체세포 복제 한우 수소의 정액 성상, 정자의 활동성 및 수정 능력 분석 (Analysis of Semen Parameters, Sperm Activity, and Fertility of Somatic Cell Cloned Hanwoo Bulls)

  • 본 연구는 체세포를 이용하여 생산된 복제 한우 수소의 번식능력을 검토하기 위해 실시하였다. 복제 한우 수소(C-38 및 C-39) 또는 일반 한우 종모우로부터 정액을 채취하여 정자의 수 및 동결 전 후의 생존성 등을 살펴보았으며, 정자의 운동성 등은 computer assisted sperm analysis(CASA)를 이용하여 측정하였다. 또한, 이들의 수정 능력을 확인하기 위하여 체외수정과 인공수정을 각각 실시하였다. 정액 성상에서는 복제 수소들과 일반 종모우 간에 정액의 양, 정자의 농도 및 동결융해 후의 생존성 등에서 차이가 나타나지 않았다. CASA를 이용한 분석에서 운동성, 곡선 운동 속도(VCL), 직선운동 속도(VSL) 및 평균 진행 속도(VAP) 등은 복제 수소의 정액이 일반 종모우의 정액에 비하여 유의적으로 높았다 (p<0.05).체외수정에 따른 수정란의 분화율 및 배반포로의 발달율은 복제 수소와 일반종모우 간에 차이가 나타나지 않았다. 복제소 정액(C-38)을 이용하여 인공수정을 한 5두의 체세포 복제 대리모에서 암 수 각각 한 두씩의 건강한 복제 후대 송아지 2두를 생산하였다. 이상의 결과를 종합하여 보면, 실험에 공시된 복제 수소 개체간의 차이가 나타나기는 하였지만, 복제 수소는 정액 성상과 정자의 운동성 등에서 일반 종모우와 차이가 없었으며, 또한 인공수정을 통해 송아지를 생산함으로써 정상적인 번식능력이 있음을 확인하였다.

정자 운동성 및 수명 보존을 위한 최적 배양에 관한 연구 (Optimized Methods to Maintain Motility and Viability in Normozoospermic Males)

  • 목 적: 본 연구는 정상 정액을 배양액으로 세척한 후 $4^{\circ}C$, $22^{\circ}C$, $37^{\circ}C$에서 5일 동안 보존하면서 정자의 운동성, 생존성을 관찰하여 정자의 운동성과 수명 유지를 위한 적정 배양 환경을 분석하고자 하였다. 연구방법: 정액검사 시 정상으로 판정된 남성의 정자를 HTALP 배양액으로 세척하여 정자의 최종 농도 $1{\times}10^6/ml$로 각 5 ml을 배양온도 $4^{\circ}C$, $22^{\circ}C$, $37^{\circ}C$에서 5일 동안 배양하였다. 1일, 3일 5일째에 CASA에 의해 운동성을 측정하였고, HOST로 정자막의 온전성을 분석하였으며, CTC pattern으로 수정능획득 상태 분석하여 최적의 배양 환경을 분석하였다. 결 과: 정자의 운동성, 생존성 및 수정능획득이 야기되지 않은 정자는 배양일에 따라 유의하게 감소하였다 (p<0.05). 또한 정자 배양 후 1일에는 정자의 운동성, 생존성 및 정자막의 온전성과 CTC pattern은 온도에 따라 차이가 없었으나, 배양 후 3일과 5일에서는 $22^{\circ}C$에서 배양된 정자가 다른 배양온도에 비해 가장 잘 보존되었다 (p<0.05). 결 론: HTALP로 세척된 정자를 $22^{\circ}C$에서 보존 시 5일까지 정자의 운동성과 수명을 보조생식술에 적합한 수준으로 유지시킬 수 있는 최적 배양 환경으로 제시할 수 있다.

