One of the important functions of skin is protection from harmful environments. Many studies have explored how to prevent skin from wrinkling and the occurrence of pigmentation changes. Skin wrinkling and pigmentation changes could be caused by unusual disruption of connective tissue, the formation of free radicals and ultraviolet radiation. In this study, extracts obtained from 254 different kinds of Jeju medicinal plants were screened for inhibitory effects on tyrosinase and elastase, and for free radical scavenging effects. Four herbs, Phormium tenax, Morus bombycis, Morus alba, and Cudrania tricuspidata, were potent inhibitors of tyrosinase ($IC_{50}$ values 4.62, 5.46, 8.17, and 64.17 ${\mu}g$/mL, respectively). Aleurites fordii [$IC_{50}$: 5.29 ${\mu}g$/mL, 1,1-diphenyl-2-picrylhydrazyl (DPPH)], Distylium racemosum ($IC_{50}$: 6.14 ${\mu}g$/mL), Acer palmatum ($IC_{50}$: 5.44 ${\mu}g$/mL), and Spiraea salicifolia ($IC_{50}$: 5.25 ${\mu}g$/mL) showed good antioxidative effects. Furthermore, Distylium racemosum ($IC_{50}$: 7.51 ${\mu}g$/mL), Diospyros kaki ($IC_{50}$: 15.1 ${\mu}g$/mL), Cornus macrophylla ($IC_{50}:$ 16.59 ${\mu}g$/mL), and Psidium guajava ($IC_{50}$: 40.25 ${\mu}g$/mL) exhibited potent inhibitory effects on elastase. These results suggest that medicinal plants possessing several biological activities may be potent inhibitors of the processes involved in pigmentation increases and aging. Further investigations will focus on in vivo assays and on the chemical identification of the major active components responsible for whitening and anti-aging activity in the screened efficacious extracts.

In this study, we evaluated anti-oxidation, whitening and anti-inflammatory effects of Elsholtzia ciliata extract for use as the cosmeceuticals. Elsholtzia ciliata extracts (30, 70 and 100% methanol extract) exhibited a significant tree radical scavenging effect (up to 80% over 0.025% concentration of 30 and 70% methanol extract, over 0.01% concentration of 100% methanol extract) against DPPH radical generation and showed a significant inhibitory effect (up to 80% over 0.1% concentration) on melanin synthesis in B-16 Melanoma cells. We separated 4 fractions from Elsholtzia ciliata extract (70% methanol extract) by MPLC. The 1st, 2nd, and 3rd fractions showed anti-oxidation (DPPH radical scavenging activity and suppressive effect on Mn-SOD), whitening(inhibitory effect on melanin synthesis) and anti-inflammatory (suppressive effects on $IL-1{\alpha}$, IL-6, COX-2, and total NO synthesis) effects.

In this study, we evaluated anti-oxidation, whitening and anti-inflammatory effects of Ardisia crenata for use as the cosmeceuticals. Ardisia crenata extract (70% MeOH) showed a significant free-radical scavenging effect (up to 90% over at the concentration of 0.01 %) against DPPH radical generation and showed a significant inhibitory effect (up to 50% over at the concentration of 0.05 %) on melanin synthesis in B16 meanoma cells. We separated 5 fractions from Ardisia crenata extract (70% MeOH) by MPLC. The 3rd, 4th, and 5th fractions showed the anti-oxidation (DPPH radical scavenging activity and supressive effect on Mn-SOD), whitening (inhibitory effect on melanin synthesis) and anti-inflammatory (supressive effects on $IL-1{\alpha}$, IL-6, COX-2 and total NO synthesis) effects.
The objective of this research was to evaluate the inhibitory activities of phenolic compounds isolated from Cheomoknosang callus on Helicobacter pylori. Total phenolic compounds of 80% ethanol extracts from callus were 15.3 mg/g. The activity of H. pylori inhibition at 80% ethanol extracts from Cheongmoknosang callus was determined as 14 mm clear zone. Isolation of inhibitory compounds was carried out on Sephadex LH-20 and MCI-gel CHP-20 column chromatography using a gradient elution procedure of increasing MeOH in $H_2O$. The chemical structure of the inhibitory compound against Helicobacter pylori was confirmed as protocatechuic acid, chlorogenic acid, caffeic acid and rosemarinic acid by spectroscopic analysis of FAB-MS, NMR and IR spectrum.

