한국산 방아잎(배초향, Agastache rugosa O. Kuntze)에서 항산화물질 로즈마린산의 분리, 동정 및 활성 (Isolation, Identification, and Activity of Rosmarinic Acid, a Potent Antioxidant Extracted from Korean Agastache rugosa)

  • 국내 식물자원 30 가지의 항산화활성을 탐색한 결과 방아잎, 형개, 택란의 methanol 추출물에서 높은 활성을 확인하였다. 그 중에서 항산화활성이 가장 높은 방아잎의 건조분말을 methanol로 환류냉각 추출하여 hexane chloroform, ethyl acetate, water 순으로 용매분획한 결과 ethyl acetate 분획에서 가장 높은 항산화활성을 확인하였으며, 이를 농축한 다음 Sephadex LH-20으로 chromatography하여 순수물질을 얻었다. 이물질을 ESIMS로 분석한 결과 분자량이 360임을 확인하였고, IR과 UV spectra에 의해서 phenol계 화합물임을 확인하였으며 NMR 분석에 의해서 이 항산화물질이 rosmarinic acid임을 확인하였다. 이 물질의 항산화활성을 검정한 결과 돈지와 팜유에 대해서 BHT나 ${\alpha}-tocopherol$보다도 우수한 지질과산화 억제효과를 나타냈다. 방아잎의 methanol추출물의 항산화력은 rosmary의 methanol 추출물과는 비슷하였으나, sage의 methanol추출물 보다는 높게 나타났다.

YS 49, a Synthetic Isoquinoline Alkaloid, Protects Sheep Pulmonary Artery Endothelial Cells from tert-butylhydroperoxide-mediated Cytotoxicity

  • Endothelium, particularly pulmonary endothelium, is predisposed to injury by reactive oxygen species (ROS) and their derivatives. Heme oxygenase (HO) has been demonstrated to provide cytoprotective effects in models of oxidant-induced cellular and tissue injuries. In the present study, we investigated the effects of YS 49 against oxidant [tert-butylhydroperoxide (TBH)]-induced injury using cultured sheep pulmonary artery endothelial cells (SPAECs). The viability of SPAECs was determined by quantifying reduction of a fluorogenic indicator Alamar blue. We found that TBH decreased cell viability in a timeand concentration-dependent manner. YS 49 concentration- and time-dependently increased HO-1 induction on SPAECs. As expected, YS 49 significantly decreased the TBH-induced cellular injury. In the presence of zinc protophorphyrin, HO-1 inhibitor, effect of YS 49 was significantly inhibited, indicating that HO-1 plays a protective role for YS 49. Furthermore, YS 49 showed free radical scavenging activity as evidenced by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and inhibition of lipid peroxidation. However, YS 49 did not inhibit apoptosis induced by lipopolysaccharide (LPS) in SPAECs. Taken together, HO-1 induction along with strong antioxidant action of YS 49 may be responsible for inhibition of TBH-induced injury in SPAECs.

배추김치의 dichloromethane 획분으로부터 항산화 물질의 분리 및 동정 (Isolation and Identification of Active Principle in Chinese Cabbage Kimchi Responsible for Antioxidant Effect)

  • 배추김치의 dichloromethane 획분의 수율은 약 1.66% 이었으며, 이로부터 silica gel column chroamtography, Sepadex LH-20 column chromatography, Sep-pack catridge 등을 이용하여 활성물질을 분리 정제하였고, IR, EI-MS, GC-MS. $^{1}H-NMR,\;^13C-NMR$로 구조를 확인하여 분자량 226인 3-(4'-hydroxyl-3',5'-dimethoxyphenyl)propionic acid로 밝혀다. 비추김치 활성성분의 DPPH 유리기 소거능은 vitamin C에 비해 3.4배 높았고, LDL 산화억제 효과는 대조군에 비해 현저히 높아 인체에서 지질저하 효과 및 동맥경화 억제 효과가 있을 것으로 예상된다.

Skin Anti-Aging Activities of Bacteriochlorophyll a from Photosynthetic Bacteria, Rhodobacter sphaeroides

  • In this work, the anti-aging skin effects of bacteriochlorophyll a isolated from Rhodobacter sphaeroides are first reported, with notably low cytotoxicity in the range of 1% to 14% in adding 0.00078 (% (w/w)) of the extracts, compared with the normal growth of both human dermal fibroblast and keratinocyte cells without any treatment as a control. The highest production of procollagen from human fibroblast cells (CCD-986sk) was observed as 221.7 ng/ml with 0.001 (% (w/w)) of bacteriochlorophyll a, whereas 150 and 200 ng/ml of procollagen production resulted from addition of 0.001 (% (w/w)) of the photosynthetic bacteria. The bacteriochlorophyll-a-induced TNF-α production increased to 63.8%, which was lower secretion from HaCaT cells than that from addition of 0.00005 (% (w/w)) of bacteriochlorophyll a. Additionally, bacteriochlorophyll a upregulated the expression of genes related to skin anti-aging (i.e., keratin 10, involucrin, transglutaminase-1, and MMPs), by up to 4-15 times those of the control. However, crude extracts from R. sphaeroides did not enhance the expression level of these genes. Bacteriochlorophyll a showed higher antioxidant activity of 63.8% in DPPH free radical scavenging than those of water, ethanol, and 70% ethanol extracts (14.0%, 57.2%, and 12.6%, respectively). It was also shown that the high antioxidant activity could be attributed to the skin anti-aging effect of bacteriochlorophyll a, although R. sphaeroides itself would not exhibit significant anti-aging activities.

