Extraction of Pectinesterase from Jalapeno Chili Pepper (Capsicum annuum) and Its Thermal Stability

  • The effect of NaCl solution concentration (from 0 to 3 M) on the extraction of pectinesterase (PE) from jalapeno chili pepper (Capsicum annuum) was studied by determining its solubilization degree from the chili tissue. All concentrations of the salt favored the solubilization of PE in jalapeno chili pepper, compared to that in water. Maximum enzyme activity was obtained with NaCl 2.0 M. The effect of temperature on the PE activity of jalapeno chili pepper in the extracts was also studied. The PE residual activity of jalapeno pepper was 75% after 60 min of incubation at $55^{\circ}C$ and 10% at $75^{\circ}C$. At $85-95^{\circ}C$, PE residual activity was 5% after 5 min of incubation.

  • In the cold and temperate regions of Korea the icing and ice coats on 25 kV overhead contact wire during winter is a very serious problem. This generates shocks at the mechanical interface of the collecting strips of the pantograph and the contact wire and extra electrical resistance, which may affect quality of current collection at the contact wire / collecting strips of pantograph interface. De-icing operations should be performed just before train operation to avoid the formation of another ice layer. This paper presents temperature analysis of the de-icing system which could be applied to the overhead contact wire of railways.

Warm Dust and Gas of Massive YSOs Revealed by Herschel PACS Spectroscopy

  • As part of the Herschel key program "Water in Star-forming Regions with Herschel (WISH)", PACS imaging spectroscopy data have been taken toward ten massive young stellar objects (YSOs): four high mass protostellar objects (HMPOs), two hot molecular cores (HMCs), and four ultracompact HII regions (UCHIIs). The spectra cover a broad range of wavelengths (55 to 210 micron) presenting various atomic and molecular lines as well as excellent dust thermal continua. By fitting the continua utilizing a modified black-body formula we estimate mass-weighted temperature and column density distributions of warm dust and find that UCHII regions are warmer and HMCs are more deeply embedded than the other types. We also estimate rotational temperature and column density distributions of warm CO gas using the rotational diagram analysis. In addition, based on the comparison of high J CO line fluxes to the RATRAN estimates of central heating envelope models, we find that majority of warm CO is originated from bipolar outflow shocks.

IEC 61131-3 국제표준언어 기반 임베디드 모션제어기의 개발 (Development of an Embedded Motion Controller based on the IEC 61131-3 International Standard Language)

  • 본 논문에서는 IEC 61131-3 국제표준언어에 근거한 PLC 프로그래밍 개발환경을 지원해 주는 임베디드 모션 제어기를 개발하였다. 이 모션제어기에는 모션시스템 개발자가 PC 개발환경은 물론, PLC 개발환경에서도 손쉽게 모션제어 프로그래밍이 가능하도록 IEC 61131-3 개발 도구인 CoDeSys가 탑재되었으며, 직선 및 원호보간 제어와 같은 다양한 모션제어 함수들이 PLCopen 표준에 부합되는 모션 펑션블록으로 구현되었다. 또한 실시간 운영체제하에서 이더넷 기반 원격제어 및 모션 프로그래머를 위한 모션 시뮬레이터가 구현되었다.

A Genome-wide Approach for Functional Analysis Using Rice Mutant

  • Rapid extension of genomic database leads to the remarkable advance of functional genomics. This study proposes a novel methodology of functional analysis using 5-methyltrytophan (5 MT) mutant together with their 2-DE analysis and public microarray database. A total of 24 proteins was changed in 5 MT mutant and four remarkably different expressed proteins were identified. Among them, three spots were converted to Affymetrix probe. A total of 155 microarray samples from Gene Expression Omnibus (GEO) in NCBI was retrieved and followed by constructing gene co-expression networks over a broad range of biological issues through Self-Organising Tree Algorithm. Three co-expressing gene clusters were retrieved and each functional categorization with differential expression pattern was exhibited from 5 MT resistance mutant rice. It was indicated new co-expression networks in the mutant. This study suggests that on investigating possibility which correspond 2-DE to microarray database with their full potential.

Synthesis, morphology and electrochemical applications of iron oxide based nanocomposites

