3D Micromorphology Producing within Poly(lactic acid) Skeleton Using Room-Temperature Ionic Liquids: From Particulate, Fibrous or Porous Scaffolds to Beads

  • We describe herein a three-dimensionally diverse micropatterning of poly(lactic acid), as a biopolymer, using 1-butyl-3-methylimidazolium-based room-temperature ionic liquids (bmim-based RTILs), [bmim]X (X = $SbF_6$, $PF_6$, $NTf_2$, Cl). Utilizing the hydrophobic bmim-based RTILs, [bmim]X (X = $SbF_6$, $PF_6$, $NTf_2$) and a phase separation technique, we were able to produce white and opaque membranes with a three-dimensional structure closely packed with particles ($10-50{\mu}m$ in diameter). The particlulate structure, made by the assistance of [bmim]$NTf_2$ and DCM, interestingly transformed to a fibrous structure by using a cosolvent, e.g., DCM/$CF_3CH_2OH$. When we used an increased amount of [bmim]$NTf_2$, the particles were effectively detached and macrosized ($100-500{\mu}m$ in diameter) and the oval-shaped beads were obtained in a powder form. By varying the counter-anion type of the imidazolium-based RTIL, for example from $NTf_2^-$ to $Cl^-$, the particulate 3D-morphology was once more transformed to a porous structure. These reserch results could be potentially useful, as a method to fabricate particulate scaffolds, fibrous or porous scaffolds, and beads as a biopolymer device in diverse fields including drug delivery, tissue regeneration, and biomedical engineering.

Isolation, Identification, and Characterization of a Bacteriocin-Producing Enterococcus sp. from Kimchi and Its Application to Kimchi Fermentation

  • A bacteriocin-producing lactic acid bacterium, which strongly inhibited the Lactobacillus plantarum recognized as an important acid spoilage microorganism in kimchi fermentation, was isolated from kimchi. From morphological, physiological, sugar fermentation, biochemical tests, and l6S rDNA sequencing results, the isolate was identified as an Enterococcus sp. and designated as Enterococcus sp. K25. The bacteriocin produced by Enterococcus sp. K25 inhibited several Gram-positive bacteria, including Lb. plantarum, whereas it did not inhibit Gram-negative bacteria and yeasts. Optimal temperature and pH for the bacteriocin production were $25^\circ{C}$ and 5.5, respectively. Enterococcus sp. K25 was applied to kimchi manufacturing alone and together with other preservatives (i.e., chitosan and fumaric acid). In addition, growth of lactic acid bacteria, pH, and titratable acidity (TA) were measured during aging at $5^\circ{C}$ and $10^\circ{C}$. Inoculation of Enterococcus sp. K25 together with fumaric acid showed the most synergistic effect on extension of kimchi shelf-life. Compared to control (no addition), the treatment prolonged the kimchi shelf-life up to 6 days, whereupon the eight-point TA value recognized as the edible limit was reached.

Stainless steel에 접종된 식중독 미생물에 대한 천연항균제 및 유기산 분무 살균효과 (Efficacy of Aerosolized Natural Antimicrobial and Organic Acids as a Sanitizer against Foodborne Pathogens on Stainless Steel)

  • 본 연구는 Escherichia coli O157:H7, Salmonella Typhimurium 그리고 Listeria monocytogenes에 대하여 천연항균제와 유기산을 이용한 분무 살균제 (aerosol sanitizer)의 살균효과를 검토하였다. 실제 가공공장의 주요 기계나 기구로 사용하는 stainless steel을 coupon으로 이용하여 인위적으로 접종한 식중독 미생물을 model cabinet에서 5분 동안 자몽종자추출물(grapefruit seed extract), acetic acid, citric acid 그리고 lactic acid로 처리 하였다. 3가지 식중독 미생물의 수는 GEF와 유기산 단독 처리시 0.34-3.77log의 균 감소를 나타내었고, 병행 처리시 1.72-3.89 log의 균 감소를 나타내었고 GEF, 유기산 그리고 알코올을 병행 처리시 1.46-5.05 log의 균 감소효과를 나타냈다. 세 종류의 유기산 중 항균력이 가장 높은 것으로 측정된 lactic acid를 이용하여 scale-up 모델에서 10분 동안 처리 한 결과는 E. coli O157:H7, S. Typhimurium 그리고 L. monocytogenes는 각각 3.42, 2.72, 2.30log의 균 감소를 나타내었다. 따라서 천연항균제와 유기산을 이용한 분무 살균제는 안전한 식품에 대한 소비자의 수요를 충족시키고, 환경 위생 방법으로 사용할 수 있다고 생각되었다.

