Antiradical Capacities of Perilla, Sesame and Sunflower Oil

  • The aim of this study is to examine the radical scavenging activity of perilla and sesame oil that Koreans traditionally consume. For DPPH radical scavenging activity, oil and its hexane/70% methanol extracts (ME) are used and for superoxide and hydroxyl radical scavenging activities, ME are used. Unrefined perilla oil, sesame oil, and refined sunflower oil are used. The yields for ME of perilla, sesame and sunflower oil are 0.57, 0.61, and 0.30%, respectively, and the amounts of phenolic compounds in ME of corresponding oil are 18.77, 88.64 and $0.05\;{\mu}g$ tannic acid/mg, respectively. $IC_{50}$ for DPPH scavenging activity of perilla, sesame and sunflower oil are 2.12, 1.91, and 3.35 mg/mL, respectively and those for ME of corresponding oils are 0.42, 0.07, and 43.11 mg/mL, respectively. In DPPH assay, the solvent used for oil sample is iso-octane and that for ME is methanol. Superoxide anion scavenging activity of ME of perilla, sesame and sunflower oil tested at 1 mg/mL concentration are 21.10, 13.25, and 3.14%, respectively. Hydroxyl radical scavenging activities of those samples tested at 1 mg/mL concentration are 86.08, 93.30, and 93.17%, respectively. In summary, the refining process seems to remove the phenolic compound during oil processing. Antiradical substances in perilla and sesame oils responsible for scavenging DPPH radicals are present in the methanol fraction, while the antiradical substances in the sunflower oil are in the lipid fraction. DPPH scavenging activity of ME of sesame oil is significantly higher than that of perilla oil (p<0.05). However, superoxide anion scavenging capacity of ME of perilla oils was found to be greater than that of both sesame and sunflower oils (p<0.05).

Antioxidant activity of ethanol extracts from the root and bark of Ulmus davidiana

  • Antioxidant activities of Ulmus davidiana root and bark extracts were evaluated by various antioxidant tests, including DPPH radical-scavenging, nitric oxide-scavenging, superoxide anion radical-scavenging, and ABTS radical-scavenging assays, and superoxide dismutase (SOD)-like activity and reducing power analysis, along with the determination of total phenolic and flavonoid contents. Both the extracts showed strong antioxidant activities by these testing methods. Ulmus davidiana root extract possessed strong reducing power and nitric oxide-scavenging activity, and high scavenging activities against free radicals including the superoxide anion, and the ABTS and DPPH radicals, but a weaker scavenging activity of SOD. In contrast, the Ulmus davidiana bark extract exhibited a strong SOD-like activity, but all the other activities were weak. It was observed that the total phenolic and flavonoid contents of the Ulmus davidiana root extract were higher than those of the Ulmus davidiana bark extract.

해양균류의 라디칼 소거활성 검색 (Screening of Radical Scavenging Activity from the Marine-Derived Fungus)

  • In order to screen new radical scavenging principle which is expected to be antiaging drug lead, we have isolated 160 strains of the marine-derived fungi and investigated 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity for their acetone extracts. The significant activities (>50% Inhibition) were observed in 8 strains of fungi (MFA006, MFA0l4, MFA040, MFA133, MFA139, MFA143, MFA148, MFA153), and among them, MFA153 (Aspergillus parasiticus) showed the most significant radical scavenging activity. The active components were purified by assay-guided isolation to yield two known benzyl alcohols, l53B3 (1) and l53B4 (2), and their structures were determined by physicochemical evidence. Two compounds (1,2) showed the significant radical scavenging activity with $IC_{50}$ values of 0.6 and $1.4{\mu}M$ against DPPH, respectively.

The Radical Scavenging Effects of Stilbene Glucosides from Polygonum multiflorum

  • The extract of the root of Polygonum multiflorum exhibited a significant antioxidant activity assessed by the DPPH radical scavenging activity in vitro. The bioassay-guided fractionation of the extract yielded a stilbene glucoside, (E)-2,3,5,4'-tetrahydroxystilbene-2-Ο-$\beta$-d-glucopyranoside (1) as an active constituent responsible for the antioxidant property. Compound 1 demonstrated a moderate DPPH radical scavenging activity ($IC_{50}$, 40 $\mu$M), while the corresponding deglucosylated stilbene 2 exhibited a much higher activity ($IC_{50}$, 0.38 $\mu$M).

