  • Jowiseungcheongtang(JST) has been in Sasang constitution medicine for many years as a therapeutic agent for cerebral disease. But the effect of Jowiseungcheongtang(JST) on neurotoxicity is not known. The purpose of this study was to determine the effects of Jowiseungcheongtang(JST) on the hippocampal cell injured by Xanthine Oxidase/Hypoxanthine. The results were as follows: 1. XO/HX decreased the survival rate of the cultured hippocampal cells on NR assay and MTT assay. 2. JST water extract have efficacy of decreasing a amount of lipid peroxidation increased by XO/HX in cultured hippocampal cells. 3. JST water extract have efficacy of increasing DNA synthesis decreased by XO/HX in cultured hippocampal cells. From the above results, It is concluded that JST has marked efficacy in preventing cultured hippocampal cells from the damages by XO/HX.

Invitro Anticancer Effect of Chinese Cabbage Kimchi Fractions (배추김치 분획물의 in vitro 항암효과)

  • In vitro anticancer effect of Chinese cabbage kimchi fractions was investigated by using human cancer cells, AGS human gastric adenocarcinoma cells and HT 29 human colon adenocarcinoma cells. The Chinese cabbage kimchi(fermented for 4 days at 15oC) was fractionated into 7 groups, methanol extract, hexane fraction(fr.), methanol soluble fr., dichloromethane fr., ethylacetate fr., butanol fr. and aqueous fr.. Chinese cabbage kimchi fractions inhibited the growth of AGS and HT 29 cancer cells as dose dependent. In particular, the dichloromethane fr. showed the highest inhibitory effect among other fractions. When the dichloromethane fr.(0.2mg/ml) was treated, the number of AGS and HT 29 survival cancer cells reduced to 12$\times$104/ml and 11$\times$104/ml compared to 166$\times$104/ml and 50$\times$104/ml of the controls, respectively. Chinese cabbage kimchi fractions also inhibited the DNA synthesis of the cancer cells. They inhibited the DNA synthesis of AGS human gastric adenocarcinoma cells more efficiently than that of HT 29 human colon adenocarcinoma cells. These results indicate that Chinese cabbage kimchi fractions show in vitro anticancer activity and the dichloromethane fr. among them reveals the highest effect.

  • Telomerase plays a pivotal role in the pathology of aging and cancer by maintaining genome integrity, controlling cell proliferation, and regulating tissue homeostasis. Telomerase is essentially composed of an RNA component, Telomerase RNA or TERC, which serves as a template for telomeric DNA synthesis, and a catalytic subunit, telomerase reverse transcriptase (TERT). The canonical function of TERT is the synthesis of telomeric DNA repeats, and the maintenance of telomere length. However, accumulating evidence indicates that TERT may also have some fundamental functions that are independent of its enzymatic activity. Among these telomere-independent activities of hTERT, the role of hTERT in gene transcription has been investigated in detail. Transcriptional regulation is a fundamental process in biological systems. Several studies have shown a direct involvement of hTERT in gene transcription. This mini-review will focus on the role of hTERT in gene transcription regulation, and discuss its possible mechanisms.

Effects of Samul-Tang Extract on Vascular Endothelial Cells from Hydrogen Peroxide-induced Injury (사물탕(四物湯)이 혈관내피세포(血管內皮細胞)에 미치는 영향(影響))

  • This study is designed to investigate the effects of Samul-Tang extract on the response of lactic dehydrogenase(LDH) release, cellular activity, lipid peroxidation, DNA synthesis and the changes of total protein of bovine pulmonary artery endothelial cells(PAEC) from hydrogen peroxide$(H_2O_2)$-induced injury. The results are as follows : 1. Samul-Tang significantly decreased $H_2O_2$-induced release of LDH from injured bovine PAEC. 2. Samul-Tang significantly repressed $H_2O_2$-induced cellular activity from injured bovine PAEC. 3. Samul-Tang significantly repressed $H_2O_2$-induced lipid peroxidation from injured bovine PAEC. 4. Samul-Tang significantly stimulated DNA synthesis in bovine PAEC. 5. Samul-Tang significantly repressed $H_2O_2$-induced changes of total protein volume from injured bovine PAEC. Above results suggest that Samul-Tang can protect bovine PAEC from $H_2O_2$-induced injury. These results can be effectively applied to the prevention and cure of cardiovascular and cerebrovascular diseases.