  • The development of hybrid systems comprising nanoparticles and polymers is an opening pathway for engineering nanocomposites exhibiting outstanding mechanical, optical, electrical, and magnetic properties. Among inorganic counterpart, iron oxide nanoparticles (IONP) exhibit high magnetization, controllable surface chemistry, spintronic properties, and biological compatibility. These characteristics enable them as a platform for biomedical applications and building blocks for bottom-up approaches, such as the layer-by-layer (LbL). In this regard, the present study is addressed to investigate IONP synthesised through co-precipitation route (average diameter around 7 nm), with either positive or negative surface charges, LbL assembled with sodium sulfonated polystyrene (PSS) or polyaniline (PANI). The surface and internal morphologies, and electrochemical properties of these nanocomposites were probed with atomic force microscopy, UV-vis and Raman spectroscopy, scanning electron microscopy, cross-sectional transmission electron microscopy, and electrochemical measurements. The nanocomposites display a globular morphology with IONP densely packed while surface dressed by polyelectrolytes. The investigation of the effect of thermal annealing (300 up to $600^{\circ}C$) on the oxidation process of IONP assembled with PSS was performed using Raman spectroscopy. Our findings showed that PSS protects IONP from oxidation/phase transformation to hematite up to $400^{\circ}C$. The electrochemical performance of nanocomposite comprising IONP and PANI were investigated in $0.5mol{\times}L^{-1}$ $Na_2SO_4$ electrolyte solution by cyclic voltammetry and chronopotentiometry. Our findings indicate this structure as promising candidate for potential application as electrodes for supercapacitors.

Hippo Signal Transduction Mechanisms in T Cell Immunity

  • Hippo signaling pathways are evolutionarily conserved signal transduction mechanisms mainly involved in organ size control, tissue regeneration, and tumor suppression. However, in mammals, the primary role of Hippo signaling seems to be regulation of immunity. As such, humans with null mutations in STK4 (mammalian homologue of Drosophila Hippo; also known as MST1) suffer from recurrent infections and autoimmune symptoms. Although dysregulated T cell homeostasis and functions have been identified in MST1-deficient human patients and mouse models, detailed cellular and molecular bases of the immune dysfunction remain to be elucidated. Although the canonical Hippo signaling pathway involves transcriptional co-activator Yes-associated protein (YAP) or transcriptional coactivator with PDZ motif (TAZ), the major Hippo downstream signaling pathways in T cells are YAP/TAZ-independent and they widely differ between T cell subsets. Here we will review Hippo signaling mechanisms in T cell immunity and describe their implications for immune defects found in MST1-deficient patients and animals. Further, we propose that mutual inhibition of Mst and Akt kinases and their opposing roles on the stability and function of forkhead box O and β-catenin may explain various immune defects discovered in mutant mice lacking Hippo signaling components. Understanding these diverse Hippo signaling pathways and their interplay with other evolutionarily-conserved signaling components in T cells may uncover molecular targets relevant to vaccination, autoimmune diseases, and cancer immunotherapies.

Responses of Guava Plants to Inoculation with Arbuscular Mycorrhizal Fungi in Soil Infested with Meloidogyne enterolobii

  • In the Northeast of Brazil, expansion of guava crops has been impaired by Meloidogyne enterolobii that causes root galls, leaf fall and plant death. Considering the fact that arbuscular mycorrhizal Fungi (AMF) improve plant growth giving protection against damages by plant pathogens, this work was carried out to select AMF efficient to increase production of guava seedlings and their tolerance to M. enterolobii. Seedlings of guava were inoculated with 200 spores of Gigaspora albida, Glomus etunicatum or Acaulospora longula and 55 days later with 4,000 eggs of M. enterolobii. The interactions between the AMF and M. enterolobii were assessed by measuring leaf number, aerial dry biomass, $CO_2$ evolution and arbuscular and total mycorrhizal colonization. In general, plant growth was improved by the treatments with A. longula or with G. albida. The presence of the nematode decreased arbuscular colonization and increased general enzymatic activity. Higher dehydrogenase activity occurred with the A. longula treatment and $CO_2$ evolution was higher in the control with the nematode. More spores and higher production of glomalin-related soil proteins were observed in the treatment with G. albida. The numbers of galls, egg masses and eggs were reduced in the presence of A. longula. Inoculation with this fungus benefitted plant growth and decreased nematode reproduction.

Assimilation of Peptides and Amino Acids and Dissimilation of Lactate During Submerged Pure Cultures of Penicillium camembertii and Geotrichum candidum

  • The behavior of Penicillium camembertii and Geotrichum candidum growing in submerged pure cultures on simple (glutamate) or complex (peptones) substrates as nitrogen and carbon sources and lactate as a second carbon source was examined. Similar to the behavior previously recorded on a simple substrate (glutamate), a clear differentiation between the carbon source and the energy source was also shown on peptones and lactate during P. camembertii growth, since throughout growth, lactate was only dissimilated, viz., used for energy supply by oxidation into $CO_2$, whereas peptides and amino acids from peptones were used for carbon (and nitrogen) assimilation. Because of its deaminating activity, G candidum preferred peptides and amino acids to lactate as energy sources, in addition to being assimilated as carbon and nitrogen sources. From this, on peptones and lactate, G candidum grew faster than P. camembertii (0.19 and 0.08 g/l/h, respectively) by assimilating the most readily utilizable peptides and amino acids; however, owing to its lower proteolytic activity, the maximum biomass was lower than that of P. camembertii (3.7 and 5.5 g/l, respectively), for which continuous proteolysis and assimilation of peptides were shown.
