A Stereochemical Aspect of Pyridoxal 5' -Phosphate Dependent Enzyme Reactions and Molecular Evolution

  • We have studied the stereospecificities of various pyridoxal 5'-phosphate (PLP) dependent enzymes for the hydrogen transfer between the C-4' of a bound coenzyme and the C-2 of a substrate in the transamination catalyzed by the enzymes. Stereospecificities reflect the structures of enzyme active-sites, in particular the geometrical relationship between the coenzyme-substrate Schiff base and the active site base participating in an $\alpha$-hydrogen abstraction. The PLP enzymes studied so far catalyze only a si-face specific (pro-S) hydrogen transfer. This stereochemical finding suggests that the PLP enzymes have the same topological active-site structures, and that the PLP enzymes have evolved divergently from a common ancestral protein. However, we found that o-amino acid aminotransferase, branched chain L-amino acid aminotransferase, and 4-amino-4-deoxychorismate lyase, which have significant sequence homology with one another, catalyze a re-face specific (pro-R) hydrogen transfer. We also showed that PLP-dependent amino acid racemases, which have no sequence homology with any aminotransferases, catalyze a non-stereospecific hydrogen transfer: the hydrogen transfer occurs on both faces of the planar intermediate. Crystallographical studies have shown that the catalytic base is situated on the re-face of the C-4' of the bound coenzyme in o-amino acid aminotransferase and branched chain L-amino acid aminotransferase, whereas the catalytic base is situated on the si-face in other aminotransferases (such as L-aspartate aminotransferase) catalyzing the si-face hydrogen transfer. Thus, we have clarified the stereospecificities of PLP enzymes in relation with the primary structures and three-dimensional structures of the enzymes. The characteristic stereospecificities of these enzymes for the hydrogen transfer suggest the convergent evolution of PLP enzymes.

Self-Organization of Dendron-Poly(ethylene glycol) Conjugates in an Aqueous Phase

  • We have prepared amide dendrons having alkyl peripheral units and various focal moieties through a convergent synthetic approach. The amphiphilic properties, due to hydrophilic amide branches and the hydrophobic peripheral units, provide an opportunity for the amide dendrons to self-organize in water. The dendritic architecture itself is also one of the critical factors in the self-organization of the amide dendrons in water. In particular, function-alization was performed at the focal point to elucidate the relationship between the focal functionality and the self-organized structures of the dendritic building blocks in the aqueous phase. The dendron having a short poly(ethylene glycol) monomethyl ether (MeO-PEG) unit (M$\_$n/ =750) as the focal moiety formed a vesicular organization in water. As the size of the hydrophilic focal MeO-PEG increased to M$\_$n/ =2,000 and 5,000, the self-organized structures became rod-type and spherical micelles, respectively. Our observation of multiple morphologies for amide dendrons is in good agreement with previous reports that indicated that the micellar structures changed from vesicles to rod-types and then to spheres upon increasing the size of the hydrophilic moiety of the amphiphiles.

  • This study was conducted to investigate the effects of communication competence and interpersonal understanding on empathy of nursing students who have experienced fundamental nursing skills course. The subjects of this study were 122 second-year nursing students in a university. Data were collected using a questionnaire from September to December 2018. The data were analyzed by descriptive statistics, correlations and hierarchical regression using the SPSS 22.0 program. Factors affecting empathy were active participation in the practice, communication skills, and understanding of interpersonal. Therefore, In order to improve the empathy of nursing students, it is necessary to introduce a step-by-step empathy education program into the curriculum that can contribute to establishing a device that can induce active participation in practice and contribute to communication and interpersonal relationships.

Convergent relationship between functional oral health literacy, oral health knowledge and oral health behavior of some university students (일부 대학생의 기능적 구강건강문해력과 구강보건지식 및 구강보건행태에 관한 융합적 관계)

  • The research had an investigation of relevance between functional oral health literacy, oral health knowledge, and oral health behaviors of college students. Survey of 410 students in K city college done from June 22 to 26, 2015 was analyzed, and the final analysis subject was 398 students. The result were 281(70.6) students with experience of oral health education, 117(29.4) students without experience of education, and there were 194(48.7) students who had difficulty several times with the material and explanation while attending the education, and 128(32.2) students who had difficulty a few times. Scores of functional oral health literacy were low for more than half of the college students. The higher the functional oral health literacy score, the higher oral health knowledge was shown, and oral health knowledge turned out to be affecting functional oral health literacy. Therefore, an effective program for delivering oral health knowledge to improve low functional oral health literacy of college students must be developed.

  • The environment of the food service industry is becoming increasingly complicated and unpredictable as modernization and industrialization become more and more common. Moreover, the development of the food service industry is rapidly changing due to the convergence of the inter-industry services. In this study, based on the high reliance on the human resources of the food service industry, it investigates relations of each component as integrated view through leadership, organizational performance, and organizational culture. In particular, the relationship between emotional/primal leadership and organizational performance(as job satisfaction and organizational commitment) was verified through mediating effects of organizational culture. The results of this study will provide an integrated perspective on the understanding and development of the food service industry structure, which requires the importance of human relations, and will help convergent thinking among various service industries.

