  • For evaluation of the prevalence of Cryptosporidium sp. infection in diarrheic and non-diarrheic humans in Iran, fecal specimens from diarrheic (n = 129) and non-diarrheic humans (n = 271) were collected and examined for the presence of Cryptosporidium sp. oocysts. The presence of Cryptosporidium sp. oocysts was determined by Ziehl-Neelsen acid-fast staining. Humans were grouped according to their age as follows: younger than 15, 16-25, 26-35, 36-50, and over 51 years. The results showed that the overall prevalence of infection in all 400 samples was 10.8%, but the prevalence (25.6%) in diarrheic humans was higher than that (3.7%) in non-diarrheic humans. Oocysts of Cryptosporidium sp. were detected in the feces of 21.4%, 9.3%, 8.8%, 6.7% and 5.7% of different age groups, respectively. The intensity of oocysts was significantly higher in diarrheic humans than in non-diarrheic ones. There was a significant association between Cryptosporidium sp. infection and occurrence of diarrhea (P < 0.05). The results indicate that Cryptosporidium sp. infection is prevalent in diarrheic humans in Iran.

  • Cryptosporidium spp. were detected in 17 of 135 swine lagoon samples from five farms by 18S ribosomal DNA locus and PCR. Seventeen positive samples identified were included two distinctive genotypes C. suis and Cryptosporidium sp. based on a small-subunit rRNA gene-based PCR-restriction fragment length polymorphism analysis. Cryptosporidium spp. were detected out of farrowing, farrowing and nursery (mix), and finishing. Prevalence rate was 12.6% with infection rates between 3.7 and 18.5%. We concluded that Cryptosporidium oocysts can persist in treated lagoon and potentially contaminate surface water through improper discharge. This study was undertaken for the evaluation of the infection status of the genotypes of Cryptosporidium spp. in swine lagoon.

  • Cryposporidium species have a wide host range. These coccidian parasites are found in close association with epithelial cells of many species of animals including mm. The gastrointestinal tract is most commonly affected in young ruminants and this parasite is thought to be considerable importance in calf diarrhea complex. Major outbreaks of cryptosporidiosis have been reported in calves, lambs, pigs and others including avian species. Cryptosporidiosis is transmitted by oocysts of Cryptosporidium species. Because cryptosporidiosis is common infection among animals, early literature considered it a zoonosis. Human infections contracted from calves, cats, and horse feces. However, the resrvoir host is longer considered the major source of infection. Mild cases of disease have been reported in farm workers. Immunosuppressed, very young and very old persons should avoid contact with this parasite because it may cause severe diarrhea. In order to detect of Crytosporidium sp infection from feces of cattle and pigs at Chonbuk Iksan and Kunsan area, sedimentation and modified acid fast stain were applied. The positive rate of Cryptosporidium sp infection from 1, 176 of cattle and 267 of pigs were 0.5 % and 16.8%, respectively. According to area in Iksan and Kunsan, the positive rates were 0.4% and 0.9% from cattle, and 18.9% and 12.1% from pigs, respectively. In cattle, positive detection rate was 0.6% in milking cows but not in Korean cattle.

  • In order to survey the prevalence of intestinal parasites in dogs, 304 fecal samples were taken from dogs in Taejon city, The prevalence and identification of intestinal parasites were determined by the fecal examinations using sheather's floating technique and sedimentation methods and then Cryptosporidium oocysts were identified by kinyoun's modified acid fast stainning method. The results were obtained as follows ; 1. Parasite eggs and oocysts were detected in 105 samples (34.5%) from 304 cases of dog feces. 2. The 11 kinds of eggs and oocysts were isolated from the feces of dog. Those were Ancylostoma caninum (12.1%, 37 dogs), Trichuris vulpis (11.5%, 35 dogs), Toxocara canis (10.2%, 31 dogs), Isospora sp (7.2%, 22 dogs), Cryptosporidium sp (3.6%, 11 dogs), Toxascaris leonine (1.9%, 6 dogs), Strongyloides sp (1.9%, 6 dogs), Taenia sp (0.6%, 2 dogs), Diphylidium caninum (0.3%, 1 dog), Spirometra sp (0.3%, 1 dog) and Clonorchis sinensis (0.3%, 1 dog). 3. It was mixed infection such as single, double, triple and quadruple, 64.8%, 25.7%, 8.6% and 0.9%, respectively. 4. In indiviually-raised 4095, the infectious late of T canis (11.4%), A Caninum(13.2%), Cryptosporidium sp (6.1%), T leontna (2.6%) were higher than those of group raised dogs. But the infectious rate of T vulpis (12.1%) in group raised dogs was higher than that of individually-raised dogs. 5. Adults of Demodex and Sarcoptes which have been found in this survey are excluded in this report.

