  • A study was designed to investigate the sorption characteristics of binary mixtures of NaCl and sucrose or glucose stored at various relative humidities ranging from 46% to 92%. At low relative humidity below RH 65%, the sorption equilibrium was easily achieved, whereas at higher relative humidity values over 73%, all of the mixtures tended to cintinously absorb moisture with increase in storage time. A linear equation of log $({\frac{dw}{dt}})$ = a log(t) + log(b) was found to be valid between the sorption rate and storage time with respect to storage humidities. In sucrose-NaCl mixture, the slope showed a increasing tendency as the percentage of NaCl increased in the mixture, while that of glucose-NaCl mixture failed to show a definite trend. Plateaus were obtained when the amount of water absorbed was plotted on the X axis and the percent composition of mixture on the Y axis at different storage time. The shape of plateau was varied with respect to the kind of sugar-NaCl mixture, composition of the mixture and relative humidities. A linearity was found between log(1-Aw) and the amount of water absorbed over the Aw range 0.73-0.92 and the slope was affected by the kind and composition of sugar-NaCl mixtures.

  • Panax Ginseng C.A. Meyer has been known for hundreds of years as the most valuable drug having mysterious effects among all the herbal medicines and plants in Korea. Also, many experimental studies have been performed recently that the various effects were identified and applied clinically. So we attempted an experimental study on the effect of ginsenoside Rg1 mixtures in an isolated rat heart with the use of the Langendorff model. The objective of this study was to determine whether this ginsenoside Rg1 mixtures would protect the myocardial injury after ischemic arrest and reperfusion. Isolated rat hearts were allowed to equilibrate for 20 minutes and were then subjected to 15 minutes of normothermic ischemia. After this ischemic period, isolated rat hearts were allowed to reperfusion for 10 minutes(Ischemic Group). In other group , isolated rat hearts were perfused for 60 minutes continuously with normothermia( Normothermic Group). Hemodynamic and biochemical parameters such as heart rate, left ventricular pressure, +dp/dt max, coronary blood flow and cardiac enzymes were measured during initial perfusion, ischemia, reperfusion period (Ischemic group) and 20, 40 and 60 minutes after continuous perfusion(Normothermic group). After completion of the experiment, this data was evaluated and the following results were obtained. 1. Heart rates showed an increase in both ischemic and normothermic experimental groups, but statistically significant differences were not identified. 2. LVP(Left Ventricular Pressure) showed statistically significant differences in both ischemic and normothermic experimental groups(p<0.005, p<0.01). 3. +dp/dt max showed statistically significant differences in both ischemic and normothermic experimental groups(p<0.01, p<0.01). 4. There were no statistically significant differences in coronary blood flow and cardiac cenzymes in all groups, but experimental groups seemed to have better protection and recovery. These results suggest that ginsenoside Rg1 mixtures has a protective effect on the myocardial injury after ischemia and reperfusion.

  • A Bi$_{4}$Ti$_{3}$O$_{12}$ (BIT) thin film is prepared by sol-gel method using acetate precursors and evaluated whether it could be applied to NVFRAM (Non-Volatile Ferroelectric RAM). The drying and the annealing temperature are 40$0^{\circ}C$ and $650^{\circ}C$, respectively and they are determined from the DT-TG (Differential Thermal-Thermal Gravimetric) analysis. The BIT thin film deposited on Pt/Ta/SiO$_{2}$/Si substrate shows orthorhombic perovskite phase. The grain size and the surface roughness are about 100 nm and 70.2$\AA$, respectively. The dielectric constant and the loss tangent at 10 KHz are 176 and 0.038, respectively, and the leakage current density at 100 ㎸/cm is 4.71 $mutextrm{A}$/$\textrm{cm}^2$. In the results of hysteresis loops measured at $\pm$250 ㎸/cm, the remanent polarization (Pt) and the coercive field (Ec) are 5.92 $\mu$C/$\textrm{cm}^2$ and 86.3 ㎸/cm, respectively. After applying 10$^{9}$ square pulses of $\pm$5V, the remanent polarization of the BIT thin film decreases as much as about 33% from 5.92 $\mu$C/$\textrm{cm}^2$ of initial state to 3.95 $\mu$C/$\textrm{cm}^2$.

  • This study was carried out to develop knock-in vector for EGFP (enhanced green fluorescent protein) expression in porcine $\beta$-casein locus. For construction of knock-in vector using porcine $\beta$-casein gene, we cloned the $\beta$-casein genome DNA from porcine fetal fibroblast cells, EGFP and SV40 polyA signal using PCR. The knock-in vectors consisted of a 5-kb fragment as the 5' recombination arm and a 2.7-kb fragment as the 3' recombination arm. We used the neomycin resistance gene ($neo^{r}$) as a positive selectable marker and the diphtheria toxin A (DT-A) gene as a negative selectable marker. To demonstrate EGFP expression from knock-in vector, we are transfected knock-in vector that has EGFP gene in murine mammary epithelial cell line HC11 cells with pSV2 neo plasmid. The EGFP expression was detected in HC11 cells transfected knock-in vector. This result demonstrates that this knock-in vector may be used for the development of knock-in transgenic pig.

