Recently, research investment in the improvement of food safety as a food source and specializing of nutritional source of edible insects is being actively conducted. Cricket especially has been attracting considerable interest in entomophagy; however, research on the safety assessment of cricket is limited. This study investigated the effects of cricket ethanol extract when orally administrated in Sprague-Dawley rats. Here, we performed a 4 wk repeated oral dose toxicity test in Sprague-Dawley rats following the Organization for Economic Cooperation and Development test guidelines 407 under Good Laboratory Practice regulation. Rats were randomly allocated 4 groups: vehicle control, 250, 500, 1,000 mg/kg test groups and administrated based on body weight for 28 d. The animals were observed for mortalities and clinical signs, body weight changes, food and water consumption. At the end of treatment period, blood and urine were collected and analyzed. Subsequently, the animals were sacrificed and subjected to gross pathological examination and organ weight measurement. The organs were preserved for histopathological examination. The results showed that there were no systemic toxicological effects related with the cricket ethanol extract in the 4 wk oral repeated dose toxicity study. It is considered that NOAEL of cricket ethanol extract is greater than 1,000 mg/kg/d and there was no target organ detected.
A cDNA encoding the protein lethal (2) essential for life [l(2)efl] was cloned from Gryllus bimaculatus and named GBl(2)efl. This protein is composed of 189 amino acids, including an N-glycosylation site and 15 phosphorylation sites. Its predicted molecular mass is 21.19 kDa, with a theoretical isoelectric point of 6.2. The secondary structure of GBl(2)efl was predicted from the identification of random coils (56.08%), alpha helices (22.22%), extended strands (17.99%), and beta turns (3.7%) through sequence analyses. A homology analysis revealed that GBl(2)efl exhibited a high similarity with other species at the amino acid level, ranging from 52% to 69%. While GBl(2)efl mRNA expression was higher in the dorsal longitudinal flight muscle following a three-day starvation and in the Malpighian tubules following a one-day starvation, no changes in expression were detected in other tissues. Furthermore, tunicamycin-induced endoplasmic reticulum (ER) stress resulted in an approximately 1.8-fold higher expression in the fat body compared with the wild type.

We describe the isolation and characterization of six different intestinal microorganisms from the digestive tract of the cricket Gryllus bimaculatus. Based on 16S rRNA gene sequences, we obtained six isolates belonging to four different genera: Staphylococcus, Bacillus, Citrobacter, and Proteus. All the isolates were resistant to ampicillin. Ampicillin is an irreversible inhibitor of the enzymeetranspeptidase, which is needed to make bacterial cell walls. None of the isolates were resistant to kanamycin, which binds to the 30S subunit of the bacterial ribosome and then inhibits total protein synthesis. Gram staining was conducted, in addition to morphological classification under a microscope. Four grampositive isolates and two gram-negative isolates were detected. The gram-positive isolates were GL1 (round shaped, 2 am in diameter), GL2 (rod shaped, $2.5{\mu}m$ in length), GL3 (rod shaped, $2{\mu}m$ in length), and GL4 (round shaped, $1.5{\mu}m$ in diameter). The gram-negative isolates were GL5 (rod shaped, $2{\mu}m$ in length) and GL6 (rod-shaped, $2.5{\mu}m$ in length). Notably, two of the isolates, GL2 and GL4, secreted specific extracellular proteins. These were determined by MALDI-TOF-MS spectral analysis to be a 87 kDa collagenase, 56 kDa hypothetical protein, and 200 kDa hypothetical protein. The six isolates in this study could be used for various biotechnological applications and pest management, both in the field and in greenhouse systems. In addition, it would be interesting to determine the relationship between these isolates and their host.
Objective: Replacing soybean meal (SBM) with cricket (Gryllus bimaculatus) meal pellets (CMP) in concentrate diets was investigated for feed efficiency, ruminal fermentation and microbial protein synthesis in Thai native beef cattle. Methods: Four male beef cattle were randomly assigned to treatments using a 4×4 Latin square design with four levels of SBM replaced by CMP at 0%, 33%, 67%, and 100% in concentrate diets. Results: Results revealed that replacement of SBM with CMP did not affect dry matter (DM) consumption, while digestibilities of crude protein, acid detergent fiber and neutral detergent fiber were significantly enhanced (p<0.05) but did not alter digestibility of DM and organic matter. Increasing levels of CMP up to 100% in concentrate diets increased ruminal ammoniacal nitrogen (NH3-N) concentrations, blood urea nitrogen, total volatile fatty acids and propionate concentration (p<0.05), whereas production of methane and protozoal populations decreased (p<0.05). Efficiency of microbial nitrogen protein synthesis increased when SBM was replaced with CMP. Conclusion: Substitution of SBM with CMP in the feed concentrate mixture at up to 100% resulted in enhanced nutrient digestibility and rumen fermentation efficiency, with increased volatile fatty acids production, especially propionate and microbial protein synthesis, while decreasing protozoal populations and mitigating rumen methane production in Thai native beef cattle fed a rice straw-based diet.
Platelets act a fundamental role in primary- and secondary-hemostasis, however, platelet activation may cause thrombosis simultaneously. Therefore, control of platelet aggregation is crucial in preventing thrombosis-mediated diseases. Recently, the development of insect materials is attracting attention. Among the highly nutritious functional food sources, insects such as two-spotted cricket (Gryllus bimaculatus). Gryllus bimaculatus (G. bimaculatus) contains high protein and unsaturated fatty acids and has been registered as a food material September 2015 by the Ministry of Food and Drug Safety of Korea. In this study, we examined whether G. bimaculatus extract (GBE) inhibits platelet aggregation, intracellular calcium mobilization, thromboxane A2 production and glycoprotein IIb/IIIa (integrin αIIb/β3) activation. We investigated whether GBE can regulate signaling molecules, such as 1, 4, 5-triphosphate receptor type I, extracellular signal-regulated kinase, cytosolic phospholipase A2, mitogen-activated protein kinases p38, vasodilator-stimulated phosphoprotein, phosphatidylinositol-3 kinase, Akt, glycogen synthase kinase-3α/β, and SYK. Taken together, GBE is a potential therapeutic drug candidate to prevent platelet-related thrombosis and cardiovascular disease.
In 2016, the two-spotted cricket was approved as a general food ingredient by the Korean Ministry of Food and Drug Safety. Additionally, the two-spotted crickets have high protein content (60%) and can be reared throughout the year even during the overwintering period. In this study, storage conditions were set in case cricket breeding was not possible due to problems such as breeding space and labor costs, and selection oviposition mats to determine high fecundity rates and low cannibalism rates for the eggs were investigated. The oviposition mat was mostly composed of soil (p < 0.05). Less than 62 crickets per 10 litters were found to be best suited for 1 day of spawning, based on the cannibalism rate of the eggs during the egg laying period. The results from the hatched eggs indicated that the hatching extended 12 to 14 days, and the best hatching rate was approximately 85% when stored at low temperatures(16℃) for 10 days. An efficient production proposed method that established the best oviposition mat and egg storage method for the cricket.

