Effect of Whitmania pigra whitman on the Allergic Inflammatory Response (수질(水蛭)이 비만세포의 알러지 염증 반응에 미치는 영향)

  • Objectives: Whitmania pigra whitman (WP) has been used in herbal medicine to treat various conditions, such as eczema, skin burns and frostbites in herbal medicine. The purpose of this study is was to investigate the effect of WP on anti-allergy mechanism. Methods: To clarify the mechanism, the effect of WP on vascular permeability of rat cutaneous tissue and histamine and cytokines (IL-6, IL-8, $TNF-{\alpha}$) released from mast cells were observed. Results: The results were 1. the pretreatment with WP significantly decreased the compound 48/80-induced degranulation and histamine release from RPMC 2. WP did not inhibit the anti-DNP IgE-induced increment of vascular permeability of rat cutaneous tissue 3. WP significantly reduced the PMA plus A23187-induced increment of expression of IL-6, IL-8, and $TNF-{\alpha}$ in HMC-1 cells. Conclusions: The present study providesevidence that WP acid inhibits mast cell-derived inflammatory allergic reactions by blocking histamine release and pro-inflammatory cytokine expression, and suggest the mechanisms of action. Furthermore, in vivo and in vitro anti-allergic effect of WP suggests a possible therapeutic application of this agent in inflammatory allergic diseases.

The Development of New Diffusive Sampler for Formaldehye in Air (기중 포름알데히드 측정을 위한 확산포집기의 개발)

  • To utilize diffuse sampling of formaldehyde in air, a new sampler was designed. A glass fiber filter was impegnated with 2,4-dinitrophenylhydrazine(DNP) and phosphoric acid and mounted 37mm cassette. The formaldehyde vapor was sampled in the dynamic chamber and measured by high performance liquid chromatograph and compared with solid sorbent tube. The results were as follows ; 1. The desorption efficiencies of diffusive sampler between 97.0% and 100%. 2. the sampling rate is constant as in $58{\sim}61.8m{\ell}/min$ when sampling times are between 120 and 360 min. 3. There was a significant relationship between concentrations of diffusive samples and active samples with the coefficient of determination(R2) of 0.92. 4. Desorbed amount of formaldehyde diffusive sampler was increased by high relative humidity. 5. Wheth diffusive samplers were stored at room temperature or at refrigerator there was no statistically significant difference in the accuracy of result. 6. When the diffusive samplers, which collected formaldehyde vapor, were exposed to clean air for three hours, there was no significant loss of formaldehyde due to reverse diffusion. In conclusion, this study suggest that developed diffusive samplers will be a reasonable substitute for the solid sorbent tube for sampling formaldehyde and practical comparative study of developed diffusive sampler should be performed at workplaces of manufacturing industry.

  • Artificial induction of nitrogen deficiency symptoms (leaf chlorosis) with two root respiratory inhibitors(DNP and Na$_2$S) was studied and regarded to be the same characteristic to red discoloration in rice var. Tongil seedling leaves as well as adult ones. Tongil(IR 667) was shown to be more nitrogen sensitive and have more distinctive appearance of the leaf discoloration than Punggwang(a native Japonica-type varity.) Conclusions were drawn from the present data that so-called red discoloration of Tongil under the natural field conditions is brought about either by insufficient nitrogen supply in soils or certain factors which may limit at any time the root absorption of nitrogen (low temperature, toxic gases or substances, poor drainage, around roots, etc.) in soils even with ample supply of it.

  • Samul-Tang (SMT) has been used for nourishing of the blood, hematopoiesis as a herbal medicine history. The purpose of this study is to find out anti-allergic inflammatory reaction of SMT. To clarify the mechanism, the effect of SMT on vascular permeability of rat cutaneous tissue and histamine and cytokines (IL-6, IL-8, TNF-${\alpha}$) release from mast cells were observed. Anti-Oxidative and Anti-Inflammatory Effect of Water Extract from Perillae Semen in RBL-2H3 Cells (RBL-2H3 세포에서 차조기씨 물 추출물의 항산화 및 항염증 효과)

