Anti-diabetic effects of the extract from Atractylodes lancea, Anemarrhena asphodeloides and Cinnamomum Cassia mixture in high fat diet-induced diabetic mice and regulation of the function in C2C12 mouse skeletal muscle cells (창출·지모·육계 복합추출물의 고지방식이 유도 당뇨병 마우스에서의 항당뇨 효능 및 C2C12 골격근세포에서의 조절기전 연구)

  • Objective : This study investigated the anti-diabetic effects of DM1, a herbal mixture with Atractylodis Rhizoma, Anemarrhenae Rhizoma, and Cinnamomi Cortex in high fat diet (HFD)-induced diabetic mice and the mechanism in C2C12 mouse skeletal muscle cells. Methods : The C57B/6 mice were fed high fat for 12 weeks, and then administrated DM1 extract (500 mg/kg, p.o.) for 4 weeks. The changes of body weight, calorie and water intakes, fasting blood glucose levels and the serum levels of glucose, insulin, triglyceride, HDL-cholesterol, AST and ALT were measured in mice. The histological changes of liver and pancreas tissues were also observed by H&E stain. C2C12 myoblasts were differentiated into myotubes and then treated with DM1 extract (0.5, 1, and 2 mg/㎖) for 24 hr. The expression of myosin heavy chain (MHC), PGC1α, Sirt1 and NRF1, and the AMPK phosphorylation were determined in the myotubes by western blot, respectively. Results : The DM1 extract administration significantly decreased the calorie and water intakes, glucose, triglyceride, AST and ALT levels and increased insulin and HDL-cholesterol in HFD-induced diabetic mice. DM1 extract inhibited lipid accumulation in liver tissue and improved glucose tolerance. In C2C12 myotubes, DM1 treatment increased the expression of MHC, PGC1α, Sirt-1, NRF-1 and the AMPK phosphorylation. Conclusion : In our results indicate that DM1 can improve diabetic symptoms by decreasing the obesity, glucose tolerance and fatty liver in HFD-induced diabetic mice, and responsible mechanism is might be related with energy enhancement.

Identification of Cinnamon Components and Quantitative Determination of Cinnamlc Acid from Crude Drug Drink Preparations (생약복방 드링크제 중 계피성분의 확인 및 계피산의 분리정량)

  • For the quality control of crude drug drink preparations, methods for identification of cinnamon components and quantitative determination of cinnamic acid were established. Cinnamon components were identified by TLC with benzene/ethyl acetate (1:1, v/v) on silica gel plate by spraying p-anisaldehyde-sulfuric acid. Cinnamic acid contents were determined at UV 280 nm by HPLC on $\mu$-Bondapak Cls column with acetonitrile/water/acetic acid (40:60:2, v/v). Recoveries of cinnamic acid in three crude drug drink preparations were between 84.1-90.2% compared to the content of the cinnamon extract.

The Effect of Plant Ethanolic Extracts on Oxidation of Soybean Oil (다류원류 식물류의 에탄올 추출물이 대두유의 산화에 미치는 영향)

  • This study was planned to investigate the effect of 40 plant ethanolic extracts on antioxidant activities in vitro. The total phenolics, $\beta$-carotene, $\alpha$-tocopherol and selenium contents were also determined . Antioxidant activities fo the ethanolic extracts(0.02%, w/w) in the soybean oil were measured both by determining the peroxide value (POV) during 35 days of storage at 4$0^{\circ}C$ in a forced draft air-incubator and by determining changes in conductivity at 11$0^{\circ}C$(Rancimat method.). Soybean oil without any additives was used as a control and that treated with 0.02% BHT was used as a positive control. Based on the POV determination, green tea extract was found to be the most effective in stabilizing soybean oil, then followed by long tea, which both of them showed higher antioxidant activities compared to the BHT treatment. The antioxidant activities of them showed higher antioxidant activities compared to the BHT treatment. the antioxidant activities of coffee, cinnamomi cortex, acanthopanacis cortex, black tea, orange peel , instant coffee, peony and crni fructus extracts were stronger compared to the control .By the Rancimat method, green tea leaf and oolong tea leaf, foxglove, acanthopanacis cortex and peony extracts. Compared to other extracts, green tea leaf, black tea leaf, foxglove, acanthopanacis cortex and peony extracts had stronger antioxidative effects in both the POV and Rancimat methods used in this study. ethanolic extracts which showed the stronger antioxidative effect also has the higher contents of total phenolics, $\beta$-carotene, and/or $\alpha$-tocopherol. The antioxidative effect of ethanolic extracts was found to be due to the combined effect of various antioxidants.

