Screening New Antihypertensive Angiotensin I-Converting Enzyme Inhibitor -Producing Yeast and Optimization of Production Condition (항고혈압성 안지오텐신 전환효소 저해제를 생산하는 새로운 효모의 선별 및 저해물질 최적 생산조건)

  • Forty eight strains of yeast were cultured in potato dextorse(PD) broth at $30^{\circ}C$ for 24 hr and centrifuged with 12,000 rpm for 20 min. After concentrated the cultures, antihypertensive angiotensin I-converting enzyme(ACE) inhibitory activities of its concentrates were investigated. Among them, the concentrates from Saccharomyces cerevisiae Y183-3 showed the highest ACE inhibitory activity of 71.8%. The ACE inhibitor from Saccharomyces cerevisiae Y183-3 was maximally produced when Saccharomyces cerevisiae Y183-3 cultured in PD broth at $30^{\circ}C$ for 36 hr.

Physiological Characteristics and Angiotensin Converting Enzyme Inhibitory Activity of Lactobacillus brevis HLJ59 Isolated from Salted Shrimp (국내 새우젓에서 분리한 Lactobacillus brevis HLJ59의 Angiotensin Converting Enzyme 저해활성 및 생리적 특성)

  • In this study, lactic acid bacteria with high angiotensin converting enzyme inhibitor activity were isolated from Korean fermented food, such as kimchi and salted seafood. The strain HLJ59, isolated from salted shrimp showed the highest angiotensin converting enzyme inhibitor activity in DeMan Rogosa Sharpe broth. Optimum growth temperature of Lactobacillus brevis HLJ59 was at $34^{\circ}C$. Acid treatment at pH 3.0 for 1.5 h decreased cell viability from $9.9{\times}10^8$ CFU/ml to $3.11{\times}10^4$ CFU/ml. The bile extract concentration of 0.3%, 0.5%, and 1.0% in MRS broth did not inhibit the growth of HLJ59. Isolated strain HLJ59 showed more sensibility to amikacin, gentamycin, neomycin, streptomycin, kanamycin, cefmetazole, cephalothin, ampicillin, ticarcillin, sulbactam+ampicillin, amoxicillin+clavulanic acid (AMC), tetracycline, and sulfamethoxazole+trime thoprim (SXT) as compare to other 7 different antibiotics. However, it showed more resistance to cefoxatin, ceftnaxone, penicillin, ciprofloxacin, nalidixic acid, lincomycin, and chloramphenicol.

Functionality and Inhibitory Effect of Soybean Hydrolysate on Angiotensin Converting Enzyme (대두 가수분해물의 혈압 강하 효과 및 기능성)

  • This studies were conducted to select optimal enzyme that produced hydrolysate from soybean, and to evaluated functionality of hydrolysate. Soybean powder was suspended with water and hydrolyzed by seven commercial proteases. Hydrolysate produced with protease from Bacillus subtilis showed the highest inhibition effect on the activity of angiotension converting enzyme(ACE), and the condition of enzymatic hydrolysis was 5cA substrate concentration, 0. l% enzyme concentration, 4 hour hydrolysis time. Under above optimum condition, soybean was hydrolyzed with protease from Bacillus subtilis yielding a DH (degree of hydrolysis) of about 49%. Hyrophobicity of hydrolysate was not correlated with the inhibition effect on ACE activity. The functionality of hydrolysate was significantly influenced by pH. Solubility of hydrolysate at alkali solution was greater than that at acidic solution.

  • A new set of functional group interaction energy descriptors relevant to the ACE (Angiotensin-Converting Enzyme) inhibitory peptide, QSAR (Quantitative Structure Activity Relationships), is presented. The functional group interaction energies approximate the charged interactions and distances between functional groups in molecules. The effective energies of the computationally derived geometries are useful parameters for deriving 3D-QSAR models, especially in the absence of experimentally known active site conformation. ACE is a regulatory zinc protease in the renin-angiotensin system. Therapeutic inhibition of this enzyme has proven to be a very effective treatment for the management of hypertension. The non bond interaction energy values among functional groups of six-feature of ACE inhibitory peptides were used as descriptor terms and analyzed for multivariate correlation with ACE inhibition activity. The functional group interaction energy descriptors used in the regression analysis were obtained by a series of inhibitor structures derived from molecular mechanics and semi-empirical calculations. The descriptors calculated using electrostatic and steric fields from the precisely defined functional group were sufficient to explain the biological activity of inhibitor. Application of the descriptors to the inhibition of ACE indicates that the derived QSAR has good predicting ability and provides insight into the mechanism of enzyme inhibition. The method, functional group interaction energy analysis, is expected to be applicable to predict enzyme inhibitory activity of the rationally designed inhibitors.