  • The present study was to investigate anti-oxidative and anti-inflammatory activity of Perillae semen in RBL-2H3 basophilic leukemia cells. Inhibitory effect of Perillae semen onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activities in vitro. Anti-inflammatory actions of Perillae semen extracts (100, 250, $500\;{\mu}g/mL$) were assessed by testing their effects on the degranulation of mast cells. For this, $\beta$-hexosaminidase released from RBL-2H3 cells was used and proinflammatory cytokines were measured by an ELISA kit. Our results indicated that Perillae semen water extracts effectively inhibited free radical generation. At the concentration of $500\;{\mu}g/mL$ of water extract, the degranulation of RBL-2H3 cells were inhibited by 42.1%. The IgE-antigen complex increased the accumulation of IL-4 and TNF-$\alpha$ secretion in RBL-2H3 cells and treatments with 250 and $500\;{\mu}g/mL$ of Perillae semen extracts suppressed the IgE induced secretion of IL-4 and TNF-$\alpha$ protein by 20.5, 26.9% and 14.5, 16.5% respectively. We observed that Perillae semen water extract reduced $\beta$-hexosaminidase, IL-4, and TNF-$\alpha$ secretion in RBL-2H3 cells. These results provide that Perillae semen may be beneficial in the treatment of allergic inflammatory disease.

Effects of Tribuli Fructus extract on inflammatory responses in IgE-stimulated RBL-2H3 mast cells (비만세포에서 백질려 추출물의 항염증효과에 대한 연구)

  • Objectives : Tribulus terrestris $Linn{\acute{e}}$ (Tribuli Fructus; TF) has been used to treat hypochondrium, agalactia, nebula, itching and vitiligo in traditional Korean medicine. In this study, we investigated the effects of TF 30% ethanol extract on inflammatory responses in IgE-stimulated RBL-2H3 mast cells. Methods : TF extract was prepared by 30% ethanol. RBL-2H3 cells, a rat mast cell line, were treated with TF extract at different concentrations for 1 hr and then stimulated with DNP-IgE/HSA for indicated times. Cell viability was measured by WST-1 assay. The expression of inflammatory cytokines (IL-4, IL-13 and $IFN-{\gamma}$) mRNA was determined by reverse transcriptase-PCR, and the phosphorylation of ERK1/2, p38 and JNK MAP kinases (MAPKs) was determined by Western blot. The nuclear expression of $NF-{\kappa}B$ p65 in the cells was detected by Western blot and immunocytochemistry, respectively. Results : The treatment of TF extract at 0.1 and $0.2mg/m{\ell}$ significantly decreased the expression of IL-4 and IL-13 mRNA in IgE-stimulated RBL-2H3 mast cells, while significantly increased the expression of $IFN-{\gamma}$ mRNA. TF extract treatment was also inhibited the phosphorylation of ERK1/2, p38 and JNK MAPKs in IgE-stimulated RBL-2H3 mast cells in a dose-dependent manner. In addition, TF extract significantly blocked the translocation of $NF-{\kappa}B$ p65 into the nuclear of cells after IgE stimulation. Conclusions : These results indicate that TF extract inhibits inflammatory response in IgE-stimulated mast cells through blocking MAPKs/$NF-{\kappa}B$ pathway. This suggests that TF extract has an anti-inflammatory activity in mast cell activation.

Antioxidative and Anti-asthma Effect of Morus Bark Water Extracts (상백피 물 추출물의 항산화 활성 및 항천식 효과)

  • This study was performed to evaluate the antioxidant activities and anti-asthma effects of Morus bark water extracts. Inhibitory effect of Morus bark onto free radical generation was determined by measuring DPPH and hydroxyl radical scavenging activities in vitro. Anti-asthma activities of Morus bark water extracts were assessed by testing their effects on the degranulation of mast cell. For this, $\beta$-hexosaminidase released from a basophilic cell line, RBL-2H3 was used and pro-inflammatory cytokines were measured by ELISA kit. The antioxidant activities of water extracts of Morus bark was 59.2% in the DPPH assay at $2,000\;{\mu}g/mL$ and 78.8% in the hydroxyl radical scavenging assay at $2,000\;{\mu}g/mL$. Our results indicated that Morus bark water extracts effectively inhibited free radical generation. Morus bark water extracts inhibited inflammation-mediating substances such as histamine and $\beta$-hexosaminidase release from RBL-2H3 cells. Cytokine release demonstrated a more effective blockading ability of the Morus bark water extracts to the release of IL-4 and TNF-$\alpha$ compared to control. These results demonstrate that Morus bark may be beneficial in the treatment of allergic inflammatory disease.