High Throughput Screening (HTS) 기법을 통한 제주 자생 해조류의 DPPH 라디칼 소거활성 평가 (Evaluation of DPPH Radical Scavenging Activity of Jeju Seaweeds Using High Throughput Screening (HTS) Technique)

  • 본 연구는 제주산 해조류의 생리활성을 대량검색하기 위해 HTS 기법을 이용한 실험과정의 제어 및 관리를 위한 자동화 시스템을 구축하고자 DPPH 라디칼을 이용하여 HTS를 이용한 자동화 시스템을 구축하고 제주산 해조류의 DPPH 라디칼 소거활성을 대량 검색하고자 하였다. 그 결과 HTS를 위한 효율적인 검색방법을 개발하였고 DPPH 라디칼 소거활성이 높은 해조류를 검색하게 되었다. 녹조류의 경우 납작파래 (20G6)의 $20^{\circ}C$ 추출물(1 mg/ml)에서 약 60%이 DPPH 라디칼 소거활성을 보였을 뿐 나머지 해조류에서는 40% 미만이 낮은 활성만을 보였다. 갈조류의 경우 꽈배기 모자반 (20B17), (70B17), 참그물바탕말 (70B1), 큰잎모자반 (70B16)과 감태 (70B26)에서 DPPH 라디칼 소거활성이 우수하였다. 홍조류 중에서는 왜떨기나무붉은실 (20R24)에서 가장 우수한 DPPH 라디칼 소거활성을 나타내었고 갈래잎 (20R17), 굵은참우뭇가사리 (20R18), 부채분홍잎 (20R23) 또한 높은 소거활성을 보였다. 따라서 우수한 효과를 나타내었던 해조류를 대상으로 용매분획 및 활성물질의 분리, 정제를 통해 해조류 유래의 기능성 신소재를 개발한다면, 해조류의 고부가가치화 및 HTS기술의 보편화에 큰 영향을 미칠 것으로 판단된다.

Antioxidant Activity of Partially Purified Extracts Isolated from Bacillus polyfermenticus SCD Culture

  • The antioxidant activity of Bacillus polyfermenticus SCD was studied by partially purified culture extracts using various methods: ammonium sulfate precipitation, adsorption to Diaion HP-20 columns using polar solvents, and extraction using non-polar solvents. The 1,1-diphenyl-2-picyrylhydrazyl (DPPH) radical scavenging activity of these partially purified fractions was then investigated. The precipitate isolated using 75%-saturated ammonium sulfate was shown to contain about 77.2% DPPH radical scavenging activity. Using the Diaion HP-20 resin adsorption method, the fraction obtained using 60% ethanol and 60% methanol possessed 76.7 and 89.5% DPPH radical scavenging activity, respectively. Fractions obtained by extracting with the non-polar solvents 80 mg/mL chloroform, 80 mg/mL n-hexane, 80 mg/mL ethyl acetate, and 80 mg/mL butanol contained 68.4, 75.0, 70.7, and 87.5% DPPH radical scavenging activity, respectively. Further study is needed to characterize the antioxidant substance(s) released by B. polyfermenticus SCD cultures.

사과락(絲瓜絡)의 항산화효과 및 3T3-L1분화 시 Cytokine류에 미치는 영향 (Experimental Study on Antioxidative Effect of Luffae Fructus Retinervus and Their Effects on Cytokines to 3T3-L1 Cell Lines)

