Whitening and Anti-wrinkle Effects of Prunus persica Flos (도화(桃花, Prunus persica Flos)의 미백 및 주름개선 효과)

  • The solvent extracts of Prunus persica Flos were investigated for the activities of whitening and anti-wrinkle effects to apply as a functional ingredient for cosmetic products. The tyrosinase inhibitory effect, which is related to skin-whitening, was 54.0, 58.3% in P. persica Flos (PPW, PPE) at 1,000 ppm. In addition, the ethanol extract of P. persica Flos (PPE) showed a potent tyrosinase inhibitory activity in the test using melanoma cell lines resulting in 40.0% inhibition at 100ppm. Furthermore, the aqueous acetone extract from the flower of P. persica Flos was found to inhibit elastase, which was more effective than ascorbic acid at 1,000 ppm. The inhibition of melanin synthesis by P. persica Flos extract (PPE) was about 56.5% at 100 ppm concentration. When compared to other extraction methods, the ethanol extract showed more potent whitening activity. For anti-wrinkle effect, the elastase inhibition activity of P. persica Flos extract (PPA) was 57.0% and higher than that of ascorbic acid at 1,000 ppm. The collagenase inhibition activity of P. persica Flos extract (PPA) was about 48.0% at 1,000 ppm. Collagen synthesis in fibroblast cell by P. persica Flos extracts (PPA) was about 41.0% at 100 ppm and its acetone extract was the best showing antiwrinkle activities. All these findings suggested that P. persica Flos has a great potential as a cosmeceutical ingredient with a whitening and anti-wrinkle effect.

Skin Whitening Effect of Sedum kamtschaticum Fisch. Solvent Fractions (기린초(Sedum kamtschaticum Fisch.) 추출 분획물의 피부 미백효과 연구)

  • Sedum kamtschaticum Fisch., a native plant of Korea, has been used in Korean traditional medicine in the form of water extract for its capacity to improve blood circulation and for its antioxidant and anti-inflammatory effects. Since previous research suggests that S. kamtschaticum Fisch. has excellent antioxidant and mushroom tyrosinase inhibition activities, in this study, the root and stem parts of S. kamtschaticum Fisch. are extracted in 70% ethanol (SKS, SKR), fractionated with and in order of n-hexane (SSH), ethyl acetate (SSE, SRE), chloroform (SSC, SRC) and water (SSW, SRW) according to the polarity of each solvent, and tested for its applicability as a cosmetic material. According to the total polyphenol, flavonoid contents and DPPH radical scavenging activity of each fraction, the contents and scavenging activity of the root extractions (SKR) were higher than those of the stem extractions (SKS), ethyl acetate fractions (SSE, SRE) being the most effective. In addition, ethyl acetate fractions had the highest tyrosinase inhibition activity and melanin synthesis inhibition activity used on B16F10 melanoma cells, at the concentration of 10 ㎍/mL. HPLC analysis detected a variety of polyphenols including gallic acid and quercetin. This study suggests the potential role of S. kamtschaticum Fisch. as a natural cosmeceutical material.

Study on Effects of Anti-oxidant and Viscoelastic on Emulsion by the Extract of Astragalus membranaceus (황기(黃芪)추출물의 항산화 효능과 에멀션 점탄성도에 미치는 영향에 관한 연구)

  • Objectives : The Astragalus membranaceus has been used to diuretic, tonic, anti-viral, anti-oxidant activities in oriental herb medicine, and in each experiment, proved in this effects. Root of A. membranaceus includes according to flavonoid, saponin, polysaccharide, thus it has been studied for anti-oxidant, anti-inflammatory, anti-cancer activities. However, previous researches have been performed with being limited mainly to food industry. The present study was conducted anti-oxidative activity of A. membranaceus extracts. In addition, The objective of the present study was to determine the possibility for the application of emulsions in the development of A. membranaceus for cosmetics. Methods : In this study, aimed SOD like activity of A. membranaceus extract, effects of A. membranaceus extract on viscoelastic properties of emulsion was measured using rheometer. Results : In the results, 1, 3-butylene glycol extract of A. membranaceus efficiently reduce the active oxygen, SOD like activity of more than 40% decrement depend on concentration. And A. membranaceus extract increase the modulus of elasticity of emulsion, therefore A. membranaceus extract have not influence on stability loss. As such, the A. membranaceus extracts will remain stable when applied to emulsions or skin products. Conclusions : In consequence of this study, A. membranaceus extract can be effectively used in cosmetic emulsions when the relation between natural product extracts and formulation of cosmetics is elucidated. It is highly recommended that rheology be applied to determine the optimal extract concentration for cosmetic formulations.

