Studies on the Chemical Synthesis of Aflatoxin-DNA Adduct (Aflatoxin-DNA Adduct의 화학합성에 관한 연구)

  • Aflatoxins are highly carcinogenic agents consistantly found as contaminants in human food supplies in many areas of the world and epidemiologically linked to incidence of human liver cancer. To examine the carcinogenic action of aflatoxin $B_1$, aflatoxin $B_1-DNA$ adducts were chemically synhtesized with the reaction of 20 mg calf thymus DNA, and 8 mg standard aflatoxin $B_1$. Since DNA molecule was too large for analysis, it was fragmented by acid hydrolysis and heat. The fragmented aflatoxin $B_1-DNA$ adducts were selectively concentrated by immunoaffinity column procedure and confirmed by HPLC method. The main component was aflatoxin $B_1-guanine$ adduct, which was quantatively measured as 5.2 mg aflatoxin $B_1$.

Effect of Housing Systems of Cage and Floor on the Production Performance and Stress Response in Layer (계사 사육 형태가 산란계의 생산성과 스트레스 반응에 미치는 영향)

  • This study was conducted to investigate the effects of housing systems on the productivity and physiological response as stress indicators in White Leghorn chickens. The chickens subjected to the conventional cages had a significantly lower viability, hen-housed egg production, egg weight and body weight compared with those to the floor pens. However, the hens housed in the conventional cages had a shorter day of the first egg and a greater egg quality compared with those housed in the floor pens. In addition, this study was also investigated to identify biological markers for assessing the physiological response of chickens under stress conditions. As biological markers, the amount of telomeric DNA was analyzed by quantitative fluorescent in situ hybridization on the nuclei of cells. The DNA damage rate of lymphocytes was also quantified by the comet assay. The amount of telomeric DNA of the lymphocytes, kidney and spleen was significantly higher in the chickens under floor pens than those under conventional cages. The DNA damage also increased in chickens raised under conventional cages, as compared to the chickens under floor pens. As results, we conclude that the chickens housed in conventional cages have a greater stressful status than those housed in floor pens.

Effects of Artemisia Capillaris Thunberg on Apoptosis in HeLa Cells (사철쑥의 HeLa 세포고사 효과)

  • This study analyzes the apoptosis of HeLa cells to see if we can use the Artemisia capillaris Thunberg for the prevention of chronic degenerative diseases. We used the HeLa cells to see what effects the A. capillaris Thunberg had on apoptosis of the cancer cells. We checked the cell activity, cell morphological change, DNA fragmentation, and DNA content after administering 0, 100, 500, 1000, and $2000{\mu}g/ml$ methanol, ethyl acetate, n-butanol extract of the A. capillaris Thunberg. As for the cell viability, the increase of concentration of methanol and ethyl acetate decreased the survival rate of the cell, but the phenomenon was much weakened in n-butanol extract and was not observed in aqueous extract. The higher the density of the methanol, ethyl acetate, n-butanol and aqueous extract was, the lower the survival rate of the HeLa cell was. These extracts obstructed the cell cohesion and caused the blebbing of he cell membrane and fragmentation of the nucleus, both of which are symptoms of apoptosis. Laddering-pattern DNA fragmentation was observed in the groups that were treated with the $1000{\mu}g/ml$ and $2000{\mu}g/ml$ of methanol extract. The DNA content of the cells apoptosis measured by fluorescent-activated cell sorter (FACS) increased as the density of the methanol, ethyl acetate and butanol extract increased. The result of the study shows that A. capillaris Thunberg fosters the apoptosis of HeLa cells, which suggests that the A. capillaris Thunberg has a great potential value as food additives, medicinal supplements for patients with chronic diseases, and preventive measures against cancer.

Antioxidant Activity and DNA Damage Protective Effect of a Robinia pseudoacacia L. Flower Extract (아까시 꽃 추출물의 항산화 활성 및 DNA 손상 억제 효과)

  • The antioxidant properties of Robinia pseudoacacia L. water and 70% ethanol extracts were evaluated by determining total phenolic content (TPC), DPPH radical scavenging activity (RSA), and reducing power (RP). The water extract showed higher TPC (9.07 mg/g gallic acid equivalents) and RP than those of ethanol extract, whereas ethanol extract had greater DPPH RSA. The R. pseudoacacia L. extracts also showed antigenotoxic effects for 200 ${\mu}M$ $H_2O_2$-induced DNA damage in human leukocytes. The 200 ${\mu}M$ $H_2O_2$-induced DNA damage decreased following treatment with the water extract. Reductions in DNA damage with 50 ${\mu}g/ml$ of the water and ethanol extracts were 46.5 and 32.4%, respectively.

Amount of Telomeric DNA on Pig Lymphocytes by Quantitative Fluorescence in situ Hybridization (양적형광접합보인법(Q-FISH)에 의한 돼지 백혈구 세포의 텔로미어 함량 분석)

  • The amount of telomeric DNA was quantified across different breeds(Landrace, Duroc, Yorkshire and Berksire), at different ages(90 days old and 180 days old) and among sexes(male and female) in pigs raised at the Performance Testing Station of Korea Swine Association, Jinkyo, Korea. The telomeric DNA amount was quantified by Quantitative Fluorescence In Situ Hybridization(Q-FISH) using a porcine telomeric DNA probe on interphase nuclei of lymphocytes. Analysis revealed that the amount of telomeric DNA on the pig lymphocytes was found to decrease with age. The quantity of telomeres significantly differed among breeds at 90 days of age. The colored breeds such as Berkshire and Duroc had higher amount of telomeric DNA than the Yorkshire and Landrace breed. In addition, the amount of telomeric DNA in male lymphocytes was significantly higher than that of females. In the correlation coefficients between the telomere quantity and their productive traits; average daily gain, loin percent and index value were positively correlated, whereas body length, feed efficiency and back fat thickness correlated negatively. However, the correlation coefficients were very low and not significant. Therefore, this study suggests that the amount of telomeres on lymphocytes can be considered as a physiological marker but not as a productive marker in pig.

