Metabolic Activation of Marijuana Constituents, Cannabinoids, in Relation to Their Toxicity for Human and Its Oxidation Mechanism

  • Many oxidative metabolites of tetrahydrocannabinols (THCs), active components of marijuana, were pharmacologically active, and 11-hydroxy-THCs, 11-oxo-${\Delta}^8$-THC, 7-oxo-${\Delta}^8$-THC, 8$\beta$, 9$\beta$-epoxyhexahydrocannabinol (EHHC), 9$\alpha$, l0$\alpha$-EHHC and 3'-hydroxy-${\Delta}^9$-THC were more active than THC in pharmacological effects such as catalepsy, hypothermia and barbiturate synergism in mice. Cannabidiol (CBD), another major component, was biotransfomred to two novel metabolites, 6-hydroxymethyl-${\Delta}^9$-THC and 3-pentyl-6, 7, 7a, 8, 9, lla-hexahydro-I, 7-dihydroxy-7, 1O-dimethyldibenzo[b, d]oxepin (PHDO) through 8R, 9-epoxy-CBD and 85, 9-epoxy-CBD, respectively. Both metabolites exhibited some pharmacological effects comparable to d9 - THe. Cannabinol (CBN), the other major component, was mainly metabolized to ll-hydroxy-CBN by hepatic microsomes of animals including humans. The pharmacological effects of the metabolite were higher than those of CBN demonstrating that II-hydroxylation of CBN is metabolic activation pathway of the cannabinoid as is the case in THCs. Tolerance and reciprocal cross-tolerance developed to pharmacological effects d8 - THC and ll-hydroxy-d8-THC , and the magnitude of tolerance development produced by the metabolite was significantly higher than that by d8-THC. The results indicate that ll-hydroxy-d8-THC has an important role not only in the pharmacological effects but also its tolerance development of d8 - THe. THCs and their metabolites competed to the specific binding of CP-55, 940, an agonist of cannabinoid receptor, to synaptic membrane from bovine cerebral cortex. The Ki value of THCs and their metabolites were closely paralleled to their pharmacological effects in mice. A novel cytochrome P450 (cyp2c29) was purified and identified as a major enzyme responsible for the metabolic activation of d8-THC at the II-position in the mouse liver. cDNA of CYP2C29 was cloned from a mouse cDNA library and its sequence was determined. The oxidation mechanism of THC by cyp2c29 was proposed.

  • In this paper, a 100 Watt high power amplifier has been implemented and performed evaluation, which is operating at UHF band ($470\;{\sim}\;806\;MHz$) for Digital TV repeater. To achieve increase of bandwidth and high power capability, 3-way power combiner and divider of Wilkinson type was adopted. In order to measure the fabricated 100 Watt power amplifier, the estimation technique function which makes equivalent mask was used. As a result of the measurement, the existence of pilot signal is confirmed and the signal transmitted at the rated output power 100 Watt is brought out the flat feature through 6 MHz bandwidth. and it resulted that its value was less than -47 dB at the edge of radiation channel and less than -110 dB at more than 6 MHz position from channel edge.

