한국고등균류기(韓國高等菌類記) (Notes on Korean Higher Fungi)

  • 1974년(年) 9월(月) 5일(日)부터 1975(年) 7월(月) 30일(日)까지 소백산(小白山) 및 안동지방(安東地方)에서 50여종(餘種)의 고등균류(高等菌類)를 채집(採集)하여 동정(同定)하였다. 그중(中) 한국(韓國) 미기록종(未記錄種)은 다음과 같다. Hygrophorus pantoleucus Hongo 흰갓주름버섯, Marasmius prasiosmus (Fr.) Fr. 흰낙엽버섯, Conocybe lactea (Lange) Metrod 노란종버섯, Pholiota spimosa(Fr.) sing. 노란갓비늘버섯, Crepidotus sulphurinus Imaz. et Toki 노란귀버섯, Pulveroboletus retipes (Berk. et Curt.) Sing. 밤색비단그물버섯, Calodon zonatus (Fr.) $Qu{\acute{e}}l$. 갈색깔대기버섯 Patella scutellata (St. Amans) Morgan 주홍접시버섯, Peziza badia Pers. ex Merat. 자주주발버섯.

진균 세포벽 유래 신물질을 이용한 감자의 전신적 획득저항성 유도 (A Natural Fungus-derived Elicitor for Induction of Systemic Acquired Resistance (SAR) in Potato)

  • 식물에 대해 병원성균이 아닌 자연계에서 존재하는 진균류로 부터 세포벽 성분을 분리하여 식물의 유도 저항성을 유도하는 엘리시터로써의 가능성을 시험하였다. 그 결과 선발된 진균 세포벽 성분이 감자에서 종래 알려진 어떤 엘리시터 보다도 아주 낮은 농도처리에서도 활성산소와 파이토알렉신 생성을 다량 유도하였다. 뿐 만 아니라 감자괴경을 이용한 병저항성 생물검정에서도 우수한 유도저항성이 발현되었음을 확인하였다. 그러므로 자연에서 분리된 일반적인 균주의 세포벽물질 엘리시터도 식물체의 유도저항성을 효과적으로 유도할 수 있는 새로운 식물병방제제로서의 개발 가능성이 있음을 시사하였다.

Antimite Activity of Cumin Volatiles Against Dermatophagoides farinae and Dermatophagoides pteronyssinus (Acari: Pyroglyphidae)

  • The antimite activities of cumin seed oil-derived cuminaldehyde and eleven commercial components of Cuminum cyminum oil were examined against Dermatophagoides farinae and Dermatophagoides pteronyssinus adults and compared with those of benzyl benzoate and N,N-diethyl-m-toluamide. Responses varied according to dose and mite species. On the basis of $LD_{50}$ values, the compound most toxic to D. farinae adults was cuminaldehyde ($2.40\mug/cm^2$) followed by benzyl benzoate ($9.32\mug/cm^2$), thymol ($9.43\mug/cm^2$), DEET ($36.84\mug/cm^2$), and 3-carene ($42.11\mug/cm^2$). Against D. pteronyssinus adults, cuminaldehyde ($1.94\mug/cm^2$) was much more effective than benzyl benzoate ($6.50\mug/cm^2$) thymol ($6.92\mug/cm^2$), DEET ($17.79\mug/cm^2$), and 3-carene ($39.85\mug/cm^2$). These results indicate that the antimite activity of cumin seed oil could be caused by cuminaldehyde. Cuminaldehyde was about 3.9 and 3.4 times more toxic than benzyl benzoate against D. farinae and D. pteronyssinus adults, respectively. Therefore, further study is needed to confirm the findings of this study and the possibility of cuminaldehyde as a house dust mite control agent or a lead compound.

Nondestructive Internal Defects Evaluation for Pear Using NIR/VIS Transmittance Spectroscopy

  • Internal defects such as browning of the flesh and blackening and rot of the ovary of pear can be easily developed because of the inadequate environmental conditions during the storage and distribution of fruit. The quality assurance system for the agricultural product is to be settled in Korea. All defected agricultural products should be excluded prior to the distribution to enhance the commercial values. However, early stage on-line defect detection of agricultural product is very difficult and even more difficult in a case of the internal defects. The goal of this research is to develop a system that can detect and classify internal defects of agricultural produce on-line using VIS/NIR transmittance spectroscopy. And Shingo pear, which is one of the famous species of Korean pear, was used for the experiment. Soft independence modeling of class analogy (SIMCA) algorithm was employed to analyze the transmittance spectroscopic data qualitatively. On-line classification system was constructed and classification model was developed and validated. As a result, the correct classification rate (CCR) using the developed classification model was 96.1 %.

Development of Polymorphic Microsatellite Markers Suitable for Genetic Linkage Mapping of Olive Flounder Paralichthys olivaceus

  • Microsatellite markers are important for gene mapping and for marker-assisted selection. Sixty-five polymorphic microsatellite markers were developed with an enriched partial genomic library from olive flounder Paralichthys olivaceus an important commercial fish species in Korea. The variability of these markers was tested in 30 individuals collected from the East Sea (Korea). The number of alleles for each locus ranged from 2 to 33 (mean, 17.1). Observed and expected heterozygosity as well as polymorphism information content varied from 0.313 to 1.000 (mean, 0.788), from 0.323 to 0.977 (mean, 0.820), and from 0.277 to 0.960 (mean, 0.787), respectively. Nine loci showed significant deviation from the Hardy-Weinberg equilibrium after sequential Bonferroni correction. Analysis with MICROCHECKER suggested the presence of null alleles at five of these loci with estimated null allele frequencies of 0.126-0.285. These new microsatellite markers from genomic libraries will be useful for constructing a P. olivaceus linkage map.

