Development and Validation of HPLC-PDA Method and Pattern Recognition Analyses Using Eight Marker Compounds for the Quality Control Between the Seeds of Cuscuta chinensis Lam. and Cuscuta japonica Choisy

  • Cuscuta chinensis Lam. and Cuscuta japonica Choisy are parasitic plants. C. chinensis seeds were traditionally used for treatment of kidney and liver deficiencies. C. japonica seeds were used as tonic medicine to improve liver function and strengthen kidneys, treatment of high blood pressure, chronic diarrhea, and sore eyes. Cuscutae Semen are seeds of only C. chinensis in Korean Herbal Pharmacopoeia (K.H.P.). The developed HPLC-PDA method easily, accurately, and sensitively quantified using eight marker compounds [hyperoside (1), astragalin, (2), quercetin (3), kaempferol (4), chlorogenic acid (5), 3,4-di-O-caffeoylquinic acid (6), 1,5-di-O-caffeoylquinic acid (7), and 4,5-di-O-caffeoylquinic acid (8)]. In addition, the method may be used to distinguish seeds between C. chinensis Lam. and C. japonica Choisy. Furthermore, the result from the current study was applied to clarify samples between steam processed and unprocessed samples of C. chinensis by pattern analysis.

Growth-Inhibitory Effects of Cuscuta japonica Choisy's and C. australis R.Be's Extracts against Propionibacterium acnes (새삼 (Cuscuta japonica Choisy) 및 실새삼 (C. australis R.Be) 추출물의 여드름 유발균 Propionibacterium acnes 증식 억제 효과)

  • According to the traditional chinese herbal Cuscuta japonica Choisy and C. australis R.Be (original plant of Semen Cuscutae in Korea) have been used to cure pimple. In order to prove their bacteriostatic effects against Propionibacterium acnes, we performed the micro broth dilution method. The minimal inhibitory concentration (MIC) of C. japonica juice was 27.3 mg/ml, but C. australis juice showed no effect. The MIC of water extracted C. japonica seed and Wontosa and Bupjetosa were $960\;{\mu}g/ml$. The MIC of both water and ethanol extracts of C. australis seed was $9240\;{\mu}g/ml$. But ethanol extracts of C. japonicaseeds, Wontosa and Bupjetosa had no bacteriostatic activity. We also performed the disk diffusion test, C. japonica juice had prominent effect. Water extracts of C. japonica seed, Wontosa and Bupjetosa showed bacterio-static effect in 10 mg/ml, and C. australis seed in 10 mg/ml. The ethanol extract of C. japonica seed, Wontosa and Bupjetosa, and C. australis seed showed bacteriostatic effect in 20 mg/ml.

Inhibitory Effects of Cuscuta japonica Extract and C. australis Extract on Mushroom Tyrosinase Activity (새삼 (Cuscuta japonica Choisy) 및 실새삼 (C. australis R.Be) 추출물의 Mushroom Tyrosinase 활성 억제 효과)

  • The purpose of this study was to evaluate mushroom Tyrosinase inhibitory activity of Cuscuta japonica Choisy and C. australis R.Be. The experimental materials were expressed juice from their stems and flowers, both water and ethanol extracts, their seeds, and two kinds of commercially available cosmetic packing Wontosa and Bupjetosa (made from seeds of C. japonica). The 50% inhibitory concentration $(IC_{50})$ of C. Japonica juice was 5.4 mg/ml. However, C. australis juice showed negligible mushroom tyrosinase inhibitory activity. The $IC_{50}$ of water extracted C. japonica seed was $54.0\;{\mu}g/ml$, water extracted product of Wontosa $50.0\;{\mu}g/ml$ and Bupjetosa $40\;{\mu}g/ml$. The $IC_{50}$ of ethanol extracted C. japonica seed was $10\;{\mu}g/ml$, Wontosa $10\;{\mu}g/ml$ and Bupjetosa $20\;{\mu}g/ml$.

Inhibitory Effects on Melanin Biosynthesis and Tyrosinase Activity; Cytotoxicity in Clone M-3 and Antioxidant Activity by Cuscuta japonica, C. australis, and C, chinensis Extracts (새삼, 실새삼 및 갯실새삼 추출물이 Clone M-3 세포주의 Melanin 생합성 및 Tyrosinase 활성에 미치는 영향과 세포독성 및 항산화효과)

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).

The Effects of Cuscuta japonica Chois on Gene Expression in RANKL-induced RAW 264.7 Cell (도사자(菟絲子)가 RANKL 유도 파골세포(破骨細胞)에 미치는 영향)

  • Objectives : This study was performed to evaluate the effect of CJ(Cuscuta japonica Chois) on osteoclast differentiation and gene expression. Methods : The osteoclastogenesis and gene expression were determined in RANKL(receptor activator of nuclear factor kappa B ligand)-stimulated RAW 264.7. The results were summarized as followes. Results : CJ decreased the number of TRAP positive cell in RANKL-stimulated RAW264.7 cell. CJ decreased the expression of RANK(receptor activator of nuclear factor kappa B), $TNF{\alpha}$, and IL-6 in RANKL-stimulated RAW264.7 cell. CJ decreased the expression of iNOS and COX-2 in RANKL-stimulated RAW264.7 cell. CJ decreased the expression of Cathepsin K in RANKL-stimulated RAW264.7 cell. Conclusions : It is concluded that CJ might decrease the bone resorption resulted from decrease of osteoclast differentiation and it's related gene expression.