This study aimed to determine a feed withdrawal duration time of Gryllus bimaculatus (Gb), for removal of intestinal feces. Finally, this species of cricket has been temporarily approved for use as a human food source in Korea. To commercialize Gb as an edible insect, we attempted to prevent the proliferation of micro-organisms and improve taste of Gb by removing intestinal feces of live crickets. Our study showed that weak crickets were damaged in 2 days after feed withdrawal in farm. The microscope findings also revealed that feces of strong crickets' intestine almost disappeared at 1 day after feed withdrawal and instead, some of arms and legs of weak crickets were observed in the former's intestine. The results suggest that 1 day is deemed to be an adequate time for feed withdrawal, when it usually takes 1 day to deliver live crickets from the farm to a destination.

Edible insects, such as the two-spotted cricket (Gryllus bimaculatus, GB), have high nutritional value but are not widely consumed because of their appearance and smell. Consequently, the development of foods containing these insects in less recognizable forms, e.g., flour-like powders, has drawn considerable attention. Herein, we investigated the quality characteristics of muffins prepared from wheat flower supplemented with fresh ginseng (5%) and GB (0, 10, 20, and 40%) powders. GB loading was negatively correlated with muffin volume, height, moisture content, and textural properties (hardness, springiness, cohesiveness, and chewiness) and positively correlated with crude protein content and antioxidant properties. Significant (p<0.05) color differences were observed between samples with different GB loadings. The contents of hexanal and nonanal, which are the major volatiles responsible for off-flavor, increased with increasing GB loading, and the number of volatiles maximized at 40% GB. Sensory preference decreased in the order of 0% GB>10% GB≈ 20% GB>40% GB. Based on these results, a GB loading of 20% offered the best trade-off between attractiveness and nutritional value. Thus, this study promotes the widespread use of GB in the food industry and the development of various edible-insect-based food products.
We evaluated the protective effects of cricket methanol extract (CME) on ultra-violet B (UVB)-induced photoaging in human skin fibroblasts. The fibroblast cells were treated with 10, 50, and 100 ㎍/mL of CME for 24 h, and then exposed to UVB (30 mJ/㎠). CME showed a dose-dependent cytoprotective effect without any observable cytotoxicity. CME reduced UVB-induced production of reactive oxygen species (ROS) by 34.4, 34.9, 40.6% at concentrations of 10, 50, 100 ㎍/mL respectively. CME inhibited the release of matrix metalloproteinase (MMP) 1 and 3. Furthermore, CME also reduced UVB-induced collagen degradation in the fibroblast cells. Taken together, our data suggests that CME has a significant protective effect on UVB-induced photoaging of the skin. This benefit occurs through multiple mechanisms. The results also suggest a potential role for CME as an ingredient in anti-photoaging cosmetic products in the future.
