Botrytis cinerea에 의한 들깨 잿빛곰팡이병의 발생 (Occurrence of Gray Mold Rot of Perilla Caused by Botrytis cinerea)

  • Gray mold rot of perilla was epidemic at Kangdong, Pusan and Miryang, Kyungnam in 1997 and 1998. The incidence of this disease ranged form 21.3 to 68.1% at Kangdong area. Leaf necrosis initially appeared on the edge of the infected leaves, and it was developed to the center of the leaves forming typical V-shaped brown necrotic lesions. Under high moisture condition, abundant mycelia of the pathogen was formed on the surface of the lesions. Infected stems became slender and were completely blighted up to the top of the plant. Two isolates, LVF12 and SD7, were isolated from diseased lesions showing typical symptoms, and the pathogenicity was tested using mycelial disks and conidial suspension inoculation. The developed symptoms were same as the naturally produced ones. These two pathogenic fungi were identified as Botrytis cinerea based on the morphological characteristics using a microscope and a scanning electron microscope, and cultural characteristics. This is the first report of gray mold or perilla in Korea.

Three Unreported Fungi Isolated From Reservoirs in Korea: Mortierella biramosa, Paraphoma radicina, and Sordaria macrospora

  • Freshwater ecosystems have a large reserve of latent biological resources that play an essential ecological role, and have significant economic and social value. Fungi in freshwater are prospective materials that can be used in the food, medicine, and biomass energy fields. In this study, three promising fungal species were isolated from freshwater ecosystems in Korea. These isolates were identified as Mortierella biramosa, Paraphoma radicina, and Sordaria macrospora, based on their cultural and morphological characteristics, as well as molecular phylogenetic analyses. These species were previously unknown in Korea. The finding allows us to explore its physiological and biochemical characteristics in more detail and use them as biological resources.

Galactomyces pseudocandidus (Dipodascaceae): An Unrecorded Yeast-like Fungus Isolated from the Digestive Tract of Procambarus clarkii

  • The unrecorded fungus Galactomyces pseudocandidus de Hoog & M.T. Sm. was isolated and cultured from an introduced species, Procambarus clarkii, in Korea. Two related species, Galactomyces citri-aurantii and Geotrichum candidum, have been reported, but no other Galactomyces spp. have been found in Korea. In this study, we isolated G. pseudocandidus from the digestive tract of American crayfish using RGY (river water-glucose-yeast) medium and then compared the morphological and molecular characteristics of this species with those of related species. As a result, we confirmed that G. pseudocandidus is an unrecorded fungus in Korea, so we described and reported the cultural characteristics and microstructures, such as conidia.

An Unrecorded Genus Lambertella Höhn. (Rutstroemiaceae) and Its Unrecorded Species in Korea

  • Lambertella corni-maris Höhn., an endophytic fungus that is an unrecorded species in Korea, was isolated and cultured from alpine coniferous Korean fir. The Lambertella sp. was found in Korean pine in Yeongwol-gun but no Lambertella spp. were recorded elsewhere in Korea. In this study, the morphological characteristics and molecular barcode sequence of L. cornimaris were compared to its related species. We tried to explain the cultural characteristics and microstructures, such as the conidia of L. corni-maris, compared to closely related taxa and present it.

청자상감운학문매병 제작 재료의 물리화학적 특성 및 제작 단계별 형상학적 특성 변화 (Physicochemical properties of the materials used for the production of celadon maebyeong inlaid with cloud-and-crane designs and changes in their morphological properties by production stage)

  • 본 연구는 전통도자기의 제작 전과 후, 소성 전과 후에 기물의 내부에서 일어나는 다양한 물리화학적 변화를 알아보고자 청자상감운학문매병의 재현품을 성형 → 조각 → 상감 → 초벌 → 유약 → 재벌 등의 6단계로 제작하여 과학적 조사 및 3차원의 정밀한 투과 이미지 촬영으로 획득한 이미지를 세부 영역별로 분석하였다. 재현편의 제작 재료는 각각의 재료가 가진 성분에 따라 다른 광물상이 존재하며, 산화철(Fe2O3)의 함량이 높은 흑상감은 어두운 색을 나타냈고, 알루미나(Al2O3)의 함량이 높은 백상감은 백색이 나타나는 것과 같이 색상에도 영향을 준다. 재현편의 색도와 전암대자율, 주요성분 등의 물리화학적 특성을 CT이미지를 통해 밀도의 차이로 확인할 수 있었으며, 미세조직과 흡수율, 겉보기기공률, 주요구성광물 등의 번조에 따른 특성 변화는 영상 분석에서 기물 내부의 기공 유무와 존재량, 균열의 발달과 확장으로 파악하였다.

