Polyphenolic compounds widely occurring in the traditional medicine plants have been reported to possess strong antioxidant activity. The antioxidative substances of water soluble extract from leaves, stem bark, root bark and fruit powder of Cudrania tricuspidata were tested in three different in vitro experimental models. In oxidation models using DPPH( , ' diphenyl picrylhydrazyl) method, Fe2+ induced linoleic acid peroxidation, and autooxidation of hepatic microsomal membranes, the antioxidative activities of water soluble extract from stem bark were strong than that from leaves and root bark. Fruits of Cudrania tricuspidata contained the highest amounts of polyphenolic compounds among the parts of this plant. The changes in polyphenolic compound contents of fruit powder caused by heat treatment (20oC, 40oC, and 60oC) were also monitored. After water blanching, contents of phenolic compounds was increased slightly in the following order; 20oC(1454mg), 40oC(1487mg), and 60oC(1511mg). These results supports that water soluble extracts from Cudrania tricuspidata contain antioxidative compounds.
High-performance liquid chromatography coupled to an online $ABTS^+$-based assay (online HPLC-$ABTS^+$) system was used to determine the principal antioxidants in Cudrania tricuspidata fruits. Six prenylated isoflavonoids (1 - 6) were isolated from C. tricuspidata fruits according to the online HPLC-$ABTS^+$ system. The structures of isolated compounds, alpiniumisoflavone (1), 6,8-diprenylorobol (2), 6,8-diprenylgenistein (3), pomiferin (4), 4'-methylalpiniumisoflavone (5), and osajin (6) were identified by their retention time, UV spectra, ESI-MS, and NMR data. Among these compounds, 6,8-diprenylorobol (2) and pomiferin (4) reduced A2E photooxidation in a dose dependent manner.
The aim of this study was to develop a new functional traditional fermented soybean food using Cudrania tricuspidata fruits and fermentation microbes isolated from traditional fermented food. Aspergillus oryzae koji, Lactobacillus sp., and Bacillus sp. were used for the selection of a suitable microbe for the fermentation of Cudrania tricuspidata fruits, and as a result, Bacillus licheniformis SCDB 1234 was selected. SCDB 1234 enhanced the concentration of kaempferol in the Cudrania tricuspidata fruits from 9.54 to $217.04{\mu}g/g$ (about 22 times). The DPPH radical scavenging activity of the fermented materials was similar to that of BHA and BHT (92~99 ppm). The tyrosinase inhibitory activity was high with arbutin (95 ppm) and kojic acid (90 ppm). Doenjang-added fermentation materials of the Cudrania tricuspidata fruits were developed, and the organic acid, reducing sugar, and free amino acid of the developed Doenjang were analyzed. The pancreatic lipase inhibitory (PLI) activity and ${\alpha}$-glucosidase inhibitory (AGI) activity of the fermentation materials of the Cudrania tricuspidata fruits and the developed Doenjang were investigated, and it was found that after fermentation, the PLI and AGI activities of the fermentation materials of the Cudrania tricuspidata fruits were higher than those before fermentation, and that the AGI activity of the developed Doenjang after aging ($91.25{\pm}0.04%$) was higher than that before aging ($84.89{\pm}0.08%$).
Macrophages play a crucial role in the host immune defense system. The current study investigated immunomodulatory activities induced by polysaccharides extracted from Cudrania tricuspidata (CTPS) fruits in murine macrophages and their role in signaling pathways. In macrophages, CTPS predominantly induced nitric oxide (NO), tumor necrosis factor-a, and interleukin-6 production. In addition, CTPS significantly up-regulated expression of the macrophage surface marker (CD80/86 and MHC class I/II). These results indicate that polysaccharides extracted from CTPS may potentially play an immunomodulatory role in macrophages via mitogen-activated protein kinases and nuclear factor-B signaling. These findings may be useful in the development of immune enhancing adjuvant materials obtained from natural sources.
We evaluated total phenolics and antioxidative activities of water extracts from different parts of Cudrania tricuspidata (specifically, the leaves, stems, roots, and fruits). The antioxidative activities of these samples were determined using five methods, including 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS) radical scavenging, reducing power, ferric reducing ability of plasma (FRAP), and $\beta$-carotene/linoleic acid system. The water extract of leaves exhibited the higher DPPH, ABTS radical scavenging activities, reducing power, and FRAP than water extract of stem, roots, and fruits. Inhibition values on linoleic oxidation of water extracts from leaves, stems, roots, and fruits were calculated as 45.98%, 33.03%, 39.73%, and 25.48% at 10 mg/mL, respectively. The water extract of C. tricuspidata leaves had the highest amount of toal phenolics (73.60$\pm$0.28 mg/g). High-performance liquid chromatography (HPLC) analysis showed that quercetin is the predominant phenolic compound in water extract of leaves. Thus, our study verified that the water extract of leaves has strong antioxidant activities which are correlated with its high level of phenolic compounds, particularly quercetin. This water extract of C. tricuspidata leaves can be used as an effective and safe source of antioxidants.