  • Adult development and fecundity of the green peach aphid, Myzus persicae $S\H{u}lzer$, were studied at $15{\sim}32.5^{\circ}C$ with $60{\sim}70%$ RH under 16L:8D and the results were put together to build a life table. The longevity shortened as temperatures rose at and below $25^{\circ}C$ whereas it remained relatively constant at $27.5^{\circ}C$. Total fecundity was not significantly different at all temperatures except at $30^{\circ}C$. Daily fecundity gradually increased from $15\;to\;22.5^{\circ}C$. It was 5.1 at $25^{\circ}C$ and as temperatures either went down or up, it decreased to 2.8 at both $15^{\circ}C\;and\;30^{\circ}C$. Fecundity model built from total fecundity at various temperatures suggests that $18.3^{\circ}C$ was the optimum temperature for the maximum progeny of 51. Net reproduction rate RO was highest of 36.5 at $27.5^{\circ}C$. The intrinsic rate of increase per day $r_m$ and the finite rate of increase per day $\lambda$ were highest of 0.4 and 1.6, respectively and the doubling time Dt was shortest of 1.6. Constructed life table suggests that the optimum temperature for adult development for the green peach aphid was $27.5^{\circ}C$ at which the mean generation time was 8.1.

  • Background: The aim of this study is to define the cardioprotective effects (hemodynamic, cytochemical and ultrastructural of the newly developed Histidine-Tryptophan-Ketoglutarate (HTK) cardioplegia compared to DelNido cardioplegia. Material and Method: Seventy-nine isolated rat hearts were divided into three groups on the basis of techniques of cardioplegia infusion. Twenty-eight hearts (Group 1) were flushed with cold DelNido cardioplegia with every 40 minutes for 2 hours. Twenty-seven hearts (Group 2) were flushed with cold HTK cardioplegia for once during the 2 hours. Twenty-four hearts (Group 3) were flushed with cold HTK cardioplegia with every 40 minutes for 2 hours. Heart rate, left ventricular developed pressure (LVDP), changes of + dp/dt max, coronary flow, and rate-pressure product value were measured at pre-ischemic, post-reperfusion 15 minutes, 30 minutes, and 45 minutes for hemodynamic study. Aspartate aminotransferase (AST), lactate dehydrogenase (LD), creatine kinase (CK), CK-MB, troponin-I, myoglobin, and lactate were measured at pre-ischemic and post-reperfusion 45 minutes for cytochemical parameters. Mitochondrial scores were counted in 3 cases from each group for ultrastructural assessment. Result: In hemodynamic study, there were no significant differences among group 1, group 2, and group 3. However, the decrease values of heart rate in group 2 and 3 exhibited significantly lower values than in group 1. In cytochemical study, there were no significant differences among group 1, group 2, and group 3. However, the increase values of lactate in group 2 and 3 exhibited significantly lower values than in group 1. In ultrastructural assessment, the mean myocardial mitochondria scores in group 1, group 2, and group 3 were 2.14$\pm$0.10, 1.52$\pm$0.57, and 2.10$\pm$0.16. Conclusion: HTK solution provides adequate myocardial protection with some advantages over DelNido solution in isolated rat hearts.

  • This study was conducted to evaluate the actual risk of fipronil on worker honey bees (Apis mellifera L.) through acute contact toxicity test, acute oral toxicity test, toxicity of residues on foliage test, and small scale field test. The $48h-LD_{50s}$ of fipronil SC on honeybee were $0.005{\mu}g$ a.i./bee in acute contact toxicity test and $0.004{\mu}g$ a.i./bee in acute oral toxicity test, respectively. In toxicity of residues on foliage test, fipronil showed over 90% of mortality during 28days after treatment at recommended application rate. The $DT_{50}$ of dislodgeable foliar residue was 9 days. Finally, In small scale field test, fipronil showed similar toxicity in the residues on foliage test. It was concluded that fipronil has very high acute toxicity and long residual toxicity to honeybee. Therefore, fipronil is highly toxic to bees exposed to direct treatment or residues on blooming crops or weeds. Do not apply this product or allow it to drift to blooming crops or weeds if bees are visiting the treatment area. To protect honeybee and wild pollinators from outdoor use of fipronil, ultimately it should need to limit for only indoor use to prevent pollinators from unintentionally exposure of fipronil.
