Sample Preparation and Stability of Human Serum and Urine Based on HPLC-DAD for Metabonomics Studies

  • Many literatures focus on the biological relevance and the identification of biomarkers for disease activity assessment while less attention has been paid to the development of standard procedures for sample preparation and storage based on liquid chromatography technique. The influencing factors including protein precipitation, storage temperature, storage time, and reconstitution by ultra pure water were analyzed employing HPLC-DAD. The effects were investigated from five participants over three months by principal components analysis (PCA) and the values of percent changes (PC). The samples with protein precipitation might slow the rate of bacterial enzymatic conversion. After protein precipitation, the average PC of urine samples ($0.136{\pm}0.013$, n = 5) is relatively less than that of the serum samples ($0.173{\pm}0.026$, n = 5) for three months. Minimal effects on metabolic profiles of serum and urine (PC < 0.15) are reasonable for metabolomic studies after protein precipitation and storage at $-20^{\circ}C$ for two months.

Analysis of Constituents in Socheongryong-tangs Fermented by Lactic acid bacteria (유산균 발효에 의한 소청룡탕의 발효 전 후 성분 변화 연구)

  • Objective : The purpose of this study was to investigate the changes in the contents of constituents in Socheongryong-tang (CY) and its fermentations (FCY) with 10 species of lactic acid bacteria. Methods : Ten strains of lactic acid bacteria, Lactobacillus casei 127, L. acidophilus 128, L. casei 129, L. plantarum 144, L. amylophilus 161, L. curvatus 166, L. delbruekil subsp. lactis 442, L. casei 693, B. breve 744, and B. thermophilum 748, were used for the fermentation of Socheongryong-tang. The increased and decreased constituents were identified using HPLC/DAD and various liquid chromatographic techniques, and the structure was elucidated using NMR and MS. These compounds were quantitatively analyzed using an HPLC/DAD system. Results : The increased constituents were identified to be liquiritigenin (1) and cinnamyl alcohol (2), and the decreased constituent was determined to be liquiritin (3). Liquiritigenin (1) and cinnamyl alcohol (2) were increased in all of the FCYs, while liquiritin (3) was decreased. The fermentation of the ten lactic acid bacteria demonstrated that the decomposable rate of these three compounds in FCYs were different. Socheongryong-tang fermented by L. plantarum 144 and L. amylophilus 161 showed the most remarkable changes. Conclusions : CY could be increased antibacterial, neuroprotective, or antiinflammatory effect by fermentation with lactic acid bacteria, especially with L. plantarum and L. amylophilus, considering their known biological activities. In addition, it is expected that this study will help to establish quality control parameters for FCY.

  • The movement of the MR loaded on the train is confined to the bidirectional movement along the rail. Therefore, the AR connected to the MR can use the address information of the neighboring ARs and configure CoA in advance before performing L2 and L3 handoff. The MR can acquire new CoA immediately from the present AR after the movement detection procedure. The performance analysis shows that the proposed scheme can provide CoA to the MR about 1.8(s) at minimum and 4.98(s) at maximum faster than the Stateless scheme because the proposed scheme does not carry out any additional CoA and DAD procedure unlike the Stateless scheme.

Analysis of Anthocyanin Glycosides in Korean Mulberry Fruit Cultivars (한국산 오디 품종별 안토시아닌 배당체 성분 분석)

  • Mulberry fruit is a superior source of polyphenols, especially anthocyanin, and has a long history of use as an edible fruit and traditional medicine. The anthocyanin composition of mulberry fruit from 15 Korean cultivars was analyzed by ultra-performance liquid chromatography diode array detector with quadrupole time of flight/mass spectrometry (UPLC-DAD-QToF/MS) based on a cyanin internal standard. The four glycosides were identified by comparison with authentic standards and published reports. The major anthocyanin was cyanidin 3-O-glucoside (71.7%), followed by cyanidin 3-O-rutinoside (26.6%). The minor components (total of 1.7%) were pelargonidin 3-O-glucoside and pelargonidin 3-O-rutinoside. The total anthocyanin content (mg/100 g, dry weight) of mulberry fruit varied by cultivar and ranged from 471.5±4.0 (Su Hong) to 4,700.2±54.0 (Gwa Sang2). Among the 15 cultivars examined, Gwa Sang2 showed the highest level of cyanidin 3-O-glucoside (3,133.4±32.6), which was 9-fold higher than that of Su Hong (351.5±3.4). In conclusion, anthocyanin profiles, including pelargonidin 3-O-glucoside and pelargonidin 3-O-rutinoside, were reported for the first time from 15 Korean mulberry fruit cultivars. The results will contribute valuable information on pharmaceutical properties, breeding superior mulberry cultivars, and food industries.

