alpha}-melanocyte$ stimulating hormone on B16F10 cells were significantly reduced in the presence of ATE (250 to $1000{\mu}g/m{\ell}$, p<0.05). ATE (125 to $1000{\mu}g/m{\ell}$, p<0.05) suppressed the tyrosinase activity but did not show any significant effect on ${\alpha}-glucosidase$ activity at the same condition. Taken together, ATE possesses tyrosinase inhibitory potential with significant antioxidant capacities. These effects of ATE might be involved in suppression of melanin synthesis, at least, in B16F10 cells. The anti-melanogenic potential of ATE will provide an insight into developing a new skin whitening product.

Background: The objective of this study was to make colloidal dispersions of the active compounds of radix of Angelica gigas Nakai that could be charaterized as nano-composites using hot melt extrusion (HME). Food grade hydrophilic polymer matrices were used to disperse these compound in aqueous media. Methods and Results: Extrudate solid formulations (ESFs) mediated by various HPMCs (hydroxypropyl methylcelluloses) and Na-Alg polymers made from ultrafine powder of the radix of Angelica gigas Nakai were developed through a physical crosslink method (HME) using an ionization agent (treatment with acetic acid) and different food grade polymers [HPMCs, such as HP55, CN40H, AN6 and sodium alignate (Na-Alg)]. X-ray powder diffraction (XRD) analysis confirmed the amorphization of crystal compounds in the HP55-mediated extrudate solid formulation (HP55-ESF). Differential scanning calorimetry (DSC) analysis indicated a lower enthalpy (${\Delta}H=10.62J/g$) of glass transition temperature (Tg) in the HP55-ESF than in the other formulations. Infrared fourier transform spectroscopy (FT-IR) revealed that new functional groups were produced in the HP55-ESF. The content of phenolic compounds, flavonoid (including decursin and decursinol angelate) content, and antioxidant activity increased by 5, 10, and 2 times in the HP55-ESF, respectively. The production of water soluble (61.5%) nano-sized (323 nm) particles was achieved in the HP55-ESF. Conclusions: Nano-composites were developed herein utilizing melt-extruded solid dispersion technology, including food grade polymer enhanced nano dispersion (< 500 nm) of active compounds from the radix of Angelica gigas Nakai with enhanced solubility and bioavailability. These nano-composites of the radix of Angelica gigas Nakai can be developed and marketed as products with high therapeutic performance.
Objectives To check marker content for appropriate quality control of Woohwangchungsimwon sterilized to ensure microbiological safety and to observe antioxidant activity for any changes in efficacy. Methods To measure any effects of sterilization on the effective compounds, 8 ingredients of Woohwangchungsimwon were screened for any changes in marker content using HPLC-DAD. Using the colorimetric method on the microplate reader any changes in total phenolic compound and flavonoid levels were observed. Antioxidant activity was measured using the DPPH, ABTS, and FRAP. Results Of the ingredients of Woohwangchungsimwon, 8 were subject to quantitative analysis before and after sterilization. 21.6 mg and 1.93 mg of Glycyrrhizin was found in Glycyrrhiza uralensis Fischer pre and post sterilization, respectively. Decursin found in Angelica gigas Nakai increased from 0.16 mg to 0.29 mg after sterilization. Bilirubin found in Gallstone of Bostaurusvar. domesticus increased from 0.24 mg to 0.33 mg. Cinnamic acid found in Cinnamomum cassia Blume increased from 0.02 mg to 0.05 mg. Ginsenoside Rb1 found in Panax ginseng C. A. Meyer decreased from 0.02 mg to 0.14 mg. Paeoniflorin found in Paeonia lactiflora Pallas increased from 1.05 mg to 1.13 mg. Amygdalin found in Armeniacae Amarum Semen increased from 2.68 mg to 2.83 mg. L-muscone found in Musk increased from 0.63 mg to 0.76 mg. As for total phenolic compound and total flavonoid content, there was a 1.22 and 4.15-fold increase. DPPH and ABTS increased by 20.45% and 20.69%, respectively. FRAP activity was 2.78 times more active post stabilization. Conclusions This study confirmed that high temperature high pressure steam sterilization, a method used to ensure microbiological safety of Woohwangchungsimwon, does not affect marker content; in other words, does not affect quality of the Woohwangchungsimwon. It could also be seen that total phenolic compound and flavonoid content increased after sterilization. An antioxidant activity test showed that there was significantly increased activity of antioxidants.