  • The EtOAc and CH$_2$Cl$_2$ soluble fractions from the fruit body of Ganoderma applanatum showed strong aldose reductase inhibitory activity. Nine compounds were isolated from both fractions. They were identified by spectral data as D-mannitol (1), 2-methoxyfatty acid (2), cerebrosides [(2S,3R,4E,8E)-1-O-${\beta}$-D-glucopyranosyl-3-hydroxy-2-[(R)-2'-hydroxypalmitoyl]amino-9-methyl-4,8-octadecadiene] (3), daucosterol (4), 2,5-dihydroxybenzoic acid (5), protocatechualdehyde (6), 5-dihydroergosterol (7), ergosterol peroxide (8), and cerevisterol (9).   • In this paper, we proposed a burst mode AGC loop and preamble detector applicable for VDL(VHF Digital Link) mode-2 using D8PSK modulation scheme and the performance analysis of proposed schemes is described. Generally the AGC scheme can be divided into two types, continuos and burst mode AGC. The continuos mode is performed well only with an analog feedback AGC loop. But the analog feedback AGC loop is not suitable for burst mode since its gain lock time is more than preamble duration, which causes the preamble detector misses preamble. Hence a fast digital AGC loop is required for burst mode. Also the AGC loop has to be designed with full gain during idle time to detect bursts although the signal level is very low. If the time to acquire gain lock is slow, the preamble detector fail to detect burst due to saturation of a lot of preamble samples. The receiver performance might be down even if the burst was detected because the preamble is used to estimate several parameters need to demodulation at receiver. In this paper we analysed relationships between the AGC loop and preamble detector. we present an AGC loop and preamble detector in burst mode.

Revers phase HPLC Separation of D-Amygdalin and Neoamygdalin and Optimum Conditions for Inhibition of Racemization of Amygdalin

  • In boiling aqueous solution, D-amygdalin usually begins to convert into neoamygdalin in 3 min and more than 30% of the initial D-amygdalin is found as neoamygdalin after 30 min. In this report, we establish methods for simple HPLC analysis and the inhibition of D-amygdalin conversion. D-Amygdalin and its conversion product, neoamygdalin, were clearly separated on reverse-phase column chromatography by an optimized eluent of 10mM sodium phosphate buffer (pH 3.8) containing 6% acetonitrile.   • The ethanol extract from the fruit of Terminalia chebula (Combretaceae) and the hook of Uncaria sinensis (Rubiaceae) exhibited significant inhibitory activity on oxidative stress and the age-dependent shortening of the telomeric DNA length. In the peroxidation model using t-BuOOH, human epidermal keratinocytes-neonatal foreskin (HEK-N/F) cells were treated with the T. chebula and U. sinensis extracts. The results showed a notable enhancing effect on the cell viability of 60.5 ${\pm}$ 3.8 and 65.0 ${\pm}$ 3.0%, respectively, by 50 $\mu\textrm{g}$/ml of the extracts. (omitted)

  • In this paper, we present an discrimination method of unknown digital modulated signals in noisy communication environment. We propose the use of an identification procedure based on time domain signal parameters. First, We extract instantaneous envelope. Frequency and difference phase as the basic feature informations from received signals. In order to identify signals using the extracted feature informations, we design the two dimensional feature space. The extracted feature infomations are mapped into2Dfeature space using 2D feature points. The procedure has been tested by simulations on a computer in noisy communication environment, and the considered signals are ASK-W, ASK-4, BPSK, QPSK, 8PSK, FSK, and QAM.

  • In this study, we investigated the molecular pathways targeted by platycodin D, which could involve apoptosis in immortalized human keratinocytes (HaCaT). We demonstrated that platycodin D-mediated apoptosis of HaCaT cells exhibited representative features, including DNA fragmentation, caspase-3, caspase-8 activation, and upregulation of Fas and FasL expression, but not p53 activation. To investigate the events involved in activation-induced FasL upregulation, we have examined mRNA accumulation, protein expression, and NF-$\kappa$B activity to elucidate transcription level in the HaCaT cell line treated with platycodin D.   • D-Amygdalin and its conversion product, neoamygdalin, were clearly separated on reverse-phase column chromatography by an optimized eluent of 10 mM sodium phosphate buffer (pH 3.5) containing 8.5% acetonitrile. Linearity for analyzing D-amygdalin and neoamygdalin was observed in the range from 0.05 to 0.5 mM. The detection limits for D-amygdalin and neoamygdalin were ca. 5 uM per injected amount. When extracting amygdalin from a whole piece of Armeniacae Semen in the boiling aqueous solution, there was almost no influence of emulsin; it resulted in higher extraction yield. (omitted)
