To examine the new practical utilization of mucilages in Opuntia humifusa, the Korean pear cactus, the polysaccharides were isolated from O. humifusa, and immuno-stimulating activities were assayed. The main polysaccharide, CNC-E, was prepared by a commercial enzyme treatment, water extraction, and ethanol precipitation. The molecular mass of CNC-E was estimated to be about 700 kDa, and it consisted mainly of arabinose, galactose and xylose in addition to two minor sugars such as rhamnose and fucose. On the other hand, CNC-E showed considerably high splenocyte proliferation activity in a dose-dependent manner. Peritoneal macrophages stimulated with CNC-E produced cytokines such as IL-6, IL-12, IL-10, and TNF-${\alpha}$. The intravenous administration of CNC-E significantly augmented the cytotoxicity of natural killer (NK) cells against Yac-1 tumor cells. Especially, NK cells obtained from the mice treated with $100{\mu}g$ of CNC-E showed threefold higher cytolytic activity than those of untreated mice. CNC-E also showed potent anti-complementary activity in a dose-dependent fashion. Identification of C3 activation products by the crossed immunoelectrophoresis using anti-human C3 and the anti-complementary activity of CNC-E in a $Ca^{2+}$-free condition suggested complement activations by CNC-E that occur via both alternative and classical pathways. These results indicate that Korean pear cactus contains selected polysaccharides that provide immuno-stimulating activities beneficial to human health.

Heme oxygenase-1 (HO-1) degrades heme to carbon dioxide, biliverdin, and $Fe^{2+}$, which play important roles in various biochemical processes. In this study, we examined the protective function of HO-1 against oxidative stress in SH-SY5Y cells and in a Parkinson's disease mouse model. Western blot and fluorescence microscopy analysis demonstrated that PEP-1-HO-1, fused with a PEP-1 peptide can cross the cellular membranes of human neuroblastoma SH-SY5Y cells. In addition, the transduced PEP-1-HO-1 inhibited generation of reactive oxygen species (ROS) and cell death caused by 1-methyl-4-phenylpyridinium ion ($MPP^+$). In contrast, HO-1, which has no ability to transduce into SH-SY5Y cells, failed to reduce $MPP^+$-induced cellular toxicity and ROS production. Furthermore, intraperitoneal injected PEP-1-HO-1 crossed the blood-brain barrier in mouse brains. In a PD mouse model, PEP-1-HO-1 significantly protected against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced toxicity and dopaminergic neuronal death. Therefore, PEP-1-HO-1 could be a useful agent in treating oxidative stress induced ailments including PD.

Agaricus bitorquis is an edible white mushroom of the genus that is cultivated at high temperature($25{\pm}1^{\circ}C$) unlike A. bisporus is cultivate at $16{\pm}2^{\circ}C$. Unlike Agaricus bisporus, an edible white A. bitorquis mushroom is cultivated at high temperature ($25{\pm}1^{\circ}C$). Most farmers cultivate this mushroom for a long cultivation period in Korea. For this reason, we made heterokayons to develop a new cultivar that generate fruitbodies for short cultivation period. Over one hundred SSIs(single spores isolates) were collected from selected A. bitorquis ASI1151 and ASI1349 strains. Seventy-three SSIs were germinated on CDA(compost dextrose agar) media after 20 days (minimum) or 83 days (maximum) incubation under different media condition. The mycelial growth rate of germinated SSIs was different. 9 homokaryons in ASI 1151 and 11 homokaryons in ASI 1349 from SSIs were selected by OPN-02 primer in RAPD analaysis. Also this primer was used to select heterokaryon that cross among each homokaryon with compatible locus. Therefore 44 compatible matings were confirmed of 99 crossed lines.

This study describes conditions for plant regeneration from protoplasts-derived from embryogenic callus of satsuma mandarin. Plants were generated via somatic embryogenesis. Protoplasts isolated directly from nucellar callus induced from immature ovule of satsuma mandarin cv. Okitsu (Citrus unshiu Marc.) were cultured in 0.6M $BH_3$ medium. Cell division and plating efficiency were affected by protoplast culture method. The liquid over solid method was the most effective for formation of microcalli. Most of microcalli grew rapidly and transferred onto embryoid formation medium. Optimum embryoid formation medium was MT medium containing 1.5 g/L malt extract, 0.146 M sucrose and the medium for plantlet regeneration was MS medium containing 0.09M sucrose, 1.0 mg/L $GA_3$. No differences were noticed in growth habits and leaf characters such as shape, thickness, and colour between protoplast-derived plants and nucellar seedlings. This plant regeneration system from protoplasts-derived from embryogenic callus provides an alternative way for producing new scion and rootstock cultivar from citrus species which can not be crossed.