This research was performed to develop a new material consisting of a mixture of Red Allium cepa (RA) Cucurbita moschata duch (CM), and Angelica gigas Nakai (AG). RA and CM have low storage stability because of their high moisture content. Therefore, their major components were extracted and used for the research after a content analysis. In order to overcome these limitations, the quercetin from RA, ${\beta}-carotene$ from CM, and decursin/decursinol angelate (D/DA) from AG were separately extracted, and the biochemical activity of each extract and mixture was compared. RA was bioconverted by the Bacillus subtillis KJ-3 (BS3) after ethanol extraction. After bioconversion, the quercetin content of RA was increased by 128.9%. ${\beta}-carotene$ was detected in the CM ethanol extract and its content was very low concentrations at 0.2 mg/g. The AG ethanol extract (1 mg) contained 0.4146 mg and 0.3659 mg of D/DA, respectively. The purity of the D/DA was found to be about 78%. The flavonoid and polyphenol content of each extract and their mixtures (mixture 1 (RA:CM:AG = 5:2:3), mixture 2 (RA:CM: AG = 3:5:2), and mixture 3 (RA:CM:AG = 3:2:5)) were measured. In addition, the cell survival rate, anti-inflammatory activity, and antioxidant ability were also evaluated. In all the results, the antioxidant activity of mixture 3 was most effective. Therefore, these findings provide basic data for future food development using a 3:2:5 mixture of RA, CM, and AG.

Background: We measured the weight characteristics, intercellular space volume as a proportion of root volume, and quantities of the functional component in Angelica gigas Nakai, collected from four cultivation areas in Pyeongchang, including Tapdong-ri and SangjinBu-ri, and tested the results for correlations. Methods and Results: Dry weight was the highest in the Tapdong-ri group ($13.37{\pm}0.13g$) and the lowest in the SangjinBu-ri group ($11.90{\pm}0.57g$). The percentage of intercellular spaces within the roots, determined by micro-CT camera, was the highest in the Tapdong-ri group ($6.44{\pm}0.76%$) and the lowest in the SangjinBu-ri group ($9.01{\pm}2.15%$). The quantity of functional components were the highest in the Tapdong-ri group ($116.13{\pm}9.53mg/g$) and the lowest in the SangjinBu-ri group ($99.77{\pm}23.23mg/g$). Conclusions: The dry weight of A. gigas significantly correlated with the volume and percentage of intercellular space. An increase in the dry weight resulted in a negative correlation between the volume and ratio of the intercellular spaces. The volume of intercellular space correlated negatively with levels of the functional components, this was significant in the case of the nodakenin, decursin and decursinol angelate content (p < 0.016).
Korean angelica(Angelica gigas Nakai), a oriental medicinal plants is important and in great demand in Korea. This crop is a biennial which is to die after flower, so as could not harvest the root. A new variety 'Anpung' was developed from the selection of the local collecting population by medicinal crop team at NCES in 2001. This showed stable bolting during its cropping year and have high quality and yield potential. The color is greenish purple in stem and red purple in flower. It was longer in plant height and root length, thicker in root diameter, more leave and latheral roots comparing with the check variety 'Manchu'. The dry root was 72 g/plant showing heavier than check variety of 65 g/plant. It showed such low bolting rate as 0.5%, on the other hand 49.3% in landrace when it cultured by transplanting with heating nursery seedlings, and $0{\sim}15%$ with field nursery seedlings. The dry root yield of the variety was averaged 3,630 kg/ha out yielding the check variety by 11% at the three regional yield test from 1999 to 2001. Finally, 'Anpung' showed higher content of extracts up to 46.3% in 'Anpung' and 45% in check variety, decursin and decursinol contents was 3.48% and 2.79%, showing slightly higher than check variety.
