Screening of Antioxidative Activity of Hot-Water Extracts from Medicinal Plants (한약재 열수추출물의 항산화효과 검정)

  • Interrelation between the antioxidative activity of hot-water extracts of 130 medicinal plants and their cellular antimutagenic activity was investigated. Antioxidative activity was evaluated by assaying electron-donation to DPPH free radical and scavenging of hydroxyl radical $({\cdot}OH)$ generated through Fenton rection, respectively. All medicinal plants examined in this study exhibited markedly electron-donating ability and radical scavenging ability in each assay system. The results demonstrated the fact that Pilbal (Piper longum L.) is the strongest in electron-donating activity, on the other hand, that Seokgok (Dendrobium moniliforme L.) is the strongest in ${\cdot}OH$ scavenging activity. When evaluated their antioxidative activities, 24 medicinal plants including Jimo (Anemarrhena asphodeloides Bunge) were found to be the medicinal plants carrying strong antioxidative activity, which exhibited more than 50% activity compared to the control group in both electron-donating and free radical scavenging. The experiment was also performed to examine whether 11 medicinal plants having significant antimutagenicity damage DNA in the presence of $Cu^{2+}$, showing the fact that all samples tested, except Taeksa (Alisma canaliculatum All. Br.), Paekjain (Nitraria sibirica Pall) and Ohyak (Lindera strychifolia Sieb. et Zucc. Villar) are capable of inducing DNA strand break. We also found that Taeksa and Paekjain strongly block DNA strand break induced by chemical mutagen mitomycin C.

Antioxidant Activities of Selenium-Treated Spinacia oleracea L. (셀레늄 강화 시금치의 항산화 활성)

  • In the present study, we investigated the anti-oxidant activities of selenium-treated Spinacia oleracea L. by utilizing experiments in vitro assays. The selenium content of non-treated spinach in this study was noted at $61.19{\mu}g/kg$, whereby the selenium-treated spinach which was treated by a 2000 mg/kg selenium was 1000-fold diluted, and was reported to be about 4 times higher than that of non-treated spinach. In this case, the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in the concentration of selenium-treated spinach, 0.1~1.0 mg/mL was measured as stronger than that of the identified non-treated spinach. By the same token, the DPPH radical activity of non-treated spinach and selenium-treated spinach was recorded as 46.05~52.75% and 49.52~59.09% respectively. It is emphasized that the 2,2'-azino-di-2-ethyl-benzthiazoline-sulphonate (ABTS) radical scavenging activity as revealed in the concentration of selenium-treated spinach, 0.1~1.0 mg/mL was noted as being stronger than that of non-treated spinach. The ABTS radical activity of non-treated spinach and selenium-treated spinach was 11.85~52.01% and 27.14~53.59% respectively. In this respect, the nitric oxide (NO) radical scavenging activity and reducing power activity in the concentration of selenium-treated spinach, 0.1~1.0 mg/mL was identified and noted as stronger than that of non-treated spinach. These results suggest that selenium-treated spinach could possibly be more useful as a potential antioxidant to improve human health outcomes, than the non-treated spinach.

Comparison of the antioxidant activity and nutritional contents of ectomycorrhizal mushroom extracts in Korea (국내 균근성 버섯류 추출물의 항산화능 및 영양성분 비교)

  • The extracts of four species of ectomycorrhizal mushrooms-Cantharellus cinnabarinus (OK1247), Lactarius parallelus (OK1264), Tricholoma matsutake (OK1282), and Ramaria botrytis (OK1283)-were prepared to determine their antioxidant activities and nutritional properties. R. botrytis extract displayed the highest 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (33.8%), ferric reducing antioxidant power (0.38), reducing power (0.35), total polyphenol (13.83 mg gallic acid equivalent/g), and flavonoid contents (2.56 mg quercetin equivalent/g). L. parallelus extract displayed the highest nitrite scavenging activity. Analysis of amino acid contents revealed that C. cinnabarinus extract had the highest total amino acid (1,046.1 mg/kg) and essential amino acid (404.9 mg/kg) contents, while R. botrytis extract had the lowest total amino acid (708.3 mg/kg) and essential amino acid (247.3 mg/kg) contents. Among the amino acid components detected in the four ectomycorrhizal mushrooms, cysteine was the most abundant, accounting for 14.3~20.7%, followed by phenylalanine, which accounted for 9.5~13.4% of all amino acids. In summary, the antioxidant activities were the highest in R. botrytis extract, and the amino acid content was the highest in C. cinnabarinus extract, among the four ectomycorrhizal mushrooms.

