Vegetative growth of four different strains of Hericium erinaceus was observed. The temperature suitable for optimal mycelial growth was determined to be $25^{\circ}C$, with growth observed in the extend temperature range of $20{\sim}30^{\circ}C$. The different strains of this mushroom showed distinct pH requirements for their optimum vegetative growth, with the most favorable growth observed at pH 6. Considering vegetative mycelial growth, PDA, YM, Hennerberg, Hamada, and Glucose peptone were the most favorable media, and Czapek Dox, Hoppkins, Glucose tryptone, and Lilly were the most unfavorable media for these mushroom strains. With the exception of lactose, most of the carbon sources assayed demonstrated favorable vegetative growth of H. erinaceus. For mycelial growth, the most suitable nitrogen source was alanine and the most unsuitable was histidine. Oak sawdust medium supplemented with $10{\sim}20%$ rice bran was the best for mycelial growth of the mushroom.
Incubation condition affecting the chlamydospore formation and isolation from mycelia and conidia of Cylindrocarpon destructanse (isolate ACY-9701), isolated from the root rot lesion of the American ginseng (Panax quinquefolium) was investigated. Chlamydospores were formed from mycilia but not from conidia on the Czapek-Dox agar without carbon or nitrogen source after 20 days incubation at 2$0^{\circ}C$. In the medium added with nitrogen and carbon sources, immatured chlamy-dospore-like cells were formed from microconidia and mycelia as well. Immatured chlamydospore-like cells were formed from mycelia as well as microconidia In corn, kidney bean, and pea root extracts after 20 days incubation at 20"C, while typical chlamydospores were formed from both of them in the root extract of Panax quinquefolium. The 3.6 log chlamydospore/mm" was converted from microconidia in the medium, which was equal to 2.5% conidia formed. Under the light condition (251.1 pmol/m" sec, 12 hrs dark and light cycle), 4.2 log/mm" of chlamydospores were converted from interracially or terminal cells of macroconidia, which was 4.0% of macroconidia produced on Potato dextrose agar (PDA). When mycelia and microconidia were stored at -7$0^{\circ}C$ for 32 days and incubated on PDA after thawing at room temperature to isolate chlamydospores from them, microconidia and mycelia were still alive. Meanwhile, microconidial lysis was found after heating them at 32$^{\circ}C$ for 7 days, but the chlamydospores converted from macroconidia were not lysed up to 13 days at 32"C. to 13 days at 32"C.ot;C.
A polymeric micelle, based on the poly(benzyl-L-histidine)-b-poly(ethylene glycol) (polyBz-His-b-PEG) diblock copolymer, was designed as a tumor-specific targeting carrier. The micelles (particle size: 67-80 nm, critical micelle concentration (CMC); 2-3 $\mu$g/mL) were formed from the diafilteration method at pH 7.4, as a result of self-assembly of the polyBz-His block at the core and PEG block on the shell. Removing benzyl (Bz) group from polyBz-His block provided pH-sensitivity of the micellar core; the micelles were physically destabilized in the pH range of pH 7.4-5.5, depending on the content of the His group free from Bz group. The ionization of His group at a slightly acidic pH promoted the deformation of the interior core. These pHdependent physical changes of the micelles provide the mechanism for pH-triggering anticancer drug (e.g., doxorubicin: DOX) release from the micelle in response to the tumor’s extracellular pH range (pH 7.2-6.5).
The objective of this study was to evaluate the reversal of multi drug resistance of human cell lines by specific inhibitors of ${\alpha}-glycosidase$ and mannosidases that had been reported to be involved in N-linked oligosaccharide processing of glycoproteins. N-methyldeoxynojirimycin, I-deoxynojirimycin, and castanospermine, which were known to be potent inhibitors of both ${\alpha}-glycosidase$ I and II, showed no activity against the multidrug resistant phenotype of the cell lines of SNU1DOX, KB-V1, and MCF-7/ADR. In contrast, I-deoxymannojirimycin, an inhibitor of mannosidase I, resulted in a slight reversal for the vinblastine resistance of the KB-V1 cell line, but did not show any activity toward the other cell lines. Parallel experiments with tunicamycin, an inhibitor of N-linked glycosylation, also resulted in no significant changes in multidrug resistant (MDR) phenotype of the cell lines tested in this work. These observations suggest that the unglycosylation of P-glycoprotein associated with the inhibitor treatments might not be correlated with the reversal of multidrug resistance of the cell lines tested in this study.

Flat form technology for constructing anticancer loaded multi-walled carbon nanotubes (mwCNTs) was introduced in this study. Conventional anticancer drugs, such as MTX (Methotrexate), cisplatin, DOX (Doxorubicin hydrochloride), DAU (Daunorubicin) and EPI (epirubicin) were bio-conjugated with folic acid (FA) for selective targeting tumor cells. Loading efficiencies of the used anticancer drugs on mwCNTs have shown different order of bindings depending on the molecular bind affinity of NH (amine) formation on mwCNTs. MTT assays have shown increased selective target efficiency of FA conjugated mwCNTs on breast cancer cell growth inhibition. All results collectively indicated promising application of mwCNTs as a smart inorganic nanomaterial for selective targeting drug delivery vehicle at tumor tissues.