Background: The cellular senescence of primary cultured cells is an irreversible process characterized by growth arrest. Restoration of senescence by ginsenosides has not been explored so far. Rg3(S) treatment markedly decreased senescence-associated β-galactosidase activity and intracellular reactive oxygen species levels in senescent human dermal fibroblasts (HDFs). However, the underlying mechanism of this effect of Rg3(S) on the senescent HDFs remains unknown. Methods: We performed a label-free quantitative proteomics to identify the altered proteins in Rg3(S)-treated senescent HDFs. Upregulated proteins induced by Rg3(S) were validated by real-time polymerase chain reaction and immunoblot analyses. Results: Finally, 157 human proteins were identified, and variable peroxiredoxin (PRDX) isotypes were highly implicated by network analyses. Among them, the mitochondrial PRDX3 was transcriptionally and translationally increased in response to Rg3(S) treatment in senescent HDFs in a time-dependent manner. Conclusion: Our proteomic approach provides insights into the partial reversing effect of Rg3 on senescent HDFs through induction of antioxidant enzymes, particularly PRDX3.
Whitening effect of the extract of Phaseolus angularis (PA) shell was investigated for its potential application as a functional ingredient for cosmetic products. The tyrosinase inhibitory effect, which is related to whitening, was 76.4% at a concentration of 1,000 ㎍/ml. Cell viability of melanoma cells was measured to test toxicity of the PA shell extract at concentrations showing at least 90% viability. In addition, western blot and RT-PCR assays of the PA shell extract showed concentration-dependent decreases of MITF, TRP-1, TRP-2 and tyrosinase protein and inhibitory effect of mRNA expression. These findings suggested that extract from PA shell has great potential as a cosmeceutical ingredient with whitening effect.
Extracts from Schizandra chinensis Baillon (Korean name: Omija) were tested for antioxidant and their inhibitory activities of ${\alpha}-amylase$ and ${\alpha}-glucosidase$. Total contents of phenolics were found as 4.35 mg/g (water extract)${\sim}$6.35 mg/g (60% ethanol extract). Electron donating ability (EDA), ABTS [2,2'-azinobis(3-ethyl-benzothiaznoline-6-sulfonic acid)] radical decolorization, antioxidant protection factor (PF) and thiobarbituric acid reactive substance (TBARS) were measured for the antioxidative activity of the extracts from S. chinensis. The water extract were determined as 97.5% at 200 ${\mu}g/ml$ while the activity of 60% ethanol extract were 96.2% at 200 ${\mu}g/ml$ in EDA. The 60% ethanol extract showed higher antioxidant activity than water extract when evaluated by ABTS radical decolorization, antioxidant PF and TBARS. ${\alpha}-Amylase$ inhibitory activity of water extract was similar with that of 60% EtOH extract. ${\alpha}-glucosidase$ inhibitory activities of water extract (97.4%) was higher than that of 60% ethanol extract (84.5%) at 200 ${\mu}g$/ml. The water extract from S. chinensis did not show an antimicrobial activity against Helicobacter pylori, but the 60% ethanol extract showed high antimicrobial activities such as 23 ${\pm}$ 1.6 mm of clear zone in 200 ${\mu}g/ml$ of phenolics. The result suggest that the water and 60% ethanol extract from S. chinensis will be useful as natural antioxidants and functional foods.

