Polyphenol Contents of Rumex crispus Root Extract with Hot Water and its Antioxidative Effect (소리쟁이 뿌리 열수 추출물의 폴리페놀 함량 및 추출물의 항산화 효과)

  • This study was conducted to investigate the extraction characteristics including total polyphenol compound content (TPC) and the antioxidative effect of Rumex crispus root extract on commercial corn oil. Extraction yield was increased with extraction temperature but decreased with extraction ratio. No significant differences in aromatics content were found among the extracts prepared with various extraction conditions; extraction ratio, extraction temperature, extraction pH and composition of extractant. Total flavonoid content of the extract was increased with extraction temperature and extraction pH, and highest value of it was found when extractant composition of ethanol in water was 75%. Total polyphenol compound content (TPC) of the extract with 10 fold extraction ratio showed the highest value, but no significant difference in TPC was found with extraction temperature. Reducing power and DPPH (${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging ability (RSA) of the extract in spectrophotometric absorbance were decreased with extraction ratio but increased with extraction temperature and showed 63.1%~98.4% and 67.6%~86.7% of those of ascorbic acid in reducing power and DPPH RSA, respectively. The antioxidative effects of the extract on corn oil were 84.8~93.0% of that of commercial butylated hydroxytoluene, the antioxidant index value was highest when the ethanol composition to water in extractant was equal ratio.

DPPH Free Radical Scavenging Effect of the Aerial Parts of Desmodium oldhami (큰도둑놈의갈고리 지상부의 DPPH Free Radical 소거효과)

  • The antioxidant activity of Desmodium oldhami (Leguminosae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of D. oldhami showed the strong antioxidant activity. Five compounds, kaempferol-3-O-rhamnopyranoside (afzeline) (1), quercetin-3-O-[$\alpha$-L-rhamnopyranosyl($1{\rightarrow}6$)-$\beta$-Dglucopyranoside] (rutin) (2), kaempferol-3-O-glucopyranoside (astragalin) (3), genistein-7-O-$\beta$-D-glucopyranoside (genistin) (4), kaempferol-3-O-rutinoside (5) were isolated from the active ethyl acetate soluble fraction of D. oldhami through repeated silica gel and Sephadex LH-20 column chromatography. Among them, compound 2 showed the most significant antioxidative effect on DPPH free radical scavenging test. Compounds 1-5 are reported for the first time from this plant.

  • Ethyl acetate-soluble neutral fraction of hot water extracts from the aerial parts of Angelica keiskei showed a 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity. Six antioxidative compounds were purified and isolated by various chromatographic procedures. Based on the analyses of FAB-MS and NMR, the isolated compounds were structurally elucidated as luteolin 7-O-${\beta}$-D-glucopyranoside (1), quercetin 3-O-${\beta}$-D-galactopyranoside (2), quercetin 3-O-${\beta}$-D-glucopyranoside (3), quercetin 3-O-${\alpha}$-D-arabinopyranoside (4), kaempferol 3-O-${\alpha}$-D-arabinopyranoside (5), and luteolin 7-O-rutinoside (6). The glycosides of flavonols and luteolin showed DPPH radical-scavenging activity. One molecule of 2, 3, 4, 6, 1, and 5 scavenged 4.2, 4.2, 4.1, 2.5, 2.2, and 1.4 molecules of DPPH radical, respectively.

Antioxidant Capacity and Quinone Reductase Activity of Methanol Extracts and Fractions from Papaya Seed (파파야씨 추출물 및 분획물의 항산화, QR 활성)