Psychometric Evaluation of a Need Scale for Cancer Patients Undergoing Follow-up Care (추후관리 받는 암환자의 요구 측정도구 개발 및 평가)

  • Purpose: The purpose of this present study was to develop and evaluate the psychometric properties of a needs scale for patients with cancer undergoing follow-up care (NS-C). Methods: A preliminary NS-C of 48 was derived from literature reviews and in-depth interviews with patients with cancer. Content validation of the items was established by oncology physicians and nurses. Each item was scored on a five-point Likert scale. The preliminary NS-C and Eastern Cooperative Oncology Group (ECOG) performance status questionnaires were administered to 873 patients with cancer recruited from three university hospitals. The data were analyzed using factor analysis, multidimensional scaling analysis, ANOVA, Pearson correlation coefficients, and Cronbach's alpha. Results: From the factor analysis, 25 significant items in six subscales were derived. The subscales were named physical symptoms, diet and exercise, support, relationship with health professionals, treatment/prognosis, and keeping mind under control. The NS-C also established item convergent and discriminant validity, and known-groups validity. Cronbach's alpha of the subscales ranged from .90 to .92. Conclusion: This study suggests that the NS-C is an easy, reliable and valid instrument to measure the needs of patients with cancer. Health professionals may use the NS-C for patients with cancer both in practice and research.

Comparison Analysis on Operation Characteristics of Design Consultancies between Korea and America (한국과 미국의 디자인컨설팅기업의 운영현황 비교 분석)

  • This study aims to consider the action plans to make the industry of Korean design consultancies accomplish qualitative growth by comparing the operation characteristics of design consulting firms between Korean design firms and American ones. For this, questionnaire survey and comparison analysis was performed for the operation characteristics of 109 Korean design firms and 97 American ones. The analysis results shows that operation characteristics of Korean design firms comparing to those of American ones keep low in all 7 areas of building long-term partnership relationship, building multi- disciplinary organization, utilizing result-based compensation system, developing design at the strategic business level of clients, executing active design business, making convergent networking with other industry and performing specialized design services. In order to accomplish stable industry infrastructure of design consulting industry with qualitative growth, it requires continuous effort to execute 7 operation characteristics of Korean design firms.

Convergent Relationship between of Positive Psychological Capital on College Life Adaptation in Freshmen of Dental Hygiene Department (치위생(학)과 신입생의 긍정심리자본과 대학생활 적응과의 관련성에 대한 융합적 연구)

  • This study aimed to investigate the effects of positive psychological capital on college life adaptation in freshmen of dental hygiene department of universities in Daegu, Busan and Ulsan. The major variables including hope, resilience, optimism, self-efficacy, academic adaptation, social adaptation, personal emotional adaptation, and university environment adaptation all showed significant positive correlations. Among the influential factors of positive psychological capital on college life adaptation, academic adaptation and social adaptation showed significant differences in optimism(p<.001) and self-efficacy(p<.001), while personal emotional adaptation and university environment adaptation showed significant differences in optimism(p<.001). Therefore, it is necessary to develop intervention programs that can improve positive psychological capital and apply them systematically to the educational field.

Validity and Reliability of the Perceived Elderly Stigma Scale and the Relationships between the Stigma and Demographic Factors (지각된 노인 낙인 척도의 타당도 검증 및 인구통계학적 특징에 따른 낙인 인식)

  • This study examined the validity and the reliability of the perceived elderly stigma scale. Moreover, we investigated the relationships between the perceived elderly stigma and demographic factors (age, gender, education, and income). The initial scale consisted of 5 factors (ability, personality, appearance, authoritarian dependency, and child-obsession) and 28 items. In order to test the validity and the reliability of the scale, this study conducted a survey with 693 adults aged 18 to 65. Based on the confirmatory factor analysis (CFA), the scale was modified to 5 factors (ability, personality, appearance, authoritarian dependency, and child-obsession) and 15 items and its convergent validity and discriminant validity were confirmed. In terms of the relationships between the perceived elderly stigma and demographic factors, age had a negative relationship with the "ability" stigma, and education was a negatively associated with the "appearance" stigma.

The Convergent Influence of MultiCultural Acceptability, Empathy and Global Citizenship in Nursing Students (간호대학생의 다문화수용성, 공감능력이 세계시민의식에 미치는 융합적 영향)

  • This study is a descriptive survey study intended to analyze the relationship between multicultural acceptability, empathy, and global citizenship of nursing students and identify the factors influencing global citizenship. The data was collected targeting 187 nursing students at universities located in the cities of B, U, and S. The collected data was analyzed using t-test, ANOVA, Pearson correlation coefficients, and stepwise multiple regression analysis. The results were as follows: global citizenship of nursing students was positively correlated with multicultural acceptability and empathy. Factors influencing the level of global citizenship were empathy, multicultural acceptability, which together explained 40.0% of the total variance in global citizenship. Consequently, this study raises the need for the development and application of steady practical programs and various learning methods for the enhancement of global citizenship, which is required in the fields of nursing education where global competence is considered to be important.