Whitening Activities of the Halophyte L. tetragonum (Thunberg) A. A. Bullock Extract in B16F10 Melanoma Cells (염생식물 갯질경 추출물의 미백 활성)

  • To identify the natural and novel cosmeceutical ingredients which have whitening function, we examined the effects of tyrosinase activity and melanin production on halophyte Limonium tetragonum (Thunb.) A. A. Bullock extracts. As a result, the extracts showed whitening effect with no cytotoxicity. Melanin contents of B16F10 melanoma cells were decreased in a dose-dependent manner at 50, 100, $200{\mu}g/mL$ treatment of the extracts. In tyrosinase activity inhibition test, L. tetragonum (Thunb.) A. A. Bullock extracts showed decreased tyrosinase activity as the concentration of ${\alpha}-MSH$ was increased. Furthermore, it was observed that the tyrosinase and MITF expression was significantly downregulated by adding L. tetragonum (Thunb.) A. A. Bullock extracts in ${\alpha}-MSH$, a melanogenesis inducing material, treated B16F10 melanoma cells. These results indicate that L. tetragonum (Thunb.) A. A. Bullock extract might be an effective whitening agent by inhibit melanin formation.

Antioxidant and Antimicrobial Activities of Trypsin-treated Pinus densiflora Ethanol Extract (트립신 처리에 따른 적송잎 추출물의 항산화 활성 및 항균 효과)

  • Objectives: We investigated the antioxidant and antibacterial activities of Pinus densiflora ethanol extracts (PDEE) treated with trypsine as a protease. Methods: Various antioxidant activities were evaluated by measuring total contents of polyphenol and flavonoid, DPPH electron-donating ability and $ABTS^+$ radical scavenging activity of test material. To compare the antibacterial activity, paper disc diffusion assay was performed against two resident bacteria in human skin (Staphylococcus aureus and Staphylococcus epidermidis). Results: As for the total contents of polyphenol and flavonoid, and the electron-donating ability and ABTS+ radical scavenging activity, both PDEE and trypsin-treated Pinus densiflora ethanol extract (T-PDEE) showed high antioxidant activity in dose-dependent manner. And the T-PDEE showed slightly higher activity than PDEE, which indicated protease treatment seemed to affect in antioxidant activity. In the result of paper disc diffusion assay, antibacterial activity was confirmed in all two types of skin resident bacteria. T-PDEE was more active than PDEE and it seems that treatment of protease may increase the antibacterial activity of PDEE. Conclusion: All of these results, we confirmed that treatment of protease to PDEE can increase the antioxidant and antibacterial activities, and it can be explained thought that this would be applicable as a cosmeceutical material in the future.

The study on pharmacologic activation as cosmetic material of Sanguisorbae radix (지유(地榆)의 화장품 소재로서의 약리활성에 관한 연구)

  • Objectives : This study was aimed at producing emulsion by using butanol fractions of Sanguisorbae radix(SRA-B) which have high antioxidative and anti-inflammatory actions, and then evaluating stabilities of the emulsion. Methods : We measured antioxidant efficacy of SRA-B by using DPPH assay. Also, we checked the expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) by using the Western blot to evaluate the anti-inflammatory effects of SRA-B3. We prepared emulsion containing SRA-B3(E-SRA-B3) and analysed its particle size distribution under a microscope. Also, we performed the test for stability of the emulsion. Results : SRA-B3 showed the highest efficacy in electronic donating abilities' activity. The Western blot's results indicated that the protein expression's amount of iNOS and COX-2 in macrophage stimulated by LPS were reduced by SRA-B3 treatment. The average particle size of E-SRA-B3 was $5{\sim}6{\mu}m$ in diameter and was $6.7{\mu}m$ in a view of the particle distribution. For a period of a observation, E-SRA-B3 has not made particular changes with storage temperature. It was observed that E-SRA-B3 could preserve its stable condition without a particular difference of viscosity during 28 days. Conclusions : From the above results, it was confirmed that SRA-B3 has potentiality enough to be applied to industrialization and could be utilized as antioxidative natural materials and anti-inflammatory cosmetics.

Anti-Oxidant and Antiinflammatory Effects of Rosa multiflora Root (찔레나무뿌리(Rosa multiflora root)의 항산화 및 항염증효과)