In order to investigate the possibility of Sesamum indicum L. (S. indicum) extract as an active ingredient for wrinkle-care cosmetics, we prepared 70% ethanolic extract of S. indicum and measured its elastase inhibitory activity and collagenase inhibitory activity. We also evaluated the effect of S. indicum extract on protein and mRNA expression of MMPs in fibroblast cell (CCD-986sk). For anti-wrinkle effects, elastase inhibition activities and collagenase inhibition activities were 37.8% and 45% at a dose of $1,000{\mu}g/mL$ of S. indicum 70% ethanol extract. For a cell viability test, measured on fibroblast cell by ethanol extract of S. indicum, results showed 96% with cell viability at $100{\mu}g/mL$ concentration. According to the results of western blot of ethanol extract from S. indicum the expression of the matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-3 (MMP-3) protein was decreased by 63%, 43%, 49% at $100{\mu}g/mL$ concentration. Reverse transcription-polymerase chain reaction (PCR) of ethanol extract from S. indicum showed that the expression of MMP-1, MMP-2, MMP-3 mRNA was decreased by 82%, 79%, 82% at $100{\mu}g/mL$ concentration. The findings suggest that 70% ethanol extract from S. indicum has potential as a cosmeceutical ingredient with anti-wrinkle effects.

Portulaca oleracea L. is an edible plant widely consumed in daily diet throughout Europe, Asia and America. In this study, protective effects of P. oleracea L. extracts against oxidative stress and matrix metalloproteinase (MMP) activity induced by ultraviolet B (UVB) radiation were investigated using HaCaT immortal human keratinocytes. In this context, the mRNA and protein productions of MMPs (MMP-1, -2, and -9) and type I procollagen, which are major markers of photoaging induced by UVB radiation in HaCaT keratinocytes, were evaluated. Furthermore, UVB-induced reactive oxygen species (ROS) generation and mRNA and protein expression levels of superoxide dismutase-1 (SOD-1), oxygenase-1 (OH-1), and nuclear factor-erythroid 2-related factor-2 (Nrf-2), all of which are associated with the antioxidant balance, were investigated. As shown by the results, UVB radiation induced ROS formation and led to increased production of MMPs and decreased collagen production in human keratinocytes, which resulted in skin photoaging or photodamage. The treatment with P. oleracea L. extracts downregulated MMP (MMP-1, -2, and -9) production and upregulated type I procollagen expression in UVB-induced HaCaT cells. Furthermore, treatment with the extracts decreased UVB-induced ROS generation and increased the expression of antioxidant enzymes, such as SOD-1 and OH-1, through the Nrf-2 pathway. Taken together, these results suggest that P. oleracea L. extracts could be a potential cosmeceutical agent for the prevention of skin photoaging or photodamage.

Recently, several researchers have been developing cosmetics from natural ingredients for skin whitening and anti-aging products. The red sea cucumber (RSC), Apostichopus japonicas, is a species of sea cucumber in the family stichopodiae, which is widely distributed in China, Japan, and Korea. To use Red Sea Cucumber as a cosmetic ingredient, its inhibitory effects on melanogenesis and the anti-aging effects of RSC extracts were investigated. First, a tyrosinase activity assay was performed, which showed that RSC inhibited tyrosinase activity at a concentration of $200{\mu}g/ml$. An MTT assay was carried out to evaluate cell toxicity, and the results showed that RSC extract has no cytotoxicity in HaCaT cells. Furthermore, the mRNA expression levels of tyrosinase, tyrosinase related protein 1 (TRP-1), tyrosinase related protein 2 (TRP-2), microphthalmia-associated transcription factor (MITF), and matrix metalloproteinase (MMPs) genes treated with RSC extract in B16F10 and HaCaT cells decreased. Moreover, a wound-healing assay was performed to identify the cell regeneration effect of RSC extracts. Also, a skin turnover effect was confirmed by creating a three-dimensional cell culture with HaCaT and human fibroblasts. Altogether, the results suggested that Red Sea Cucumber may possess a high ability to induce whitening and anti-wrinkle effects as a cosmeceutical ingredient.