Optimum Condition of Polymerase Chain Reaction Techniques for Randomly Amplified Polymorphic DNA of Strawberry (딸기의 RAPD를 위한 PCR의 최적조건)

  • This study was performed to select marker which can identify genetic variation between mother plant and in vitro cultured plantlets of strawberry by PCR using random primer. When 'Yeobong' DNA extracted was treated with proteinase-K and RNase-H, clear DNA bands were shown. The optimal condition for RAPD in strawberry was to use 50ng of template DNA, 10pmol of primer,37oC of annealing temperature, and 45 cycles of PCR. After establishing above PCR optimal condition, RAPD pattern was investigated by using UBC primers. PCR was performed, and 46 of 90 primers produced PCR product showing 158 total bands. GC content was compared between the primers forming bands and no bands. The GC content showing bands was average 67.4%, whereas primers showing no bands 58%.

Comparison of Physiological Characteristics, Stomata and DNA Content between Seedling and 5-year-old Somatic Plant (Somatic Embryo Derived-plant) in Liriodendron tulipifera (백합나무 5년생 실생묘 및 체세포묘 (체세포배 유래 식물체) 간의 생리적 요인, 기공 및 DNA 함량 비교)

  • Field performance of somatic plants (somatic embryo derived-plants) of yellow-poplar (Liriodendron tulipifera) produced from somatic embryogenesis was compared with that of seedlings at age 5. In comparison of photosynthetic rate (seedling, $10.67{\mu}mol$ $CO_2m^{-2}s^{-1}$; somatic plant, $9.04{\mu}mol$ $CO_2m^{-2}s^{-1}$), stomatal conductance rate (seedling, 0.2 $H_2Om^{-2}s^{-1}$; somatic plant, 0.166 $H_2Om^{-2}s^{-1}$) and respiration rate (seedling, 1.71 mmol $H_2Om^{-2}s^{-1}$; somatic plant, 1.513 mmol $H_2Om^{-2}s^{-1}$), no significant differences were found between plants. The seedlings were a little higher in comparison of stomatal density (seedling, $23.33/mm^2$; somatic plant, $22.43/mm^2$), length (seedling, $25.83{\mu}m$; somatic plant, $23.46{\mu}m$) and width (seedling, $15.87{\mu}m$; somatic plant, $15.3{\mu}m$). In comparison of DNA content of the leaves using flow cytometry, no differences in ploidy level were found between the seedlings and somatic plants.

Cytogenetic Analysis of Three Hemibarbus Species (Cypriniformes) from Korea (한국산 누치속 어류 3종의 세포유전학적 연구)

  • Cytogenetic characteristics of three Hemibarbus species (H. labeo, H. longirostris and H. mylodon) were analyzed based on erythrocyte measurement, flow cytometric estimation of cellular DNA content, and karyological analysis. Average nuclear volumes for H. labeo, H. longirostris and H. mylodon were 22.5, 21.7 and $26.0\;{\mu}m^3$, respectively. The estimated genome sizes of those three species were not significantly different from one another, being recorded as 2.51, 2.33 and 2.35 pg/cell for H. labeo, H. longirostris and H. mylodon, respectively. Modal chromosome numbers of the three species were the same as 2n = 50. However, their karyotypes and fundamental numbers (FN) were different among species; 16M+16SM+18T/A (FN = 82) for H. labeo, 18M+16SM+16T/A (FN = 84) for H. longirostris and 18M+24SM+8T/A (FN = 92) for H. mylodon.

  • Bagrid catfish hybrid between Korean bullhead (Pseudobagrus fulvidraco) and Ussurian bullhead (Leiocassis ussuriensis) was produced by artificial fertilization. Modal chromosome numbers of Korean bullhead, Ussurian bullhead and their hybrid were the same as 2n=52. The karyotypes were 22 metacentric (M), 22 submetacentacentric (SM), and 8 acrocentric (A) chromosomes for Korean bullhead, 22M+14SM+16A for Ussurian bullhead and 22M+18SM+12A for their hybrid. The hybrid bullhead karyotype consisted of each haploid set of parental species chromosomes. Erythrocytic sizes and DNA contents of hybrids were intermediate between two parental species.

  • The DNA damage mechanism by fish oil peroxidation was investigated through the model system of a DNA-mackerel lipid at $37^{\circ}C$. Mackerel lipid peroxidation products induced a great DNA damage with the increment of its concentration, and such DNA damage in all systems examined occurred below $100millieq{\cdot}/kg$ in POV (peroxide value) Singlet oxygen $(^1O_2)$ and superoxide anion${\cdot}O_2^-$ greatly participated in the DNA damage during peroxidation of mackerel lipid, while hydrogen peroxide$(H_2O_2)$ and hydroxyl radical $({\cdot}OH)$ did little show the DNA damage. From the results of the addition of several active oxygen scavengers to the DNA-lipid systems, singlet oxygen ana superoxide anion greatly affected to the increase of POV ana to the DNA damage by mackerel lipid peroxidation, respectively. It indicates that there was a close relationship between the effects of active oxygens in the mackerel lipid peroxidation and its DNA damage mechanism.