  • Rosa multiflora thunberg belonging to Rosaceae is widely distributed in East Asia including Korea and Japan, and has been reported to have tormentic acid and rosamultin. To develop a new natural anti-inflammatory agent for cosmetics, we investigated the inhibitory effects of inflammation in Rosa multiflora root (R. multiflora root). The biological activity and anti-inflammatory effects were investigated by water, ethanol, methanol and acetone extracts of R. multiflora root. The measurements of polyphenol content from R. multiflora root were highest in water and acetone extracts, at 57.48 ${\pm}$ 0.88 mg/g and 67.05 ${\pm}$ 0.56 mg/g, respectively. The result of DPPH, ABTS and superoxide anion radical scavenging effects showed over 50% efficacy at 50 ${\mu}g/ml$ in ethanol, methanol and acetone extracts. Hyaluronidase inhibition effect showed over 60% efficacy at 500 ${\mu}g/ml$ in ethanol, methanol, and acetone extracts. Nitric oxide radical inhibition effect of R. multiflora root ethanol extracts showed over 30% efficacy at 500 ${\mu}g/ml$. We investigated the effect of R. multiflora root extracts on nitric oxide (NO) production of inducible nitric oxide synthase (iNOS) in LPS-induced RAW 264.7 macrophage cells. The result showed that R. multiflora root extracts have an inhibitory effect on NO production and iNOS expression and also can be used as an anti-inflammatory agent. These antioxidant and anti-inflammatory effects of R. multiflora root show applicant potential application as a functional cosmetic material.

Studies on Antioxidant Effect of Mushroom Complex (버섯복합물의 항산화 효과)

  • In this study, we evaluated the antioxidant effects of Cordyceps, Ganoderma lucidum (Curtis) P. Karst and Phellinus linteus complexes. Three mushrooms were extracted using 70% ethanol, respectively, in ratios of 80:10:10(?). The electron-donating ability was measured using the DPPH radical mushroom complexes and showed effects of 66% in 500 ${\mu}g/ml$. ABTS radical cation decolorization measured 73% in 50 ${\mu}g/ml$ and had an effect similar to BHA in 500 ${\mu}g/ml$. In their superoxide anion radical scavenging ability, the mushroom complexes showed an effect of 41% in 10 ${\mu}g/ml$. The effect on the control of BHA was more than three times as high as BHA in 50 ${\mu}g/ml$ and 100 ${\mu}g/ml$ concentration. The results showed that lipid oxidation is an excellent inhibitor of lipid oxidation $Cu^{2+}$, compared to the $Fe^{2+}$ group. The results of superoxide dismutase (SOD) activity in the mushroom complexes showed a low effect of 13% in 500 ${\mu}g/ml$. Thus, the results of this study showed that the antioxidant effect of the mushroom complexes was excellent, indicating their potential as an antioxidant material.

Anti-wrinkle effect of berberine by inhibition of MMP-2 and MMP-9 activity in fibroblasts (섬유아세포에서의 MMP-2 및 MMP-9 활성 억제에 의한 베르베린의 항주름 효과)

  • We analyzed the antioxidant and anti-wrinkle activities of berberine, isolated from dried rhizome of Coptis japonica Makino, to determine its cosmetic potential. We performed the 3-[4,5-dimethylthiazol]-2-yl]-2,5-diphenyl-tetrazoliumbromide (MTT) assay to evaluate the toxicity of the berberine. We also measured the ROS and hyaluronic acid production, and expression of MMP-2, MMP-9, TIMP-1, TIMP-2, and tumor necrosis factor-alpha ($TNF-{\alpha}$) to evaluate the antioxidant and anti-wrinkle activities of berberine, respectively. The cytotoxicity of ultraviolet light, in presence of berberine, was measured by the MTT assay using CCD-986sk fibroblasts, and no cytotoxicity was observed at concentrations less than $25{\mu}g/mL$. We also found that berberine decreased ROS production in a concentration-dependent manner and promoted the synthesis of hyaluronic acid. Further, berberine reduced the protein levels and mRNA expression of MMP-2 and MMP-9, which are associated with wrinkle formation, and increased the expression of TIMP-1 and TIMP-2. In addition, the inhibitory effect of berberine on $TNF-{\alpha}$, known as pro-inflammatory cytokine, was inhibited by $TNF-{\alpha}$ gene in a concentration-dependent manner. These results suggest that berberine holds cosmetic value owing to its antioxidant activity, by inhibiting ROS production and anti-wrinkle activity by reducing MMP-2 and MMP-9 and increasing TIMP-1 and TIMP-2 expression.

In vitro activities of Grape Pruning Stems for Application of Cosmetic Ingredients (포도 전정가지의 화장품 소재로서의 응용)

  • Grape pruning stems constitute a scarcely investigated class of byproducts with limited reports on their bioactive polyphenol content and/or industrial applications. Herein we present the outcome of our investigation on grape pruning stems extracts, concerning the assessment of their total polyphenolic content and the detailed evaluation of their antioxidant properties. Results obtained indicate that grape pruning stems are particularly rich in flavonoids and trans-resveratrol. The antioxidant activities was analyzed and expressed as electron donating ability, ABTS cation radical decolorization, hydrogen peroxide scavenging activity and superoxide anion radical scavenging activity, the antioxidant activity of Vitis labrusca L. pruning stems extracted (GPSE) was higher than that of BHA and L-ascrobic acid. The whitening and anti-wrinkle activities display an capability. Results herein grape pruning stems used as a valuable resource for the extraction of resveratrol, which would be added to functional cosmetics and food materials.
