Inhibitory Effect of Solvent Fraction of Various Kinds of Kimchi on Ultraviolet B Induced Oxidation and Erythema Formation of Hairless Mice Skin (김치종류별 용매 획분의 자외선 B 조사에 의해 유도된 피부산화 및 홍반 생성 억제 효과)

  • Kimchi is well known as a traditional Korean food containing various antioxidative compounds and it has been demonstrated that dietary kimchi inhibit the tissue oxidation and aging in many related studies. Ultraviolet B (UVB) radiation is the most potent among the many related factor to skin photo-aging. This experiment is designed to elucidate the inhibitory effect of solvent fractionation of various kimchi on UVB induced oxidation and erythema formation. Among solvent fractions, phenol content mostly existed in dichloromethane / ethylacetate fraction of buchu kimchi, dichloromethane fraction of Korean cabbage kimchi and ethylacetate / dichloromethane fraction of mustard leaf kimchi. Free radical scavenging activities were higher in dichloromethane and ethylacetate fraction from buchu kimchi, dichloromethane fraction from Korean cabbage kimchi, and ethylacetate fraction from mustard leaf kimchi. When the damage of photo-oxidation by UVB irradiation was tested in the presence of solvent fractions of kimchi, dichloromethane / ethylacetate fraction of buchu kimchi, ethylacetate fraction from Korean cabbage and from mustard leaf kimchi showed the higher protective activities than others. The erythema formations which were induced by UVB irradiation were decreased in solvent fractions of kimchi that hate higher antioxidant activities. In conclusion, kimchi solvent fractions having antioxidant compounds mostly inhibited photo-oxidation and erythema formation by UV-irradiation, therefore kimchi consumption can retard skin aging due to the presence of antioxidative compounds.

A Study on Fire Explosion Characteristics via Physico-chemical Analysis of Petroleum Residues (석유 부산물의 물리화학적 분석을 통한 화재폭발 특성연구)

  • In this study, the physical and chemical analyses of petroleum residues (pyrolized fuel oil, PFO) were conducted and major components were selected to investigate their fire and explosion characteristics. Major component distribution areas of the PFO were identified via the GC-SIMDIS and MALDI-TOF analyses. In addition, the qualitative analysis of major component distribution areas was performed by GC-MS analysis. Major components of pyrolysis residue were selected based on the results of various analyses such as EA, SARA and TGA. As a result, benzene, toluene and xylene were selected as major components. Finally, the process hazard analysis software tool (PHAST) analysis was performed to investigate the range of maximum damage effect in case of fire and explosion. Toluene presented the highest risk due to the radiation effect of $227kW/m^2$ and 118 m in the case of jet fire. Xylene and benzene showed the maximum radiant heat values of 114 and $151kW/m^2$, respectively. It was also confirmed from the analysis of pasquill stability and wind speed that the radiant heat increased up to 55% according to wind speed in benzene, which was considered to be a main factor increasing the influence range.

Schisandrae Fructus ethanol extract attenuates particulate matter 2.5-induced inflammatory and oxidative responses by blocking the activation of the ROS-dependent NF-κB signaling pathway

