Antioxidative and Cellular Protective Effects of Jeju Native Plant Extracts against Reactive Oxygen Species (I) (제주 자생 식물 추출물의 항산화 및 세포 보호작용에 관한 연구(I))

  • In this study, we investigated the anti-oxidative, anti-wrinkle and whitening effects of 36 plant extracts collected from self-growing plants in Jeju island. Their anti-oxidant activities were measured by free radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl radical), reactive oxygen species (ROS) scavenging activities on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay, and cell protecting activities using the rose-bengal sensitized photohemolysis of human erythrocytes. In addition, the inhibitory activities of tyrosinase for whitening effect and elastase for anti-wrinkle were investigated. The results showed that the Rumex crispus (all grass) extract has the most significant free radical scavenging activity ($FSC_{50};\;10{\mu}g/mL$), Plantago asiatica and Rumex crispus extracts for the prominent ROS scavenging activity ($OSC_{50};\;0.006{\mu}g/mL$, $0.04{\mu}g/mL$ respectively), Rumex crispus ($\tau_{50};\;1,140 min $at $50{\mu}g/mL$), Machilus thunbergii leaf (216 min), and Celastrus orbiculatus (200 min) for cell protecting effects, Morus alba stem for the inhibitory activity on tyrosinse (94.8% at $200{\mu}g/mL$), Rumex crispus (81.8% at $200{\mu}g/mL$), Morus alba (74.6%), and Celastrus orbiculatus leaf/stem/flower (63.1%) for the activity on elastase. These results indicated that the extracts of Rumex crispus, Plantago asiatica, Machilus thunbergii leaf, Morus alba stem, Celastrus orbiculatus leaf/stem/flower could have the functional effects when they are added as ingredients in cosmetics. Thus, it is concluded that further experiments are needed to apply for cosmetic products.

Antioxidative and Neuroprotective Effects of Enzymatic Extracts from Leaves of Perilla frutescens var.japonica

  • The antioxidative activity of various enzymatic extracts from leaves of Perilla frutescens var. japonica was evaluated by measuring 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, and alkyl radical scavenging activity using an electron spin resonance (ESR) spectrometer. For this study, the leaves were enzymatically hydrolyzed by 8 carbohydrases (Dextrozyme, AMG, Promozyme, Maltogenase, Termamyl, Viscozyme, Celluclast, and BAN) and 9 proteases [Flavourzyme, Neutrase, Protamex, Alcalase, PP-trypsin (trypsin from porcine pancreas), papain, pepsin, $\alpha$-chymotrypsin, and BP-trypsin (trypsin from bovine pancreas)]. The DPPH radical scavenging activities of Promozyme and Alcalase extracts were the highest, and the $IC_{50}$ values were 77.25 and $109.66\;{\mu}g/mL$, respectively. All enzymatic extracts of the leaves scavenged hydroxyl radical, and the $IC_{50}$ values of Celluclast and pepsin extracts which were the highest activity were 243.34 and $241.86\;{\mu}g/mL$, respectively. The BAN and $\alpha$-chymotrypsin extracts showed the highest scavenging activities, and the $IC_{50}$ values were 21.13 and $33.23\;{\mu}g/mL$, respectively. The pepsin extracts from the leaves showed protective effect on $H_2O_2$-induced DNA damage. In addition, the pepsin extracts decreased cell death in PC-12 cells against $H_2O_2$-induced oxidative damage. The findings of the present study suggest that enzymatic extracts of the leaves possess antioxidative activity.

Antioxidant effect of ethanol extract from Plantaginis Herba (차전초 에탄올 추출물의 항산화 효능)

  • Objectives : Butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA) were well known as anti-oxidant, but they were limited to use because of toxicity. So, many studies are being done to develope natural anti-oxidant. Total phenolic and flavonoid contents along with total antioxidant capacity of the ethanolic extract of Plantaginis Herba (PH) were evaluated to explore the reliable and potential sources of novel natural antioxidants. Methods : Total polyphenol contents and total flavonoid contents in PH ethanol extract were determined by colorimetric method. And DPPH(1.1-diphenyl-2-picrylhydrazyl), ABTS(2'-azino-bis (3-ethylbenzothiazoline-6-Surfonicacid)) free radical scavenging capacity and reducing power inhibition activities of PH ethanol extract were measured at 100, 500, 1000, $5000{\mu}g/m{\ell}$ concentrations by spectrometric assay. Results : The total polyphenol contents and total flavonoid contents of the extract were 161.99 mg/g, 144.05 mg/g, respectively. Also, DPPH, ABTS free radical scavenging capacity and reducing power of PH ethanol extract in treated concentrations (100, 500, 1000, $5000{\mu}g/m{\ell}$) increased dose dependently. In particular, DPPH free radical scavenging capacity of PH ethanol extract from $500{\mu}g/m{\ell}$ was significantly increased compared to positive control (BHA). ABTS free radical scavenging capacity of PH ethanol extract from $1000{\mu}g/m{\ell}$ was significantly higher than BHA. Also, reducing power showed that PH ethanol extract from $500{\mu}g/m{\ell}$ was significantly increased compared to BHA. Conclusions : These results suggest that PH ethanol extract has effects to scavenge free radicals, thus PH has potential and applicable benefits for development of materials and products to have anti-oxidation functions.