Differential gene expression pattern in brains of acrylamide-administered mice

  • The present study was performed to evaluate the relationship between the neurotoxicity of acrylamide and the differential gene expression pattern in mice. Both locomotor test and rota-rod test showed that the group treated with higher than 30 mg/kg/day of acrylamide caused impaired motor activity in mice. Based on cDNA microarray analysis of mouse brain, myelin basic protein gene, kinesin family member 5B gene, and fibroblast growth factor (FGF) 1 and its receptor genes were down-regulated by acrylamide. The genes are known to be essential for neurofilament synthesis, axonal transport, and neuroprotection, respectively. Interestingly, both FGF 1 and its receptor genes were down-regulated. Genes involved in nucleic acid binding such as AU RNA binding protein/enoyl-coA hydratase, translation initiation factor (TIF) 2 alpha kinase 4, activating transcription factor 2, and U2AF 1 related sequence 1 genes were down-regulated. More interesting finding was that genes of both catalytic and regulatory subunit of protein phosphatases which are important for signal transduction pathways were down-regulated. Here, we propose that acrylamide induces neurotoxicity by regulation of genes associated with neurofilament synthesis, axonal transport, neuro-protection, and signal transduction pathways.

Study on the Anti-Cancer Effect of Acanthopanax Sessiliflorus (오가피(五加皮)의 항암효과에 관한 연구)

  • Objectives: Hepatoma is a very serious disease in Korea and vvorldwiclc. Hepatitis B vims (HBV) has proved the most significant cause of hepatoma. We canied out this study to investigate the effect of Acanthopanax sessilifloms on inhibiting cell proliferation and DNA synthesis in HepG2.2.15 cell line and on inhibiting phosphorilation of oncogene (MAP kinase) in NIT/3T3-HBx ceIl. Methods: To investigate the anti-cancer effect of Acanthopanax sessiliflorus, we did the CellTiter 96 Aqueous Non-radioactive Cell Proliferation assay (Promega); MTS/PMS assay, [$^3H$]-thymicline incorporation assay, and we measured the gene expression through westem blotting. Results: Acanthopanax sessiliflorus showed an inhibiting effect on the increase of HepG2.2.15 in the NTS/PMS assay. It also showed an inhibiting effect on DNA synthesis of HepG2.2.15 in the [$^3H$]-thymidine incorporation assay. Acanthopanax sessiliflorus showed an inhibiting effect of phosphorilation of MAP kinase in HBV - X genes. too. Conclusions: The results suggested that this herb had an anti cancer effect. We may discover an effective anti-cancer herb medicine through further studies on this herb medicine.

New anti-wrinkle cosmetics

  • In the aged skin especially in the face and eyelid, deep and slight wrinkles are one of the remarkable phenomena of aging and the cause of wrinkle is various. Among the cause of wrinkles an oxidative stress plays an important roles in wrinkle formation process. It caused the lipid peroxidation of cell membrane, the increase of the MMPs(MatrixMetalloProteinase) gene expression and cellular DNA damage. These ROS induced materials may cause the degradation of collagen matrix system in the dermis and cause the formation of skin wrinkle. So, it is very important for protecting skin wrinkle formation to regulate ROS activity. In this study, we developed one active ingredient having multi functional activities such as activation of collagen synthesis, inhibition of MMPs activity, lipid peroxidation and free radical scavenging activity and inhibition of free radical induced DNA damage in vitro. Pericarpium castaneae extracts showed collagen synthesis increase in Normal Human Fibroblast and the inhibition of elastase activity (IC$\_$50/ of Elastase: 43.9$\mu\textrm{g}$/㎖). It showed also anti-oxidative activity (IC$\_$50/ : 48$\mu\textrm{g}$/㎖) and free radical scavenging activity(IC$\_$50/: 7.6$\mu\textrm{g}$/㎖). Conclusively, Pericarpium castaneae extracts may be used as an ingredient for new anti-wrinkle cosmetics.

Influence of Panax Ginseng on DNA Synthesis of Submandibular Gland in Mice Receiving ACTH(II) (고려인삼이 ACTH를 받은 마우스의 악하선 조직 DNA 합성능에 미치는 영향( II ))