  • BACKGROUND/OBJECTIVES: Schisandrae Fructus, the fruit of Schisandra chinensis Baill., has traditionally been used as a medicinal herb for the treatment of various diseases, and has proven its various pharmacological effects, including anti-inflammatory and antioxidant activities. In this study, we investigated the inhibitory effect of Schisandrae Fructus ethanol extract (SF) on inflammatory and oxidative stress in particulate matter 2.5 (PM2.5)-treated RAW 264.7 macrophages. MATERIALS/METHODS: To investigate the anti-inflammatory and antioxidant effects of SF in PM2.5-stimulated RAW 264.7 cells, the levels of pro-inflammatory mediator such as nitric oxide (NO) and prostaglandin E2 (PGE2), cytokines including interleukin (IL)-6 and IL-1β, and reactive oxygen species (ROS) were measured. To elucidate the mechanism underlying the effect of SF, the expression of genes involved in the generation of inflammatory factors was also investigated. We further evaluated the anti-inflammatory and antioxidant efficacy of SF against PM2.5 in the zebrafish model. RESULTS: The results indicated that SF treatment significantly inhibited the PM2.5-induced release of NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. SF also attenuated the PM2.5-induced expression of IL-6 and IL-1β, reducing their extracellular secretion. Moreover, SF suppressed the PM2.5-mediated translocation of nuclear factor-kappa B (NF-κB) from the cytosol into nuclei and the degradation of inhibitor IκB-α, indicating that SF exhibited anti-inflammatory effects by inhibiting the NF-κB signaling pathway. In addition, SF abolished PM2.5-induced generation of ROS, similar to the pretreatment of a ROS scavenger, but not by an inhibitor of NF-κB activity. Furthermore, SF showed strong protective effects against NO and ROS production in PM2.5-treated zebrafish larvae. CONCLUSIONS: Our findings suggest that SF exerts anti-inflammatory and antioxidant effects against PM2.5 through ROS-dependent down-regulating the NF-κB signaling pathway, and that SF can be a potential functional substance to prevent PM2.5-mediated inflammatory and oxidative damage.

  • High impervious surfaces increase the surface runoff during rainfall and reduces the underground infiltration thereby leading to water cycle distortion. The distortion of water cycle causes various urban environmental problems such as urban flooding, drought, water pollutant due to non-point pollution runoff, and water ecosystem damage. Climate change intensified seasonal biases in urban rainfall and affected urban microclimate, thereby increasing the intensity and frequency of urban floods and droughts. Low impact development(LID) technology has been applied to various purposes as a technique to reduce urban environmental problems caused by water by restoring the natural water cycle in the city. This study evaluated the contribution of hydrologic characteristics and water cycle recovery after LID application using long-term monitoring results of various LID technology applied in urban areas. Based on the results, the high retention and infiltration rate of the LID facility was found to contribute significantly to peak flow reduction and runoff delay during rainfall. The average runoff reduction effect was more than 60% at the LID facility. The surface area of the LID facility area ratio(SA/CA) was evaluated as an important factor affecting peak flow reduction and runoff delay effect.

  • The military technology currently receiving the most attention is the hypersonic missile. hypersonic is faster than the speed of sound or Mach 5+. The vast majority of the ballistic missiles that it inspired achieved hypersonic speeds as they fell from the sky. Rather than speed, today's renewed attention to hypersonic weapons owes to developments that enable controlled flight. These new systems have two sub-varieties: hypersonic glide vehicles and hypersonic cruise missiles. Hypersonic weapons could challenge detection and defense due to their speed, maneuverability, and low altitude of flight. The fundamental question of this study is: 'What effect will the hypersonic missile have on the maritime strategy?' It is quite prudent to analyze and predict the impact of technology in the development stage on strategy in advance. However, strategy is essential because it affect future force construction. hypersonic missiles act as a limiting factor in securing sea control. The high speed and powerful destructive power of the hypersonic missile are not only difficult to intercept, but it also causes massive ship damage at a single shot. As a result, it is analyzed that the Securing sea control will be as difficult as the capacity of sea denial will be improved geographically and qualitatively. In addition, the concept of Fortress Fleet, which was criticized for its passive strategy in the past, could be reborn in a modern era. There are maritime power projection/defence, SLOC attack/defence in exploiting sea control. The effects of hypersonic missiles on exploiting sea control could be seen as both limiting and opportunity factors.