내열성 포도당 이성화효소를 생산하는 Streptomyces chibaensis J-59의 분리 및 동정

  • A bacterial strain J-59 was isolated from a humus soil, which produced simultaneously a thermostable glucose isomerase as well as xylanase. The morphological, cultural and physiological characteristics of the isoisomerase strain J-59 were detemined by the use of the media and methods described in International Streptomyces Project. The chemotaxonomic characteristics of the isolated strain J-59 were determined by the analysis of G+C molar % of DNA, diaminipimelic acid, composition of fatty acid and menaquinone. As the results of various examinations, the strain J-59 was identified to be Streptomyces chibaensis. This strain produced glucose isomerase intracellularly and xylanase extracellularly when grown in a medium containing xylan, but it was not able to utilize the xylose or xylan as a carbon source. The glucose isomerase of S. chibaensis J59 was highly thermostable, which retained more than 75% activity in the presence of Co$^{2+}$ at 80$\circ $C for 72 h.

Podospora leporina (Podosporaceae): An Unrecorded Endophytic Fungus Isolated from Cypripedium guttatum

  • Here, Podospora leporina was isolated and cultured from Cypripedium guttatum Sw. (spotted lady's slipper), which was designated as an endangered wildlife class 1 by the Ministry of Environment. Further, Podospora sp., which is found in Calanthe discolor Lindl. (Japanese hardy orchid) root is recorded as an endophytic fungus in Jeju Island, Korea. However, there is no existing record of Podospora spp. Thus, in this study, we analyzed the morphological characteristics and two molecular markers, internal transcribed spacer (ITS) and large ribosomal subunit (LSU), of P. leporina and compared these data with other relative species. Consequently, this endophytic fungal species was confirmed as an unrecorded species in Korea, so we presented cultural characteristics and microstructures, such as the conidia of P. leporina.

Occurrence of Anthracnose on Welsh Onion Caused by Colletotrichum circinans

  • Anthracnose occurred frequently on leaf sheaths of Welsh onions grown in Gangwha island, Korea in November, 2007. The disease incidence was as high as 30% in five fields investigated. A total of 20 single spore isolates of Colletotrichum species were obtained from the affected plants, and all the isolates were identified as Colletotrichum circinans based on their morphological and cultural characteristics. Three isolates of the fungus caused anthracnose symptoms on the leaf sheaths of Welsh onions by artificial inoculation, which were similar to those observed during the field survey. In this study, the mycological and pathological characteristics of C. circinans identified as causing anthracnose of Welsh onions are clarified.

산느타리버섯의 품종별 재배특성 (Cultural characteristics of fruit body on Pleurotus pulmonarius cultivars)

  • 국내에서 육성된 산느타리버섯 8품종을 병재배 방식으로 재배특성을 조사하였다. 8품종의 자실체 형태는 전체적으로 갓직경이 크고 대는 짧은 형태였다. 대는 편심생으로 갓에 결합되어 있다, 자실체의 길이가 가장 긴 품종은 여름2호(72.8mm)이고 가장 짧은 품종은 여름1호(33.8mm)이다. 대의 두께는 여름2호, 호산의 순서로 두겁고 15.5mm, 12.8mm 였다. 갓의 직경은 45.8-62,3mm로 약산이 가장 작고 여름2호가 가장 큰 갓을 지닌 것으로 나타났다. 전체 무게(수확량)은 여름2호가 80.7g으로 가장 많은 생산량을 보였고 여름1호가 35.5g으로 가장 낮은 수치를 보였다. 가장 갓색이 밝은 품종은 여름2호로 55.6이었고 호산도 54.1이었다. 특징적으로 갓색이 가운데 부분은 엷은 편이었으며 가장자리가 더 진하게 착색되었다. 평균적 발이소요일과 생육소요일은 각각 4.6일과 8.0일이어서 발이 후 3.4일 만에 수확을 완료하였다.

Comparison of Characteristics of Ganoderma lucidum According to Geographical Origins : Consideration of Morphological Characteristics(II)

  • Nine species of genus Ganoderma different in cultural characteristics each other were separated by histo-anatomical differences. Differences among the species and geographic distribution of G. lucidum were also analyzed to provide the criteria for the classification of Korean cultivation or wild type strains. Korean cultivation and wild type strains were quiet different from other species as well as Taiwan and North American G. lucidum strains in histo-anatomical traits of Ganoderma. Pore color of Korean G. lucidum strains was less brown than those of Taiwan and North American strains. Shapes of pores were round or circular in Korean strains but ellipsoidal or angular in Taiwan or North American strains. Pore numbers of Korean strains were more than those($4{\sim}6/mm$) of Taiwan or North American strains. Hardness of the pileus of Korean strains was much more than that of Taiwan or North American strains. Such characteristics of Korean strains were different from those of other species of Ganoderma. Korean G. lucidum strains could be classified into the other group because they had many different traits in growth characteristics of fruiting bodies and histo-anatomical characteristics from those of Taiwan or North American G. lucidum and other species of G. lucidum complex.
