Comparison of physiological activity of medicinal mushrooms produced in Korea and China using different extraction solvents (한국 및 중국산 약용버섯류의 추출용매에 따른 생리활성 성분 비교)

  • In the present study, physiological activities of hot water extracts and 70% ethanol extracts of three types of medicinal mushrooms (Hericium erinaceus, Cordyceps militaris, and Ganoderma lingzhi) produced in Korea and China were investigated. Both the hot-water and 70% ethanol extracts of H. erinaceus and C. militaris from Korea displayed the highest 2,2-diphenyl-1-picrylhydrazy radical scavenging activities. Nitrite scavenging activities of hotwater extracts of G. lingzhi from Korea and China (41% and 39%, respectively) were higher than the activities of 70% ethanol extracts. Total polyphenol contents of hot water extracts of H. erinaceus and C. militaris from Korea were higher than those of 70% ethanol extracts. The ethanol extracts of G. lingzhi from Korea displayed the highest total polyphenol content. C. militaris from Korea displayed the highest ${\beta}-glucan$ level (45.11%). ${\beta}-glucan$ content of H. erinaceus from China (30.87%) was higher than H. erinaceus from Korea (16.94%). The findings indicate that healthy ingredients can be maximally extracted using the optimal solvents for each mushroom. These results will be useful in understanding the difference in physiological activities between the solvents used for the extraction of medicinal mushrooms from Korea and China.

Antioxidant activities of noni juice extracts using various ethanol ratios and quality characteristics of Konjak jelly added with noni juices (에탄올 농도에 따른 노니주스 추출물의 항산화활성 및 노니곤약젤리의 품질특성)

  • This study examined the antioxidant activities of noni juice (NJ) extracts using various ethanol ratios and quality characteristics of Konjak jelly (KJ) supplemented with NJ. The total polyphenol content of 0% EtOH extract (EE) was higher than that of the other samples. The 100% EE contained the highest amount of total flavonoids. The 50% EE showed higher DPPH radical scavenging activity (SA) than the other samples. ABTS+ radical SA was the highest at 100% EE. The pH and moisture content of KJ decreased as the amount of NJ increased. The sugar content of KJ increased with an increase in the amount of NJ. Hardness and gumminess showed an increasing tendency, while L and b values decreased with the addition of NJ. Regarding consumer acceptability, 25% noni KJ was evaluated to be high in color and flavor. As a result, KJ containing 25% NJ was suitable for increasing consumer acceptability and functionality of KJ.

Physicochemical characteristics and antioxidant activity of kimchi during fermentation (발효 단계별 김치의 이화학적 특성 및 항산화 활성)

  • In the present study, we investigated the physicochemical characteristics and antioxidant activity of kimchi during the fermentation process. Kimchi was fermented at 18.5 ℃, then after one day, the storage temperature was changed to 5 ℃ without fresh kimchi (Fresh; pH 5.6, total acidity 0.3%), which obtained optimum-ripened kimchi (OptR; pH 4.3, total acidity 0.64%), and over-ripened kimchi (OvR; pH 3.8, total acidity 1.24%). As a result, the glucosinolates content of the kimchi was increased during the fermentation process. Among the glucosinolates, glucoraphanin possesses the highest amounts in kimchi. In addition, the contents of sulforaphane and total polyphenol, which are common antioxidant compounds, were increased during the fermentation process. To evaluate the antioxidant activities of Fresh, OptR, and OvR, we measured 1,1-diphenyl-2-picrylhydrazyl (DPPH) and hydroxyl (·OH) radicals radical scavenging activity in vitro. Fresh, OptR, and OvR exerted DPPH and ·OH radical scavenging activities dose-dependently. In particular, the ·OH radical scavenging activities of OptR and OvR were higher than that of Fresh. Therefore, we suggest that kimchi at the ripe and over-ripe stage is considered to have high antioxidant activity by increasing glucosinolate, sulforaphane, and total polyphenols, compared with fresh kimchi.

