홍화잇꽃의 여드름피부 개선효과 및 세안용 생약식물소재 응용 (Efficacy of Safflower on the Acne Skin and Its Application for Facial Cleansing Biomedical Material)

  • 홍화는 예로부터 '사람의 건강에 도움이 된다'고 해서 '잇꽃'으로 불려져 왔으며 동의보감에서는 꽃잎이 홍색을 나타내기 때문에 홍화(紅花)로 소개되어 있다. 예로부터 홍화는 꽃잎을 압착후 정제된 색소를 이용하여 옷감에 천연색소를 입히는 염료와 연지의 재료로 사용되기도 하였다. 현대에 와서는 홍화의 주성분인 폴리페놀 화합물(Polyphenol cocktail) 성분이 건강식품업계에서 항노화 및 항산화물질로 알려지면서 관심의 대상이 되고 있다. 금번연구에서는 홍화의 주성분인 폴리페놀 화합물이 청소년기 혹은 장년기에서도 고민거리가 되고 있는 여드름균의 생성을 억제시킬 수 있는 항균, 항염효능이 있음을 DDT(Disk Diffusion Test) assay, MTT assay, NF-${\kappa}$B Luciferase activity inhibition assay의 in vitro법으로 확인하고 더불어 인체안면에 대한 임상시험에 사용할 세안화장품 제형의 화장비누 시제품을 제조하여 여드름피부의 개선 효과가 있음을 시험하고자 한다. 또한 이번 연구는 여드름용 화장비누 개발에 진일보하여 약용화장품 산업의 발전에 기여할 것임을 확신한다.

Rosa multiflora Thunberg fruit 추출물로부터 Helicobacter pylori 억제물질의 분리 및 동정 (Isolation and Identification of Inhibitory Compounds on Helicobacter pylori from Rosa multiflora Thunberg Fruit Extracts)

  • 영실에 존재하는 페놀성 화합물의 추출 최적 조건을 알아보고자 추출용매를 달리하여 phenol성 물질의 함량을 비교한 결과, 70% ethanol을 용매로 하여 24시간 교반 추출하였을 때 phenol성 물질의 용출량이 13.8 mg/g 이상으로 가장 높았다. Helicobacter pylori에 대한 항균효과는 영실추출물을 150 ${\mu}g$/ml의 농도로 첨가하였을 때 생육 저해환이 12 mm, 200 ${\mu}g$/ml 일 때 14 mm의 생육저해환이 나타나 항균활성이 우수한 것을 알 수 있었다. H. pylori에 대한 저해물질을 정제하기 위하여 영실을 70% 에탄올로 추출한 후 Sephadex LH-20 column과 $C_{18}$ cartridge column을 이용하여 항균활성을 가지는 물질을 순수 정제하고 구조를 동정한 결과 protocatechuic acid, chlorogenic acid 및 quercetin으로 동정하였으며, 단일물질에 의한 H. pylori에 대한 저해효과 보다 혼합물질에 의한 synergy 효과에 의해 저해 효과가 높아지는 것을 알 수 있었다.

매화꽃봉오리의 추출용매별 항산화 효과 (Antioxidant Activities of Plunus mume flower buds Extract by Various Solvents)

  • This study investigated the antioxidant activities of distilled water, ethanol and methanol extracts of Plunus mume flower buds (PFB). The various solvent extracts of PFB were evaluated for their total polyphenol, flavonoid, reducing power and free radical scavenging activities by FRAP and DPPH analysis. The ethanol extract of PFB contained significantly higher amounts of total polyphenols (145 mg GAE/g) and flavonoids (25.43 mg QE/g) than methanol (132 and 25.42) and distilled water (113.6 and 18.04). Among solvent extracts of PFB, the ethanol extract showed the highest antioxidant activities. The 100% ethanol extract of PFB contained significantly higher amounts of total polyphenols and flavonoids than 70% and 50% ethanol extracts. Moreover, the 100% ethanol extract of PFB showed high efficacy in DPPH radical scavenging activity and in collagenase inhibition activity. This results suggest that 100% ethanol extract of PFB has the most effective antioxidant capacity compared to the methanol and water extracts tested in the present study. Thus, it can be applied for the effective extraction of functional material from PFB for usage of cosmeceutical and/or food industries.

