Progesterone assays as an aid for improving reproductive efficiency in dairy cattle I. Use of milk progesterone profiles in the confirmation of estrus detection and early pregnancy diagnosis (Progesterone 농도측정(濃度測定)에 의한 유우(乳牛)의 번식효율증진(繁殖效率增進)에 관한 연구(硏究) I. 유즙(乳汁)중 progesterone 농도측정(濃度測定)에 의한 발정확인(發情確認) 및 조기임신진단(早期姙娠診斷))

  • Milk progesterone concentrations were measured in 111 dairy cows for confirming the estrus observation and for the early pregnancy diagnosis. Of the 56 cows inseminated, 52 cows(92.8%) were an ovulatory estrus, 2 cows(3.6%) were an unovulatory estrus, and 2 cows(3.6%) were the error of estrus observation, respectively. Milk progesterone concentrations at 21 and 24 days after artificial insemination were significantly higher in 23 pregnant cows compared with those in 5 non-pregnant cows(P<0.05). The accuracy rate for early pregnancy diagnosis in 27 cows achieved when the discriminatory concentration at 21 days after artificial insemination was placed at 2.0 ng/ml skim milk, was 91.3% for positive diagnosis and 100% for negative diagnosis, respectively. These results indicated that milk progesterone determination at 0, 6 and 21 days after artificial insemination can be utilized for confirming the estrus observation and for early pregnancy diagnosis. In conclusion, milk progesterone determination is useful diagnostic tool for monitoring the reproductive performance.

Effect of Prostaglandin $F_2\alpha$ on Infertility Cows (Prostaglandin $F_2\alpha$에 의한 불임우 치료)

  • Estrus was induced and the therapeutic effect was examined with 25mg of PGF2$\alpha$ intramuscular injection and 3mg of PGF2$\alpha$ intraovarian injection to Holstein and Simmental cows which were raised in the large size stockfarms and the dairy farms, and were diagnosed to anestrus. The results obtained were summarized as follows: 1. The estrus inducing effect observed in the cows treated by PGF2$\alpha$ was 83.3% with 25/30 heads in the intramuscular injection group, and 86.7% with 26/30 heads in the intraovarian injection group. 2. Average duration from PGF2$\alpha$ administration to the onest of estrus was 2.7 day in the intramuscular injection group, and 2.6 days in the intraovarian injection group. 3. In status of estrus in the cows treated by PGF2$\alpha$, vigorous estrus was 12.0%, normal estrus 60.0% and silent estrus was 28.0% in the intramuscular injection group, and vigorous estrus was 15.4%, normal estrus 61.5%, and silent estrus 23.1% in the intraovarian injection group. 4. Conception rate in the cows induced to estrus was 64.0% in the intramuscular injection group and 65.4% in the intraovarian injection group.

Relationship between the Conception Rate after Estrus Induction using CIDR and Other Parameters in Dairy Cows (젖소에서 CIDR 투여에 의한 발정 유도 후 수태율과 다른 인자와의 관계)

  • The purpose of this study was to determine the relationship between conception rate and other parameters (body condition score; BCS, progesterone concentrations and follicle size) before estrus induction with CIDR(intravaginal progesterone-releasing controlled internal drug release). The conception rate in cows with < 2.75, 2.75 to 3.25, and 3.25 <, BCS regardless of AI (artificial insemination) time was 46.6%, 63.3%, and 46.6% at CIDR insertion, respectively. The conception rate regardless of BCS was 54.9% in cows inseminated based on detected estrus, and 48.7% in cows inseminated at 72 to 80 hours (timed artificial insemination, TAI) after removal of CIDR. The conception rate regardless of AI time was 40.0% in cows with low progesterone concentrations (less than 1.0 ng/ml), and 56.6% in cows with high progesterone concentrations (more than 1.0 ng/ml) at CIDR injection. The conception rate regardless of progesterone concentrations was 53.8% in cows inseminated based on detected estrus, and 38.0% in cows of TAI after removal of CIDR. The conception rate regardless of AI time was 43.3% in cows with small follicle (less than 5 mm), 53.3% in cows between 5 mm to 10 mm of follicle, and 63.3% in cows with large folliclc (more than 10 mm) at CIDR injection, respectively. The conception rate regardless of follicle size was 58.4% in cows inseminated based on detected estrus, and 45.9% in cows of TAI after removal of CIDR. These results indicated that if the cows with BCS 2.75 to 3.25, active corpus luteum, and/or large dominant follicle (more than 10 mm) are used for estrus induction, the conception rate will be greater.

