The Food and Agriculture Organization (FAO) has been estimating the potential of insects as human food and animal feed for convincing food security since 2010. On account of this reason, Tenebrio molitor larvae have been gaining considerable attention as an alternative protein source for various foods. However, consumers do not prefer them because of their disgusting appearance. Therefore, it is necessary to develop a method that improve their appearance as to lead to T. molitor larvae consumption. In this study, T. molitor larvae which is prepared for optimally sampling conditions (shade drying, 30 min boiling after hot air drying, 30 min steaming after hot air drying), were cultured with Cordyceps militaris mycelia, and measured growth and density of mycelial. Also analyzed the cordycepin and adenosine content compared with commercial C. militaris. As a result, T. molitor larvae cultivated with C. militaris mycelia showed the highest cordycepin content (13.75 mg/g) was observed in optimal sampling conditions (shade drying). Therefore, we report that the methods which T. molitor larvae cultivated with C. militaris mycelia not only improve their appearance but also increase cordycepin content, which can be contributed to lead T. molitor larvae consumption.
Cordyceps cardinalis was reported in Japan and the USA in 2004, and its fruiting bodies have recently been cultured in Korea. Herbarium specimens preserved at the Cordyceps Research Institute, Mushtech, Korea were revised and identified as C. cardinalis, based on morphological characters and conidial structures. Most of the C. cardinalis specimens were collected from Mt. Halla in Jeju-do. The effects of various nutritional sources and environmental conditions such as temperature and pH on mycelial growth of C. cardinalis were studied. Oatmeal agar, Martin's peptone dextrose agar, and Schizophyllum (mushroom) genetics complete medium plus yeast extract resulted in the best mycelial growth. Among carbon sources, cereals, and nitrogen sources, maltose, oatmeal, and peptone resulted in the best mycelial growth respectively. Mineral salts helped to increase growth rate but only resulted in thin mycelial density, similar to water agar. A temperature of $25^{\circ}C$ and a pH of 7 resulted in the highest mycelial growth. Based on these results, a Cordyceps cardinalis composite medium (CCM) was formulated with 1% maltose, 2% oatmeal, 1% peptone, and 2% agar. Use of the CCM resulted in slightly better mycelial growth than that of other commonly used agar media. Only organic nitrogen sources imparted a reddish pigmentation to the agar media, but this character diminished after several subcultures. A 7 day culture duration resulted in the best mycelial growth.

Light is an important factor for cordycepin production in Cordyceps militaris. We investigated the effects of different light-emitting diode (LED) conditions including various LED wavelengths and their combinations on cordycepin production in Cordyceps militaris cultivated in submerged culture. The results of our study showed that the combinations of LED wavelengths were more beneficial than single LED sources for cordycepin production. Among the three tested wavelength combinations, the greatest effects for cordycepin production were observed for the red:blue light combination at the wavelength ratio of 5:5 or 3:7. The optimal culture conditions were 19.2278 h/day of illumination time; 9.19497 g/50 mL of glucose content in the media; and 53.112 h of cultivation time. Our model predicted a maximum yield of 2860.01 ㎍/mL cordycepin. Finally, to verify the calculated maximum, we performed experiments in the culture media representing the obtained optimum combination and the cordycepin yield of 2412.5 ㎍/mL.

Cordyceps is a traditional Chinese medicinal herb well-known in China, Korea and Japan since B.C. 2,000. The original entomopathogenic fungus, Cordyceps sinensis belonging to the genus Cordyceps could not be found inside Korean peninsula due to the absence of the host insect for the corresponding entomogenous fungus. The development of artificial production methods of Korean type Cordyceps using the silkworm Bombyx mori as in vivo culture medium for the the entomopathogenic fungus Paecilomyces tenuipes is the first, and wonderful occasion in the research history of insect industry of this global world. The aim of this article is to review the historical research background, mass-production methods, and pharmacological effects of the silkworm-dongchunghacho (Paecilomyces tenuipes) which is a newly developed Korean medicinal insect-borne mushroom, and another non-insect-borne medicinal mushroom (Cordyceps militaris and Cordyceps pruinosa). Their biological actions include anti-tumor, immunostimulating, anti-fatigue, anti-stress, anti-oxidant, anti-aging, anti-diabetic, anti-inflammatory, anti-thrombosis, hypolipidaemic and insecticidal effects. The bioactive principles are protein-bound polysaccharides (hexose, hexosamin), cordycepin, D-manitol, acidic polysaccharide etc. Protein-bound polysaccharides and n-butanol fractions were demonstrated to show a significant anti-tumor activities but did not show a cytotoxicities. D-mannitol exhibited a significant prolongation of the life span in tumor bearing mice. Ergosterol did not show an efficient anti-tumor activity, but showed a significant phagocytosis enhancing activity. Anti-tumor activity of silkworm-dongchunghacho might be attributed to immuno-stimulating activities rather than cytotoxic effects [164]. Also this review comprises the breeding of Dongchunghacho varieties, optimization of culture conditions, improvement of learning and memory by Dongchunghacho, application of them as foods and chemical constituents.