광대싸리 줄기 추출물의 항산화 활성 (Antioxidative Activity of Securinega suffruticosa Extract)

  • In this study, the anti oxidative effects, inhibitory effects on elastase, and components of Securinega suffruticosa extracts were investigated. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}$) and reactive oxygen species ROS) scavenging activities ($OSC_{50}$) of extract/fractions from Securinega suffruticosa were measured. The aglycone fraction ($9.04\;{\pm}\;0.51\;{\mu}g/mL$) and 50 % ethanol extract ($1.05\;{\pm}\;0.41\;{\mu}g/mL$) showed the most effective scavenging activities. The protective effects on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The aglycone fraction exhibited the most prominent cellular protective effect (${\tau}_{50},\;102.77\;{\pm}\;5.37$ min at $10{\mu}g/mL$). The inhibitory effect of ethyl acetate fraction on tyrosinase were of examined ($210.0\;{\pm}\;1.02\;{\mu}g/mL$). The inhibitory effect of aglycone fraction on elastase were also investigated ($17.6\;{\pm}\;1.26\;{\mu}g/mL$). These results indicate that extract/fractions of Securinega suffruticosa can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Securinega suffruticosa extract could be used as a new cosmeceutical ingredient for whitening and anti-wrinkle products.

진생베리 발효추출물의 화장품 약리활성 (Study on Cosmeceutical Activities from Fermented Ginseng Berry Extracts)

  • 진생베리는 진세노사이드 Re을 다량함유하고 있고 항염, 항암, 혈당강하 및 미백효과가 있다. 본 연구에서는 Rhizopus Oligosporus 균주를 이용하여 진생베리 발효공정을 확립하였으며 진생베리 발효물에 대한 화장품 약리활성을 분석하였다. 발효에 의한 인삼 베리 추출물의 전자 공여 능력은 1,000 ㎍/mL 농도에서 81%를 나타냈다. ABTS+ 라디칼 소거능 1,000 ㎍/mL 농도에서 100.2% 효과를 나타내었다. 피부 미백과 관련된 티로시나 아제 억제 효과는 1,000 ㎍ /mL 농도에서 57%였다. 피부 주름과 관련된 엘라스타제 억제효과는 1,000 ㎍/mL 농도에서 47%였다. 또한, 콜라게나제 억제 효과는 1,000 ㎍/mL 농도에서 33%였다. 이상의 결과로부터 진생베리 발효물이 항염과 미백, 주름개선 효능을 가진다고 판단된다. 따라서 진생베리 발효물이 향우 항염 및 항노화 화장품 원료로서의 이용 가능성이 매우 높을 것으로 판단된다.

한국 자생 석곡 메탄올 추출물의 미백 및 주름개선 효과 (Whitening and Anti-wrinkle Effects of Korean Native Dendrobium moniliforme Methanol Extract)

  • Dendrobium moniliforme (DM) is a valuable and versatile herbal medicine with the anecdotal claims of antioxidant and anti-inflammation. In the present study, we investigated the whitening and anti-wrinkling effects of DM under various conditions with B16F10 melanoma cells and human dermal fibroblasts. The DM extract inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. Treatment of the DM extract effectively suppressed the ${\alpha}$-MSH-stimulated melanin formation, tyrosinase activity and dendrite outgrowth. Moreover, the ${\alpha}$-MSH-induced mRNA expressions of tyrosinase-related protein-1 (TRP-1) and tyrosinase-related protein-2 (TRP-2) and protein expression of tyrosinase were significantly attenuated by DM treatment. We also investigated the DM increased the production of type I procollagen and inhibited TNF-${\alpha}$-induced mRNA expressions of MMP-1, -3 in the human dermal fibroblast. These results indicate that DM may be a great cosmeceutical ingredient for its whitening and anti-wrinkle effects.