Mounting Activity Detection of Cows by Radiotelemetry (무선원격측정에 의한 소의 승가행위 검출)

  • To increase the production efficiency in dairy industry, proper artificial insemination is the most important operation. For the successful artificial insemination, accurate estrus detection is required. The Korean dairy farmers usually depended on visual observation for estrus detection of cows. Mounting behaviour is one of major inidications observed when cows are at an estrus. A mounting activity detection system for cows using radiotelemetry was developed. This system included a transmitter with a pressure switch, a receiver, a serial communication interface, a personal computer and a computer software. All components and a whole system were tested both in a laboratory and in a farm. The results of this study are as follows: 1. All components including transmitter, receiver and serial interface were operated according to the design specification. 2. A whole system tested with simulated mounting activity of 400 times showed 100% of success rate. 3. In the farm test for 4 days with three cows expecting estrus, one cows experiencing mounting activities showed correct response with this system. However two cows did not show mounting activities because of weak estrus and cold weather during the testing period.

Development of Differential Diagnosis and Treatment Method of Reproductive Disorders Using Ultrasonography in Cows IV. Confirmation of Estrus Detection and Early Pregnancy Diagnosis (초음파검사에 의한 소의 번식장애 감별진단 및 치료법 개발 IV, 발정확인 및 조기 임신진단)

  • Plasma progesterone (P$_4$) concentrations were measured for confirming the estrus observation and for the early pregnancy diagnosis in 130 cows of small farmers. Ultrasonographic examinations were performed from day 30 after artificial insemination to establish the characteristic ultrasonographic appearances of gestational structures in each pregnant stages. Of the 130 cows inseminated, 111 cows (85.4%) were an ovulatory estrus, 12 cows (9.2%) were an unovulatory estrus, and 7 cows (5.4%) were the error of estrus detection, respectively. The accuracy for early pregnancy diagnosis in 111 ovulatory estrus cows achieved when the discriminatory concentration at day 21 after artificial insemination was placed at 3.0 ng-/ml in plasma, was 86.7 % for positive diagnosis and 100% for negative diagnosis, respectively. Pregnancy diagnosis by ultrasonography were performed to evaluate gestational structures from day 30 after artificial insemination in 83 cows. Pregnant cows were 72 of 83 cows. The characteristic ultrasonography of gestational structures in each gestational stages was as follows. The embryo proper was observed within anechoic fetal fluid between 28 and 40 days after insemination, and amnion and embryonic heartbeat was also detected in this period. Between days 41 and 50, embryo proper was detected as an discriminated from head and body, and forelimb buds and hindlimb buds were also observed in this period. Between days 51 and 60, an embryo proper was clearly discriminated from head and body, and fetal movement, forelimb buds and hindlimb buds were observed in this period. Between days 61 and 70, fetus was completely developed, and fetal skeleton, organs and cotyledon were observed. After day 71, each organs of fetus were rapidly developed and a fetus was partially observed in screen because fetus was too big and larger, These results indicate that plasma P$_4$ determination at days 0,6 and 21 after artificial insemination can be utilized for confirming the estrus observation and for early pregnancy diagnosis. Also, ultrasonography was reliable method for early pregnancy diagnosis at day 30 after artificial insemination.

Programmed Reproductive Management Including Presynch-Ovsynch-Resynch Protocol in Dairy Cows (젖소에서 일련의 Presynch-Ovsynch-Resynch Protocol을 이용한 번식관리 예)