Screening on Radical Scavenging Activity of Marine Microalgae (해양미세조류의 라디칼소거활성 검색)

  • In order to screen new radical scavenging principle which is expected to be antiaging drug lead, we have investigated 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of the marine microalgae, greenalgae(10 speices), diatom (10 speices) and blue-green algae (10 speices). The significant activities$(IC_{50}:\;<100\;{mu}g/ml)$ were observed in 4 species of green algae (MA002, 006, 009, 010), 1 species of diatom (MA015) and 5 species of blue-green algae (MA017, 018, 019, 024, 025). Within the scope of family tested, MA009 $(IC_{50}:\;=78\;{mu}g/ml)$, MA015 $(IC_{50}:\;=38\;{mu}g/ml)$ and MA019 $(IC_{50}:\;=41\;{mu}g/ml)$ displayed the most significant activity. Among the marine microalgae tested at family level, cyanophycean blue-green algae was shown to be the most active family on screening of new bioactive compounds.

Antioxidant activities of phenolic compounds from Ziziphus jujuba Mill extract using subcritical water (대추 아임계수 추출물의 폴리페놀 함량 및 산화방지 평가)

  • Subcritical water extraction (SWE) is an eco-friendly extraction method that uses only purified water as a solvent under high temperature and high pressure conditions. In this study, total phenolics, and antioxidant activity were evaluated in Ziziphus jujuba Mill extracts from subcritical water obtained by varying the extraction temperature (110-190℃) and extraction time (1-20 min). Total phenolics was maximized with extraction at 190℃ for 15 min (67.79±3.45 mg gallic acid equivalent/g jujube). The 2,2-diphenyl-1-picrylhydrazyl free radical scavenging activity (48.84±4.74%) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) free radical scavenging activity (84.75±1.15%) were maximal at extraction conditions of 190℃, for 20 min. All jujube extracts prepared using SWE had higher total phenolics and antioxidant activities than extracts prepared using organic solvent extraction (60℃, 120 min), including methanol and ethanol. SWE could be an excellent alternative to organic solvents for extracting phenolics and antioxidant compounds.

Assessment of antinociceptive property of Cynara scolymus L. and possible mechanism of action in the formalin and writhing models of nociception in mice

  • Background: Cynara scolymus has bioactive constituents and has been used for therapeutic actions. The present study was undertaken to investigate the mechanisms underlying pain-relieving effects of the hydroethanolic extract of C. scolymus (HECS). Methods: The antinociceptive activity of HECS was assessed through formalin and acetic acid-induced writhing tests at doses of 50, 100 and 200 mg/kg intraperitoneally. Additionally, naloxone (non-selective opioid receptors antagonist, 2 mg/kg), atropine (non-selective muscarinic receptors antagonist, 1 mg/kg), chlorpheniramine (histamine H1-receptor antagonist, 20 mg/kg), cimetidine (histamine H2-receptor antagonist, 12.5 mg/kg), flumazenil (GABAA/BDZ receptor antagonist, 5 mg/kg) and cyproheptadine (serotonin receptor antagonist, 4 mg/kg) were used to determine the systems implicated in HECS-induced analgesia. Impact of HECS on locomotor activity was executed by open-field test. Determination of total phenolic content (TPC) and total flavonoid content (TFC) was done. Evaluation of antioxidant activity was conducted employing 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay. Results: HECS (50, 100 and 200 mg/kg) significantly indicated dose dependent antinociceptive activity against pain-related behavior induced by formalin and acetic acid (P < 0.001). Pretreatment with naloxone, atropine and flumazenil significantly reversed HECS-induced analgesia. Antinociceptive effect of HECS remained unaffected by chlorpheniramine, cimetidine and cyproheptadine. Locomotor activity was not affected by HECS. TPC and TFC of HECS were 59.49 ± 5.57 mgGAE/g dry extract and 93.39 ± 17.16 mgRE/g dry extract, respectively. DPPH free radical scavenging activity (IC50) of HECS was 161.32 ± 0.03 ㎍/mL. Conclusions: HECS possesses antinociceptive activity which is mediated via opioidergic, cholinergic and GABAergic pathways.