Antioxidant Potential of Enzymatic Extracts from Blueberry (Vaccinium corymbosum L.) (블루베리 (Vaccinium corymbosum L.) 유래 효소 추출물의 항산화성)

  • Enzymatic extracts were prepared from the blueberry (Vaccinium corymbosum L.) collected in Jeju, Korea. Five carbohydrases namely AMG, Celluclast, Termamyl, Ultraflo and Viscozyme, and five proteases namely Alcalase, Flavourzyme, Kojizyme, Neutrase and Protamex were used to prepare the enzymatic extracts. Antioxidant properties of each extracts were studied using stable 1,1-diphenyl 2-picrylhydrazyl (DPPH), reactive oxygen species (ROS), nitric oxide (NO) scavenging, metal chelating assays and lipid peroxidation inhibitory activity in hemoglobin-induced linoleic acid system. The phenolic content of all enzymatic extracts was in the range of 517.85-597.96 mg/100 g dried sample. DPPH and NO${\cdot}$scavenging, and metal chelating assays exhibited prominent activities. Viscozyme showed the highest DPPH activity $(0.046{\pm}0.002\;mg/mL)$ while AMG Showed the highest activity in NO${\cdot}$scavenging $(0.339{\pm}0.011\;mg/mL)$. All the extracts exhibited strong metal chelating activities. Blueberry enzymatic extracts also showed relatively good activity in hydrogen peroxide scavenging. AMG showed the highest lipid peroxidation inhibitory activity $(0.28{\pm}0.01\;mg/mL)$ in hemoglobin-induced linoleic acid system. In this results, the blueberry, which has potential antioxidant components, may be a good candidate as a natural antioxidant source.

Changes in Antioxidant and Nitrite Scavenging Activities of Angelica keiskei and Brassica loeracea var. acephala Vegetable Juices Treated with UV Irradiation during Storage (UV 조사한 신립초 및 케일 녹즙의 항산화 활성 및 아질산염 소거작용의 변화)

  • To elongate the shelf-life of Angelica keiskei and Brassica loeracea var. acephala vegetable juices, UV irradiation was used and the changes of antioxidant activity and nitrite scavenging ability were investigated. The content of polyphenols of vegetable juices were slightly reduced by UV treatment and/or storage period. The DPPH radical-scavenging activities of the vegetable juices treated by UV were higher than that of control but were not changed during storage. However, $ABTS{\cdot}^+$ reducing activities of the vegetable juices were reduced by UV treatment. The $ABTS{\cdot}^+$ reducing activity of Brassica loeracea var. acephala juice was lower when the flow rate was slower. The ferrous ion chelating effects of Angelica keiskei vegetable juices were reduced by UV treatment. In contrast, the ferrous ion chelating effects of Brassica loeracea var. acephala vegetable juices were not different from those of right after manufacturing. The ferrous ion chelating effects on both vegetable juices increased during storage periods. The inhibitory activity of lipid oxidation was decreased slightly by UV treatment on vegetable juices. The nitrite scavenging ability of Angelica keiskei and Brassica loeracea var. acephala vegetable juices treated by UV irradiation was not different from that of control. The nitrate scavenging abilities of vegetable juices in pH 1.2 were higher than those in pH 3.0 and 4.2.

Changes of Bioactive Compounds and Antioxidant Activities in Korean Green Tea (Camellia sinensis) with Different Harvesting Periods (국산 녹차의 채취시기별 활성물질 및 항산화능 변화)

  • Korean green tea has been claimed to have health-promoting effects, which may be related to the antioxidant activity in vitro. Korean green teas (Woojeon, WJ ; Sejak, SJ ; Jungjak, JJ ; Daejak, DJ) were subjected to different harvested times and yet little research has examined their bioactive compounds. To assess the effect of this different harvested times on nutritional and health-related properties such as Korean green tea polyphenols, flavonoids, theanine and free amino acids, antioxidant activities and radical scavenging activities were determined. Total polyphenols in JJ (37.16 mg/g) was higher than in other products (WJ, 19.55 ; SJ 24.65 ; DJ, 23.28 mg/g). Contents of flavonol and flavone glycosides were the highest at DJ (350.83 mg%) as compared to WJ (220.81), SJ (256.88) and JJ (270.36 mg%). Contents of theanine and total free amino acids were the highest at WJ (14.11, 23.62 mg/g, respectively), but decreased thereafter. Antioxidant activities were higher in JJ and DJ, using the linoleic acid peroxidation, DPPH and ABTS free radical-scavenging activities. However, WJ and SJ had less active antioxidant activity and free radical-scavenging activity. Reducing powers were increased depend on the concentration of extracts. Antioxidant activity and free radical-scavenging activity of JJ and DJ seemed to relate with total polyphenol and flavonoid contents.