돼지 정자의 운동역학 변수와 산자수 간의 상관 관계 (Correlation of Motion Kinematics of Spermatozoa and Litter Size in Porcine)

  • Male factor infertility or sub-fertility contributed half of all cases of infertility while the semen abnormality is the current topic of argument. Conventional analysis of semen showed poor correlation with fertility. Therefore, evaluation of current semen analysis method is necessary to improve standards of semen assessment. The goal of this study was to investigate that correlation between motion kinematic before and after capacitation and litter size in porcine. Sperm motility and kinematics were measure by computer-assisted sperm analysis (CASA). The motility of spermatozoa was positively correlated with curvilinear velocity (VCL), average path velocity (VAP), and mean amplitude of head lateral displacement (ALH) (p<0.05). Where as VCL positively correlated with VSL, VAP and ALH (p<0.01). Straight-line velocity (VSL) was positively correlated with VAP and ALH (p<0.01). VAP was significantly positively correlated with ALH (p<0.01). Also, we found significant positive correlation among variation of VSL, VAP and ALH (p<0.05). No motility and kinematic parameter are correlated with litter size. However, litter size was significantly correlated with breed (p<0.05). Our results suggested that analysis of sperm motility and kinematics using CASA is questionable for prediction of litter size. However, it has some practical importance to evaluate semen commercially.

뱀장어(Anguilla japonica) 정자 활성에 따른 정액 분석 및 인공정장액 개선 (Semen Analysis and Improvement of Artificial Seminal Plasma According to Sperm Activity in Eel Anguilla japonica)

  • This study examined the characteristics of eel Anguilla japonica sperm using the CASA (computer-assisted sperm analysis) system and attempted to provide the composition for artificial seminal plasma by regulating of inorganic elements. Sperm samples were first divided into four groups based on motility and progressive motility: (A) 0-10%, (B) 10-40%, (C) 40-70%, and (D) 70-100%. For observing sperm velocity variations, VCL, which is curve motion velocity, showed the highest values in all groups. The directional factor, beat cross frequency, was lower in higher activity groups, showing an opposite correlation with sperm activity. The head sizes of spermatozoa in higher activity groups were significantly larger than those in lower activity groups. The Na+ and K+ ions were important in the inorganic composition of seminal plasma in this species. Furthermore, regulating the composition in artificial seminal plasma improved the formula compared to the existing element, exhibiting 120 mM Na and 30 mM K when the sperm was conserved for a long time and 120 mM NA and 40 mM K when the sperm was conserved for a short time.

Effects of Different Concentrations of Escherichia coli and Days of Preservation on Boar Sperm Quality

  • The objective of this study was to investigate the effect of bacterial contamination on elapsed time after preservation on boar semen. Known numbers of Escherichia coli (E. coli) were inoculated to freshly ejaculated semen and sperm parameters such as viability, motility, agglutination, acrosome integrity and hypo-osmotic swelling test were performed during 7 days of liquid preservation. Semen samples were prepared using antibiotic free BTS extender and 4 different levels of E. coli were treated to semen with following concentrations; 3,000, 5,000, 7,000, 10,000 CFU/ml of sperms. Semen samples were preserved at $17^{\circ}C$ for 7 days in semen storage until analyzed. Aliquots were subjected to measure the sperm viability, motility and agglutination using computer assisted sperm analysis (CASA) system, acrosome integrity was performed using chlortetracycline (CTC) staining method and hypo-osmotic swelling test was performed using hypotonic solution from day 1 (day of semen collection) to 7. Detrimental effects on sperm motility and viability were observed 3 days after preservation at the level of 5,000 CFU/ml (p<0.05). Percentage of sperm abnormality was higher (p<0.05) in over 5,000 CFU/ml groups. Sperm agglutination rate was also significantly higher (p<0.05) in groups of 5,000 and 7,000 CFU/ml. The rate of acrosome reacted sperm was higher as preservation time goes in all the samples but the pattern was clearly higher among E. coli contaminated groups (p<0.05). The sperm membrane integrity in terms of hypo-osmotic test, E. coli affects little compared to other sperm parameters. The deleterious effects observed due to the bacterial contamination in semen suggest that importance of hygiene protocol to minimize the bacterial contamination during semen collection and processing.