Antioxidant Activities of Solvent Extracts from Pomegranate Endocarp (석류 내피 용매별 추출물의 항산화 활성)

  • This study was designed to investigate antioxidant activities of pomegranate endocarp extracts. Pomegranate endocarp extract contains the highest antioxidant function compared to pomegranate extracts from other parts. Pomegranate endocarp extract was fractionated with hexane, dichloromethane, ethyl acetate, butanol extract, and water, followed by evaluation for antioxidant activity. During this experiment, various antioxidant tests such as nitrite scavenging activity, reducing power, superoxide anion scavenging activity, $ABTS^+$ scavenging activity, SOD like-activity, and DPPH radical scavenging activity were conducted on the $CH_2Cl_2$, EtOAc, BuOH, $H_2O$ fractions of pomegranate endocarp extract. Results showed that the ethyl acetate fraction contained the highest DPPH radical scavenging activity among the samples. For SOD like-activity, the dichloromethane fraction had the highest antioxidant activity. For superoxide anion scavenging activity, the ethyl acetate fraction had the highest antioxidant activity. In the $ABTS^+$ scavenging activity test, the ethyl acetate fraction $IC_{50}$ was 39.26 ${\mu}g$/mL, whereas that of the butanol fraction was 40.95 ${\mu}g$/mL. In testing reduction power at 0.1 mg/mL, the O.D. of the ethyl acetate fraction was highest at 1.404 and showed higher activity than ascorbic acid at 1.332. The results of the nitrite scavenging activity test were very similar to those of the SOD like-activity test. These results suggest that pomegranate endocarp extract may have value as a natural antioxidant.

Physiological Functionality and Cytotoxic Effect of Korean Traditional Noble Wine, Samhaeju, and Commercial Rice Wine on Various Tumor Cell Lines (삼해주와 시판 곡주의 생리 기능성 및 세포 독성 효과)

  • This study was conducted to investigate the antioxidant activity, fibrinolytic activity and cytotoxic effect of Korean traditional noble rice wine made using different methods (A-C) and commercial rice wine (D-H) on various tumor cell lines. The antioxidant activity of rice wine was measured by DPPH (2,2-dipicryl-1-picrylhydrazyl), ABTS [2,2'-azinobis(3-ethylbenzthiazoline-6-sulphonic acid)] and NO (nitric oxide) radical scavenging assay. In this study, Samhaeju showed the greatest fibrinolytic activity of 13-17U and exhibited the highest antioxidant activity. Among the different Samhaeju, the sample prepared using method C had the highest antioxidant activity. The cytotoxic effect of rice wine were also examined against the human cancer cell line (A549 cells and HeLa cells) based on the results of a WST-1 assay and morphological changes. Rice wine induced the inhibition of cell proliferation and morphological changes in tumor cell lines in a concentration-dependent manner, with Samhaeju diluted 10 fold having the strongest effect on these factors. These findings suggest that Korean rice wine has antioxidant activity and cytotoxic effect, and that these factors are influenced by the method of preparation.

Biological Analysis of Enzymatic Extracts from Capsosiphon Fulvescens Using the Microbulbifer sp. AJ-3 Marine Bacterium (해양미생물 Microbulbifer sp. AJ-3을 이용한 매생이 효소분해산물의 생리활성 연구)

  • $Microbulbifer$ sp. AJ-3 was used to acquire the degrading products from $Capsosiphon$ $fulvescens$ (DPCF), which were investigated to determine its physiological activities. A crude enzyme extract from $Microbulbifer$ sp. AJ-3 hydrolyzes polysaccharide substrates such as agar, agarose, alginic acid, fucoidan, laminaran, starch, and chitin. Among them, agarose, laminaran, and alginic acid showed higher activities, especially alginic acid. The antioxidant activity of DPCF was measured by using 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and superoxide dismutase (SOD)-like activities and were about 32% and 93% at 2 mg/ml, respectively. In addition, the nitrite-scavenging activity of DPCF was about 82%, 53%, and 12% at pH levels of 1.2, 3.0, and 6.0, respectively. To determine the influence of DPCF on alcohol metabolism, the generating activity of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) was measured. The facilitating rate of ADH activity by DPCF was 130% at 2 mg/ml. The tyrosinase inhibitory activity of DPCF was slightly increased in a dose-dependent manner and was about 28% at 2 mg/ml. These results indicated that the enzymatic products from DPCF have a strong antioxidant, nitrite scavenging, and alcohol metabolizing activity.