시판수산물에서 분리한 Vibrio parahaemolyticus의 병원성 인자와 항균제 내성 현황 (Virulence Factors and Antimicrobial Resistance of Vibrio parahaemolyticus Isolated from Commercial Fisheries Products)

  • Vibrio parahaemolyticus causes food poisoning, mainly via marine fisheries products. We investigated the virulence factors and drug resistance of V. parahaemolyticus isolated from fisheries products purchased from the Yeosu Fisheries Market. The isolates were identified using a variety of biochemical tests and the detection of toxR and hns gene. The presence of the virulence factor-encoding genes tdh and trh in the isolates was also investigated by PCR. The resistance of the isolates to 13 antibacterial agents was tested using the disc-diffusion method and carriage of β-lactamase genes and class 1 integrons by ampicillin-resistant isolates was investigated by PCR. Four of seventeen isolates identified as V. parahaemolyticus by biochemical tests produced a species-specific PCR band. Those isolates showed >98% 16S rRNA gene sequence homology with V. parahaemolyticus and only one isolate harbored the tdh gene. All of the V. parahaemolyticus isolates were resistant to ampicillin and amoxicillin; moreover, VPA0477, a class A β-lactamase gene, and class 1 integrons were detected. Therefore, V. parahaemolyticus from fisheries products represents a low risk to human health. Also, V. parahaemolyticus is likely to develop multidrug resistance because it has class 1 integrons.

강원도 양양군 일대 관속식물의 분포와 자원특성에 관한 연구 (Distribution of Vascular Plants and Characteristics of Plant Resources in Yangyang-gun, Gangwon-do)

  • Distribution of vascular plants in Yangyang-gun were investigated 12 times from Apr. to Sept., 2008. In the result of survey, the flora of this area consist of 412 taxa totally; 89 families, 259 genera, 366 species, 5 subspecies, 37 varieties and 4 forms. There were remarkable plants such as 11 taxa of Korean endemic plants, 58 taxa of specific plants which were designated by the Ministry of Environment, 2 taxa of endangered plants of Ministry of Environment, 7 taxa of rare, endangered plants of Korea Forest Service and 28 taxa of afforestation plants and 296 taxa of useful plants. Furthermore, 36 taxa of naturalized plants (8.74%) were also observed in this area. Vegetation of investigated area was conserved well. However, vegetation has been destroyed rapidly due to construction of commercial complex for human recreation such as condominiums and stores. Therefore, it is need to efficient management of the water quality and vegetation by conservation strategies from huge visitors.

Plant Defence Responses: Current Status and Future Exploitation

  • Plants have developed a sophisticated battery of defence responses to protect themselves against attempted pathogen ingress. Manipulation of these defence mechanisms may provide significant opportunities for crop improvement. While plant resistance genes have had a long service history in plant breeding, they possess significant limitations. Recent advances are now providing significant insights into strategies designed to increase the field durability of this class of genes. Hypersensitive cell death is a common feature underlying the deployment of plant defence responses against biographic pathogens. In contrast, necrotrophic pathogens actively kill plant cells. Recently, transgenic plants have been developed that either promote or suppress cell death, providing resistance against either biotrophic or necrotrophic pathogens respectively. Methyl-jasmonate is a key signalling molecule in the establishment of resistance against some fungal pathogens. Increasing the concentration of this molecule in plant cells has been shown to increase resistance against Botrytis cineria, without significantly imparting plant growth or development. Due to the multifarious infection strategies employed by plant pathogens, how-ever, it is unlikely a single commercial product will prove a panacea for global disease control. Future stategies will more likely entail an integrated disease management approach.

Alstroemeria plants and its biotechnological applications

  • Alstroemeria plants are widely cultivated in many countries especially in Western Europe and North America and popularity has increased in recently due to its long-base life, large variety of colors and low energy requirement during cultivation period. So far, more than 60 species have been released on the commercial market in the world. To meet the demand of consumer and develop the elite Alstroemeria cultivars, conventional breeding including cross-hybridization and selection as well as mutation breeding were used. However, as other important ornamental plants such as lily, rose, carnation and orchids accepted the biotechnological methods, this newly-born approach should be applied and developed an optimized the genetic transformation system. Then, this biotechnological approach can be fused with the conventional breeding methods and thus can be contributed to the production of elite Alstroemeria plants containing agriculturally good genetic traits which are useful for the both farmers and consumers in the future. In this paper, we reviewed the botanical and genetical features of Alstroemeria plants and its biotechnological approaches in the last decades.

Detection of Korean Native Honey and European Honey by Using Duplex Polymerase Chain Reaction and Immunochromatographic Assay

  • Korean native honey (KNH) is much more expensive than European honey (EH) in Korea, because KNH is a favored honey which is produced less than EH. Food fraud of KNH has drawn attention of the government office concerned, which is in need of a method to differentiate between KNH and EH which are produced by the Asiatic honeybee, Apis cerana and the European honeybee, Apis mellifera, respectively. A method to discriminate KNH and EH was established by using duplex polymerase chain reaction (PCR) in this study. Immunochromatographic assay (IC) was examined to analyze the duplex PCR product. The DNA sequences of primers for the duplex PCR were determined by comparing cytochrome C oxidase genes of the two honey bee species. Chelex resin method was more efficient in extracting genomic DNA from honey than the other two procedures of commercial kits. The duplex PCR amplifying DNA of 133 bp were more sensitive than that amplifying DNA of 206 bp in detecting EH in the honey mixture of KNH and EH. Agarose gel electrophoresis and IC detected the DNA of 133 bp at the ratios of down to 1% and 5% EH in the honey mixture, respectively and also revealed that several KNH products distributed by internet shopping sites were actually EH. In conclusion, the duplex PCR with subsequent IC could also discriminate between KNH and EH and save time and labor.