Pentoxifylline treatment of frozen pig sperm affects sperm motility and fetal numbers

  • The objective of this study was to investigate whether supplementation of pentoxifylline (PTX; phosphodiesterase inhibitor) to thawed boar semen improves the post-thaw motility of sperm and affects the efficiency of artificial insemination (AI) and further development. To determine the concentration of PTX for AI, frozen-thawed semen was incubated with 0, 5, 10, and 20 mM PTX in an extender freezing medium, respectively, after thawing. Kinematic properties of sperm were examined with a computer-assisted semen analysis (CASA) system. In addition, viability and mitochondrial activity were also tested by LIVE/DEAD and a MitoTracker kit. There were no significant differences in the kinetic parameters of thawed sperm between control and treatment groups, but overall assessment parameters such as motility and rapid progressive were higher in the 10 mM PTX group. In the viability and mitochondrial assay, there were no significant differences observed in the PTX treatment, compared to the control. For further analysis, artificial inseminations were performed using frozen semen and 10 mM PTX treated cryopreserved semen, respectively. There were no differences in pregnancy rates and fetus weights among the groups until 30 and 40 days, but litter size was reduced and relatively low-birth weight was observed in the PTX group. In summary, our findings suggest that enhancement of in vitro sperm quality or non-toxicity supplemented by PTX may have detrimental effects on fetus development.

불임시술의 전환이 정맥상태에 미치는 영향 (Effects of Conversion of Infertility Treatment on Semen Quality)

  • 목 적: 불임 부부에서 자궁강내 정자주입술 시행 후 연속하여 시행한 자궁강내 정자주입술 및 전환하여 시행한 체외수정시술이 정액검사소견에 미치는 영향을 알아보고자 하였다. 연구방법: 진단적 목적으로 컴퓨터정액분석기를 이용한 정액검사 (computer-assisted semen analysis (CASA))를 시행한 후 자궁강내 정자주입술을 시행하였으나 임신에 실패한 53예를 대상으로 하였다. 자궁강내 정자주입술 시행 후 38예의 환자에서는 두 번째 자궁강내 정자주입술을 시행하였고 (Group 1), 15예의 환자에서는 체외수정시술을 시행하였다 (Group 2). 이들에서 두 번째 불임시술을 위해 채취한 정액의 양 (volume), 정자의 농도 (concentration), 운동성 (motility), 총운동성정자수 (total motile sperm count) 등의 컴퓨터정액분석소견을 이전에 자궁강내 정자주입술 시 시행한 정액검사소견과 비교하였다 결 과: 대상 불임 부부간의 남편 연령, 첫 번째 시술과 두 번째 시술 사이의 시간 간격, 불임의 원인 등에는 유의한 차이가 없었다. Group 1에서 첫 번째 시행 시의 정액검사소견의 정상 여부와 관계없이, 정자의 운동성은 첫 번째 자궁강내 정자주입술 시행 시보다 두 번째 자궁강내 정자주입술 시행 시에 통계적으로 유의하게 감소하였다. Group 2에서는 첫 번째 자궁강내 정 자주입술 시보다 체외수정시술 시 정자의 농도, 정자의 운동성, 총운동성정자수에서 통계적으로 유의하게 감소하였다. Group 2의 하위그룹 분석에서는 자궁강내 정자주입술 시행 시 정상 정액소견을 보인 경우는 체외수정시술 시 정자의 농도와 총운동성정자수가 유의하게 감소하였으나, 자궁강내 정 자주입술 시행 시 비정상 정액소견을 보인 경우에는 체외수정시술 시 정자의 운동성이 유의하게 감소하였다. 결 론: 자궁강내 정자주입 술 시행 후 임신에 실패하여 체외수정시술을 시행하게 된 환자의 정액검사소견은 자궁강내 정 자주입술을 계속 시행 받은 환자군에 비해 더 악영향을 받는다. 그 이유를 설명하기 위해 심리적 스트레스에 대한 후속적인 연구가 필요할 것으로 사료된다.