Effects of Daegangwhal-Tang Hot Aqueous Extract on Anti-inflammation and Anti-oxidation in RAW 264.7 Macrophage

  • Background: The objective of this study was to determine the effects of Daegangwhal-Tang (DGHT) hot aqueous extract on production of inflammatory mediators and antioxidants in RAW 264.7 macrophage. Methods: DGHT was extracted with water, filtered, concentrated and freeze-dried to perform. Cytotoxicity of DGHT extract was performed by MTT assay. Activated macrophages were treated with varying concentrations of DGHT extract (10, 50, 100 and $200{\mu}g/mL$), and nitric oxide (NO) and prostaglandin E2 ($PGE_2$) concentrations were measured to detect anti-oxidative effects. Interleukin-6 (IL-6), interleukin-1 beta ($IL-1{\beta}$) and tumor necrosis factor-alpha($TNF-{\alpha}$) concentrations were also measured to detect inflammatory responses to DGHT Results: Cytotoxicity of DGHT extract at concentrations of 10, 50, 100 and $200{\mu}g/mL$ were not observed. NO production was significantly decreased in the DGHT hot aqueous extract $200{\mu}g/mL$ concentration group. $PGE_2$, IL-6, $IL-1{\beta}$ and $TNF-{\alpha}$ production was significantly decreased in the DGHT hot aqueous extract 100 and $200{\mu}g/mL$ concentration groups. DGHT hot aqueous extract appeared to have DPPH free radical scavenging capability at all of concentrations, but did not exceed 50%. Conclusion: These results suggest that DGHT hot aqueous extract has concentration-dependent anti-inflammatory and anti-oxidative effect.

올리고당을 활용한 당절임 야생 복숭아의 품질특성과 항산화 효과에 미치는 영향 (Effect of Oligosaccharides on Quality Characteristics and Antioxidant Activities of Prunus persica Batsch var. davidiana Max. preserved in Sugar)

  • In order to investigate the quality characteristics of Prunus persica Batsch var. davidiana Max. preserved in sugar, the quality characteristics were examined by dividing them into four groups: PS-A (P. persica preserved in sugar mixed with sucrose and oligosaccharides at a ratio of 100:0, w/w), PS-B (P. persica preserved in sugar mixed with sucrose and oligosaccharides at a ratio of 60:40, w/w), PS-C (P. persica preserved in sugar mixed with sucrose and oligosaccharides at a ratio of 40:60, w/w) and PS-D (P. persica preserved in sugar mixed with sucrose and oligosaccharides at a ratio of 0:100, w/w). The results were as follows: pH levels and soluble solid contents of PS-D sample were lower than those of the others, however total acidity value were higher. In case of Hunter's value, the 'L', 'a' and 'b' values of all samples were affected by addition of oligosaccharides. No great difference occurred among samples in reducing sugar contents. Organic acid content was significantly decreased with the lower addition of sucrose. The predominating organic acid components analyzed in all samples were lactic acid 156.15 g/100 g, tartaric acid 72.75 g/100 g and citric acid 32.90 g/100 g. The highest contents of total phenol and flavonoid, and DPPH and ABTS radical scavenging activity were $10.79{\pm}0.55mg\;GAE/mL$, $8.71{\pm}0.06mg\;CE/mL$ and 71.8%, 71.0% for PS-D, respectively. The results of this study indicated the use of oligosaccharides could increase the quality and antioxidant potentials of P. persica preserved in sugar.

쑥 분말을 첨가한 양갱의 품질특성 (Quality Characteristics of Yanggaeng Incorporated with Mugwort Powder)

  • 쑥 분말의 첨가량을 달리하여 양갱을 제조한 후 이화학적 특성 및 관능적 특성을 알아보았다. $L^*$값과 $b^*$값은 유의적으로 감소하였고(p<0.05), 반면 $a^*$값은 점차 증가하는 경향을 나타내었다. 양갱의 경도(hardness), 점착성(gumminess), 씹힘성(chewiness) 및 응집성(cohesiveness)은 쑥 분말의 첨가량이 증가할수록 증가하는 경향을 보였고(p<0.05), 탄력성(springiness)은 유의적으로 감소하는 경향이 나타났다(p<0.05). 항산화활성을 나타내는 총 폴리페놀 함량과 전자공여능은 쑥 분말 첨가량이 증가함에 따라 유의적으로 증가하는 경향을 나타내었고(p<0.05), 전자공여능에서 보다 현저한 증가를 보였다. 소비자 검사결과 2% 첨가군이 모든 평가항목에서 유의적으로 높은 평가를 받아 양갱의 관능적 품질을 최대한 유지하면서 쑥 분말의 기능적 잇점을 최대한 활용하기 위한 최적 첨가농도는 2%가 가장 적합한 것으로 판단된다.