Protective Effects of Jisilhaebaekgyeji-tang and Constituents Extract on Cultured Rat Myocardial Cell treated by XO/HX (XO/HX에 의해 손상된 배양 심근세포에 대한 지실해백계지탕과 구성약물 추출물의 방어효과)

  • To certify the protective effect of herbal medicine against oxygen free radical-induced myocardiotoxicity, cytotoxicity was measured using LDH activity and TBARS assay in the presence of Jisilhaebaekgyejitang(JHGT) extracts or single constituents of this prescription, In the present study, xanthine oxidase/hypoxanthine (XO/HX) resulted in a cell damage such as increases in LDH activity in culture medium and lipid peroxidation in cultured myocardial cells. In the effect of JHGT extract and its single constituents, which are Fructus Ponciri Seu Aurantii Immaturus (FPSAI), Cortex Magnoliae Officinalis (CMO), Bulbus Allii Macrostemi (BAM), Ramulus Cinnamomi (RC) and Fructus Trichosanthis (FT), they showed the prevention from the XO/HX-induced cardiotoxicity by the decrease of LDH activity and lipid peroxidation. From these results, they show that XO/HX is cardiotoxic in cultured myocardial cells derived from neonatal rat, and it suggests that JHGT, FPSAI, PT, CMO, BAM, RC and FT extracts are positively effective in the blocking in XO/HX-induced cardiotoxicity.

  • 80% extract of Gamigwaruhaebaekbaekju-Tang (GGHBT), Gagamgwaruhaebaekbaekju-Tang (GGGHBT) and Gamigwaruhaebaekwhanggum-Tang (GGHWT) remarkably showed inhibitory effects on HMG-CoA reductase, lipid peroxidation of rat liver and LDL oxidation, and DPPH free radical scavenging effect in a dose-dependent manner. Especially, GGHWT which is formulated with Trichosanthis Fructus, Pinelliae Tuber, Aurantii Immatures Fructus, Magnoliae Cortex, Allii Macrostemi Bulbus, Cinnamomi Ramulus and Scutellariae Radix on the basis of Gwaruhaebaekbaekiu-Tang listed on the traditional medicinal references showed more effective hypocholesterolemic activities in vitro bioassay than the other prescriptions.

Studies on the Development of Antihyperlipidemic Drugs from Oriental Herbal Medicines(II) - Antihyperlipidemic Effects of Oriental Herbal Medicines - (한방약물로부터 항고지혈증 치료약물개발(2) - 수종 한약재의 항고지혈증 효과 -)

  • In the previous reports, we selected 80% MeOH extract of 7 herbs including Scutellariae Radix(SR), Paeoniae Radix Rubra(PRR), Moutan Cortex(MC), Angelicae Gigantis Radix(AGR), Crataegi Fructus(CF), Bambusae Caulis in Taeniam(BCT) and Cinnamomi Ramulus(CR), which exhibited the inhibitory effect on HMG-CoA reductase and DPPH free radical scavenging effect in vitro, and antihyperlipidemic effects on antihyperlipidemic rats induced by Triton WR 1339 in vivo. Among them, SR, MC, AGR and BCT showed significant suppression of elevated serum LDL-cholesterol level, and AGR and CF showed significant liver weight increase on high cholesterol diet induced hyperlipidemic mice. And, SR, PRR, AGR, BCT and CR significantly suppressed the elevated serum total cholesterol and triglyceride levels on corn oil induced hyperlipidemic rats. Then, in order to research new antihyperlipidemic agents from the oriental medicinal herbs, we chose SR, AGR, CR and BCT which have the antihyperlipidemic effect in vitro and in vivo, and those herbs were systematically fractionated with organic solvent. EtOAc fraction of SR, hexane fraction of BCT, AGR and chloroform fraction of CR exhibited remarkably inhibitory effect on HMG-CoA reductase activity.

