  • In this study, we investigated the antioxidant effects and active ingredients of Gynura Procumbens extracts obtained through various extraction methods for the development of natural-based cosmetics and pharmaceutical materials. The contents of compounds, total flavonoids, chlorogenic acid, and caffeic acid were compared at different concentrations, revealing the highest content of active ingredients in the 100% ethanol extract. Antioxidant activity assessed by DPPH and ABTS radical scavenging assays showed a concentration-dependent increase in antioxidant activity with the ethanol concentration. Additionally, we validated the DNA damage inhibition and anti-inflammatory effects of Gynura Procumbens extracts through UVB irradiation on Hs68 cell models. The 100% ethanol extract demonstrated significant inhibition of the expression of p-p53, γ-H2AX, iNOS, and COX-2 induced by UVB, indicating its potential in alleviating photodamage effects. Consequently, the efficient extraction of Gynura Procumbens for skin functional material development was confirmed, suggesting the suitability of ethanol or alcohol-based solvents.

  • In this work, lipid nanovesicles (LNV) containing niacinamide were prepared by changing the concentration of lecithin, fatty acids (palmitic acid), and surfactant (polysorbate 60), and physicochemical characteristics analysis and skin permeability evaluation were performed. The present LNVs were measured with an average diameter of 77 ~ 160 nm, zeta potential -63 ~ -31 mV depending on the concentration of lecithin and polysorbate 60. As a result of observing the stability over time for 12 weeks, LNV1 showed much better colloidal stability than others. In addition, in vitro skin penetration using Franz cell was evaluated for LNV. In this work, niacinamide was selected as the model drug. As a result of skin penetration, it was confirmed that the skin penetration of fatty acid containing LNV1 was much superior than control. These results confirm that LNVs containing lecithin, fatty acids, and surfactants can support the skin absorption of active ingredients in cosmetics.

  • Properties of fucoidan used for functional cosmetic ingredients and the effect of fucoidan molecular weight on the cosmetic functions were studied. Fucoidan was extracted from Undaria pinnatifida sporophylls and molecular weight (35~160 kDa) of fucoidan was controlled by contact glow discharge electrolysis (CGDE). To test possibility of fucoidan as a cosmetics material, tyrosinase inhibition property, water-holding property, elastase activity inhibition property and DPPH free radical scavenging property were measured. Water-holding property of fucoidan was higher than that of hyaruronic acid, which is known as the one of the best water-holding material. The water-holding strength of fucoidan slightly increase as molecular weight of fucoidan decrease. Elastase activity inhibition (anti wrinkle effect) of fucoidan was higher than that of adenosine using standard material for anti wrinkle test. Optimum molecular weight of fucoidan to obtain highest tyrosin inhibition effect, elastase inhibition effect and radical scavenger effect is 100 kDa.

  • Chitin deacetylase (CDA; EC 3.5.1.41) catalyzes the hydrolysis of N-acetamide bonds of chitin, converting it to chitosan. Chitosan has several applications in areas such as biomedicine, food ingredients, cosmetics, pharmaceuticals, and agriculture. In this paper, occurrence, assay and purification protocols, enzymatic characteristics, substrate specificity, and mode of action of microbial CDAs have been described. Several lines of evidence have substantiated the biological roles involved in cell wall formation and plant-pathogen interactions for fungal CDAs. The gene structure of CDAs has been compared with other family 4 carbohydrate esterases which deacetylate a wide variety of acetylated poly/oligo-saccharides. The use of CDAs for the conversion of chitin to chitosan, in contrast to the presently used chemical procedure, offers the possibility of a controlled, non-degradable process, resulting in the production of well-defined chitosan oligomers and polymers. Insect pathogen that can secrete high levels of chitin-metabolizing enzymes including CDA can be a possible alternative for new pest management tools.

  • To develop cosmetics using Jindalae flowers (Rhododendron mucronulatum), the surface tensions of extracts were measured and the properties and stability of cream with extracts were investigated. The surface tension of 0.1% ethanol extract was 30.42 mN/m and that of distilled water was 72.2 mN/m. The surface tension of cream with 0.1% ethanol extract was similar to that of sample cream and the measured pH were weakly alkalic. The surface tension of 1% ethanol extract was the lowest value of 24.98 mN/m, the measured pH of cream with 1% ethanol extract was weakly acidic and the particle size of cream was stable. According to an oscillatory test, linear viscoelastic region was extended by adding of 1% water extract and 1% ethanol extract to cream, indicating that the cream had greater enhanced resistance for preserving inner structure as compared to outside stress. Besides, as a result of the diminished loss angle of ethanol extract cream, the elasticity of cream was increased more than that of sample cream and cream with 0.1% ethanol extract. In contrast, in the case of the increased loss angle of water extract cream, the viscosity of cream was increased. In conclusion, Rhododendron mucronulatum can be deliberated as a cosmetic material because 0.1% water and ethanol extracts showed efficacious physiological activities and cream with 1% extracts could extend linear viscoelastic region.