Estimation of the Hydrological Design Frequency of Local Rivers Using Bayesian Inference and a Sensitivity Analysis of Evaluation Factors (평가인자 가중치에 대한 베이지안 추론과 민감도 분석을 통한 적정 하천설계빈도 결정)

  • In Korea, annual precipitation and its variability have gradually increased since modern meteorological observations began, and the risk of disasters has also been increasing due to significant regional variations and recent abnormal climate conditions. Given that damage from storms and floods mainly occurs around rivers, it is crucial to determine the appropriate design frequency for river-related projects. This study examined existing design practices used to determine hydrological design frequencies and suggested a new method to determine appropriate design frequencies. The study collected available data pertaining to seven evaluation factors, specifically the basin areas, shape parameters, channel slopes, stream orders, backwater effect reaches, extreme rainfall frequencies, and urbanized flood inundation areasfor 413 local rivers in Chungcheongnam-do in Korea. The estimated weights for areas of extreme rainfall frequencies and urbanized flood inundation were found to be 18, having a great effect on determining the design frequency. Compared with the established design frequency in previous government reports, the estimated design frequency increased for 255 rivers and decreased for 158 rivers.

Evaluation of Immune Enhancing Activity of Luthione, a Reduced Glutathione, in RAW 264.7 Macrophages (RAW 264.7 대식세포에서 환원형 glutathione인 luthione의 면역 증강 활성 평가)

  • Although glutathione (GSH) has been shown to play an important role in the prevention of oxidative damage as an antioxidant, studies on immune regulation by it have not been properly conducted. In this study, we investigated whether luthione®, a reduced GSH, has an immune enhancing effect in murine macrophage RAW 264.7 cells. The results of flow cytometry and immunofluorescence experiments indicated that luthione increased phagocytic activity, a representative function of macrophages, compared to the control cells. According to the results of the cytokine array, the expression of interleukin (IL)-5, IL-1β, and IL-27 was significantly increased in the luthione-treated cells. Luthione also enhanced the production of tumor necrosis factor-α and IL-1β through increased expression of their proteins, and increased release of the immune mediators such as nitric oxide (NO) and prostaglandin E2 was associated with increased expression of inducible NO synthase and cyclooxygenase-2. In addition, the expression of cluster of differentiation 86, an M1 macrophage marker, was dramatically enhanced in RAW 264.7 cells treated with luthione. Furthermore, as a result of heat map analysis, we found that cytokine signaling 1/3-mediated signal transducer and activator of transcription/Janus tyrosine kinase signaling pathway was involved in the immunomodulatory effect by luthione. In conclusion, our data suggested that luthione could act as a molecular regulator in M1 macrophage polarization and enhance immune capacity by promoting macrophage phagocytic function.

The Association between Pulmonary Function Test Result and Combustible Cigarette Smoking or Electrical Cigarette Smoking in Korean Adults : Using the 2014-2019 Korean national health and nutrition examination survey data (한국 성인에서 일반담배 또는 가열 전자담배를 이용한 흡연 형태와 폐 기능 검사 결과와의 관련성: 2014-2019년도 국민건강영양조사 자료를 이용하여)

  • Purpose : Smoking is a major factor in chronic obstructive pulmonary disease (COPD), but the effect of electrical cigarette smoking on COPD development is still uncertain. This study aimed to compare the functions of airways and lungs exposed to combustible cigarettes and electrical cigarettes based on the pulmonary function test (PFT) results from the Korean National Health and Nutrition Examination Survey (NHANES). Methods : This study used data from 8,942 participants with PFT results out of 47,309 total subjects from the 6th to 8th Korean NHANES (2014-2015, 2016-2018, and 2019, respectively). Individuals with diseases such as cancer, ex-smokers, and dual tobacco users were excluded. The PFT results were analyzed according to the COPD diagnostic criteria. After adjusting for confounding variables, a complex sample generalized linear model ANOVA test was performed to investigate the association between PFT results and combustible smoker or electrical cigarette user groups. Results : In an analysis based on the obstructive ventilatory disorders (forced expiratory volume in 1 second[FEV1]/forced vital capacity[FVC]<.7), combustible cigarette smokers showed a 3.46 times higher risk of COPD compared to non-smokers, while electrical cigarette smokers exhibited no significant difference in terms of COPD-related risks compared to non-smokers. FEV1 showed a negative relation with combustible cigarette smokers as reported elsewhere (B=-.07, p<.001). FEV1/FVC was negatively related to both combustible cigarette smokers (B=-.03, p<.001) and electrical cigarette smokers (B=-.02, p<.001). Conclusion : FEV1/FVC decreases were observed in the long-term exposure to both combustible and electrical cigarettes. The lower FEV1 in the combustible cigarette group implies the worsening of the severity of COPD, suggesting more damage to the airways and lungs in the short term. Therefore, the temporary electrical cigarettes use for the transition period in order to smoking cessation potentially aids to reduce the harmful effect of combustible cigarettes in COPD development.

