Isolation and Characterization of Calmodulin Gene from Panax ginseng C. A. Meyer

  • $Ca^{2+}$ and calmodulin (CaM), a key $Ca^{2+}$ sensor in all eukaryotes, have been implicated for defense responses of plants. Eukaryotic CaM contains four structurally and functionally similar $Ca^{2+}$ domains named I, II, III and IV. Each $Ca^{2+}$ binding loop consists of 12 amino acid residues with ligands arranged spatially to satisfy the octahedral symmetry of $Ca^{2+}$ binding. To investigate the altered gene expression and the role of CaM in ginseng plant defense system, cDNA clone containing a CaM gene, designated PgCaM was isolated and sequenced from Panax ginseng. PgCaM, which has open reading frame of 450 nucleotides predicted to encode a precursor protein of 150 amino acid residues. Its sequence shows high homologies with a number of other CaMs, with more similarity to CaM of Daucus carota (AAQ63461). The expression of PgCaM in different P. ginseng organs was analyzed using real time PCR. The results showed that PgCaM expressed at different levels in young leaves, shoots, and roots of 3-week-old P. ginseng. In addition, the expressions of PgCaM under different abiotic stresses were analyzed at different time intervals.

  • 당근 진탕배양세포로부터 분리한 원형질체에 A.rhi-zogenes를 처리하여 bacteriad의 부착과 부착과정에 영향을 주는 여러 요인들을 조사하였다. 또한 당근뿌리 절편에 strain이 다른 bacteria와 heat-inactivated bacteria를 전처리한 후 A.rhizogenes strain A4를 접종하여 식물세포 표면에 있어서 bacterial specific receptor sites존재 유무를 살펴보았다. Bacteria의 부착은 pH 6.0, 24-35$^{\circ}C$ 조건하에서 120분동안 원형질체막에 유의적인 증가를 나타내었으며, 또한 세포벽합성에 따르는 bacteriaqnckr의 영향은 볼 수 없었다. Strain이 다른 bacteria나 heat-inactivated bacteria의 전처리에 의한 A..rhizogenes의 당근조직에서 hairy root형성 억제 현상은 식물세포 표면에 specific receptor sites가 존재 한다기보다는 부착에 있어서 전처리한 bacteria의 경쟁적 저해작용으로 형질전환이 다소 억제되는 것으로 사료되어졌다.

벼멸구에 대한 식물추출물의 살충활성과 기피효과 (Insecticidal Activities and Repellent Effects of Plant Extracts against the Brown Planthopper (Nilaparvata lugens Stäl))

  • 식물추출물(20과 35종)을 사용하여 미량국소처리방법과 분무방법으로 벼멸구 암컷성충에 대한 살충활성과 기피효과에 대해 실험을 수행하였다. 살충활성과 기피효과의 결과는 식물종과 그 부위에 따라 다양한 차이를 보였다. 인면과수, 구리광, 미국자리공의 줄기 추출물, 그리고 취춘과 석류나무의 잎+줄기 추출물이 벼멸구에 대하여 높은 살충활성을 보였다. 식물추출물들의 기피효과는 남학슬의 전부위 추출물, 캐슈나무의 열매추출물, 석류나무와 등나무의 잎+줄기 추출물이 높은 기피활성을 보였다. 이 실험에 사용한 식물추출물들에서 기피효과가 높은 식물추출물들의 살충활성이 불규칙적으로 나타나 기피효과와 살충률의 관계가 성립되지 않았다. 따라서 처리된 식물추출물의 벼멸구에 대한 살충활성과 기피 효과는 상관관계가 없음을 알 수 있다.

제주특산물을 이용한 향토국수의 개발 (Development of Regional Noodles Using Agricultural and Fishery Products of Cheju Island)