The effects of carbon and nitrogen sources on the mycelial growth and sporulation of microconidia and chlamydospores of five isolates of Cylindrocarpon destructans (Zinssm.) Scholten causing root rot of Panax ginseng were studied. For the carbon sources, fructose, glucose, maltose, and sucrose in Czapek-Dox broth showed good mycelial growth of 178∼201 mg in dry weight compared with 64 mg of the control. The best carbon sources tested for conidial formation were sucrose and maltose with 2.75 and 3.03 log conidia/ml, respectively. For the nitrogen sources, aspartic acid, NaNO3, KNO3, arginine, threonine, and leucine increased mycelial growth of the fungi to 208∼231 mg in dry weight without significant difference (p=0.05) among them. Meanwhile the growth with cystine was poor (26.3 mg dry weight), and no conidium and chlamydospore were formed. Maximum microconidial formation was observed in the media with NaNO3 and KNO3 as 3.37 and 3.35 log conidia/ml, and for the chlamydospore formation the (NH4)2SO4-containing medium and the nitrogen-absent medium were the best as 3.40 and 3.57 log chlamydospores/ml, respectively. No conidium was found in the medium without nitrogen sources, in which chlamydospore formation increased 6 times more than in the nitrogen-amended medium. However, deletion of carbon source in the medium did not affect on the formation of conidia and chlamydospores of C. destructans.

In view of the warmth and hunidity in Korea, there is little possibility to avoid the fungal contamination of rice soybean paste which are major foodstuffs in this country. In order to isolate aflatoxin producing fungi from these foodstuffs, 6 samples of rice were collected from the farmhouses at Cbangsung and 40 samples of soybean paste in Seoul areas. The peptone glucose chloramphenicol, Czapek-Dox agar and malt agar were used for the isolation and identification of fungi, and Adye and Meteles medium and/or Sahouraud medium for the production of toxins. The cultures for the toxin production were done at $25^{\circ}C$ for $5{\sim}7$ days. Eleven Penicillium species were isolated. from 6 samples of rice, and 37 Aspergillus flavus, 48 Aspergillus species, 1 Penicillium species and 105 others were isolated from 40 samples of soybean paste. Among these, one Penicillium species isolated from rice was found to be aflatoxin producing strain, and the toxin was quantitatively measured. Aflatoxin appeared to be more favorably produced in the glucose Sabouraud medium than in the Adye and Meteles medium.
The objective of this study was to isolate and identify the chemical structure of a biosurfactant produced by Nocardia farcinica strain BN26 isolated from soil, and evaluate its in vitro antitumor activity on a panel of human cancer cell lines. Strain BN26 was found to produce glycolipid biosurfactant on n-hexadecane as the sole carbon source. The biosurfactant was purified using medium-pressure liquid chromatography and characterized as trehalose lipid tetraester (THL) by nuclear magnetic resonance spectroscopy and mass spectrometry. Subsequently, the cytotoxic effects of THL on cancer cell lines BV-173, KE-37 (SKW-3), HL-60, HL-60/DOX, and JMSU-1 were evaluated by MTT assay. It was shown that THL exerted concentration-dependent antiproliferative activity against the human tumor cell lines and mediated cell death by the induction of partial oligonucleosomal DNA fragmentation. These findings suggest that THL could be of potential to apply in biomedicine as a therapeutic agent.
Lactobacillus casei KC-324 was tested for its ability to inhibit aflatoxin production and mycelial growth of Aspergillus flavus ATCC 15517 in liquid culture. flatoxin $B_{1}$ biosynthesis and mycelial growth were inhibited in both simultaneous culture and individual antagonism assays, suggesting that the inhibitory activity was due to extracellular metabolites. produced in cell-free supernatant fluids of the cultured broth of L. casei KC-324. In cell-free supernatant fluids of all media tested, deMan, Rogosa and Sharpe broth, potato dextrose broth, and Czapek-Dox broth+1% yeast extract showed higher antiaflatoxigenic activity. In these case, fungal growths, however, was not affected as measured by mycelial dry weight. The antiaflatoxigenic metabolites from L. casei KC-324 were produced over wide range of temperatures between $25^{\circ}C$ and $37^{\circ}C$. However, these metabolites were not thermostable since the inhibitory activity of the supernatant was inactivated within 30 minutes at $100^{\circ}C$ and $121^{\circ}C$. The inhibitory activity was not influenced by changing pH of supernatant between 4 and 10. However, the antiaflatoxigenic activity was slightly reduced at pH 10.
인삼뿌리썩음병균(Cylindrocarpon destructans)후막 포자의 형성은 CD(Czapek`s dox broth)와 SNY(Spezieller Nahrtoffarmer yeast extract broth)배지에서 진탕배양(2$0^{\circ}C$, 120 rpm)하면 양호하였다. 특히 CD배지에서 2$0^{\circ}C$로 30일간 진탕배양하면 5.99 log 후막포자/ml로 형성되었다. 후막포자의 발아율은 12.9~23.3%로 CD배지와 SNY배지에서 좋았으며 발아적온은 5~1$0^{\circ}C$였고, 최적 pH는 6이었다. 그러나 2$0^{\circ}C$이상에서는 전혀 발아하지 않았으며 pH 7.0이상에서는 발아율이 급속히 저하되었다. 인삼추출물 3%, GA 10 ppm, IAA 10 ppm, NOVOZYM\ulcorner234 20 ppm을 처리 할 경우 대조구에 비해 발아율이 증가하였다. 특히 NOVOZYM\ulcorner234 20 ppm 처리구는 발아율이 무처리에 비해 2배 증가하였다. CD배지(pH5)에 NOVOZYM\ulcorner234 20 ppm과 GA 10 ppm을 조합처리하고 5$^{\circ}C$로 정치배양 할 때 후막포자의 발아율은 49.4%로 가장 높았다.
