Statement of problem. The effects of surface roughness have not or insufficiently been analyzed on earlier events such as cell adhesion though cell behavior most germane to implant performance is cell adhesion. Purpose. The purpose of this study was to evaluate cell adhesion of osteoblast-like cells (MG63) onto three types of titanium disks with varying roughness using the Elisa assay. Materials and methods. Representative disks from each group (SLA, HA, machined) were subjected to surface analysis and surface roughness was measured by the optical interferometer (Accura 2000, Intekplus Co., Seoul, Korea). Following this, MG63 cells were cultured on the titanium disks and released. Cell adhesion measurements using the Elisa assay were performed specifically at three points: after 24, 48, and 72 hours of culture. Results. Among the 3 types of surface analyzed, the SLA surface was the roughest with a Ra value of $1.114{\mu}m$ followed by HA coated surface and machined surface, consecutively. The optical density values for the SLA surface group was significantly higher than that of the machined and HA coated surface groups following 24 and 48 hours of culture. The cell culture on HA coated surface showed significantly higher values compared to the machined surface following 24, 48 and 72 hours of culture. Conclusion. The results suggest that surface treatment of titanium surfaces enhanced cell adhesion of human osteoblast-like cells (MG63).
Spin-filters are used as cell separation devices for achieving high cell density and high productivity in animal cell culture. We have proposed a model for the cell growth in a spin-filter perfusion culture and examined the effects on cell growth by several parameters including ammonia inhibition, specific growth rate, specific feeding rate, and cell retention. Results from computer simulation and sensitivity analysis indicate that the cell retention affects the cell growth mostly while there is a significant inhibition on cell growth by the ammonia accumulated during the culture. The specific feeding rate has minimal effects on cell growth, which is consistant with the fact that the cell growth with a step feeding is quite similar to that with a continuous feeding.
Sunflower (Helianthus annuus L.) protoplasts were isolated from seven-day-old etiolated hypocotyls of 10 A line and four-week-old fully expanded young leaves of PI 441983 line in vitro seedlings using an enzymatic method. Purified protoplasts were collected by filtration and floatation in sucrose solution. Semi-solid protoplast culture was performed using the L4 regeneration protocol with various culture media and plating densities to achieve the highest efficiencies for protoplast culture of hypocotyl and mesophyll protoplasts of 10 A and PI 441983 lines, respectively. The concentrations in liquid L'4M medium and different plating densities were evaluated in two types of cytokinins, the adenine-type 6-benzyladenine (BA) and the phenylurea-type thidiazuron (TDZ). The highest colony formation was achieved in both sunflower lines when 0.5 mgL-1 BA and 0.5 mgL-1 TDZ were applied with a high plating density (3 × 105 protoplasts mL-1). These conditions led to 38.45% and 39.40% colony formation for hypocotyl protoplasts of the 10 A line and mesophyll protoplasts of the PI 441983 line, respectively. Moreover, many hypocotyl protoplast-derived colonies developed into micro-calli. In addition, superior development of both sunflower protoplasts was observed with all plating densities when BA was used in combination with TDZ. This finding will be applicable to future sunflower hybrid production via somatic hybridization.
We investigated the effects of salinity on an estuarine cladoceran, Diaphanosoma celebensis as a substitute live food organism of Artemia, which has been used for the next step of rotifer, in the course of seedling production of marine fishes, Culture density for growth, possibility of stable culture (RPGI: Relative Population Growth Index) and food feeding activity (RCN: Remaining Cell Numbers) of D. celebensis were investigated at intervals of 5 ppt under 10 steps of different saline conditions from 0 to 45 ppt. According to the results, the experimental group, which was regarded as the best condition of culture density for growth, stable culture possibility (RPGI) and good food feeding activity (RCN), was salinity culture condition of 20 put, and relative stable culture conditions were observed under saline concentrations $15\~35\;ppt$. Their functions under culture conditions of lower or higher salinities than 20 ppt appeared remarkably to be decreased, while all individuals were dead under the saline condition of 0 ppt after the beginning of the experiment. From these results of this study, it is assumed that estuarine cladoceran, D. celebensis can be cultured easily as a substitute live food organism of Artemia in the course of seedling production of marine fishes.
A growth study was conducted to investigate the effect of stocking density on performance of juvenile fat cod (Hexagrammos otakii Jordan et Starks.). Fish averaging 13 g were stocked into duplicate tanks ($250\;{\ell}$ each) at four different densities of 330 g/$100\;{\ell},\;660\;g/100\;{\ell},\;990\;g/\;100\;{\ell}$, and $1330\;g/100\;{\ell}$, and fed MP diet containing $50\%$ frozen horse mackerel and $50\%$ commercial binder meal for 2 months. Weight gain, feed efficiency, survival rate, and protein retention were decreased as density increased. Whereas these parameters were not significantly different (P>0.01) fish stocked at initial stocking densities of $330\;g/100\;{\ell},\;660\;/g/100\;{\ell}\;and\;990\;g/100\;{\ell}$. Moisture and lipid content of whole body were not affected by stocking densities (P>0.01). These results indicate that stocking density should be considered, and $1{\~}2$ kg fish/$100\;{\ell}$ final stocking density for juvenile fat cod could be used to decrease production cost for the indoor tank (culture) system.