  • It was planned to investigate, in mice that received ACTH, the influence of Panax Ginseng upon DNA synthesis of submandibular gland by observing incorporation of $[^3H]$ thymidine into the tissue cells. Thirty male mice $(body\;weight:\;18{\sim}20\;g)$ were divided equally into the ginseng-ACTIH and the saline-ACTH groups. Each animal of the ginseng-ACTH and the saline-ACTH groups received every day (subcutaneously) 0.05 m1/10 g body weight of ginseng extract(4 mg of ginseng alcohol extract in 1 ml of saline) and the same amount of saline, respectively, for 5 days. On the 5th experimental day, all animals received 0.01 unit of ACTH intraperitoneally one hour after the last medication, and $1\;{\mu}Ci/g$ body weight of $[^3H]$ thymidine after one more hour. Five animals, at a time, of each group were sacrificed 1, 10, and 24 hr after $[^3H]$ thymidine administration, and the radioactivity of cells in their mandibular gland was measured autoradiographically in terms of the % number of radioactive cells in 1,000 cell counts (Radioactive Index, R.1.). Results obtained were as follows: 1. The radioactive indices obtained from submandibular gland of the saline-ACTH group 1, 10 and 24 hr after $[^3H]$ thymidine administration were $15.2{\pm}0.32,\;20.1{\pm}0.30,\;and\;24.5{\pm}0.52(mean{\pm}S.D.)$ in the mucous cells, $13.0{\pm}0.22,\;10.2{\pm}0.05,\;and\;7.5{\pm}0.42$ in the serous cells. and $10.5{\pm}0.40,\;13.6{\pm}0.32,\;and\;15.9{\pm}0.42$ in the duct cells, while the $mean{\pm}S.D.$ of the values obtained from the 3 cell types 1, 10 and 24 hr after $[^3H]$ thymidine were $10.9{\pm}0.28,\;12.4{\pm}0.31,\;and\;10.0{\pm}0.39.$ Thus the radioactive indices obtained from the ginseng-ACTH group were generally lower than those obtained from the saline-ACTH group. It is inferred from the above results that the ginseng tends to promote the suppressive action of ACTH upon DNA synthesis of cells in the mandibular gland.

The combination effects of PDGF and IGF-I on the proliferation and cellular activity of periodontal ligament cells (PDGF와 IGF-I 병용 사용시 치주인대세포의 증식과 세포활성에 미치는 영향에 관한 연구)

  • Current acceptable methods for promoting periodontal regeneration are based on removal of diseased soft tissue. root treatment, guided tissue regeneration, introduction of new graft materials and biological mediators. Insulin-like growth factor-I(IGF-I) and Platelet-derived growth factor-BB(PDGF-BB), the members of the polypeptuyde growth factor family have been reported as the biological mediators which regulate a variety cellular matrix biologic activities of wound healing process including the cell proliferation, migration and extracellular matrix synthesis.The purposes of this study is to evaluate the combination effects of IGF-I and PDGF-BB on the cellular activity of the periodontal ligament cells to act as a regeneration promoting agent of periodontal tissue. Human periodontal ligament cells were prepared from the first premolar tooth extracted for the orthodontic treatment and were cultured in DMEM containing 10% FBS at the $37^{\circ}C$, 5% CO2 incubator. Author measured the DNA synthetic activity, and total protein, collagen and noncollagenous protein synthetic activities according to the concentration of 10,100ng/ml IGF-I and1,10 ng/ml PDGF-BB in combination. The results were as follows: Significantly increased in the 1 ng/ml PDGF-BB alone compared to the 10 ng/ml PDGF-BB alone(P<0.01) and in the 1 ng/ml PDGF-BB and 10, 100ng/ml IGF-I in combination compared to the 1 ng/ml PDGF-BB alone(P<0.05, P<0.0l). The synthetic activity of the total protein and collagen is significantly increased like to the synthetic activity of the DNA(P<0.05). The synthetic activity of the noncollagenous protein is increased according to the concentration of IGF_I, but not statistically statistically significant(P>0.05). The percent of the collagen is significantly in the 1ng/ml PDGF-BB and 10ng/ml IGF-I in combination compared to the 1ng/ml PDGF-BB alone(P<0.05) and in the 10ng/ml IGF-I in combination compared to the 10ng/ml PDGF-BB alone(P<0.05). The synthetic activity of the DNA is In conclusions, the percent study shows that PDGF-BB and IGF-I in combination have a potentiality to enhance the DNA synthesis and the total protein and collagen synthesis of The periodontal ligament cells, especially it is more significant in the low concentration of PDGF-BB compared to the high one. Thus, the PDGF-BB and IGF-I in combination may have important roles in promotion of periodontal litgment healing, and consequently, may useful for clinical application in periodontal regenerative procedures.

Molecular Cloning of a cDNA Encoding Putative Calreticulin from the Silkworm, Bombyx mori

  • We describe here the cloning of a cDNA encoding putative calreticulin (CRT) from the silkworm, Bombyx mori. The CRT cDNA comprised of 1,194 bp encoding 398 amino acid residues. B. mori. CRT has a HDEL sequence at the end of the C-domain. The B. morl, CRT showed 88% protein sequence identity to the G. mellonella CRT, 71 % to A. aegypti CRT, and 63% to H. sapiens CRT, Phylogenetic analysis revealed that the deduced amino acid sequences of the B. mori CRT formed a highly inclusive subgroup with other insect CRTs. Northern blot analysis exhibited an expression of the B. mori CRT gene in the fat body, evidencing the fat body as a major site for CRT synthesis.