Effects of Grape Extracts on Free Radical Scavenging Activity and Inhibition of Pro-Inflammatory Mediator Production in Mouse Macrophage Cells (포도 추출물들의 자유 라디칼 소거 작용 마우스 대식세포주의 염증 발현 매개 인자들에 대한 생성 억제 효과)

  • Antioxidant and anti-inflammatory potentials of various grape extracts were evaluated. Extracts from Kyho seed, Kyho stem, and Campbell seed showed potent 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities compared to resveratrol $(IC_{50}=16.9,\;21.5,\;21.9,\;34.6\;{\mu}g/mL,\;respectively)$, among which, antioxidant effect of Kyho seed extract were similar to that of vitamin C $(IC_{50}=12.2\;{\mu}g/mL)$. These extracts also exhibited inhibitory activities on lipopolysaccharide (LPS)-induced prostaglandin $E_2$ production and nitrite formation in mouse macrophage RAW 264.7 cells at $50\;{\mu}g/mL$. Kyho stem and seed extracts showed growth inhibitory activities in human lung and colon cancer cells. These results suggest the potential roles of grape extracts as antioxidants and anti-inflammatory agents.

Purification and Identification of Antioxidant Peptides from Enzymatic Hydrolysate of Spirulina platensis

  • The aim of this study was to isolate antioxidant peptides from an enzymatic hydrolysate of Spirulina platensis. A novel antioxidant peptide was obtained by ultrafiltration, gel filtration chromatography, and reverse-phase high-performance liquid chromatography, with the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay used to measure the antioxidant activity, and the sequence was determined to be Pro-Asn-Asn (343.15 Da) by electrospray ionization tandem mass spectrometry. This peptide was synthesized to confirm its antioxidant properties, and it exhibited 81.44 ± 0.43% DPPH scavenging activity at 100 μg/ml, which was similar to that of glutathione (82.63 ± 0.56%). Furthermore, the superoxide anion and hydroxyl free-radical scavenging activities and the SOD activity of the peptide were 47.84 ± 0.49%, 54.01 ± 0.82%, and 12.55 ± 0.75%, respectively, at 10 mg/ml. These results indicate that S. platensis is a good source of antioxidant peptides, and that its hydrolysate may have important applications in the pharmaceutical and food industries.

Comparison of Oxidative Stability for the Thermally-oxidized Vegetable Oils using a DPPH Method (DPPH법에 의한 식용유지의 열산화 안정성 비교)

  • The 2,2-diphenyl picrylhydrazyl (DPPH) method, which can be used to predict the oxidative stability of edible oils, was previously reported by our research group. Not only free radical scavenging antioxidants but also radicals from oxidized oils are capable of reacting with DPPH radicals, thereby reducing the absorbance of DPPH. In this study, the optimum sample size of edible oils for the DPPH method was determined, and the oxidation of the edible oils was monitored via DPPH, coupled with other conventional methods. The optimum sample size was determined as 1.5 g using soybean oil. Soybean, corn, virgin olive, and refined olive oils were thermally oxidized for 3 hr at $180^{\circ}C$ and analyzed via DPPH, conjugated dienoic acid (CDA) value, and p-anisidine value (p-AV) protocols. Soybean and corn oils were found to be more sensitive to thermal oxidation than virgin and refined olive oils, on the basis of the CDA value and p-AV measurements. The DPPH method can indicate the inherent radical scavenging activity of unoxidized samples, the time required for the depletion of antioxidants, and the rate of degradation of the antioxidants. The soybean and corn oils evidenced higher levels of free radical scavenging compounds, required more time for the consumption of inherent antioxidants, and also manifested steeper antioxidant degradation rates than olive oils, based on the results of DPPH analysis. The DPPH method, accompanied by other conventional methods, may prove useful in predicting the degree of oxidation of vegetable oils.

Microbial Change and Fermentation Characteristics during Samjung-Hwan Natural Fermentation (천연발효 경과에 따른 삼정환의 미생물 변화 및 발효특성)

  • Objectives: Samjung-hwan (SJH), a well-known traditional fermented herb formula recorded in Dongui Bogam, has been commonly used for prolonging life for four hundred years in Eastern Asia. However, fermented SJH has not been investigated in terms of microbial ecology until present time. Methods: SJH was fermented for five weeks and fermentation characteristics during SJH fermentation were performed including pH, acidity and microbial profiling. Also, we measured total polyphenol and total flavonoid contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity. In order to select starter candidate, several lactic acid bacteria were isolated from fermented SJH. Results: pH of fermented SJH was decreased from 4.7 to 3.0 and acidity was increased from 0.45% to 1.72%. Also, fermented SJH increased antioxidant indicator such as total polyphenol and total flavonoid as well as DPPH free radical scavenging activity. Lactobacillus brevis was increased, Pseudanabaena sp. was decreased, and Lactococcus lactis subsp. lactis was stable during 5-week fermentation of SJH. L. brevis and L. plantarum were isolated from fermented SJH. Conclusions: Fermented SJH for four weeks had optimal effect on antioxidant and fermentation characteristics such as pH, acidity and microbial profile. Further studies are required to develop starter and analyze functional compounds in oder to produce standardized SJH.