Biotransformation of Valdecoxib by Microbial Cultures

  • Microbial biotransformations can be used to predict mammalian drug metabolism. The present investigation deals with microbial biotransformation of valdecoxib using microbial cultures. Thirty-nine bacterial, fungal, and yeast cultures were used to elucidate the biotransformation pathway of valdecoxib. A number of microorganisms metabolized valdecoxib to various levels to yield nine metabolites, which were identified by HPLC-DAD and LC-MS-MS analyses. HPLC analysis of biotransformed products indicated that a majority of the metabolites are more polar than the substrate valdecoxib. Basing on LC-MS-MS analysis, the major metabolite was identified as a hydroxymethyl metabolite of valdecoxib, whereas the remaining metabolites were produced by carboxylation, demethylation, ring hydroxylation, N-acetylation, or a combination of these reactions. The hydroxymethyl and carboxylic acid metabolites were known to be produced in metabolism by mammals. From the results, it can be concluded that microbial cultures, particularly fungi, can be used to predict mammalian drug metabolism.

Analysis of the Effect of Mordants on the Degradation of Alizarin in Silk Dyed with Natural Madder Dye

  • This research investigated the effect of mordants on the degradation of madder dye in silk when silk was treated by the H2O2/UV condition as a laboratory simulation of burial induced degradation. Alum, iron, and alum/iron composite mordanting methods were applied to silk before dyeing with madder dye. Dye extracted from silk was examined using HPLC-DAD-MS analysis. The abundance of the chromatogram peak at 8.88 min retention time was used as the concentration of alizarin pigment in silk. K/S values, CIE $L^{\ast}a^{\ast}b^{\ast}$ values; in addition, Munsell HVC values were obtained using a spectrocolorimeter. The findings indicated that alizarin degraded most severely in silk mordanted by alum/iron composite mordanting than alum mordanting or iron mordanting. Mordanting with alum alone provided a relatively lower dye fixation at the point of dyeing; however, it provided a better survival of alizarin after 12 hours of degradation treatment.

Isolation and Quantitative Analysis of Tricin from Ears of Phragmites communis (갈대이삭으로부터 Tricin의 분리 및 함량 분석)

  • The aim of this study was to investigate a validation method for determination of tricin in Phragmites communis ears. Tricin was isolated with chromatographic methods and used as the standard substances for quantitative analysis. The structural determination was characterized by comparing their NMR spectral data with values reported in the literature. For validation, the specificity, linearity, precision, accuracy, detection limits, and quantification limits of tricin was measured by HPLC. The results show that the specificity was satisfied with retention time and diode array detector (DAD) spectrum by analysis of tricin using HPLC. The limits of detection (LOD) for tricin was 0.84 mg/ml. Recovery of tricin was 98.80~108.22% with R.S.D values less than 2%. Intra-day and inter-day precisions of tricin in P. communis ears were 99.96~100.96% and 99.01~100.40%, respectively. Therefore, application of tricin was validated by an analytical method as major compound in P. communis ears.

Effect of Extraction Condition on the Content of EGCG and Caffeine of Green Tea: Comparison with the Inhibitory Activity on Pancreatic Lipase

  • Caffeine and epigallocatechin gallate (EGCG) are major constituents of green tea, the leaves of Camellia sinensis (Theaceae). Although EGCG is well known for diverse beneficial effect, caffeine is sometimes harmful with adverse effects. Therefore, the extraction efficiency was investigated using different extraction method such as extraction solvent, extraction time, extraction method, and repeated extraction. The content of EGCG and caffeine in green tea extract was quantitated by HPLC analysis. The extraction condition exerted difference on the extraction yield. The content of EGCG was also affected by different extraction condition. Especially, the extraction solvent greatly affected the content of EGCG in the extract. However, the content of caffeine was less affected compared to that of EGCG. The inhibitory effect of green tea extract on pancreatic lipase was almost similar regardless of extraction condition. Taken together, optimization of extraction condition will provide best efficacy for further development of green tea as anti-obesity therapeutics.

Analysis of the Components of Guibitang and Fermented Guibi-tang and their Ability to Inhibit Angiotensin-converting Enzyme

  • Guibi-tang is a traditional medicine used for the treatment of colds. We investigated the levels of several compounds in Guibi-tang before and after fermentation with Lactobacillus and tested their ability to inhibit angiotensin-converting enzyme. Six known compounds (decursin, decursinol angelate, nodakenin, liquiritin, formononetin, and 6-gingerol) and 2 unidentified compounds were detected in Guibi-tang (GB) and fermented Guibi-tang (FGB) by an established HPLC-DAD method. The levels of the 6 known compounds were decreased after fermentation. FGB showed more potent inhibition of angiotensin-converting enzyme activity than GB. In conclusion, fermentation with Lactobacillus affects the content of several compounds in GB and improves its angiotensin-converting enzyme inhibitory activity.

  • A new approach to texture classification for quantitative ultrasound liver diagnosis using run difference matrix was developed. The run difference matrix comprised the gray level difference along with a distances. From this run difference matrix, we defined several vectors and parameters such as DOD, DGD, DAD vector, SHP, SMO, SMG, LDE, LDEL etc. Each parameter values calculated in fatty, cirrhotic, normal and chronic hepatitic liver images were plotted in a plane and we found that RDM method was more sensitive to small structural changes than the conventional run length method and showed improved classification ability between the diseases.
