In the present study, we investigated the antioxidant activity of Aucklandia lappa Decne (AL). Cell viability was measured in an MTT assay. Antioxidant effects were evaluated based on total polyphenol/flavonoid contents, ABTS radical scavenging activity, DPPH radical scavenging activity, SOD activity, and ROS content. AL was found not to be toxic at concentrations of 1 ㎍/mL, 10 ㎍/mL, and 100 ㎍/mL, respectively. The phenolic content was higher in AL-D than in AL-E, while the flavonoid content was higher in AL-E than in AL-D. AL-E exhibited higher ABTS radical scavenging activity than AL-D, and the EC50 values for BHA were 217.1 ㎍/mL in AL-D and 180.5 ㎍/mL in AL-E. AL-E also showed the highest DPPH radical scavenging activity. EC50 values for BHA were 114.2 ㎍/mL in AL-D and 95.8 ㎍/mL in AL-E. The SOD-like activity of AL-E was higher than that of AL-D. The EC50 values for ascorbic acid were 48.5 ㎍/mL in AL-D and 72.9 ㎍/mL in AL-E, indicating that both AL extracts have a SOD activity higher than that of ascorbic acid. AL-E reduced relatively more ROS than AL-D. With 100 ㎍/mL AL-E, the reduction level was almost similar to that of dexamethasone. Our results demonstrate that AL have antioxidant effects, and we believe that they could be very valuable as raw materials for anti-aging products, based on their antioxidant activity.
The purpose of this study is to evaluate the free radical scavenging ability and xanthine oxidase inhibitory activity of a complex herbal bath consisted of Artemisiae argyi folium, Angelicae sinensis radix, Ligustici wallichii radix and Angelicae tuhuo radix, and its potential irritation response were also tested for safety use in the rabbits. For antioxidative activity, the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of the complex herbal bath were examined at five different concentrations (0, 250, 500, 1000 and 2000 ㎍/ml). The concentration of the complex herbal bath required for scavenging DPPH free radical by 50% was 897.2 ㎍/ml. In the inhibition of xanthine oxidase (XO) activity, the concentration of the complex herbal bath required for 50% of inhibition was 221.4 ㎍/ml. In the skin irritation study in rabbit, all animals survived for the duration of the study and the examined skin exhibited no edema, erythema, and eschar formation. In the ocular irritation study in rabbit, after application of the sample to eyes, all of the eyes were normal. In summary, the complex herbal bath has potent antioxidant effects against the DPPH radical and XO and was considered to be a non-irritation bath for safety use.
The principal objective of this study was to assess the radical scavenging activities and total polyphenols, flavonoids, and flavonol contents of takju, a Korean traditional rice wine. The antioxidant properties of the wine and takju samples were evaluated using five distinct assays: specifically, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2-azino-di-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS); hydroxyl; superoxide anion; and nitric oxide (NO) radical-scavenging activity assays. In this study, the takju evidenced strong scavenging activities against the hydroxyl, superoxide anion, and NO radicals. Furthermore, the total polyphenol contents of T-2 were similar to those previously observed in white wine (p<0.05). However, the flavonoids and flavonol contents of all takju samples were significantly (p<0.05) lower than that of white wine. The results of this study show that takju possesses a powerful radical scavenging activity against a variety of oxidative systems. The findings of this study also indicate that takju should be considered a useful antioxidant, and that their functional compound reduces oxidative stress.
The proximate composition, free amino acid composition and mineral contents from Undariopsis peterseniana were determined, and the antioxidant activities of ethanol (EtOH) and hot water extracts of U. peterseniana were investigated using DPPH (1,1-diphenyl-1-picrylhydrazyl), ABTS (2,2'- azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)) radical scavenging effects and reducing power. The contents of moisture, ash, protein, lipid and carbohydrate were 12.5%, 23.1%, 9.7%, 0.2% and 54.5%, respectively, and alginic acid content was 12.3%. The major free amino acid contents were alanine, phenylalanine, aminoethanol, valine, glutamic acid and phosphoserine. Ca (1589.1 mg) was the largest mineral followed by Na (344.6 mg), Mg (74.3 mg), Zn (10.2 mg) and Fe (1.5 mg). The total phenolic contents of EtOH and hot water extracts were exhibited at 15.7 and 4.3 mg GAE/g sample, respectively. The antioxidant activity of EtOH extract exhibited strong ABTS and DPPH radical scavenging activities with reducing power, and hot water extract also demonstrated strong ABTS radical scavenging effects. Therefore, the results of this study suggest that U. peterseniana contained an abundance of naturally occurring nutrients (free amino acids and minerals), and the strong antioxidant activities of EtOH and hot water extracts from U. peterseniana could be good sources of natural antioxidants for healthcare products.
The antioxidative effect of the extracts from the leaves and fruits of Acer ginnala against free radicals was studied by two different methods using DPPH radical-generating system, and hydroxyl radical-generating system $(Cu^{++}/H_2O_2\;system)$ which induces DNA strand breaking. Compared with well known antioxidative plants, green tea, Scutellaria baicalensis, the Acer ginnala extracts showed excellent radical-scavenging activity in DPPH radical-generating system and inhibited effectively hydroxyl radical induced-DNA strand breaking in a concentration-dependent manner in $Cu^{++}/H_2O_2$ system whereas the green tea extract stimulated the strand breaking at a low concentration. These results suggest that he extracts from the leaves and fruits of Acer ginnula could be good antioxidative agents.