클로렐라 처리에 의한 유기농 콩나물 생육촉진 및 항산화 능력 증진효과 (Effect of Chlorella sp. on Improving Antioxidant Activities and Growth Promotion in Organic Soybean Sprout Cultivation)

  • The purpose of this study was to estimate the growth promoting effects and improvement of antioxidant activity of the soybean sprouts treated with Chlorella sp. culture solution. The soybean sprout treated with 0.1% and 0.2% Chlorella sp. culture solution was significantly increased the length (more than 43.0%), the thickness (more than 0.5~0.7 mm), fresh weight (more than 2.9~3.7 g) compared to non-treated control in vitro. In organic soybean sprouts farm, the 0.2% chlorella culture solution applied to mass culture of soybean sprout and the fresh weight of soybean sprouts increased by more than 25% and the yield was very high as 598.33% compared to untreated control. In addition of sensory test, there is no fishy odor and better crunchy texture and nutty flavor for the treatment soybean sprouts compared to untreated soybean sprouts. Particularly, free-radical scavenging activity (DPPH) and superoxide dismutase activity (SOD) of the soybean sprouts were significantly increased more than 26.1% and 40.4%, respectively by treated with 0.1% and 0.2% Chlorella culture solution. Consequently, the treatment of chlorella culture solution to grow soybean sprouts is also promoting quality and antioxidant activity as well as promoting the growth of sprouts. Therefore, chlorella is considered to be worth as functional materials for high-quality sprouts grown.

당근가루를 첨가한 들깨다식의 품질 특성 연구 (Effect of Carrot Powder on Anti-Oxidative and Quality Characteristics of Perilla-Dasik)

  • ${\omega}-3$ 지방산이 풍부한 들깨를 이용하여 고령자용 건강간식을 만들기 위해 다식을 제조하고 들깨의 산패 억제를 위해 당근가루를 0, 5, 10 15, 20% 첨가하여 들깨가루 다식을 제조한 결과, 당근가루 첨가량에 따른 들깨 다식의 수분 함량의 변화는 없었으며, 당근 내 카로티노이드에 의해 당근가루 첨가량이 많아질수록 다식의 붉은색과 황색도는 증가하였다. 당근가루 첨가량이 많을수록 다식의 경도와 응집성, 씹힘성은 증가하였다. 다식의 과산화물가와 산가를 측정한 결과 당근가루 첨가량이 많을수록 항산화 효과는 증가하였고, 라디칼 소거능 역시 효과적으로 증가하였다. 지방을 함유한 식품에는 산패 억제를 위해 항산화제를 사용하고 있으며, 특히 합성 항산화제를 대체할 천연 항산화제에 대한 요구가 계속적으로 증가하고 있는데 열처리(데치기)에 의해 ${\beta}-carotene$을 활성화시킨 당근가루는 들깨다식의 항산화 활성을 증진시켜 질적 향상을 유도할 수 있는 좋은 천연 항 산화제로서의 기능을 갖고 있는 것으로 조사되었다. 관능검사 결과 당근가루 첨가 여부에 대한 선호는 뚜렷했으나, 첨가 수준에 따른 선호도의 차이는 없었다. 본 연구 결과 20% 수준의 당근가루 첨가는 항산화 특성을 효과적으로 향상시키면서 전체적인 기호도에 영향을 미치지 않아 들깨다식 제조 시 적정 첨가 수준으로 생각된다.

여정자 헥산추출물의 멜라닌합성 억제효과 (Inhibitory Effect of Fructus Ligustri Lucidi Hexane Extract on Melanin Biosynthesis)

  • The purpose of this study was to investigate the effects of Fructus Ligustri Lucidi (FLL) on the process of melanogenesis. Cell viability of B16F10 cells was measured by MTT assay and melanin content was assessed using the method of Hosoi with some modifications. Methanol extract of Fructus Ligustri Lucidi (MFLL) significantly inhibited melanin synthesis in a concentration-dependent manner, and it reduced the activity of tyrosinase, the rate-limiting melanogenic enzyme. Additionally, $\alpha$-melanocyte stimulation hormone ($\alpha$-MSH)-induced hyperpigmentation was down-regulated by MFLL. MFLL was fractionated by organic solvent. In the present study, Hexane extract of Fructus Ligustri Lucidi (HFLL) inhibited tyrosinase activity and melanin production of B16F10 cells in the absence or presence of $\alpha$-MSH. Ethyl acetate, butanol and water layers did not affect tyrosinase activity and melanin production. Meanwhile ethyl acetate, butanol and water layers showed DPPH free radical scavenging activity. These results suggest that extract of FLL could be used as functional biomaterial in developing a skin whitening agent having the antioxidant activity.
