Effects of Amomum xanthiodes on the Mast Cell-Mediated Allergic Reaction (비만세포 유래의 알레르기 반응에 대한 사인의 효과)

  • The discovery of drugs for the treatment of mast cell-mediated allergic disease is a very important subject in human health. The Amomum xanthiodes (Zingiberaceae) has been used for centuries as a traditional medicine in Korea and is known to have an anti-inflammatory effect. However, its specific mechanism of action is still unknown. In this report, we investigated the effect of hot water extract from Amomum xanthiodes (EAX) on the mast cell-mediated allergic reaction and studied its possible mechanisms of action. EAX inhibited compound 48/80-induced systemic anaphylaxis and serum his­tamine release in mice. EAX decreased the passive cutaneous anaphylaxis reaction activated by anti-dinitrophenyl (DNP) IgE antibody. EAX dose-dependently reduced histamine release from rat peritoneal mast cells activated by compound 48/80 or anti-DNP IgE. EAX increased cAMP and decreased compound 48/80-induced intracellular $Ca^{2+}$ levels. Our findings provide evidence that EAX inhibits mast cell-derived allergic reactions, and also demonstrate the involvement of cAMP and intracellular $Ca^{2+}$ in these effects.

  • We studied the inhibitory effect of Green Life Enzyme (GLE) on compound 48/80-induced anaphylactic response in a murine model. GLE inhibited compound 48/80-induced systemic anaphylactic shock at the dose of 10 g/kg by 87.5%. When GLE was given as pre-treatment at concentrations ranging from 0.01 to 1.0 g/kg, it inhibited passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. In addition, GLE (0.1 mg/ml) inhibited anti-DNP IgE-induced tumor necrosis $factor-{\alpha}$ production from mast cells by 69% compared to saline value. These results indicate that GLE may possess anti-anaphylactic and anti-inflammatory activity.

  • In a bioassay-guided search for anti-allergic compounds from higher plants of Korea, poly-methoxyflavones, 3',4',5,6,7,8-hexamethoxyflavone (I), 5-hydroxy-3',4',6,7,8-pentamethoxyflavone (II) and 3',4',5,7,8-pentamethoxyflavone (III) have been isolated from the immature peels of Citrus unshiu. Structures of these compounds were elucidated on the basis of spectroscopic techniques. Compounds I and II inhibited dose-dependent histamine release from the rat peritoneal mast cells activated by compound 48/80 or anti-DNP IgE.

Effect of modified-Okbyungpoongsan on mast cell-mediated allergic responses in RBL-2H3 mast cells (가미옥병풍산(加味玉屛風散)의 비만세포에서의 알레르기 반응에 대한 효과 연구)

  • Objectives : In this study, we investigated the effect of modified-Okbyungpoongsan (mOP) on mast cell-mediated allergic response in basophilic leukemia cell line, RBL-2H3 mast cells. Methods : Cells were stimulated with anti-DNP-IgE after the treatment of DNP-HSA (AI/D), and then incubated with different concentrations of mOP (0.1, 0.2, 0.5 and 1 mg/$m{\ell}$) in RBL-2H3 cells. Cell toxicity was determined by WST-1 assay. The degranulation of mast cells was observed by microscope with toluidine blue staining and also the levels of beta-hexosaminidase, histamine and TNF-alpha were measured in culture supernatants by enzyme-linked immunosorbant assay. Results : mOP inhibited anti-DNP-IgE-imduced degranulation of mast cells in RBL-2H3 cells. mOP also significantly decreased the levels of histamine and inflammatory cytokine, TNF-alpha in RBL-2H3 cells, but slightly decreased the level of beta-hexosaminidase. Conclusions : These results indicate that mOP, an oriental prescription could be inhibit the allergic response through suppressing the mast cell activation.

Inhibitory Effects of Allergic Reaction of Aqueous Extract of Salviae Radix Root by Anal Therapy (항장요법(肛腸療法)에 의한 단삼추출액(丹蔘抽出液)의 알레르기반응 억제효과(抑制效果))

  • Dansam, the root of Salvia miltiorrhiza BGE. (Labiatae), has a bitter and a slightly 'cold' property. and is nontoxic. It has been used for the treatment of diseases such as tumors, bruises, gynecologic diseases, menoxenia, anemia and so forth. As an oriental medicine pathway, anal therapy has many kinds of treatments, Retention enema is one the most useful, advantageous methods of anal therapy. This experiment was performed in order to study the effect of an aqueous extract of Salviae radix root(SRRAE) on Immediate type allergic reaction by Retention enema. The results were obtained as follows, 1. SRRAE inhibited compound 48/80-induced Immediate type allergic reaction 100% with the dose of 0.1g/kg by anal treatment. However, SRRAE showed no significant inhibitory effect on the same reaction by oral treatment. When mice were pretreated with SRRAE at a concentration ranging from 0.001 to 1.0g/kg, by intra-anal treatment, the serum histamine levels were reduced in a dose-dependent manner. 2. SRRAE also inhibited by 79.8% local cutaneous allergic reaction activated by anti-dinitrophenyl (DNP) IgE. 3. SRRAE close-dependently inhibited the histamine release from rat peritoneal mast cells(RPMC) by anti-DNP IgE., but SRRAE not inhibited compound 48/80-induced histamine release. 4. SRRAE dose-dependently inhibited tumor necrosis $factor-\;{\alpha}$ $(TNF-\;{\alpha}$) production from RPMC by anti-DNP IgE. Moreover, the level of cAMP in RPMC, when SRRAE was added, significantly increased compared with that of a normal control. According to the above results, anal therapy(Retention enema) of SRRAE may be beneficial in the treatment of systemic and local Immediate type allergic reactions.