  • Cryptosporidium parvum oocysts were isolated from a child suffering from acute gastroenteritis and successfully passaged in a calf and mice (designated hereafter SNU-H1) in the Republic of Korea; its molecular genotype has been analyzed. The GAG microsatellite region was amplified by a polymerase chain reaction (PCR), with a 238 base pair product, which is commonly displayed in C. parvum. The isolate was shown to be a mixture of the genotypes 1 (anthroponotic) and 2 (zoonotic). To study its infectivity in animals, 2 calves and 3 strains of mice were infected with the SNU-H1; in these animals, the propagation of both genotypes was successful. In immunosup-pressed (ImSP) BALB/c and C57BU6 mice the number of oocysts decreased after day 10 post-infection (PI); but in ImSP ICR mice, they remained constant until day 27 PI. The results show that both the C. parvum genotypes 1 and 2 can be propagated in calves and ImSP mice.

  • This study investigated the effect of breast-feeding in protection against protozoan infection in infants with persistent diarrhea. Infants were classified into 2 groups; 161 breast-fed infants and the same number of non-breast-fed infants. Microscopic examinations of stool were done for detection of parasites and measuring the intensity of infection. Moreover, serum levels of IgE and TNF-${\alpha}$ were measured by ELISA. Cryptosporidium spp., Entamoeba histolytica/Entamoeba dispar, Giardia lamblia, and Blastocystis sp. were demonstrated in infants with persistent diarrhea. The percentage of protozoan infections was significantly lower in breast-fed infants than that in the non-breast-fed infants. The levels of IgE and TNF-${\alpha}$ were significantly lower in the breast-fed group than in the non-breast-fed group. There were significant positive associations between the serum levels of IgE and TNF-${\alpha}$ and the intensity of parasite infection in the breast-fed group. It is suggested that breast-feeding has an attenuating effect on the rate and intensity of parasite infection.

  • In most developing countries, Dientamoeba fragilis infection is an obscure protozoan infection. We aimed to determine a frequency and clinical importance of D. fragilis infection in Taif, Saudi Arabia. A 1-year case control study included patients with gastrointestinal (cases, n=114) or non-gastrointestinal symptoms (controls, n=90). The fecal samples were examined with the classical parasitological methods for intestinal protozoa, and by real time PCR for D. fragilis. The infection by D. fragilis was detected in 5.8% by PCR and in 4.4% patients by microscopy. The infection was identified more in control group (n=9) than in cases (n=3); a sole infection in 11 patients and mixed with Giardia in 1 patient. The other enteric parasites detected were Blastocystis sp. (8.3%), Giardia sp. (5.3%), Cryptosporidium sp. (2.9%), Entamoeba histolytica (1.4%), Entamoeba coli (0.9%) and Hymenolepis nana (0.4%). Our results tend to reinforce the need to increase awareness of D. fragilis infection in Saudi Arabia.

  • An epidemiological survey was performed to know the status of Cwptespori, mum sp. infection among the people in Seoul and Chollanam-do in 1992. One village of Chollanam-do (Hwasun-gun) which showed the highest oocyst positive rate was re-surveyed in 1995 for human infection and for cattle also. The subjected areas consisted of 8 urban villages (: dongs) of Seoul and 4 urban (: dongs) and 7 rural (: myonsl villages of Chollanam-do. A total of 3,146 fecal samples was collected randomly, and smears were made from formalin-ether sediments. They were examined for Cwptosporinium oocysts by modified acid fast staining. The overall oocyst positive rate was 79% (248/3,146), but the rate was remarkably different between Seoul and Chollanam-do, 0.5% (4/853) and 10.6% (244/2,293), respectively The average size of oocysts was 4.8 ± 0.5 by 4.2 ± 0.5 ㎛, compatible with C. pcnlum. In Chollanam-do, rural villages showed significantly higher rate (14.0%) than urban villages (3.7%). Especially the people in Iyang-myon, Hwasun- gun, a typical rural village, revealed a very high rate of 40.0% (74/185). Adults aged 51-70 years revealed the highest positive rate among all age groups. At the re-survey of the same village of Hwasun-fun in 1995, 44 (35.2%) of 125 villagers and 14 (93.3%) of IS cattle examined were positive for C. pnnpum oocysts. The results suggest that C. pnnpum is highly prevalent in rural areas of Chollanam-do, and an important source or mode of infection seems to be contaminated water or contact with the feces of infected cattle.