Correlations between Sperm Motility, SCSA (Sperm Chromatin Structure Assay), Reproductive Performance and Heterospermic Fertility in Boars

  • The objective of this study was two folds: to investigate the relationship between paternal identification rate and sperm quality parameters such as motility and sperm chromatin structure assay after heterospermic insemination; to see if mutual complement between tests and development of useful technique to enhance the fertility in artificial insemination. In individual boar's fertilizing ability, 3 high fertility boars showed significantly high fertility (p<0.05) compared to 3 low fertility boars, but there was no difference in litter size between two groups. Sperm motility test in pooled and individual semen using computer assisted sperm analysis (CASA) revealed that no significant difference among boars. The high fertile boar showed tendency of low %Red (High red fluorescence/green+red fluorescence) in sperm chromatin structure assay (SCSA) but paternal identification rate from piglets did not differ after heterospermic insemination. The correlation coefficient between individual or pooled semen function test and farrowing rates were well correlated as follows: %Red with litter size (r= - 0.53, p=0.03); %Red with paternal identification rates (r=-0.51, p=0.03); paternal identification rates with litter size (r=0.57, p=0.02). These results indicate that sperm chromatin structure assay and sperm quality parameter test in pooled semen are useful method to predict and evaluate the fertilizing capacity after heterospermic insemination in boars.

동결보존액에 울금에서 추출한 Curcumin의 첨가가 정자의 운동성과 활성산소 생성에 미치는 영향 (Effects of Curcumin from Turmeric Supplementation in Freezing Buffer on Sperm Motilities and Reactive Oxygen Species Generation)

  • In this experiment, we determined the effect of curcumin supplementation in freezing buffer for miniature pig sperm cryopreservation. Each ejaculate was diluted with modified Modena B extender and mixed with lactose-egg yolk (LEY extender, 80% v/v lactose solution [310 mM], 20% v/v egg yolk, and $100{\mu}g/mL$ kanamycin sulfate) and LEY-glycerol Orvus ES Paste (LEYGO, 89.5% v/v LEY, 5% v/v glycerol, 1.5% v/v Orvus ES Paste), 100 mM trehalose supplemented with 0, 10, 50, 100, and $500{\mu}M$ of curcumin from turmeric, respectively. Following equilibration, the 0.5 mL French straws were frozen and plunged into $LN_2$ tank for 7 days at least. Sperm parameter and oxidative byproducts were determined by the computer assisted sperm motility analysis (CASA) and fluorescence-activated cell sorting (FACS) as compared with each groups. Supplementation of curcumin had no effect on sperm motility, progressive motility and curvilinear velocity. However, average-path velocity and straight-line velocity were significantly higher in $10{\mu}M$ curcumin group ($100.9{\pm}8.8{\mu}m/s$, $61.7{\pm}2.9{\mu}m/s$, respectively) than control group ($77.8{\pm}3.9{\mu}m/s$, $46.4{\pm}3.0{\mu}m/s$, respectively) (p < 0.05). In addition, the level of the O2 radical and H2O2 were comparatively decreased in curcumin groups by evaluation of ethidium and DCF fluorescence. According to the results, curcumin can improve sperm kinetic variables and alleviate ROS induced cryoinjury to pig sperm.

Analysis of Semen Parameters in α1,3-Galactosyltransferase-/- Boars

  • It is very difficult to get the information about semen quality analysis in transgenic pigs because of limited numbers and research facilities. Therefore, in the present study, we analyzed the semen quality of transgenic boars generated for xenotransplantation research. Briefly, the semen samples were collected from 5 homozygous ${\alpha}1,3$-Galactosyltransferase knock-out ($GalT^{-/-}$) transgenic boars and immediately transported to the laboratory. These semen samples were decupled with DPBS and conducted to analyze semen parameters by a computer-assisted semen analysis (CASA) system. The boar semen were examined all 12 parameters such as total motility (TM), curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), and hyperactivated (HYP), etc. In results, among the 5 $GalT^{-/-}$ boars, three boars (#134, 144, and 170) showed normal range of semen parameters, but #199 and 171 boars showed abnormal ranges of semen parameters according to standard ranges of semen parameters. Unfortunately, #171 boar showed azoospermia symptom with rare sperm counts in the original semen. Conclusively, assessment of semen parameters by CASA system is useful to pre-screening of reproductively healthy boar prior to natural mating and artificial insemination for multiplication and breeding.