  • This study was to evaluate the usefulness of a programmed reproductive management including Presynch-Ovsynch-Resynch protocol in lactating dairy cows. Nine hundred and thirty four cows calved during February to May 2008 were used for this study. Cows received im injections of 25 mg $PGF_{2{\alpha}}$ at $45{\pm}3$ and $59{\pm}3$ days in milk (DIM). Fourteen days later, Ovsynch was initiated with an im injection of $100\;{\mu}g$ GnRH ($73{\pm}3$ DIM), 25 mg $PGF_{2{\alpha}}$ 7 days later ($80{\pm}3$ DIM), $100\;{\mu}g$ GnRH 56 h later and timed artificial insemination (TAI) 16 h after the GnRH injection. Cows showed estrus during the Presynch-Ovsynch, were inseminated artificially according to am-pm rule. Pregnancy was determined on 32 days after AI using ultrasonography. Cows diagnosed pregnant were re-examined for pregnancy by rectal palpation at 60 days after AI. Non-pregnant cows were resynchronized by receiving $100\;{\mu}g$ GnRH, 25 mg $PGF_{2{\alpha}}$ 7 days later, $100\;{\mu}g$ GnRH 56 h later and TAI 16 h after. Cows with estrus since the first AI before the completion of Resynch protocol were also inseminated artificially according to am-pm rule. Pregnancy was determined by the same manner as following the first AI. Fifty five percents of the cows treated showed estrus and received AI before completion of the Presynch-Ovsynch protocol, while 45% received TAI, regardless of estrus exhibition following the completion of the protocol. The pregnancy rate following the first AI was higher in cows that showed estrus and received AI (43.3 and 38.5%) during the Presynch-Ovsynch than cows with TAI (34.6 and 29.6%) on 32 and 60 days after AI (P < 0.01). Sixty six percents of cows diagnosed non-pregnant following the first AI showed estrus before the completion of Resynch protocol and received AI, while 34% received TAI, regardless of estrus exhibition following the completion of the protocol. The pregnancy rate following the second AI was higher in cows that showed estrus before the completion of Resynch protocol and received AI (40.2 and 36.8%) than cows with TAI (21.2 and 18.2%) on 32 and 60 days after AI (P < 0.01). The cumulative pregnancy rates following the first and second AIs were 60.8 and 53.9% on 32 and 60 days after AI, respectively. These data suggest that inclusion of Presynch-Ovsynch-Resynch protocol and estrus detection into the programmed reproductive management might be an alternative option to control dairy cattle breeding.

Reproductive Performance according to Ovarian Status, Postpartum Uterine Treatment, and Calving Season in Estrus Synchronized Dairy Cows

  • A study on estrus synchronized dairy cows using progesterone intravaginal device was done to classify each cow's reproductive status from calving to synchronization and to evaluate the reproductive performance according to ovarian and uterine status, and calving season. From calving to estrus synchronization, silent heat or error of estrus detection among ovarian status and endometritis among uterine disorders were exposed in the most distribution (75.4% and 48.3%, respectively). The pregnancy rate of cows with inactive ovaries was lower than those in the follicular and luteal phase. And according to the uterine status before estrus synchronization, the pregnancy rate was similar in three groups; normal, endometritis, and pyometra (70.9, 69.1 and 100%, respectively). The interval from calving to conception was shorter (p<0.05) in cows calved during autumn than in cows calved during spring and winter.

Improvement of conception rate on Hanwoo; The key hormones and novel estrus detector

  • Two field experiments were conducted to improve the conception rate of Hanwoo cow. The first experiment aimed to investigate the physiological condition of Hanwoo cows on estrus, including metabolic profiles and body condition score (BCS). The second experiment investigated the effect of a novel estrus detector on the artificial insemination (AI) conception rate for Hanwoo cows. For the first experiment, 80 Hanwoo cows (2.5 ± 0.10 of parity), approximately one month before estrus, were housed in 16 pens and offered the experimental diets twice daily with free water access. The BCS were recorded, and blood was collected from the jugular veins just before AI. The collected blood was used to measure physiological conditions, such as metabolite and hormone levels. For the second experiment, each cow was equipped with a neck-mounted estrus detector collar, which had a sensor connected through the internet. Approximately one month before estrus, three hundred sixty Hanwoo cows (2.4 ± 0.21 of parity) were assigned into groups with or without W-Tag collar treatments. The animals were managed the same as in the first experiment. The pregnancy rate reached 55% in the first experiment. The concentration of luteinizing hormone (LH) was higher (p < 0.012; 1.56 vs. 1.08 ng/mL) in cows that were not pregnant (NPG) than in cows that were pregnant (PG) after AI. The BCS and other concentrations of metabolites and hormones in the blood were not different in both NPG and PG cows. The ranges of estrogen, LH, and follicle-stimulating hormone for PG cows were 11.9 to 39.0 pg/mL, < 0.25 to 1.98 ng/mL, and < 0.50 to 0.82 ng/mL, respectively. In the second experiment, cows with the estrus detector had lower days open (p < 0.001; 78.1 vs. 84.8 d), insemination frequency (p < 0.001; 1.26 vs. 2.52), and return of estrus (p < 0.001; 70.9 vs. 79.1 d) than those in cows without the estrus detector. In conclusion, the present study indicated that lower LH concentration just before AI potentially increased the pregnancy rate of Hanwoo cows. Furthermore, the application of estrus detectors to Hanwoo cows could improve the conception success rate for AI.