Objective: This study was aimed to investigate the effect of fresh and dried hydrolyzed Cordyceps militaris (CM) mushroom with proteolytic enzymes; bromelain (CMB), flavorzyme (CMF), and mixture of bromelain: flavorzyme (CMBF) on quality properties of spent hen chicken. Methods: Mushroom extract (CME) were combined with three proteolytic enzyme mixtures that had different peptidase activities; stem bromelain (CMB), flavorzyme (CMF), and mixture of stem bromelain:flavorzyme (CMBF) at (1:1). The effect of these hydrolysates was investigated on spent hen breast meat via dipping marination. Results: Hydrolyzation positively alters functional properties of CM protease. in which bromelain hydrolyzed group (CMB) displayed the highest proteolytic activity at 4.57 unit/mL. The antioxidant activity had a significant increment from 5.32% in CME to 61.79% in CMB. A significantly higher emulsion stability index and emulsification activity index compared to CME were another result from hydrolyzation (p<0.05). Texture properties along with the shear force value and myofibrillar fragmentation index were notably improved under CMB and CMBF in fresh condition. Marination with CM mushroom protease that was previously hydrolyzed with enzymes was proven to also increase the nucleotide compounds, indicated by higher adenosine 5'-monophosphate (AMP) and inosine 5'-monophosphate (IMP) in hydrolysate groups (p<0.05). The concentration of both total and insoluble collagen remained unchanged, meaning less effect from CM protease. Conclusion: This study suggested the hydrolyzation of CM protease with bromelain or a mixture of bromelain:flavourzyme to significantly improve functional properties of protease and escalate the taste-related nucleotide compounds and texture profiles from spent hen breast meat.
Mushroom is known for anti-inflammatory and anti-oxidative potential. This study provides evidence that the inhibitory effect of mushroom on the expression of pro-inflammatory cytokines in human keratinocytes, HaCaT cells. To define the underlying mechanisms of action, tumor necrosis factor${\alpha}$/$IFN{\gamma}$-activated human keratinocytes model was used. Mushroom significantly inhibited the expression of cytokines in HaCaT cells. Taken together, the results demonstrate that mushroom inhibited inflammtion, suggesting that mushroom (DW extract: Grifola frondosa Cordyceps militaris), (Ethanol extract: Ganoderma lucidum, Lentinus edodes, Cordyceps militaris, Flammulina velutipes) might be a candidate for the treatment of skin inflammation.
Mushroom jellies using extracts of Garnoderma lucidum, Lentinus edodes, Pacilomyces ten tenuipes and Cordyceps militaris were prepared, and Investigated the colors, texture and sensory characteristics of mushroom jellies. G. lucidum jelly mixed 85% mushroom, 10% jujube(Zizyphus jujuba Miller) and 5% hwanggi (Astragaslus membranaceus) extract, L. edodes jelly mixed 80% mushroom, 10% jujube, 5% gamcho( Glycyrrhiza uralensis) and 5% omija (Schizandrae chinensis Ruprecht) extract. and P. tenuipes and C. militaris jelly mixed 85% mushroom. 10% jujube and 5% gamcho extract were most effective in overall acceptability. The Jellying ability of carrageenan was better than other jelling agents. According to increase carrageenan content, color of mushroom Jellies were not effect. however hardness, gumminess and chewiness were increased. Sensory evaluation of mushroom Jellies were most preferable at the 0.6% carrageenan content.
This study was carried out to investigate anti-inflammatory effects of mushroom, Cordyceps militaris. Anti-inflammatory effects analysis was followed by peroxynitrite inhibition activity. Cordyceps militaris mushrooms extracts were screened about inhibition effects of nitric oxide for Raw 264.7 cell treated by lipopolisaccharide(LPS) and inhibition of cyclooxygenase-2(COX-2) for inflammatory effects. In our result, Cordyceps militaris mushrooms were good resource for anti-inflammatory effects and to be followed more research about related anti-inflammatory effects.
Cordyceps (vegetable wasp and plant worm), an entomopathogenic fungi, has been used as a herbal medicine in Asian countries since ancient times. Cordyceps nutans is common but there is little research on this species. This study investigated the optimal culture conditions of C. nutans and the inhibitory effect on nitric oxide (NO) production in RAW 264.7 cell treated culture broth. The optimal conditions for the mycelial growth were $25^{\circ}C$ and pH 7.0-8.0. Mycelial growth was highest on mushroom complete medium (MCM), V8 juice agar (V8A), and yeast malt dextrose (YMD) medium. Mycelial growth on mushroom minimal medium (MMM) did not occur, so nutrient source was essential. Dextrose and sucrose as carbon sources, and ammonium citrate as a nitrogen source were satisfactory for mycelial growth. Cytotoxicity of C. nutans culture broth was not found in RAW 264.7 cells. C. nutans culture broth suppressed NO production of lipopolysaccharide (LPS)-stimulated RAW 264.7 cell in a dose-dependent manner. Thus, our results provided the optimal conditions for cultivation of C. nutans and showed that C. nutans may have excellent physiological activities.
Interest in commercial cultivation and product development of Cordyceps species has shown a recent increase. Due to its biochemical and pharmacological effects, Cordyceps militaris, commonly known as orange caterpillar fungus, is being investigated with great interest. Cultivation of C. militaris has been practiced on a large scale in order to fulfill a demand for scientific investigation and product development. Isolates of C. militaris can be easily established from both spores and tissue. For isolation of spores, ascospores released from mature stromata are trapped in sterile medium. Multi-ascospore isolates, as well as combinations of single ascospore strains, are used for production of fruiting bodies. Progeny ascospore strains can be isolated from artificial fruiting bodies, thus, the cycle of fruiting body production can be continued for a long period of time. In this study, we examined fruiting body production from multi-ascospore isolates and their progeny strains for three generations. $F_1$ progeny strains generally produced a larger number of fruiting bodies, compared with their mother multi-ascospore isolates; however, $F_2$ and $F_3$ progeny strains produced fewer fruiting bodies. Optimum preservation conditions could help to increase the vitality of the progeny strains. In order to retain the fruiting ability of the strains, further testing of various methods of preservation and different methods for isolation should be performed.