This study was conducted to investigate bone density and food behavior of middle-aged (<65-years-of-age) and oldaged women (${\geq}$65-years-of-age) in the Seoul area of Korea. Average height and weight, body mass index, body fat percentage, and waist-hip ration (WHR) of the middle-aged participants were 158.0 cm, 58.4 kg, 23.3, 29.7%, and 0.88. Corresponding measurements for old-aged participants were 152.6 cm, 56.3 kg, 24.1, 33.1%, and 0.93. T- and Z-scores for middle-aged women were -1.20 and -0.21, and those of old-aged women were -2.02 and -0.49. Osteopenia and osteoporosis percentage of middle- and old-aged women were 41.2% and 14.7%, and 45.2% and 37.1%, respectively. Significant differences were evident concerning between-meal, overeating, exercise, and nutritional supplementation among the two groups. Middle-aged women displayed a positive relationships between bone density and the parameters of height (p<0.05), weight (p<0.05), exercise (p<0.05), consumption of soybean and soybean-processed food (p<0.05), and negative relationships between bone density and the parameters of WHR (p<0.001), consumption of meat and meatprocessed food (p<0.05), and tea drinking (p<0.05). In old-aged women group, positive relationships were evident for bone density and the parameters of height (p<0.05), weight (p<0.05), exercise (p<0.05), nutritional supplementation (p<0.05), and consumption of soybean and soybean-processed food (p<0.05). The results reveal a link between healthy bones and desirable food behavior, indicating the importance of a practical and systematically organized food behavior education program for middle- and old-aged Korean women.

To enhance the productivity and activity of nitrile hydratase in Rhodococcus rhodochrous M33, a glucose-limited fed-batch culture was performed. In a fed-batch culture where the glucose was controlled at a limited level and cobalt was supplemented during the fermentation period, the cell mass and total activity of nitrile hydratase both increased 3.3-fold compared to that in the batch fermentation. The productivity of nitrile hydratase also increased 1.9-fold compared to that in the batch fermentation. The specific activity of nitrile hydratase in the whole cell preparation when using a fed-batch culture was 120 units/mg-DCW, which was similar to that in the batch culture.
The experiment was carried out to investigate the productivity of freshwater rotifer, Brachionus calyciflorus and marine rotifer, B. rotundiformis at various temperatures, initial inoculation and pH in a 5 L semi-continuous high density culture. Rotifers were fed by commercial condensed freshwater Chlorella. When pH was not controlled, average daily productions of freshwater and marine rotifers increased with temperature. The highest production, $44\times10^6$ rotifers, was achieved of B. calyciflorus at $32^{\circ}C$ and the possible production lasting period of B. calyciflorus was shorter than that of B. rotundiformis. Under the adjustment of pH at 7, the possible production lasting periods of B. calyciflorus and B. rotundiformis inoculated with 5,000 inds./mL were longer than those of rotifers inoculated with 10,000 inds./mL, and the daily production rate of the former was higher than that of the latter. The results from this study indicated that optimum density of the initial inoculation for the cultivation of B. calyciflorus and B. rotundiformis was 5,000 inds,/mL under the controlled conditions of pH 7 and at $32^{\circ}C$ in a semi-continuous high density culture, in terms of production rate and food cost aspects.

For the establishment of an efficient embryogenesis from microspore culture in Brassica napus L. domestic cultivar 'Tammiyuchae', four different factors affecting microspore embryogenesis and plantlet regeneration were investigated. The highest embryogenesis rate was achieved when microspores at late uninucleate to early binucleate stage were isolated from flower buds with a length of 3.0~3.5 mm. On average, 388 embryos generated from 1 ml of microspores media. The highest number of embryos was obtained when microspores were subjected to $32.5^{\circ}C$ for 2 days. Embryogenesis of 'Tammiyuchae' was increased with increasing microspore culture density up to about $5{\times}10^4ea/mL$. Gradually higher culture density repressed embryogenesis of microspores. Regeneration rate of shoots from microspore-derived embryos was observed in MS solid medium supplemented with $0.5mg{\cdot}L^{-1}$ NAA and $1.0mg{\cdot}L^{-1}$ BA, and grew well in MS solid medium without plant growth regulators.