During monocarpic senescence in higher plants, functional stay-green delays leaf yellowing, maintaining photosynthetic competence, whereas nonfunctional stay-green retains leaf greenness without sustaining photosynthetic activity. Thus, functional stay-green is considered a beneficial trait that can increase grain yield in cereal crops. A stay-green japonica rice 'SNU-SG1' had a good seed-setting rate and grain yield, indicating the presence of a functional stay-green genotype. SNU-SG1 was crossed with two regular cultivars to determine the inheritance mode and identify major QTLs conferring stay-green in SNU-SG1. For QTL analysis, linkage maps with 100 and 116 DNA marker loci were constructed using selective genotyping with $F_2$ and RIL (recombinant inbred line) populations, respectively. Molecular marker-based QTL analyses with both populations revealed that the functional stay-green phenotype of SNU-SG1 is regulated by several major QTLs accounting for a large portion of the genetic variation. Three main-effect QTLs located on chromosomes 7 and 9 were detected in both populations and a number of epistatic-effect QTLs were also found. The amount of variation explained by several digenic interactions was larger than that explained by main-effect QTLs. Two main-effect QTLs on chromosome 9 can be considered the target loci that most influence the functional stay-green in SNU-SG1. The functional stay-green QTLs may help develop low-input high-yielding rice cultivars by QTL-marker-assisted breeding with SNU-SG1.
An experiment was conducted to examine the effect of a high environmental temperature on the performance, conformation, and carcass characteristics of pigs and the influence of diet. Thirty-six three-way crossed castrated male pigs with average initial body weight of 50.4 kg were used in the experiment. The pig were allocated to the following treatments: two environmental temperatures (20 and $30^{\circ}C$)${\times}$three dietary energy levels (2,980, 3,300 and 3,600 kcal/kg)${\times}$three protein levels (12.8, 15.2 and 17.2%). Daily weight gain was lighter (p<0.01) and feed: gain ratio lower (p<0.05) in pigs at $30^{\circ}C$ than for pigs at $20^{\circ}C$ The pigs at $30^{\circ}C$ were significantly taller with deeper bodies (p<0.05) and significantly longer (p<0.05) both vertically horizontally in the planum nasal when kept at $30^{\circ}C$. The width of body and the circumference of the neck were greater (p<0.05) at $20^{\circ}C$. The lean meat of the loin, middle section, ham, and hind section were significantly greater (p<0.05) in pigs kept at $30^{\circ}C$ and the belly was significantly heavier. The total unsaturated free fatty acids were significantly higher (p<0.05) in the body fat of pigs kept at $20^{\circ}C$ than in that of pigs at $30^{\circ}C$. The results indicated that when pigs are given very restricted same amounts of feed, they may need less energy to maintain their body temperature under moderately high environmental temperature ($30^{\circ}C$); consequently, their performance is better than that of pigs under optimum environmental temperature. The results showed very clearly that weight gain of pigs increased with increasing of dietary protein and energy content. The increasing of dietary protein content seemed more significant when content increasing to 17.2% compared with the 12.8 and 15.2% protein content treatments. The increasing of dietary energy content was more significant when content increasing to 3,600 compared with the other low energy content treatments.
Fresh elephant grass was replaced by urea treated rice straw (UTRS) to evaluate the effects on milk production of crossed lactating cows. A total of 16 crossbred F1 cows (Holstein Friesian ${\times}$ Vietnamese Local Yellow), with a body weight of about 400 kg and lactation number from three to five, were used in the experiment. The experimental cows were blocked according to the milk yield of the previous eight weeks and divided into 4 homogenous groups. The experiment was conducted with a Latin Square design with 4 treatments and 4 periods. Each period was 4 weeks, with 2 weeks of feed adaptation and 2 weeks for data collection. The ratio of concentrate to roughage in the ration was 50:50. All cows were given constant amounts of elephant grass dry matter (DM), with ratios of 100% grass without UTRS (control treatment 100G), and 75% grass (75G), 50% grass (50G) and 25% grass (25G) with ad libitum UTRS. Daily total DM intake on 100G, 75G, 50G and 25G was 12.04, 12.31, 12.32 and 11.85 kg, and the daily ME intake was 121.6, 121.5, 119.4 and 114.3 MJ, respectively. The daily CP intake was similar for all treatments (1.85-1.91 kg). There was a difference (p<0.05) in daily milk yield between the 25G and the 100G and 75G (11.7 vs. 12.6 and 12.5 kg, respectively). Milk protein concentration was similar for all treatments, while a tendency to increased milk fat concentration following the increase of UTRS ratio was observed. The cows gained 4-5 kg body weight per month and showed first oestrus 3-4 months after calving. The overall feed conversion for milk production was not affected by ratio of UTRS in the ration. It is concluded that replacement of green grass by UTRS with a ratio of 50:50 for crossbred lactating cows is as good as feeding 100% green grass in terms of milk yield, body weight gain and feed conversion. UTRS can preferably replace green grass in daily rations for crossbred dairy cows in winter to cope with the shortage of green grass, with the ratio 1:1.