  • In recent year, We are concerned in anti-aging, disease-protection, long-life, many method are used in solving this problem. Recently, We heard that Luffae Fructus Retinervus(LFR) has effect of anti-aging, disease-protection, long-life. So I let made a experiment for this result. The purpose of this study is to; 1) the anti-oxidant effect of Luffae Fructus Retinervus(LFR) used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity, Superoxide anion radical scavenging activity, 2) cultivation 3T3-L 1 Preadipocytes and Protein chip used for ProteoPlexTM 16-Well Murine Cytokine Array Kit. We measured level of DPPH radical scavenging activity. And we experienced that the ability of DPPH radical's elimination was increased by rising concentration of LFR. When the concentration of LFR was 5 mg/ml, the ability of DPPH radical's elimination was Maximum. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was significant when concentration of LFR was from 1.25 mg/ml to 2.5 mg/ml. We measured level of Superoxide anion radical scavenging activity. And we founded that the ability of Superoxide anion radical's elimination was maximum when concentration of LFR was 0.3125 mg. When we inspected Antioxidative Effects with BSA, we experienced that ability of defense was increased by rising concentration of LFR. We known the immunity of LFR about 3T3-L1 Preadipocytes and gained the increase of Cytokines(IL-2, IL-4, GM-CSF) without IL-12p70, $INF-{\gamma}$, $TNF-{\alpha}$ So I guess that Luffae Fructus Retinervus(LFR) has effects of anti-aging, disease-protection, long-life, etc.

두리안 추출물의 항산화 활성 (Antioxidative Activity of the Durian (Durio zibethinus) Extract)

  • The purpose of this study was to examined the antioxidant activities by water and 70% ethanol extract from durian (Durio zibethinus.) seed, sarcocarp and peel. Durian extract were studied for reducing sugar content, polyphenol content, superoxide dismutase (SOD) like activity, electron donating ability, nitrite scavenging ability, flavonoid content, hydroxy radical scavenging activity. Reducing sugar content were increased peel > sarcocarp > seed. Total polyphenol, flavonoid content, DPPH radical scavenging ability and SOD like activity were increased seed > peel > sarcocarp. Total polyphenol content was relatively high as $21.90{\pm}0.50mg/g$ in the ethanol extract of the seed. DPPH radical scavenging ability was relatively high as $62.08{\pm}2.63%$ in the water extract of the seed. Nitrite scavenging ability was no significant difference. Hydroxy radical scavenging activity was increased seed > peel > sarcocarp, was relatively high as $58.27{\pm}1.13%$ in the water extract of the seed.

Antioxidant Studies on the Methanol Stem Extract of Coscinium fenestratum

  • The methanol extract of Coscinium fenestratum, commonly own as tree turmeric, which is widely used in the indigenous system of medicine was studied for its in vitro scavenging activity in different methods viz DPPH scavenging, nitric oxide scavenging, iron chelation activity, superoxide scavenging, ABTS radical scavenging and lipid peroxidation. The results were analyzed statistically by regression method. Its antioxidant activity was estimated by $IC_{50}$ value and the values are $57.1\;{\mu}g/ml$ for DPPH radical scavenging, $36.5\;{\mu}g/ml$ for iron chelating activity, $51.7\;{\mu}g/ml$ for nitric oxide scavenging, $53.63\;{\mu}g/ml$ for ABTS scavenging, $44.2\;{\mu}g/ml$ for superoxide scavenging, and $40\;{\mu}g/ml$ for lipid peroxidation. In all the methods, the extract showed its ability to scavenge free radicals in a concentration dependent manner. The results indicate that C. fenestratum has potent antiofidant activity.

Phytotoxic Effect, DPPH Radical Scavenging Activity and Chlorogenic Acid Level of Methanol Extracts from Aerial Parts of Several Korean Salad Plants

  • A series of aqueous or methanol extracts from four different Korean salad plants were assayed to determine their allelopathy and antioxidant activity. The extracts applied on filter paper in a Petri-dish bioassay significantly inhibited root growth of against alfalfa (Medicago sativa) seedlings. Leaf extracts from 40 g dry tissue $L^{-1}$ of Aster yomena was most phytotoxic to alfalfa root growth, and followed by that of Cirsium japonicum, Taraxacum officinale, and Ixeris dentate. Methanol extracts of plants dose-dependently increased DPPH free radical scavenging activity in vitro. Antioxidant activity of methanol extracts from the same plant species was investigated, and the result showed high DPPH free radical scavenging activity in Cirsium japonicum, Aster yomena, and Ixeris dentate, however, in Taraxacum officinale was least activity. By means of HPLC analysis, chlorogenic acid, p-coumaric acid, and total phenolics with 7.68, 17.47 and 18.64 mg, $100g^{-1}$, respectively, showed the highest amounts in methanol extracts from Cirsium japonicum leaves. These results suggest that Compositae salad plants contain water-soluble substances with allelopathic potential as well as antioxidant activity.