Antioxidant and Cytoprotective Effects of Socheongja and Socheong 2, Korean Black Seed Coat Soybean Varieties, against Hydrogen Peroxide-induced Oxidative Damage in HaCaT Human Skin Keratinocytes (HaCaT 인간 피부 각질세포에서 과산화수소 유도 산화 손상에 대한 소청자 및 소총2호의 항산화 및 세포보호 효능)

  • Black soybeans are used as food sources as well as for traditional medicines because they contain an abundance of natural phenolic compounds. In this study, total phenolic contents (TPCs) of Korean black seed coat soybean varieties Socheongja (SCJ), Socheong 2 (SC2) and Cheongja 2 (CJ2) as well as their antioxidant capacities were investigated. Among them, TPCs were abundantly present in the order of CJ2$H_2O_2$-stimulated HaCaT human keratinocytes. Our results revealed that treatment with SCJ and SC2 prior to $H_2O_2$ exposure significantly increases the viability of HaCaT cells, indicating that the exposure of HaCaT cells to SCJ and SC2 conferred a protective effect against oxidative stress. SCJ and SC2 also effectively inhibited $H_2O_2$-induced apoptotic cell death through the blocking of mitochondrial dysfunction. SCJ and SC2 also attenuated the phosphorylation of Histone H2AX. Furthermore, they effectively induced the levels of thioredoxin reductase (TrxR) 1, a potent antioxidant enzyme, which is associated with the induction of nuclear transcription factor erythroid-2-like factor 2 (Nrf2); however, the protective effects of SCJ and SC2 were significantly reversed by Auranofin, a TrxR inhibitor. These results indicate that they have protective activity through the blocking of cellular damage related to oxidative stress via the Nrf2 signaling pathway. In conclusion, our study indicated that SCJ and SC2 might potentially serve as novel agents for the treatment and prevention of skin disorders caused by oxidative stress.

Cytoprotective Effects and Gene Expression Patterns Observed Based on the Antioxidant Activity of Lonicera japonica Extract (금은화 추출물의 항산화 효과를 통한 세포 보호효과 및 유전자 발현 양상)

  • In this study, based on the antioxidative effects in organic solvent fractions obtained from the main methanolic extract of L. japonica, the protective cellular effects and gene expression patterns of ethyl acetate fractions on $H_2O_2$-induced Raw 264.7 cell death ($IC_{50}$) were analyzed. The antioxidant activity of the fractions measured using DPPH free radical scavenging activity increased in a dose-dependent manner, and the $ED_{50}$ exhibited the highest $39.56{\mu}g/ml$ in the ethyl acetate fraction. In addition, the ethyl acetate fractions' cell viability on $H_2O_2$-induced Raw 264.7 cell damage increased in a concentration-dependent manner, showed a visible cell survival rate of 82.49% at a concentration of $100{\mu}g/ml$. The gene expression patterns related to the ethyl acetate fractions' cytoprotective effect in $H_2O_2$-induced Raw 264.7 cell damage presented similar patterns to those of BHA. In comparative analysis for antioxidant activity-related genes affected by ethyl acetate fractions and BHA in $H_2O_2$-induced Raw 264.7 cells, both ethyl acetate fractions and BHA showed very similar gene expression patterns, but the gene expression level of the heme oxygenase 1 (Hmox1) gene making antioxidant enzymes in cells was four times higher in ethyl acetate fractions than BHA. In inflammation-related genes in $H_2O_2$induced Raw 264.7 cells, the T-box transcription factor (Tbx21) gene was expressed about two times more frequently in the ethyl acetate fraction treatment group, while it was expressed half as frequently in the BHA treatment group.