A series of dihydroxylated chalcone derivatives with diverse substitution patterns on a phenyl ring B and the para-substituents on a phenyl ring A were prepared, and their radical scavenging activities were evaluated by simple DPPH test to determine quantitative structure-activity relationship in these series of compounds. The chalcone compounds with the ortho- (i.e. 2',3'- and 3',4'-) and para- (i.e. 2,5'-) substitution patterns show an excellent antioxidant activities (80-90% of control at the concentration of 50 $\mu$M) which are comparable to those of ascorbic acid and $\alpha$ -tocopherol as positive reference materials. On the contrary, the compounds with meta- (i.e. 2',4'-, 3',5'-) substitution pattern demonstrate very dramatic decrease in activities which are around 25% of the control even at the concentration of 200 $\mu$ M (IC50 > 200 $\mu$ M). These dramatic differences could be interpreted in terms of the ease formation of fairly stable semiquinone radicals from the ortho- and parasubstituted chalcone molecules through facilitating electron delocalization. Our results indicate that the substitution patterns of two hydroxyl groups on ring B are very important structural factors for their radical scavenging activity enhancement. Meanwhile, the substituents at para-position of the phenyl ring A of chalcones have no influence on the activity.
본 연구에서는 백두구(A. cardamomum L.) 메탄올 추출물(ACME)의 항산화 및 항비만 활성을 DPPH radical 소거능과 췌장 lipase 효소 활성 억제능, 그리고 세포실험계를 이용하여 분석하였다. 그 결과 ACME는 DPPH radical을 농도 의존적으로 소거하였으며 DPPH radical 소거능의 50% 저해농도($IC_{50}$)는 $25.15{\mu}g/ml$로 나타났다. 또한 ACME는 농도 의존적으로 lipase 효소 활성을 유의적으로 억제시켰으며, 3T3-L1 preadipocyte를 이용하여 지방세포 분화 및 지방생성, 생성된 지방의 분해에 미치는 영향을 분석한 결과 ACME는 지방세포 분화, 세포 내 지방 축적, TG 함량 등을 독성 없이 농도의존적으로 억제하였으며 지방세포 내 중성지방을 유의적으로 분해시키는 것으로 나타났다. 이러한 백두구의 지방세포 분화 억제능은 핵심 작용 인자인 $C/EBP{\alpha}$, $C/EBP{\beta}$, 그리고 $PPAR{\gamma}$의 유전자 및 단백질 발현조절에서 기인함을 확인하였다. 이러한 결과는 백두구가 보유한 항산화능과 췌장 lipase 활성 저해능, 지방세포 분화 억제능, 지방세포 내 지방 분해능을 통한 항비만 활성을 처음으로 밝혀낸 것이며 추후 계속적인 연구를 통해 활성 물질의 규명이 필요할 것으로 판단된다.
Antioxidant and anti-diabetic activities of acorn were evaluated by its potential for scavenging stable DPPH free radical, inhibition of lipid peroxidation, reducing power, and inhibiton of ${\alpha}-glucosidase\;and\;{\alpha}-amylase$. The water extract of acorn exhibited strong antioxidant and antidiabetic related activities in the tested model systems. Solvent fractionation of the water extract revealed that the water fraction and the EtOAc fraction had strong antioxidant activity, and inhibitory activity on ${\alpha}-glucosidase\;and\;{\alpha}-amylase$. The water fraction exhibited higher DPPH radical scavenging activity ($EC_{50}=7.19{\mu}g/mL$) than that of ${\alpha}-tocopherol\;(EC_{50}=7.59{\mu}g/mL)$. It is considered that water extract of acorn has the potential for natural antioxidant and anti-diabetic products.
The potential antioxidant activities of different fractions from methanolic extract of Curcuma longa L. were assayed in vitro. All of the fractions exception of n-hexane and $H_2O$ showed a strong antioxidant activity, especially the ethylacetate (EtOAc) fraction, which showed the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity ($IC_{50}=9.86\;{\mu}g/mL$). The results of 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity and ferric reducing antioxidant power (FRAP) assay showed concentration dependency, the EtOAc fraction demonstrating a better result than the other fractions at the same concentration in this studies. Additionally, when the total phenolic contents of fractions were measured, the EtOAc fraction contained the highest level. Meanwhile, correlation analysis indicated a high correlation between the antiradical activity and the total phenolic contents, suggesting that fractions obtained from the methanolic extract of Curcuma longa L. have wide potential for use as sources of antioxidant material.
The aim of this study is to utilize the algae extracts with functionalities such as antioxidant and antimicrobial properties for preparing lip balm. Functional properties of extracts from Dictyota coriacea and Ulva pertusa were evaluated in terms of DPPH radical scavenging activity and antimicrobial activity. Lip balms contained the algae extracts were tested its efficacy by measuring the moisture retention of lip skin after applying human subjects. According to the results of skin moisture retention measurement, lip balm was effective to increase the moisture retention of lip skin. It is considered that the utilization of colorants extracted from algae which are verified of antioxidant effect and antimicrobial activity is valuable and useful in developing the cosmetic products.