  • The portion of Cuscuta japonica haustorium which lies external to the host tissues, the upper haustorium, was investigated at the light- and electron-microscopic levels. The haustorium lightly contacted with the host was formed by the expansion of the epidermis and cortex of the stem at the contact side, and to have a group of meristematic cells within the haustorial cortex. When such a haustorium was closely contacted with the host, the meristematic region transformed into a primordial structure of the endophyte (endophyte primordium, EP) which may penetrate into the host tissues. EP consisted of the three kinds of cell group: dividing cells at the adaxial or proximal side; large, elongate cells (idioblasts) at the middle portion,; compressed cells at the abaxial or basal side. the idioblasts were characterized by the presence of large nucleus, dense cytoplasm, several small vacuoles, and abundant cell organelles including the multilamellar structures and cytosegresomes, and thus suggested to have a high metabolic activity. The features of the EP were discussed in relation to the possibility of the penetrating into host tissues.

  • The portion of Cuscuta japonica haustorium which lies internal to the host tissues, the endophyte, was examined at the ultrastructural level. The endophyte consisted of mainly small parenchymatous cells and large, slightly elongate cells at the tip. The tip cells were characterized by the presence of large and lobed nucleus, several small vacuoles, dense cytoplasm, abundant rough endoplasmic reticulum, dictyosomes, and mitochondria, and thus suggested to have a high metabolic activity. The shape, arrangement, and cytological characteristics of the parenchymatous and tip cells consisting the endophyte were very similar to those of the dividing cells and idioblasts, respectively, which appeared in the endophyte primordium of the upper haustorium. The tip cells with the thickened-apical wall were observed to grow intrusively through the host cell walls and to engulf the remnants of the degenerated host cells. In the former case intrusive growing cell was regarded to develop into the filamentous cell, the hypha. Plasmodesmata through the cell wall were not observed between host and parasite cells. Some host cells that in contact with the penetrating tip cells of the endophyte, showed the degenerating features such as a loss of cytoplasm, a beaded fashion of small vesicles, and deformation of chloroplasts.

Effect of Essential Oil from Cuscuta japonica CHOISY on Proliferation and Migration in Human Skin Keratinocyte (새삼(Cuscuta japonica CHOISY) 유래 정유의 피부 각질형성세포 증식 및 이주에 미치는 효과)

  • The migration and proliferation of keratinocytes are key events in re-epithelization, itself a major phase in the wound healing process. Cuscuta japonica Choisy (CJC) is used as a traditional medicine to improve liver, heart, and intestinal function, and its extracts are reported to have various biological properties such as whitening, anti-oxidancy, and an anti-acne effect. However, it is not yet known in particular whether or not CJC essential oil (CJCEO) affects skin regeneration. In the present study, we isolated CJCEO by solvent extraction and tested its effect on wound healing responses using normal human keratinocytes, namely HaCaT cells. We found that CJCEO induced proliferation as well as migration in HaCaT cells in a dose-dependent manner. Compared with a control group, CJCEO treatment at 250 ㎍/ml increased proliferation by 239.98±5.51% in HaCaT cells in a dose and migration by 124.86±6.06%. Moreover, the oil induced sprout outgrowth and, at 250 ㎍/ml, increased collagen synthesis by 148.56±15.47% in HaCaT cells. These results demonstrate that CJCEO may promote skin regeneration and wound healing by increasing the migration, proliferation, and collagen synthesis of HaCaT cells. We therefore suggest that CJCEO could be used as a cosmetic material.

Effect of Water Extracts of Cuscuta Japonica Chois in RANKL-induced Osteoclast Differentiation (파골세포 분화에서 토사자 물 추출물의 효과)

  • Osteoclasts are bone-resorbing multinucleated cells derived from the monocyte/macrophage lineage. The differentiation of osteoclasts are regulated by osteoblastic cells expressed RANKL, which is the most critical molecule for osteoclast differentiation. In this study, we found that water extracts of cuscuta inhibited RANKL-mediated osteoclast differentiation by direct action on bone marrow macrophages (BMMs) without cytotoxicity. In BMMs, water extracts of cuscuta inhibited the mRNA expression of c-Fos, NFATc1, TRAP, and OSCAR. Also, the protein expression of c-Fos and NFATc1 was inhibited by water extracts of cuscuta treatement. Water extracts of cuscuta inhibited the phosphorylation of p38, ERK, and JNK in BMMs treated with RANKL. However, water extracts of cuscuta did not inhibit RANKL-induced I-${\kappa}B$ activation. Water extract of cuscuta failed to inhibit bone resorption by osteoclasts cultured on hydroxyapatite plates. These results suggest that cuscuta may be a promising drug for use against bone disorders such as osteoporosis and rheumatoid arthritis.