  • A peptide that inhibits angiotensin-converting enzyme (ACE) was isolated from a hydrolysate of Manila clam (Ruditapes philippinarum) proteins prepared with thermolysin. Amino acid sequence of the peptide was determined to be Leu-Leu-Pro. Chemically synthesized Leu-Leu-Pro had an $IC_{50}\;value\;of\;158\;\mu{M}$. Peptides related to the Manila clam-derived peptide were synthesized to study the structure-activity relationships. The tetrapeptide, Leu-Leu-Pro-Pro, had a very weak effect on the enzyme. However, Leu-Leu-Pro-Asn showed no inhibitory activity.

Effects of Herb Medication on Dry Cough, a Common Side Effect Caused by Angiotensin Converting Enzyme (ACE) Inhibitor (한약치료가 Angiotensin Converting Enzyme Inhibitor를 복용중인 고혈압환자에게 부작용으로 나타나는 건해(乾咳)에 미치는 영향)

  • Objectives: In order to confirm a remedial effect and related influence of the clinic treatment by prescribing herb medicines to hypertensives experiencing angiotensin converting enzyme (ACE) inhibitor dosage and suffering from common side effect generally known as dry cough. Methods: 1. We selected the 19 patients who visited National Oriental Medical Center, from August 21, 2007 to August 16, 2008 and suffering from dry cough caused by taking ACE inhibitor, with no other possible diseases causing dry cough. 2. We separated the 19 patients into two groups (Type 1: Bi-Qi hie (脾氣虛) group prescribed Samchuljojung-tang & Type 2: Qi-hie dam-wul (氣虛痰鬱) group prescribed Samsoumgamibang). 3. We then observed the symptom level and post-treatment effect, and recorded changes of dry cough intensity level for each group. Results: 1. Type 1: In the survey of 12 patients, initial level recorded 16.33 at entry diagnosis, and next level meant changing of symptoms, recorded as 2.75 at Stage 1 and reaching 3.33 at Stage 2. 2. Type 2: 7 patients, with initial level recorded as 18.71 at entry diagnosis, and 1.86 at Stage 1 and reaching to 3.29 at Stage 2. 3. No additional prescriptions were issued at Stage 2 or afterwards, and final result indicates that the mean value ended at 3.95 in the total group. Conclusions: It is concluded that there is a significant remedial effect and related influence of the clinic treatment between the Oriental medicine treatment and one of the common side effects of ACE inhibitor, dry cough.

  • Chunkookjang, Korean traditional fermented soybean food emerges as a functional food to improve intestinal function and blood circulation. During Chungkookjang fermentation, microorganisms, enzymes, and diverse bioactive compounds increase sharply. Chungkookjang contains diverse oligo-peptides. Formation of peptides was confirmed by SDS-PAGE. Solube fermented soybean in our sample contained Tyr, Gln-Lys, Trp, Gln, and Lys-Pro as major components. Lys-Pro (0.083 mg/100 g sample) was purified by HPLC analysis. Angiotensin I­converting enzyme (ACE) causes hypertension by converting angiotensin I to angiotension II. ACE inhibitory activity of Lys-Pro was determined to be $IC_{50}=32.1\;{\mu}M.$ Whether or not eating Chungkookjang can lower blood pressure was also determined. Sistolic blood pressure dropped by 15 mmHg, and diastolic blood pressure by 8 mmHg 2 hr after a single administration of 20 g of fermented soybean. Chungkookjang might be helpful in improving blood circulation since it has ACE inhibitor and antihypertenisve effect.