  • Acceptable fish noodles of better nutritional and sensory values than conventional noodle made only wheat flour were prepared by mixing wheat flour and ground tile fish (Branchioste gus japonicus). In oder to mask a distinctive fish odor, various herbs and spices, such as ginger (Zingiber officinale), curry (Chalcas koenigii), nutmeg (Myristica Pagrans), garlic (Ailium sativum), black pepper (Piper nigrum), lemon (Cirtus limon) or sodachi (Citrus sudachi) were added to the basic tile fish noodle. Also, for the purpose of enhancing eating qual ify and nutritional value of basic noodle were combined some agricultural or fishery products of Cheju island, citrus fruits; danyooja (Citrus danyooga), hagul (C. natfudaidai), medical plants; angelica utilis (Angelica keiskei), ginseng (Panax ginseng), cactus (Opuntia dillenii), vegetable; carrot (Daucus carota), dropwort (Oenanthe jnvanica), seaweeds; fusiforme (Hizkia fusiforme), gulfweed (Sargaceae hlvelium). The optimal mixing ratio for preparing the basic tile fish noodle amounted to ground tile fish 135 g: tile fish stock 139 $m\ell$: wheat flour 450 g: salts 10 g. The mixture of curry powder (2.5 g) and nutmeg powder (2.5 g) was proved to be the most effective combinations for masking unfavorable fish odor. The optimal amounts of materials to be added to the prepared basic noodle were 25 g citrus zest and 80m1 citrus juice for citrus fruits noodles, and ca. 140 g puree for noodles from medical plants, vegetables and seaweeds, respectively. The preference score obtained from consumer preference test, on a 9-point scales, were in oder of i) danyooja > carrot, angelica, ginseng > fusiforme, ii) hagul > dropwort, cactus > gulfweed. The shelf-life of tile fish noodle based on bacterial counting was estimated to be 7-days at 5$^{\circ}C$.

  • Agorobacterium rhizogenes에 의하여 유도된 당근(Daucus carota L. )의 hairy root를 배양하였으며 opine 유무에 대한 형질전환 확인 및 배지조성을 달리하여 성장률에 따른 색소함량을 비교하였고, 재분화된 식물체의 형태적 차이를 관찰하여 몇가지 결론을 얻었다. Hairy root는 균 접종 2-4주 후에 형성층 부위를 중심으로 유도되었다. 유도된 hairy root의 초기 배양에는 R.C.M 배지가 적합하였으며 MSO(2, 4-D $10^{-4} ml/ l, pH6, sucrose 5%, 질소원 0.03M 등)에서 최대 성장을 보여주었고 성장의 증가에 따른 색소의 형성은 비교적 안정하였다. 재분화된 식물체는 정상 식물체에 비하여 형태적으로 차이를 나타내었으며 형질전환된 hairy root 및 재분화된 식물체에서 mannopine 분석으로 Ri-plasmid에 의한 형질전환이 이루어졌음을 확인하였다.

Growth-Inhibiting Effects of Vegetable Extracts on Beneficial and Harmful Human Intestinal Bacteria

  • Ethanol extracts of 38 vegetables were subjected to an in vitro screening for their growth-inhibitory activities towards Bifidobacterium bifidum, B. longum, Clostridium perfringens, Lactobacillus acidophilus, L. casei, and Escherichia coli using paper disc agar diffusion methods under anaerobic conditions. The responses varied with both bacterial strain and vegetable species. In a test with 20 mg/disc, Zingiber officinale extracts showed significant growth-inhibitory responses against B. bifidum, and strong inhibitions against L. casei were detected in the extracts of Chrysanthemum coronarium var. spatiosum and Lactuca sativa. The extracts of Allium sativum, Capsicum annuum, L. esculentum, L. esculentum var. cerasiforme, and Z. officinale showed strong inhibitory activities against C. perfringens, while moderate growth-inhibitory responses were observed in the extracts of C. ffutescens, Cucurbita moschata, Daucus carota var. sativa, and Rubus coreanus. However, all vegetable extracts showed no inhibitions against B. longum, L. acidophilus, and E. coli. In tests with 5 mg/disc, moderate inhibitions were observed in the extracts of C. coronarium var. spatiosum and L. sativa against L. casei and Z. officinale against B. bifidum. Vegetables extracts, except for C. coronarium var. spatiosum, L. sativa, and Z. officinale, did not affect the growth of beneficial bacteria. Strong inhibitory responses against C. perfringens were detected in the extracts of C. annuum and L. esculentum var. cerasiforme. Daily intake of vegetables may be important in the prevention of human diseases caused by the intestinal bacteria.

Biological Control of Meloidogyne hapla Using an Antagonistic Bacterium

  • We examined the efficacy of a bacterium for biocontrol of the root-knot nematode (RKN) Meloidogyne hapla in carrot (Daucus carota subsp. sativus) and tomato (Solanum lycopersicum). Among 542 bacterial isolates from various soils and plants, the highest nematode mortality was observed for treatments with isolate C1-7, which was identified as Bacillus cereus based on cultural and morphological characteristics, the Biolog program, and 16S rRNA sequencing analyses. The population density and the nematicidal activity of B. cereus C1-7 remained high until the end of culture in brain heart infusion broth, suggesting that it may have sustainable biocontrol potential. In pot experiments, the biocontrol efficacy of B. cereus C1-7 was high, showing complete inhibition of root gall or egg mass formation by RKN in carrot and tomato plants, and subsequently reducing RKN damage and suppressing nematode population growth, respectively. Light microscopy of RKN-infected carrot root tissues treated with C1-7 showed reduced formation of gall cells and fully developed giant cells, while extensive gall cells and fully mature giant cells with prominent cell wall ingrowths formed in the untreated control plants infected with RKNs. These histopathological characteristics may be the result of residual or systemic biocontrol activity of the bacterium, which may coincide with the biocontrol efficacies of nematodes in pots. These results suggest that B. cereus C1-7 can be used as a biocontrol agent for M. hapla.