A methylene chloride soluble fraction of the fruits of Cudrania tricuspidata significantly inhibited the mouse brain monoamine oxidase (MAO). Three known prenylated isoflavones were isolated and identified by activity-guided fractionation. Gancaonin A (1), 4'-O-methylalpinumisoflavone (2), and alpinumisoflavone (3) inhibited MAO activity in a concentration-dependent manner with $IC_{50}$ values of 19.4, 23.9, and 25.8 $\mu$M, respectively. Of these, gancaonin A (1) showed a selective and potent inhibitory effect against MAO-B ($IC_{50}$ 0.8 $\mu$M) than MAO-A ($IC_{50}$ >800 $\mu$M). The kinetic analysis using Lineweaver-Burk plots indicated that gancaonin A (1) competitively inhibited MAO-B.
This study was conducted to classify the variation in leaf and fruit morphology among natural grown 16 Cudrania tricuspidata stands. As results of the analysis in C. tricuspidata stands total 134 Species were found and as results of the analysis in leaf morphology, it was grouped as Group IV. By the analysis of variance in length, width, and weight of fruits were differed in 15 C. tricuspidata stands. The length, width, and weight of fruits were higher in Jinan and Sunchang, than Gochang, Gimje, and Goheung. By the cluster analysis of 3 fruit morphology were classified as 3 Groups. According to the results of this study C. tricuspidata leaves and fruit morphological characteristics for each group were compared by analysis there were differences between the morphological characteristics and the characteristics of the fruit did not match.
The current study examined antioxidant and neuronal cell protective effects of the crude polysaccharide fraction in Cudrania tricuspidata fruits (CTP). The radical scavenging activities of (1,1-diphenyl-picrylhydrazyl and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid)) and reducing power and FRAP of CTP were increased dose-dependently. In addition, the expression of neuroprotective effect of CTP was tested in HT22 mouse hippocampal cells. CTP treatment exhibited non cytotoxicity at dose levels below $500{\mu}g/mL$. Within this optimal concentration range, CTP treatment significantly increased cell viability in $H_2O_2-treated$ HT22 cells. Furthermore, CTP treatment increased superoxide dismutase (SOD) activity and decreased malonaldehyde (MDA) levels in HT22 cells. Therefore, these results indicate that the crude polysaccharide fraction from Cudrania tricuspidata fruits (CTP) possesses antioxidant activities and displays therapeutic potential as a useful source material in the development of brain disorder treatments targeting oxidative stress in neuronal cells.

In this study, an analytical method to evaluate the quality of isoflavonoid compounds purified and isolated from the fruits of Cudrania tricuspidata was developed and validated using Ultra Performance Liquid Chromatography (UPLC). The fruits of C. tricuspidata were extracted with methanol and further fractioned with n-hexane, ethyl acetate and water. The resulting ethyl acetate extract separated into four isoflavonoid compounds by a combination of silica gel and sephadex LH-20 column chromatography. The structures of the compounds were elucidated as alpinumisoflavone, 6,8-diprenyl genistein, 6,8-diprenyl orobol, 4'-O-methylalpinumisoflavone by various techniques such as UV-Vis, ESI-MS, $^1H\;NMR$ and $^{13}C\;NMR$ spectroscopy. Finally, a method to characterize the compounds was developed by using the UPLC equipped with a $C_{18}$ column and a gradient mobile phase system consisting of 2% acetic acid in water (solvent A) and 2% acetic acid in methanol (solvent B). The developed method was validated with the parameters such as selectivity, linearity, limit of detection, limit of quantitation, accuracy, and precision, which are defined by the ICH (International Conference on Harmonization). Using the validated method, the compounds in the fruits harvested in different months were also quantitatively analyzed. We propose this approach this approach can readily be utilized as an efficient evaluation method to quantify the extracts of C. tricuspidata.
To raise the added value of the fruits of Cudrania tricuspidata, Cudrania tricuspidata vinegar was produced and examined for its fermentation conditions. Forty nine acetic acid bacteria with resistance against acetic acid, ethanol, and sulfide as high acetic acid producers were isolated from fermented foods and identified as Acetobacter indonesiensis, A. cerevisiae, A. orientalis, A. tropicalis, A. fabarum, A. pasteurianus, and A. syzygii based on the results of the analysis of the 16S rRNA gene sequences. Among them, two GRAS strains, A. pasteurianus SCMA5 and SCMA6, were finally selected for the production of acetic acid. Optimal vinegar productions were obtained from the medium containing 40% (v/v) fruit juice of Cudrania tricuspidata and 5% (v/v) ethanol at $25^{\circ}C$ for 72 hr. The sensory panel preferred the vinegar fermented with the SCMA06 to that with the SCM05 strain. The radical scavenger capacity of DPPH was 53% higher than that of the control in the vinegar fermented with the SCMA06 strain. The ${\alpha}$-glucosidase inhibitor activity as an index of the antidiabetic drug showed 91% inhibition, which is higher than that of acabose. This study will be helpful for the scale-up production of vinegar with the fruit of Cudrania tricuspidata.