Effects of Days Open on the Subsequent Reproductive Performance Following to CIDR-Based Estrus Synchronization in Lactating Dairy Cows

  • The purpose of this study was to evaluate the effects of days open on subsequent reproductive performance following to estrus synchronization in the 114 lactating dairy cows. The animals were divided into two groups according to the time of estrus synchronization; viz, ${\leq}$ 85 days, and > 85 days postpartum, respectively. The estrus synchronization protocol consisted of insertion of a controlled internal drug release (CIDR) device containing 1.9 g progesterone with an injection of 250 ${\mu}g$ gonadorelin (Day 0), an injection of $PGF_2{\alpha}$ and removal of the device on Day 7, an injection of 250 ${\mu}g$ GnRH on Day 9, and TAI 17 h later. Pregnancy diagnosis was determined at 30 to 60 days after TAI using both ultrasonography and rectal palpation. The body condition score (BCS) gradually increased over the postpartum period. In estrus synchronized cows until 85 days, conception rate on first service, number of service per conception, interval from estrus synchronization to conception, and interval from calving to conception were not significantly different among two farms (P>0.05). In estrus synchronized cows after 85 days postpartum, conception rate on first service, number of service per conception and interval from calving to conception were significantly different ($P{\leq}0.05$) between herds A and B (26.8 vs 50.0%; $2.1{\pm}1.35$ vs $1.37{\pm}0.54$ times, $237.3{\pm}97.8$ vs $164.7{\pm}69.3$ days, respectively). In estrus synchronized cows after 85 days postpartum interval from estrus synchronization to conception was greater (P<0.01) in herd B than in herd A ($63.6{\pm}57.4$ vs $26.1{\pm}24.9$). These results indicate that the time of estrus synchronization for maximized the reproductive performance is before 85 days postpartum and feeding and management is important factor for high reproductive performance.

The Estrus Manifestation according to the Follicle Size and the Growth Phase after $PGF_{2{\alpha}}$ Injection in Korean Native Cows (한우에서 $PGF_{2{\alpha}}$ 투여 후 난포 크기 및 발달 과정에 따른 발정 증상의 발현)

  • This study was operated to establish induction using ultrasonography by estimating the relation of follicle size and estrus manifestation. Clinical estrus symptoms were observed 97.4% in cows and 87.5% in heifers when overall 55 cows were induced to estrus in a single dose of $PGF_{2{\alpha}}$ after verifying CL through ultrasonography, which means estrus hours among those 52 cows showing the clinical estrus symptoms were estimated 2.39 days on cows and for 2.37 days on heifers which showed no differences (p>0.05). The estrus manifestation hours according to the follicle size in cows didn't have any significance each other (p>0.05), though estrus hours was 54 hours (the shortest) with follicle size bigger than 10 mm and were made up within 69 hours. The estrus manifestation hours according to the follicle size in heifers didn't have any significance each other (p>0.05) and took around 42 hours (the shortest) with follicle size of 5mm (the smallest) and were made up within 66 hours. Follicles after $PGF_{2{\alpha}}$ injection were ovulated and assigned to many phases as follows; Group 1 (growing phase) - continuously growing into ovulation, Group 2 (growing and static phase) - delaying in growth after the growth of follicles, Group 3 (static and growing phase) - growing after growth delay, Group 4 (regressing and new growing phase) - the follicle is closed and a new follicle grows. In addition, the process of follicle development and estrus hours had no significance each other (p>0.05), though estrus manifestation hours in Group 1 and 2 was relatively short, and in Group 3 and 4 for a relatively long time. In the result of all above, the estrus manifestation hours after $PGF_{2{\alpha}}$ injection has no differences accoring to the follicle size in cows and heifers. Therefore, High pregnancy rate is obtained when practicing artificial insemination within 3 days in estrus or TAI in 72 to 80 hours after adminitrating $PGF_{2{\alpha}}$.