Effect of Carrot Intake in the Prevention of Gastric Cancer: A Meta-Analysis

  • Purpose: Gastric cancer is the third leading cause of cancer-related mortality, with the incidence and mortality being higher in men than in women. Various studies have shown that eating carrots may play a major role in the prevention of gastric cancer. We conducted a meta-analysis to determine the relationship between carrot consumption and gastric cancer. Materials and Methods: We searched multiple databases including PubMed, Cochrane Library, Scopus, ScienceDirect, and Persian databases like Scientific Information Database (SID) and IranMedx. The following search terms were used: stomach or gastric, neoplasm or cancer, carcinoma or tumor, and carrot. Statistical analyses were performed using Comprehensive Meta Analysis/2.0 software. Results: We retrieved 81 articles by searching the databases. After considering the inclusion and exclusion criteria, 5 articles were included in this study. The odds ratio (OR) obtained by fixed effects model showed that a 26% reduction in the risk of gastric cancer has been associated with the consumption of carrots) OR=0.74; 95% confidence interval=0.68~0.81; P<0.0001). According to funnel graph, the results showed that the possibility of a publication bias does not exist in this study. Conclusions: The findings of this study showed an inverse relationship between the consumption of carrots and the risk of gastric cancer.

한국에서의 오이모자익 바이러스의 기주범위에 관한 연구 (Studios on the Host Range of Cucumber Mosaic Virus in Korea)

  • 가. CMV의 기주조사결과, 43과 145종을 공시하여 27과 71종이 기주로 판명되었으며 그중 24두 57종이 Systemic mosaic을 나타내었다. 나. CM V의 새로운 기주로 확인된 식물은 쇠별꽃 쇠무릎, 아게라탐, 중대가리풀, 천인국, 지칭개, 씀바귀, 그늘취, 유채, 다닥냉이, 수염가래꽃, 좀명아주, 실사초, 깨풀, 새콩, 벌노랑이, 팥, 큰기린초, 들깨풀, 들깨, 개곽량, 아마, 주름잎, 버어베나의 24종이 판명되었다. 다. 다른 연구자의 보고에는 감수성으로 나타났으나 본 시험에서 음성으로 나타난 식물은 양파, 맨드라미, 당근, 쑥, 과꽃, 망초, 해바라기, 만수국, 봉선화, 양배추, 배추, 스위트알리슘, 무, 고구마, 대두, 강남콩, 나팔백합, 양귀비, 수수, 밀, 옥수수, 소루쟁이, 채송화 등 23종이었다.

Antioxidant and Anti-inflammatory Properties of Raw and Processed Fruits and Vegetables

  • Reactive oxygen species (ROS) generated from metabolic reactions cause oxidative DNA damage, which results in oxidative tissue injury. Therefore, there is an increasing demand in the intake of high antioxidant sources in order to maintain a healthy environment in cells. In this study, we investigated the antioxidant and anti-inflammatory activities of Malus domestica (apple), Pyrus communis L. (pear), Daucus carota L. (carrot), Brassica oleracea var. (broccoli), Brassica oleracea var. capitata (cabbage), and Raphanus sativus L. (radish) obtained from the local market. Since these are common fruits and vegetables that are widely consumed, we aimed to investigate their beneficial properties, placing particular emphasis on their antioxidant and anti-inflammatory properties. The samples were processed via an indirect heating method and their properties were compared to their raw forms. Based on DPPH and ABTS assays, processed samples showed better antioxidant activities when compared to raw samples and processed pear samples exhibited the best antioxidant activity. The anti-inflammatory activities of the samples were also investigated in LPS-treated RAW 264.7 cells. mRNA expression of pro-inflammatory mediators and cytokines (iNOS, COX-2, $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6) was assessed using RT-PCR. As expected, processed samples exhibited better iNOS inhibition when compared to their raw forms and processed broccoli and cabbage samples exhibited outstanding anti-inflammatory effects. The samples, up to 1 mg/mL concentration, did not exhibit cytotoxicity against RAW 264.7 cells as demonstrated by cell viability assays. Altogether, processed broccoli and cabbage samples exhibited the strongest anti-inflammatory properties.