  • In this study, the antioxidant activity of methanol extracts and fractions from papaya seed were investigated in vitro. Total polyphenol contents of methanol extracts and fractions from papaya seed varied from 17.74 to 125.99 ${\mu}g/mg$ and total flavonoid contents varied from 1.60 to 32.69 ${\mu}g/mg$. Contents of polyphenol and flavonoid in ethyl acetate (EtOAc) fraction was found to be extremely high (compared with the other fractions examined). Radical-scavenging activities of methanol extracts and fractions were examined using ${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) radicals, 2,2'-azino-bis (3-ethyl-benzthiazoline-6-sulfonic acid) (ABTS) and hydrogen peroxide assay. As a result, ethyl acetate fraction of papaya seed showed the highest radical-scavenging activity in various antioxidant systems. The EtOAc fraction from papaya seed induced QR activity in concentrations of 12.5 to 50 ${\mu}g/ml$ with a maximum of a 3.3-fold induction at 50 ${\mu}g/ml$ of fraction. Therefore, the most effective QR inducer among these fractions can be said to reside in the EtOAc fraction, indicating that strong constituents responsible for QR induction potency in the papaya seed extract are largely contained in the EtOAc fraction.

Rheological Properties and Sensory Characteristics of Roll Bread with Chungkukjang Powder (청국장 분말을 첨가한 롤빵의 물성 및 관능적 특성)

  • The purpose of this study was to investigate quality characteristics of roll bread with the addition of Chungkukjang powder(0, 5, 10, 15, 20%). The antioxidative activity of antioxidative substances produced from several bacterial strains isolated from fermented foods were tested by DPPH $(\alpha,\alpha’-diphenyl-\beta-picrylhydrazyl)$ free radical scavenging activity. The most optimal medium condition for the production of antioxidative substance from Bacillus sp. FF-7 was 2% galactose as carbon source and 1% tryptone as nitrogen source. In color values, with increasing Chungkukjang powder content, the "lightness" (p<0.05) decreased in the crumb bread, while the redness(p<0.05) and yellowness(p<0.05) increased. The volume and specific volume of roll bread with 5.0% Chungkukjang powder were the highest(p<0.05). In the rheology measurements for the roll bread, the hardness (p<0.05) decreased with the addition of 5.0% Chungkukjang powder, but then increased with further additions of Chungkukjang up to 10.0%. Highest sensory scores for color, flavor, taste(p<0.05), texture(p<0.05) and overall acceptance(p<0.05) were obtained from roll bread with 5.0% Chungkukjang powder and control. From the results of this study, it was assumed that the development of food products using Chungkukjang is prospective in response to health-oriented consumers.

Antioxidants and Anti-inflammatory Effects of Fermented Houttuynia cordata Thunb and Scoria Mixture Extract

  • In this study, antioxidant and anti-inflammatory of fermented Houttuynia cordata Thunb and scoria mixture extract were investigated in vitro. Radical-scavenging activities of the ethanol extracts were examined by using ${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) and 2,2-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS)) radicals assay. Consequently, we confirmed that fermented Houttuynia cordata Thunb and scoria mixture extract dependent removed DPPH and ABTS radical. Also, to confirm anti-inflammatory activity of ethanol extract, we treated fermented Houttuynia cordata Thunb and scoria mixture extract on BV-2 cell with LPS. The result showed that fermented Houttuynia cordata Thunb and scoria mixture extract concentration-dependent inhibited NO production. Therefore, fermented Houttuynia cordata Thunb and scoria mixture extract showed inhibition radical oxygen activities and inflammatory and have available for a pharmacological composition on neuritis-protection and treatment.

Anti-Oxidative and Anti-Neuroinflammatory Effect of Ethanol Extracts from Walnuts's (Juglans regia L.) Shell

  • In this study, antioxidant and anti-neuroinflammatory of ethanol extracts from walnuts's (Juglans regia L.) shell were investigated in vitro. Radical-scavenging activities of the walnuts's shell ethanol extracts (WSE) were examined by using ABTS radicals and ${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) radicals assay. In the ABTS and DPPH radical scavenging activity, $RC_{50}$ of WSE were measured as 15.74 and $40.13{\mu}g/mL$, respectively. Also, to evaluate the anti-neuroinflammatory effects of WSE in lipopolysaccharide (LPS)-stimulated BV-2 microglial cells. The production of proinflammatory cytokines NO were examined by LPS in BV-2 cell. BV-2 cells activated with LPS were treated with various doses (10, 25, 50, $100{\mu}g/mL$) of WSE. Supernatants were analyzed for the production of NO using Griess reagent. WSE up to $10{\mu}g/mL$ still required to inhibit NO induced by LPS. These results showed that walnuts's (Juglans regia L.) shell can be used as an easily accessible source of natural anti-neuroinflammatory and natural antioxidants.