Study on Anti-allergic Effect and Safety of Bangpung-galgeun-tang (방풍갈근탕(防風葛根湯)의 항알레르기효과에 관한 연구)

  • The purpose of this study was to examine the anti allergic effect in vivo and in vitro, and to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated anti DNP IgE-mediated passive cutaneous anaphylaxis in rodents and compound 48/80-induced active systemic anaphylatic shock in mice after oral administration with BGT of 0.4 g/kg and 0.8 g/kg for 8 days, and also examined MTT assay, ${\beta}-hexosaminidase$ activity, IL-4 and $TNF-{\alpha}$ from RBL-2H3 and $TNF-{\alpha}$ from Raw264.7 after pre-treatment with BGT of 0.25 mg/ml, 0.5 mg/ml, 1 mg/ml and 2 mg/ml. To ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg /p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. BGT inhibited passive cutaneous anaphylaxis and active systemic anaphylatic shock by oral administration. All the concentrations of BGT from 0.25 to 2 mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosaminidase$ release, IL-4 and $TNF-{\alpha}$, and in Raw264.7, $TNF-{\alpha}$ were significantly reduced by treated all concentrations of BGT. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.

  • (1) Relatively active polyphenol oxidase influence was seen at $0{\circ}C$, and the optimal pH level for the enzyme from the seeds of a small type sweet pepper Zairaisisi is 6.5. (2) The starting stage of the brown coloration with low-temperature injuries showed a strong activity of polyphenol oxidase, and the activity drops to 0 as the entire seed became brownish. (3) The browning effect with enzyme solution of polyphenol extracts suggested that the brown coloration continues in vitro even if polyphenol oxidase activity is nil. (4) Although cytochrome oxidase activity dropped when an abnormality occurrs in electron pathways of respiration at the starting stage of the browning with low temperature injuries, there was no marked influence of it on the total respiration, indicating the fact that polyphenol oxidase can take place of terminal oxidase in the compensatory respiration process.

Inhibitory Effects of Several Korean Traditional Herbs on Anaphylactic Reaction and Mast Cell Activation (아나필락시스 반응과 비만세포 활성화에 대한 한국전통약제의 억제효과)

  • Korean traditional herbs, especially Anemarrhena asphodeloides (A. asphodeloides), Salvia miltiorrhiza (S. miltiorrhiza), and Terminalia chebula (T. chebula) have been known to have the immunomodulatory, anti-tumor, and anti-inflammatory effects. However, direct cellular mechanism underlying the mast cell-mediated anaphylaxis-like reaction has poorly been understood. In the present study, the effects of the methanol extracts of A. asphodeloides (MEAA), S. miltiorrhiza (MESM), and T. chebula (METC) on anaphylactic reaction were investigated. Using in vitro and in vivo experiments, the influences of MEAA, MESM, and METC on the compound 48/80-induced anaphylaxis-like reaction and mast cell activation, and IgEmediated PCA were examined. Results are below; 1) MEAA, MESM, and METC significantly inhibited systemic anaphylaxis-like reaction, ear swelling response, and IgE-mediated PCA. 2) the compound 48/80-induced mast cell degranulation, histamine release of rat peritoneal mast cells (RPMC) were significantly inhibited by the pretreatment with MEAA, MESM, and METC, and 3) the compound 48/80-induced calcium influx in RPMC was significantly inhibited by the pretreatment with MEAA, MESM, and METC. These results suggest that MEAA, MESM, and METC may be an activity to inhibit the compound 48/80-induced or anti-DNP IgE-mediated anaphylactic reactions by blocking histamine release and calcium uptake into RPMC. MEAA, MESM, and METC potentially may serve as an effective therapeutic tool for allergic diseases.
