Screening of Antioxidant Activity of Acanthopanax species in vitro (오가피류의 시험관내 항산화활성 검색)

  • Acanthopanax species (Araliaceae) has been traditionally used as tonic, analgesic, stimulant of immune system, and replenishment of body function. The antioxidant activities of leaf and root bark of Acanthopanax species were determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and thiobarbituric acid reactive substances (TBARS) assay and relative electrophoretic mobility (REM) on human plasma low density lipoproteins (LDL). Acanthopanax divaricatus var. albeofructus and Acanthopanax for. nambunensis showed potent antioxidant activities.

  • This study was carried out to investigate the antioxidant activities of domestic trees grown in Korea. Based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity method, the methanolic extracts of 23 species were screened in order to search for natural antioxidants. Among these species, Acer ginnala, Cotinus coggygria, Acanthopanax koreanum, Thea sinensis and Pinus densiflora showed stronger antioxidative activity comparing with reference compound, ascorbic acid.

  • Citrus essential oil were extracted from citron peel of cultivated in Gohong (CCP) using environmentally friendly supercritical carbon dioxide method. Antioxidant activity and aroma composition of the essential oils extracted by the SC-$CO_2$ method were evaluated by comparing with those extracted by organic solvent method. Fatty acid composition, DPPH scavenging, and antioxidant activity of the oils obtained by different extraction condition of SC-$CO_2$ method were investigated and their antioxidant activities were compared with commercially available lavender, eucalyptus and tea tree oils. As the results, linoleic acid was most abundantly found from CCP oil extracted by the SC-$CO_2$ method and cis-11,144-eicosadienoic acids was second abundantly found. Radical scavenging ability of DPPH was 98% when the concentration of CCP oil was 50 mg/mL. This scavenging ability increased with the increase of oil concentration. On the other hand, flavonoids content (84 mg/100g) of the CCP oil extracted by the SC-$CO_2$ method was slightly higher than that (75 mg/100g) by hexane extraction. The results, obtained from CCP oil by GC-MS, indicated that, among 66 components, the content of terpenes was 55.8% and limonene was 27.3%.

Antioxidative effects of traditional medicinal plants on lipid peroxidation (지질 과산화에 대한 전통약용 식물의 항산화 효과)

  • To assess the antioxidative activity of 12 medicinal plants on lipid peroxidation, twelves traditional medicinal plants extracted with 95% methanol were investigated the antioxidative activity using DPPH, thiocyanate acid method, and thiobarbituric acid (TBA) methods. Out of 12 medicinal plants extracted with methanol, the extraction yields of Sedum kamtschaticum was the highest values (49.46%) among them and Geranicum sibiricum, Saururus chinensis root (R), Agrimonia pilosa leaf (L), Agrimonia pilosa root was the lowest value (9.97%). Radical scavenging effect of the selected traditional medicinal plants extracted from different extract solution were examined by 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical method. Antioxidative activity of methanolic extracts was higher than those of ethanol and n-hexane extracts. Scavenging effects in Sedum kamtaschaticum (R) determined by DPPH radical showed the highest among the 12 plants. The antioxidative effects of the first four medicinal plants were similar to those of butylated hydroxyanisole (BHA), and butylated hydroxytoluene (BHT), but higher than that of tocopherol, which was used as a handled control. Antioxidative effects of each indicated concentration of the methanolic extracts on linoleic acid by thiocyanate method was the highest in Sedum kamtschaticum and followed by Geum japonicum and Agrimonia pilosa and their antioxidative effect were similar to those of BHA, and BHT, but higher than that of tocopherol. Antioxidative effects of the selected medicinal methanolic extract on linoleic acid by thiocyanate acid method were examined for 15 days. Peroxidation of control and tocopherol group occurred on days 5 and 9, respectively, but BHA, BHT, selected medicinal methanolic extract group did not occur until on day 15. Antioxidative effects of the selected medicinal methanolic extract on linoleic acid by TBA method were examined for 15 days. Antioxidative activity was similar to those obtained by thiocyanate acid method.

Equivalence of Mixed and Individual Preparation of Galgeun-tang in terms of Polyphenol Contents and DPPH Radical Scavenging Activity (폴리페놀 및 DPPH 라디칼 소거활성 관점에서의 복합갈근탕 및 단미혼합갈근탕의 동등성)

  • Traditional medicines usually use combinational formula that are prepared by mixing different varieties of medicinal herbs and boiling them in water to yield a decoction. In recent years a modified method has been proposed and practiced wherein the individual herbs are boiled with water separately and later these extracts are mixed together for use. We attempted to evaluate their equivalence in terms of its polyphenol contents and DPPH radical scavenging activity. The polyphenol contents as well as the DPPH radical scavenging activities were very similar to each other.