동과자 추출물의 항산화, 항염 및 미백 효능 (Anti-oxidant, anti-inflammatory and Whitening effect of Benincasa hispida seed extract)

  • 본 연구는 피부 관리에 사용되는 천연화장품 원료로서 동과자 추출물의 활용 가능성을 조사하기 위해 항산화, 항염 그리고 미백 효능을 평가하였다. DPPH 자유라디컬 소거 활성은 농도 의존적으로 감소하였으며, 총폴리페놀 함량은 메탄올추출물(22.42±0.002 mgGAE/g)이 열수추출물(9.77±0.002 mgGAE/g)보다 더 높게 나타났다. MTT assay 결과 동과자 추출물에 의한 RAW264.7 세포와 B16F10 세포에서의 세포독성은 없었다. 또한 LPS 처리에 의해 활성화된 RAW264.7 세포에서 NO 생성과 TNF-α 분비 억제능을 확인하였다. B16F10 melanoma 세포에서 동과자 추출물은 α-MSH에 의한 멜라닌 합성을 유의하게 저해하는 것으로 나타났다. 게다가, westernblot 분석결과 동과자 메탄올추출물은 MITF, TRP-1 및 TRP-2 단백질의 발현 수준을 현저하게 감소시켰다. 이상의 결과로 동과자 추출물은 항산화, 항염 그리고 미백 효능을 가진 기능성 화장품 소재로써의 가치가 높을 것으로 사료된다.

Human sebocyte-based assay system for the screening of compounds to lower the lipid synthesis in sebaceous gland

  • SZ95 cell is an immortalized human sebaceous gland cell line that shows the morphologic, phenotypic and functional characteristics of normal human sebocytes. Sebocytes may play crucial parts in the pathophysiologic processes and disorders of the pilosebaceous unit. The secretory activity of the sebaceous gland is remarkably species-specific and acne is an exclusively human disease. Thus, this SZ95 cells offer possibilities for investigations on the physiology of the sebaceous gland and its role in sebum-associated skin disease such as acne. In this study, we investigated the effects of 13-cis-retinoic acid (13-cis-RA) and spironolactone, frequently used as therapeutic agents of acne, on the lipid synthesis and proliferation of human sebocytes. Cell proliferation was determined by MTT assay and cytoplasmic lipid droplets was shown by Oil-red a staining. Total lipid levels were biochemically estimated by the sulfo-phospho-vanilline reagent. 13-cis-RA and spironolactone significantly inhibited proliferation and lipid levels in a dose-dependent manner. Combined treatment with testosterone and 13-cis-RA or spironolactone resulted in a lower total lipid levels than that with androgen alone. These observations indicate that 13-cis-RA and spironolactone are potent inhibitors of both cell proliferation and lipid synthesis in human sebocytes. We will provide experimental evidence that this human sebocyte cell line serves as an adequate tool for evaluating the anti-lipogenic activity of various compounds potentially useful for the bioactive cosmeceutical ingredients on acne skin, and studying the intracellular biochemical markers depending on the types of compounds from various sources.

Pectin Micro- and Nano-capsules of Retinyl Palmitate as Cosmeceutical Carriers for Stabilized Skin Transport

  • Retinyl palmitate (RP)-loaded pectinate micro- and nano-particles (PMP and PNP) were designed for stabilization of RP that is widely used as an anti-wrinkle agent in anti-aging cosmeceuticals. PMP/PNP were prepared with an ionotropic gelation method, and anti-oxidative activity of the particles was measured with a DPPH assay. The stability of RP in the particles along with pectin gel and ethanolic solution was then evaluated. In vitro release and skin permeation studies were performed using Franz diffusion cells. Distribution of RP in each skin tissue (stratum corneum, epidermis, and dermis) was also determined. PMP and PNP could be prepared with mean particle size diameters of $593{\sim}843{\mu}m$ (PMP) and 530 nm (i.e., $0.53{\mu}m$, PNP). Anti-oxidative activity of PNP was greater than PMP due largely to larger surface area available for PNP. The stability of RP in PMP and PNP was similar but much greater than RP in pectin bulk gels and ethanolic solution. PMP and PNP showed the abilities to constantly release RP and it could be permeated across the model artificial membrane and rat whole skin. RP was serially deposited throughout the skin layers. This study implies RP loaded PMP and PNP are expected to be advantageous for improved anti-wrinkle effects.