L-Lactide 폴리머의 내열성 향상을 위한 핵제의 효과 연구 (The Effect of Nucleating Agent for Improving Heat Resistance Properties of L-Lactide Polymer)

  • 본 논문은 poly(lactic acid)의 내열성을 향상시키기 위한 결정화 핵제의 검토 및 연구에 대한 것이다. 네 종류의 미세분말 핵제인 금속염 2,2'-methylene bis(4,6-di-tert-butylphenol) salt를 결정화 핵제로 사용하였다. 폴리머 컴파운드의 열적 및 기계적 특성은 DSC, HDT, UTM을 통해 측정하였다. 이 연구의 결과로서, 컴파운드 한 폴리머 샘플들의 내열특성은 핵제의 함량 증가와 미세분말 일수록 선형적으로 증가 하였다. 그 중에서MPZ2의 함량이 2 wt%일때 가장 높은 내열특성을 나타내었으며, PL98Z2 컴파운드의 열변형 온도는 $116^{\circ}C$를 나타내었다 (ASTM D 648, 0.455MPa).

Effect of Lactic Acid Bacteria-Fermented Mulberry Leaf Extract on the Improvement of Intestinal Function in Rats

  • This study examined the laxative effects of mulberry leaf extract (MLE) fermented by lactic acid bacteria (LAB), which contains high levels of polyphenolic and flavonoid compounds, against loperamide-induced constipation in rats. Sprague-Dawley rats were divided into a normal group (N) and three experimental groups; loperamide treated group (C), loperamide and LAB-fermented MLE 300 mg/kg treated group (MLEL), and loperamide and LAB-fermented MLE 600 mg/kg treated group (MLEH). After 33 d, fecal pellet amount, fecal weight, water content of fecal, gastrointestinal transit time and length, and serum lipid profiles were measured. Constipation was induced via subcutaneous injection of loperamide (2.0 mg/kg b. w., twice a day) for the final 5 d of the experiment. After loperamide administration, the LAB-fermented MLE groups showed a significantly increase in the fecal pellets number, wet weight, and water content in rats compared with the C group. Moreover, increases in the intestinal length and viable Lactobacillus numbers in the feces were observed in the LAB-fermented MLE groups. The intestinal transit time was shorter in the LAB-fermented MLE groups than in the C group. In addition, the LAB-fermented MLE groups showed a significant decrease in triglyceride and total cholesterol levels and an increase in HDL-cholesterol level. These results indicated that oral administration of LAB-fermented MLE shows laxative effect in loperamide-induced constipated rats.

  • 이 연구에서는 촉매로서 tetraphenyltin을 사용하여 D,L-lactide (2-LA)와 ${\beta}$-methyl-${\delta}$-valerolactone(MVL)의 공중합체를 합성하고 그 물성을 조사하였다. 공중합체중의 lactic acid반복단위(LA)의 조성은 단량체 공급에서의 LA조성보다 높았다. 공중합체중의 그 조성은 공중합시간의 증가와 더불어 감소하였다. 그 공중합체의 수율과 분자량은 공급조성중의 LA의 증가와 더불어 증가하였다. 이들 결과는 공중합에 있어서 2-LA의 반응성이 MVL의 반응성보다 크다는 것을 뜻한다. 공중합체의 수평균분자량은 54,000에서 63,000의 범위에 있었고 다분산성지수는 1.7에서 2.1의 범위에 해당하였다. 공중합체는 융점을 나타내지 않았고 유리전이온도만 나타내었다. Lipase에 의한 공중합체의 분해성의 경향은 L-lactide-MVL 공중합체의 분해성의 경향과 거의 같았다.