In this paper, an optical touch panel device is newly proposed, with operating principle based on the optical coupling between a pointing pen having a built-in light source and perpendicularly crossed optical waveguide arrays. In order to enable an external light to couple into a waveguide core, the auxiliary pyramidal structures are introduced into all intersecting points located periodically along optical waveguides. The shape is optimized for minimizing the unwanted propagation loss due to the same structure by a ray tracing method. For the optical waveguide with the size of $50{\times}50{\mu}m^2$, the bottom width, height, and slope angle of the optimized pyramidal structure are $50{\mu}m$, $22.5{\mu}m$, and $42^{\circ}$, respectively. The optical coupling efficiency of about 97.8% and the average propagation loss of 0.3 dB/mm were achieved for the optimized touch panel. Finally, it is found from the tolerance analysis that tilting of the pen up to ${\pm}12^{\circ}$ can be allowed.
This study was conducted to see the feasibility of breeding for sprout soybean cultivar with minimum benny flavor using lipoxygenase-less lines. Lipoxygenase-less cultivar Jinpumkong2 was crossed by lipoxygenase containing Gwangankong, Sebaeknamulkong, and Pureunkong as paternal parent and 24 lipoxygenase-less lines derived from those 3 combinations were selected and those lines were evaluated with their parental cultivars. Germination rate showed no difference between lipoxygenase-less lines and their parental cultivars, however, rates of normal sprout of those and Jinpumkong2 were 63 and $56\%$, and were lower than that of paternal parents. Hypocotyl length of those was same as Jinpumkong2, however, shorter than paternal parents. Texture characteristics including hardness, cutting force and mastication of 96 hour-cultured sprout of lipoxygenase-less lines showed higher value than that of their parental cultivars. Lipoxtgenase isozyme was not detected in the sprout cotyledon of lipoxygenase-less lines, however it was observed in the sprout hypocotyl of all the used genotypes. Though lipoxygenase activity in the seed of lipoxygenase-less lines was lower than that of Jinpumkong2(0.477, ${Delta}A$ 234 nm min-1 mg meal-1),2 lines revealed more than 0.5 value. Lipoxygenase-activity of 2 day-cultured sprout(both cotyledon and hypocotyl) was the highest, decreased in 3 days after culture and re-increased thereafter. Several lipoxygenase-less lines with lower lipoxygenase activity of sprout than Jinpumkong2 were selected and this suggested the possibility of breeding lines for soy-sprout with low benny flavor.
This experiment was carried out to obtain the information on the crossability, variation of chromosome number in pollen mother cell (PMC) and somatic cell of the progeny from the cross between hexaploid triticale cv. Sinkihomil and two diploid rye varieties. Seed set was 39.3 to 41.6% (averaged 40.5%) in the cross between triticale (P$_1$) and rye(P$_2$), which resulted in 0.33% in F$_2$(selfed F$_1$), 2.69% in F$_1$/P$_1$ 5.47% in F$_1$/P$_2$ respectively. However, seed set was extremely low in both reciprocal crosses when triticale was used as male. Germination rate of the crossed seed was 94.0% in F$_1$ 40.8% in F$_2$(selfed F$_1$), 59.5% in F$_1$/P$_1$ and 65.9% in F$_1$/P$_2$ from the cross between triticale and rye, respectively. Pollen fertility of F$_1$ plant was averaged 18.7% in the cross between triticale and rye. Number of Uni-, Bi-, and Trivalent in PMC was 12. 6, 6.94, and 0.53, respectively, in the F$_1$ between the triticale and rye. There were 28 chromosomes in F$_1$, 21 to 34 in F$_2$, 34 to 38 in F$_1$/P$_1$ and 19 to 23 in F$_1$/P$_2$ from the cross between the triticale and rye, respectively.