Screening of Antioxidants and Cosmeceuticals from Natural Plant Resources in Jeju Island (제주 자생식물로부터 항산화 및 화장품 기능성 소재 탐색)

  • Fifty-four natural plants from Jeju Island, Korea were extracted by 70% methanol. The extracts containing the highest total phenolic contents (TPC) (>250 mg gallic acid equivalents/g dry sample) were obtained from Ostrya japonica, Geranium thunbergii, Malus sieboldii, Ardisia japonica, and Agrimonia pilosa. DPPH inhibition activity was greatest in Ardisia crenata at 94.1%. A high correlation was observed between DPPH inhibition activity and TPC ($R^2=0.87$). Tyrosinase inhibition activities of more than 85% were obtained from the extracts of Persicaria filiformis, Rhus javanica, Alnus firma, and Myrica rubra. On the other hand, the P. filiformis and M. rubra extracts each showed more than 90% XOD inhibition activity. The five natural plants with the highest biological activities were also extracted by pressurized liquid (PLE, 100% methanol, 13.6 MPa, $40^{\circ}C$). The DPPH and tyrosinase inhibition activities were almost the same in both the 70% methanol and PLE extracts. The $IC_{50}$ for tyrosinase and elastase inhibition activities in PLE were 802 and 88 ppm in A. japonica, and 959 and 66 ppm in M. rubra, respectively.

Characteristics of Quality and Antioxidant Activation of the Cookies adding with Mealworm (Tenebrio molitor) and Black Bean Powder (식용 밀웜(Tenebrio molitor)분말과 검은콩 분말을 첨가한 쿠키의 품질특성 및 항산화 활성)

  • The purpose of this study was to examine the physicochemical and antioxidant properties of cookies with mealworm (Tenebrio molitor) and black soybean powder. The leavening rates, and loss ratios were significantly increased with adding up the amount of mealworm powder (p < 0.05). The total polyphenol, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging ability, FRAP activity (ferric reducing antioxidant power) and ABTS radical scavenging ability (2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid) were all significantly increased as mealworm powder contents increased. In addition, cookies containing both mealworm and black soybean powder were higher in total polyphenol, DPPH and ABTS radical scavenging abilities than cookies containing only mealworm powder (p < 0.05).

Anti-Oxidative and Anti-Obesity Activities of Tetrapanax papyriferus and Siegesbeckia pubescens Extracts and their Synergistic Anti-Obesity Effects (통초.희렴 추출물의 항산화.항비만 활성 및 혼합물의 항비만 시너지 효과)

  • In this study, the anti-oxidative and anti-obesity activities of two medicinal herb extracts, Tetrapanax papyriferus (TP) and Siegesbeckia pubescens (SP), were evaluated using DPPH radical scavenging activity assay, lipase enzyme inhibition assay, and the cell culture model system. Both methanol extracts of TP and SP showed DPPH radical scavenging activities dose-dependently, and the $IC_{50}$ of DPPH radical scavenging activities of the two medicinal herbs were 65.23 and 47.79 ${\mu}g/ml$, respectively. Furthermore, both extracts suppressed effectively lipase enzyme activity dose-dependently. Moreover, TP and SP extracts significantly suppressed adipocyte differentiation, lipid accumulation, triglyceride (TG) contents on 3T3-L1 preadipocytes in a dose-dependent manner without cytotoxicity. Their anti-obesity effect was modulated by cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$ and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) gene and protein expressions. Furthermore, TP and SP possessed a synergistic effect on anti-obesity activity. The identification of the active compounds that confer the anti-obesity activity of TP and SP might be needed.

The Antioxidant Capacity and Whitening Effects of the Extract from the Mixture of 7 Different Vegan Cosmetic Ingredients (비건(Vegan)인증 화장품 원료 7종 혼합추출물의 항산화능 및 미백효과 연구)

  • This study attempted to investigate the antioxidant capacity and whitening effects of the extract from the mixture of 7 different vegan cosmetic ingredients. To examine radical scavenging capacity, DPPH, ABTS and FRAP assays were performed. In addition, polyphenol and flavonoid concentrations were measured to check antioxidant substances. Specifically, in a DPPH test, 105.74 mg ascorbic acid/g antioxidant capacity was observed. In an ABTS assay, 85.31 mg ascorbic acid/g antioxidant capacity was found. Lastly, in an FRAP assay, 1mg of the extract revealed ascorbic acid 198.01±5.50 ㎍ reducing power. Polyphenol and flavonoid concentrations were 30.19±0.75 mg/g and 9.12±0.36 mg/g respectively. In a cell-based assay, cytotoxicity and whitening activity were examined. In terms of cytotoxicity, '20% or less' was observed. Furthermore, the inhibition of melanin synthesis was '34.70±2.97%' at 100 ㎍/mL, confirming the possibility of the extract from the mixture of 7 different substances as a cosmetic ingredient.