Lifespan Extension Property of Quercetin-3-O-${\beta}$-D-glucopyranoside-7-O-${\alpha}$-L-rhamnopyranoside from Curcuma longa L. In Caenorhabditis elegans (강황지하부 부산물에서 분리한 Quercetin-3-O-${\beta}$-D-glucopyranoside-7-O-${\alpha}$-L-rhamnopyranoside가 선충의 수명연장에 미치는 영향)

  • After harvesting the medicinal parts of Curcuma longa, the remaining underground parts were discarded. From the remaining underground parts of Curcuma longa quercetin-3-O-${\beta}$-D-glucopyranoside-7-O-${\alpha}$-L-rhamnopyranoside (Q37) was isolated. The antioxidant activities in vitro and lifespan-extension effect of Q37 were elucidated using the Caenorhabditis elegans. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging effect of Q37 showed similar potent activities in comparison with vitamin C. Q37 also showed potent superoxide quenching activities as measured by the riboflavin- and xanthine-originated superoxide quenching activity tests. Q37 prolonged lifespan of worms under normal culture condition. In terms of protective effect of Q37 on the stress conditions such as thermal and oxidative stresses, Q37-treated worms exhibited enhanced survival rate, as compared to control worms. To know the possible mechanism of Q37-mediated increased lifespan and stress resistance of worms, we examined the activities of Q37on superoxide dismutase (SOD), and invested intracellular reactive oxygen species (ROS) levels. The results revealed that Q37 was able to elevate SOD activity of worms and reduce intracellular ROS accumulation in a dose-dependent manner.

  • A phytochemical investigation on the methanolic extract of Cosmos caudatus has led to the isolation of quercetin $3-O-{\beta}-D-arabinofuranoside$ (1), quercetin $3-O-{\beta}-D-rhamnoside$ (2), quercetin $3-O-{\alpha]-D-glucoside$ (3) and quercetin (4). These compounds were shown to be the antioxidative constituents of the plant when evaluated using the ferric thiocyanate (FTC) and thiobarbituric acid (TBA), and radical scavengers based on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assays.

Constituents of Flowers of Carthamus tinctorius L. and Their Antioxidant Activity (홍화의 성분 분리 및 항산화 활성)

  • As part of our ongoing study focused on the discovery of antioxidants from natural products by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, methanol extract of flowers of Carthamus tinctorius L. was found to show potent antioxidant activity. Activity-guided fractionation of the methanol extract lead to the isolation of twenty compounds including two flavonol glycosides, quercertin-3-O-${\beta}$-D-glucopyranoside (12) and kaempferol-3-O-${\alpha}$-L-rhamnopyranosyl-${\beta}$-D-glucopyranoside (18), two flavanone glycosides, (2S)-4',5,6,7-tetrahydroxyflavanone 6-O-${\beta}$-D-glucopyranoside (15) and (2R)-5,7,8',4-tetrahydroxyflavanone 8-O-${\beta}$-D-glucopyranoside (16), and two acetylenic glycosides, 8Z-decaene-4,6-diyne-1-O-${\beta}$-D-glucopyranoside (13) and 4,6-decadiyne-1-O-${\beta}$-D-glucopyranoside (14). Their chemical structures were identified by using spectroscopic analysis. Among them, compounds 12-18 were tested in DPPH assay. Compounds 13-16 were first reported to their antioxidant activity. Quercertin-3-O-${\beta}$-D-glucopyranoside (12) showed the most potent inhibitory effect on DPPH with $IC_{50}$ value of 56.7 ${\mu}M$.