Effects of SosokMyeongTang extract on the Regional Cerebral Blood Flow and Blood Pressure in Rats (소속명탕(小續命湯)이 백서(白鼠)의 국소뇌혈류량(局所腦血流量)과 혈압(血壓)에 미치는 영향(影響))

  • SosokMyeongTang(SMT) have been used in oriental medicine for many centuries as a therapeutic agent for cerebral disease. The effects of SMT on the vascular system is not known. The purpose of this study was to investigate the effects of SMT on the changes in blood pressure(BP) and regional cerebral blood flow(rCBF) of rats. SMT consists of the following components : Radix Ledebouriellae(防風), Radix Cocculi or Stephaniae(防己), Semen Armeniacae(杏仁), Cortex Cinnamomi(肉桂), Radix Scutellariae(黃芩), Radix Paeoniae Lactiflorae(白芍藥), Radix Ginseng(人蔘), Rhizoma Cnidll(川芎), Herba Ephedrae(麻黃), Radix Glycyrrhizae(甘草), Radix Aconiti(附子?), Fructus Zizyphi Jujubae(大棗), Rhizoma Zingiberis(生薑) and the changes of BP and rCBF were tested Leser-Doppler Flowmetry(LDF) The experimental results were as follows ; BP was not affected by SMT in rats, but rCBF was increased significantly by SMT in a dose dependent manner. SMT increased previous decreasd rCBF due to pretreatment methylene blue, but did not increased previous decreasd rCBF due to pretreatment L-NNA, indomethacin. Pretreatment with indomethacin decreased BP compaired with control group. These results suggest that SMT causes a diverse response of blood pressure and regional cerebral blood flow(rCBF). The increased rCBF is mediated by nitric oxide synthease.

Study on the Effect of Gamissanghwa-tang and each Medicinal Plant Extract for the Hair Growth of the Mice using In vivo and In vitro Test (In vivo와 In vitro 실험에서 가미쌍화탕 및 구성한약재가 마우스의 모발 성장에 미치는 실험적 연구)

  • To screen the effective materials for hair loss treatment, the Gamissanghwa-tang extracts were tested. As a result we found that the Gamissanghwa-tang extracts have the hair growth promoting effect. After topical application of each test materials to the back of CS7BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. In the experiments of 5α-reductase type II inhibition assay, Radix Paeoniae Alba, Semen Cuscutae showed effective potential to inhibit the activity of 5α-reductase type II. And hair growth index of the Gamissanghwa-tang extracts ranked as 1.2, especially the hair growth index of Fructus Rubi is highest as 1.8. But there were no plant extracts which have effect on the DNA proliferation of hair dermal papilla cell measured by [³H]thymidine incorporation, the expression of growth factors such as IGF-I, KGF, HGF estimated by RT-PCR and protein synthesis of vibrissae hair follicle measured by [/sup 35/S] cysteine incorporation. Cortex Cinnamomi showed anti-bacterial effect on P. ovale, Radix Paeoniae Alba has the highest radical scavening activity and Radix Glycyrrhizae has the highest effects of NO synthesis. These results suggest that Gamissanghwa-tang can be used as a potent treatment agent for helping hair growth stimulation.

Ability of Lipophilic Extract Obtained from Plants to Inhibit Tyrosinase Activity in Reverse Micelles (역미셀계를 이용한 지용성 식물체 추출물의 tyrosinase 저해효과 분석)

  • The abilities of petroleum ether-extracts prepared from 75 plants to inhibit tyrosinase activity were evaluated in reverse micelles composed of isooctane/AOT(100 mM)/phosphate buffer(20 mM, pH 8.0) containing tyrosinase(105.3 units/mL) and 3,4-dihydroxyphenylalanine(0.18 mM). Compared with control which has no plant extracts, garlic could completely inhibit in vitro melanogenesis by tyrosinase, and Chinese quince, sweet potato, onion, radish bud and apple did more than 60%. Lipophilic extracts of medicinal plants and herbs such as rosemary, coriander, cinnamomi ramulus, crataegii fructus, ramulus biotae folium, mume fructus, menthae herba, eucommiae cortex and clove also inhibited tyrosinase activity more than 60%. When the extraction yield of lipophilic materials was considered together with their inhibition effect on tyrosinase, it was possible to select plants of which tyrosinase inhibitors could be produced in high quantity from unit weight. Using reverse micelles, the analysis of the capacity of lipophilic materials to inhibit tyrosinase activity which was difficult up to present could be possible.