Comparative Radical Scavenging and Nitric Oxide Production Capacity of Water Extracts of Acanthopanax divaricatus var, Albeofructus and Eleutherococcus divaricatus var. Chiisanensis (지리산오가피와 흰털오가피 열수추출물의 라디칼 제거 활성 및 대식세포에서의 산화질소 생성능 비교)

  • In the present study, water extracts of Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis were prepared and their antioxidant and anti-inflammatory effects were compared. The water extract of Acanthopanax divaricatus var, albeofructus showed significantly higher activity in total polyphenol and flavonoid contents, while there were no significant differences in ABTS/DPPH radical scavenging activities and ferric reducing antioxidant power. However, the water extract of Eleutherococcus divaricatus var. chiisanensis at the highest dose showed significantly lower capacity in nitric oxide production than that of Acanthopanax divaricatus var, albeofructus. Taken together, there were differences between Acanthopanax divaricatus var, albeofructus and Eleutherococcus divaricatus var. chiisanensis in total polyphenol/flavonoid contents and anti-inflammatory effect, however, the antioxidant effect was almost similar.

Anti-oxidative and Anti-inflammatory Activities of Decaisnea insignis Ethanol Extract (Decaisnea insignis 에탄올 추출물의 항산화 및 항염증 활성)

  • This study was conducted to explore new nutraceutical resources from the plant kingdom possessing biological activities. To fulfill this purpose, the anti-oxidative and anti-inflammatory activities of Decaisnea insignis ethanol extract (DIEE) were evaluated. First, DIEE possessed potent scavenging activity against 1,1-diphenyl-2-picryl hydrazyl (DPPH), similar to ascorbic acid used as a positive control. Moreover, DIEE inhibited lipopolysaccharide (LPS)- and hydrogen peroxide ($H_2O_2$)-induced reactive oxygen species (ROS) in RAW 264.7 cells. Furthermore, DIEE induced the expression of an anti-oxidative enzyme, heme oxygenase 1 (HO-1), and its upstream transcription factor, nuclear factor-E2-related factor 2 (Nrf2), in a dose-dependent manner. The modulation of the HO-1 and Nrf2 expressions might be regulated by mitogen-activated protein kinases (MAPKs) and their upstream signaling pathways. On the other hand, DIEE suppressed LPS-induced nitric oxide (NO) formation without cytotoxicity. The inhibition of the NO formation was the result of the downregulation of inducible NO synthase (iNOS) by DIEE. The suppression of NO and iNOS by DIEE might be modulated by their upstream transcription factors, nuclear factor ${\kappa}B$ ($NF-{\kappa}B$), and activator protein 1 (AP-1) pathways. Taken together, these results provide important new insights that D. insignis possesses anti-oxidative and anti-inflammatory activities. Therefore, it might be utilized as a promising material in the field of nutraceuticals.

Quality Characteristics and Antioxidant Activities of Peach Makphyun (복숭아 막편의 품질 특성 및 항산화성)

  • The objective of this study was to develop peach Makphyun, a kind of rice cake, added with peach and Makgeolli. The effects of peach paste (0, 5, 10, or 20%) on quality characteristics of Makphyun were evaluated during storage at $20{\pm}2^{\circ}C$ for 3 days. As the concentration of peach paste increased, pH decreased and acidity increased. Reducing sugar contents (%) increased with the amount of peach paste. Hunter color b (yellowness) value of rice cake increased with increasing amount of peach paste, whereas L (lightness) and a (redness) values decreased. Contents of total phenols increased with increasing amount of peach paste. Antioxidant activities such as DPPH and hydroxyl radical scavenging activities of peach Makphyun decreased with increasing amount of peach paste. The sensory evaluation results showed that overall preference of Makphyun containing peach paste content of 20% is higher than other groups.