  • This study was aimed to investigate skin whitening effects of Nomura's jellyfish (Nemopilema nomurai) hydrolyzed extracts (NHE). The extracts were prepared through the hydrolysis of N. nomurai using commercial proteolytic enzymes such as Protamex, Alcalase, Flavourzyme and Neutrase with optimum pHs (pH 5-8) at $55^{\circ}C$. Their whitening activities were examined from the inhibition of melanin synthesis using B16-F1 cell lines. Among the examined samples, Neutrase-treated extract (N-NHE) showed the most significant inhibition effect on melanin synthesis by 89.9% at a concentration of $100{\mu}g/mL$. This sample decreased the expression of tyrosinase and TRP-1 (tyrosinase related protein-1) proteins as well. These results suggested the potential application of NHE as whitening ingredients in cosmetic preparation.

  • Objectives : Under normal condition melanin protects the skin from extracellular stimuli including ultraviolet (UV)-induced oxidative skin damages, but excess production and accumulation of melanin can induce hyperpigmentation causing esthetic problems. Therefore, in this study we tried to search for natural skin whitening materials from marine natural resources. Methods : Water and ethanol extracts of marine natural resources were prepared from Porphyra thalli (PT), Laminariae thallus (LT), Ostreae concha (OC), Sargassum thallus (ST), Undaria thallus (UT), Codium thalli (CT), Enteromorpha thalli (ET), Syngnathoides biaculeatus (SB), and Hippocampus coronatus (Hc). Their effects against UVB and ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis were investigated based on melanin formation in B16 mouse melanoma cells. The mRNA and protein expression of enzymes involved in the melanogenic process were further examined by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, respectively. Results : Water extract of Ostreae concha (OCW/E) effectively inhibited UVB and ${\alpha}$-MSH-induced melanin production in B16 melanocytes, which seemed to be mediated by inhibition of mRNA expression of tyrosinase and tyrosinase-related protein 1 (TRP-1). In another experiment, ethanol extracts from Porphyra thalli (PTE/E), Laminariae thallus (LTE/E), Sargassum thallus (STE/E), Undaria thallus (UTE/E), Codium thalli (CTE/E), Syngnathoides biaculeatus (SBE/E), and Hippocampus coronatus (HcE/E) significantly suppressed UVB and ${\alpha}$-MSH-induced melanin formation. Furthermore, ethylacetate fraction isolated form LTE/E (LTE/EEt) decreased UVB and ${\alpha}$-MSH-elevated extracellular melanin levels via inhibition of tyrosinase protein expression. Conclutions : These results suggest that marine natural resources such as Porphyra thalli, Laminariae thallus, Ostreae concha, Sargassum thallus, Undaria thallus, Codium thalli, Syngnathoides biaculeatus and Hippocampus coronatus have anti-melanogenic effects, thereby exhibiting high potentials to be utilized as one of the ingredients for the development of new whitening functional cosmetics.

  • Objectives : In order to confirm whether extracts of different parts of Zostera marina (ZM), a marine flowering plant, can be used as cosmetic ingredients, this study evaluated their cytotoxicity and cytoprotective effects against ultraviolet B (UVB). Inflammatory responses induced by UV stimuli are also associated with the aging of the skin. Methods : We investigated the effects of ZM extracts on cells through the water soluble tetrazolium salt-1(WST-1) assay for cell viability. In order to investigate the anti-inflammatory effects, we evaluated the suppression of Cyclooxygenase-2 (COX-2) expression by ZM extracts in HaCaT cells with UVB-induced damages, and also evaluated the production of Prostaglandin E2 (PGE2) in RAW 264.7 cells with LPS-induced damages. Results : High cell viabilities above 90% were observed in all types of ZM extracts, except for whole ZM extract at 0.5 mg/ml; in keratinocytes with UVB-induced damages, the cell viabilities were above 80% when treated with all types of ZM extracts. We confirmed their anti-inflammatory effects by investigating the suppression of inflammatory mediators. In keratinocytes with UVB-induced damages, COX-2 expression decreased in the experimental group treated with ZM extract. Similarly, in RAW 264.7 cells where inflammation was induced with LPS, the biosynthesis of PGE2 was inhibited. Conclusion : These results suggest that ethanol extracts from Zostera marina may have value as the potential anti-inflammatory medicinal plant. Also based on the abovementioned results, ZM extract protects skin cells from UV-induced damages, and thus can be used in topically applied products for skin protection.