Survival of Double-Microencapsulated Bifidobacterium breve in Milk in Simulated Gastric and Small Intestinal Conditions

  • Bifidobacteria are probiotic organisms that provide both flavor and health benefits when incorporated as live cultures into commercial dairy products. Because bifidobacteria are very sensitive to environmental conditions (acids, temperature, oxygen, bile salts, the presence of other cultures, etc.), their viability in human gastrointestinal tract is limited. The microencapsulation of bifidobacteria is a process to protect them against harsh environmental conditions, thereby increasing their viability while passing through human gastrointestinal tract. To confirm the survival rate of microencapsulated Bifidobacterium breve CBG-C2 in milk, their survival rate was compared with several kinds of free bifidobacteria and lactic acid bacteria in commercial yogurt products under simulated gastric and small intestinal conditions. Double-microencapsulation of the bacteria was employed to increase the survival rate during digestion. The outer layer was covered with starch and gelatin to endure gastric conditions, and the inner layer was composed of a hard oil for the upper small intestinal regions. Almost all microencapsulted bifidobacteria in the milk survived longer than the free bifidobacteria and lactic acid bacteria in the commericial yogurt products under the simulated gastric conditions. Numbers of surviving free bifidobacteria and lactic acid bacteria in the commercial products were significantly reduced, however, the viability of the microencapsulated bificobacteria in the milk remained quite stable under gastric and small intestine conditions over 3$\sim$6 hrs. Thus double-microencapsualtion of bifidobacteria in milk is a promising method for improving the survival of bifidobacteria during the digestive process.

직렬배열 다중전극 고전압 펄스 전기장 처리장치를 이용한 약주의 살균 (Sterilization of Yakju(Rice Wine) on a Serial Multiple Electrode Pulsed Electric Field Treatment System)

  • 7개의 전극을 직렬로 배열한 다중전극 고전압펄스 전기장(PEF) 처리장치를 사용하여 전기장 세기와 주파수를 달리한 square wave 펄스(너비 $1\;{\mu}s$)로 약주의 연속 살균을 시도하였다. 살균 전 약주의 총균수는 $1.88{\times}10^3{\sim}2.13{\times}10^4\;CFU/mL$, 효모는 $1.72{\times}10^3{\sim}2.39{\times}10^4\;CFU/mL$, 젖산균은 $1.55{\times}10^3{\sim}2.85{\times}10^4\;CFU/mL$이었다. 약주를 유량 1 mL/s의 속도로 PEF장치에 투입하여 처리한 결과 미생물 사멸기구는 1차반응으로 해석 될 수 있었고, 주파수 및 전기장세기가 증가함에 따라 살균율이 높아졌다. $D_{Hz}$값 및 $D_{PEF}$값은 전기장 세기가 증가함에 따라 감소하였으며, 효모는 세균에 비하여 낮은 값을 보였다. 젖산균은 30 kV/cm 이하의 전기장에서는 일반 호기성 세균에 비하여 낮은 $D_{PEF}$값을 보였으나, 40 kV/cm 이상에서는 높은 $D_{PEF}$값을 나타냈다. 미생물 종류별 $Z_{PEF}$값은 일반 호기성 세균 39.4 kV/cm 젖산균 49.3 kV/cm, 효모 47.6 kV/cm로 일반 호기성 세균의 전기장세기에 대한 의존성이 가장 큰 것으로 나타났다. PEF처리에 의한 약주의 색택 변화는 열처리에 비하여 미미하였으며, PEF살균 약주의 품질은 가열살균 약주에 비하여 월등히 우수하였다. 약주를 본 연구에서 사용한 PEF장치에 2회 통과시킬 경우 상업적 살균이 달성되었다.