Inhibitory Effect of Mast Cell-Mediated Anaphylactic Reactions and Tumor Necrosis $Factor-{\alpha}$ Production by Aqueous Extract of Sinomenium acutum stem (방기 전탕액의 비만세포 매개성 아나필락시반응 및 종양괴사인자알파 생성 억제효과)

  • Objectives: The root and stem of Sinomenium acutum has been used for treatment of arthritis and neuralgia in oriental medicine. To find new substances of the anti-anaphylactic drugs, we studied Sinomenium acutum. Methods: To investigate the effect of this plant, the effect on anaphylactic reaction, plasma histamine level, and tumor necrosis $factor-{\alpha}-(TNF-{\alpha})$ production were measured after the aqueous extract of Sinomenium acutum stem (SSAE) was administrated to mice and rats. Results: The SSAE (0.1 to 1000 mg/kg) dose-dependently inhibited systemic anaphylactic reaction induced by compound 48/80 in mice. Especially, SSAE reduced compound 48/80-induced anaphylactic reaction with 50% at the dose of 1000 mg/kg. SSAE (100 to 1000 mg/kg) also significantly inhibited local anaphylactic reaction activated by anti-dinitrophenyl (DNP) IgE. When mice were pretreated with SSAE at a concentration ranging from 0.1 to 1000 mg/kg, the plasma histamine levels were reduced in a dose-dependent manner. SSAE (1 to 1000 g/ml) dose-dependently inhibited histamine release from the rat peritoneal mast cells (RPMCs) activated by compound 48/80 or anti-DNP IgE. The level of cAMP in RPMCs, when SSAE was added, increased compared with that of a normal control. In addition, SSAE (0.1 g/ml) had a significant inhibitory effect on anti-DNP IgE-induced $TNF-{\alpha}$ production. Conclusions: These results indicate that SSAE inhibits mast cell-mediated anaphylactic reactions and $TNF-{\alpha}$ production from mast cells.

Fermentation-Mediated Enhancement of Ginseng's Anti-Allergic Activity against IgE-Mediated Passive Cutaneous Anaphylaxis In Vivo and In Vitro

  • Ginseng (the root of Panax ginseng Meyer) fermented by Lactobacillus plantarum has been found to attenuate allergic responses in in vitro and in vivo experimental models. Ginseng has been reported to also possess various biological functions including anti-inflammatory activity. The present study was aimed at comparing the anti-allergic effect of ginseng and fermented ginseng extracts on IgE-mediated passive cutaneous anaphylaxis in vitro in a murine cell line and in vivo in mice. Fermented ginseng extract (FPG) showed higher inhibitory effect against in vitro and in vivo allergic responses when compared with ginseng extract (PG). The secretion of ${\beta}$-hexosaminidase and interleukin (IL)-4 from the IgE-DNP-stimulated RBH-2H3 mast cells were significantly (p < 0.05) inhibited by FPG treatment, and this effect was concentration-dependent. Further, MKK4 activation and subsequent JNK phosphorylation were attenuated by FPG treatment. The inhibitory effect of FPG on the in vitro allergic response was verified in vivo against IgE-DNP-induced passive cutaneous anaphylaxis in a mouse model. These data indicated that the fermentation of ginseng with L. plantarum enhanced its anti-allergic effects both in vitro and in vivo. We predict that compositional changes in the ginsenosides caused by the fermentation may contribute to the change in the anti-allergic effects of ginseng. The results of our study highlight the potential of the use of FPG as a potential anti-allergic agent.

Suppressing Effects of Ethanol Extract from Dioscoreae Rhizoma on atope Dermatitis (부채마 에탄올 추출물의 아토피 피부염 억제 효과)

  • This study was conducted to investigate the effect of 70% ethanol extract (DR) on the mast cell-mediated allergic contact dermatitis induced by dinitrofluorobenzene in BALB / c mice, which affects the cell activity by antigen in RBL-2H3 mast cells Respectively. The ethanol extracts of RBL-2H3 cells activated by DNP-HSA and anti-DNP IgE antibodies inhibited the secretion of ${\beta}$-hexosaminidase, histamine, and IL-4 and $TNF-{\alpha}$ Production was suppressed. In the DNFB-induced contact allergic dermatitis animal model, treatment with ethanol extract reduced ear swelling and inhibited serum histamine and IL-4 secretion, and DR treatment effectively prevented mast cell infiltration in dermatitis-induced areas. As a result, the ethanol extract may be used as a therapeutic agent for mast cell-mediated allergic diseases such as atopic dermatitis.

Inhibitory Effect of Saururus chinensis (Lour.) Baill Extracts on Allergy in Mouse Models (Mouse 모델 알레르기 반응에서 삼백초(三白草)가 미치는 영향)

We conformed compound 48/80-induced mesenteric mast cell degranulation, active systemic anaphylatic shock and histamine release. Also observed acetic acid-induced vascular permeability and anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. SCB inhibited mesenteric mast cell degranulation and active systemic anaphylatic shock induced by compound 48/80 dose-dependently. When SCB was pretreated by intra-peritoneal injection, the plasma histamine levels were reduced. SCB also significantly inhibited acetic acid-induced vascular permeability and anti-DNP IgE-mediated passive cutaneous anaphylaxis. In addition, SCB reduced IL-10 mRNA expression of the lung on ovalbumin-induced allergy. These results indicate that SCB inhibits allergy.