  • The methanol extract of Coscinium fenestratum, commonly own as tree turmeric, which is widely used in the indigenous system of medicine was studied for its in vitro scavenging activity in different methods viz DPPH scavenging, nitric oxide scavenging, iron chelation activity, superoxide scavenging, ABTS radical scavenging and lipid peroxidation. The results were analyzed statistically by regression method. Its antioxidant activity was estimated by $IC_{50}$ value and the values are $57.1\;{\mu}g/ml$ for DPPH radical scavenging, $36.5\;{\mu}g/ml$ for iron chelating activity, $51.7\;{\mu}g/ml$ for nitric oxide scavenging, $53.63\;{\mu}g/ml$ for ABTS scavenging, $44.2\;{\mu}g/ml$ for superoxide scavenging, and $40\;{\mu}g/ml$ for lipid peroxidation. In all the methods, the extract showed its ability to scavenge free radicals in a concentration dependent manner. The results indicate that C. fenestratum has potent antiofidant activity.

Change in Icariin Contents and Antioxidant activity during Ripening Period of Epimedium Wine (알코올 농도 및 숙성 온도에 따른 삼지구엽초 담금주의 이카린 함량 및 항산화 활성)

  • This study was designed to investigate the contents of icariin during ripening of Epimedium koreanum with sugar and Soju (25, 30, and 35% alcohol contents) using a homemade method. Contents of icariin of Epimedium wine were examined using high performance liquid chromatography. Icariin levels in Epimedium wine increased during the first 3 days of storage; after 6 days, icariin contents decreased gradually until 30 days. Contents of icariin in Epimedium wine by storage temperature (5 and $20^{\circ}C$) were not different during storage. After 30 days, icaiin was not detected in all tested Epimedium wines. Epimedium wines were assessed for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) and (ABTS) radical scavenging activity. DPPH and ABTS radical scavenging activities increased in early storage periods, however, after 6 or 9 days, activities decreased gradually.

  • This study was carried out to investigate the antioxidative activities of domestic plants grown in the area of Kangwon-do province. Through the examination of methanol extracts from 15 plants for radical scavenging effects using DPPH method, the extracts from Pinus densiflora Sieb. et Zucc. And Euphorbia humifusa Willd. Showed strong antioxidative activity. Because of its highest antioxidative activity among 15 domestic plants, radical scavenging effects of four different extract compartments from leaves of P.densiflora, n-Hexane, EtOAc, BuOH and $H_2O$ extracts. Were examined by DPPH method. Antioxidative activities of EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioBfdants (BHA) , suggesting that major fraction for the antioxidative activity of P. densiflora was the EtOAc and BuOH extract compartments.

Screening for Antioxidative Activity in Soybean Local Cultivars in Korea

  • Sixty local soybean cultivars were evaluated on the antioxidative activity by superoxide dismutase (SOD), 1, 1-diphenyl-2-picrylhydrazyl (DPPH), thiobarbituric acid(TBA), and chemiluminescence using the FI-CL system. Soybean were collected throughout the country, and were grown over two years (1997 and 1998) for measuring antioxidative activity in soybean seeds. There were differences in antioxidative activity depen-ding on the method of measurement and variation of the crop year. Soybeans from Kwangyang-shi-1 (76.78%) in 1997 and Kangjin-gun-3(79.14%) in 1998 showed the highest SOD activity, whereas those from Hwasoon-gun (80.43%) in 1997 and Kangjin-gun-2 (49.82 %) in 1998 exhibited the highest DPPH activity. Soybeans from Chongup-gun-2 (75.77%) in 1997 and from Yochon-shi-5 (69.17%) in 1998 exhibited the highest TBA activity, and those from Jinahn-gun (48.99%) in 1997 and Kohung-gun (49.73%) in 1998 exhibited the highest activity using the chemiluminescence method. These results suggest that it may be possible to develop soybean varieties with higher antioxidative activity.

Comparative Composition of Orgainc and Inorgainc Arabica Beans and Their Antioxidant Characteristics (아라비카 원두의 산지 및 수확방법에 따른 이화학적 특성 및 항산화성)

  • The objective of this study was to evaluate the antioxidant and physiological The objective of this study was to evaluate the antioxidant and physiological activities of organic and inorganic coffee based on flavonoid composition. Organic and inorganic coffee varieties were prepared with Brazil arabica bean and Columbia arabica bean, respectively. Color values (Lightness, redness, and yellowness), mineral contents, total phenol content, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, individual flavonoids, and sensory characteristics were then measured in the espresso of each coffee bean. The pH, Brix, and color difference between organic coffee and inorgainc coffee did not correlate with the harvest method. Coffee varieties were more influential on physiological properties, and were markedly more influential than the mineral content. Total phenolics had a higher correlation with DPPH radical scavenging activity ($r^2$=0.832). Depending on the harvesting method, significant differences (p<0.05) were shown in aroma, color, and overall acceptability in sensory evaluation.
