소금 첨가량에 따른 바질 데미글라스 소스의 관능평가 분석 (Sensory Characteristics of Demi-glace Sauce Prepared by Fresh Basil with Various Levels of Salt Compositions)

  • This study was carried out to examine the sensory characteristics of Demi-glace sauce prepared by fresh basil with different amount of salt added. The evaluation of the sensory quality of Demi-glace sauce prepared by fresh basil was conducted by unexperienced and trained panel with regard to color, odor, taste, appearance, texture, viscosity, and overall acceptability while varying the quantity of salt added, the time of preparation, and the conditions of sauce offered in which each panel was supposed to evaluate both sauce itself and sauce with meat. It was found that Demi-glace sauce prepared by fresh basil with 0.3% salt added was gained the highest score from both panels in most properties and the second was the one with 0.1% salt added. In the sensory properties correlation analysis according to the time of preparation and the conditions of sauce offered, there was no significant correlation in most sensory properties for the unexperienced panel, except some significantly positive correlation between taste and overall acceptability of the sauce. Whereas there was a positive correlation in the majority of sensory properties for the trained panel regardless of the time of preparation and the conditions of sauce offered. The sauce with 0.3% salt added ranked as the best recipe by both panels among four different ratios of salt adding regardless of the time of preparation and the conditions of sauce offered. Triangle test showed that the sauce B with 0.1% salt added before the preparation was more highly appreciated in terms of taste than the sauce A with 0.1% salt added in the final step of preparation. In conclusion, it could be suggested that Demi-glace sauce prepared by fresh basil with 0.3% salt added is the best recipe for commercial products. Also, there seems to be the need of further sensory evaluation studies on the Demi-glace sauce according to the time of addition during the process of preparation.

Antimicrobial Efficacy of Penicillium amestolkiae elv609 Extract Treated Cotton Fabric for Diabetic Wound Care

  • Diabetes mellitus is a chronic disorder which affects millions of population worldwide. Global estimates published in 2010 reported the world diabetic prevalence as 6.4%, affecting 285 million adults. Foot ulceration and wound infection are major forms of disabilities arising from diabetic diseases. This study was aimed to develop a natural antimicrobial finishing on medical grade textile that meets American Association of Textiles Chemists and Colorists (AATCC) standard. The textile samples were finished with the ethanolic extract of Penicillium amestolkiae elv609, an endophytic fungus isolated from Orthosiphon stamineus Benth (common name: cat's whiskers). Endophyte is defined as microorganism that reside in the living plant tissue, without causing apparent disease symptom to the host. The antimicrobial efficacy of the ethanolic extract of P. minioluteum was tested on clinical pathogens isolated from diabetic wound. The extract exhibited significant inhibitory activity against 4 bacteria and 1 yeast with the minimal inhibitory concentration ranged from 6.25 to 12.5 mg/mL. The results indicate different susceptibility levels of the test microorganism to the ethanolic extract. However, the killing activity of the extract was concentration-dependent. The finished medical textile showed excellent antimicrobial efficacy on AATCC test assays. All the microbial cultures treated with the textile sample displayed a growth reduction of 99.9% on Hoheinstein Challenge Test. The wash durability of the finished textile was found good even after 50 washes with commercial detergent. Besides, the gas chromatography mass spectrometry analysis showed that 6-octadecenoic acid and diethyl phthalate were the main bioactive constituents of the extract. In conclusion, the developed medical textile showed good antimicrobial efficacy on laboratory tests. This work can be extended to in vivo trials for developing healthcare textile products for antimicrobial applications.

철도터널 화재시 구난역 내의 연기거동에 미치는 배연효과에 관한 연구 (A Study on Ventilation Effects on Smoke Behavior in Rescue Station for Tunnel Fires)

  • 본 연구는 구난역을 갖는 철도 터널 내에서 화재가 발생하였을 때 연기 전파 특성에 미치는 배연효과를 다루고 있다. 터널 화재 실험은 4cm의 정사각형 형태의 n-heptane$(C_7H_{16})$ 풀화재(pool fires)에 대해서 수행되었고 다양한 화재 발생위치에 따른 발열량을 측정하였다. 또한, 본 연구에서는 상용코드인 Fluent(Ver.6.3)를 사용하여 구난역을 갖는 철도 터널 내의 연층 전파 특성을 분석하였으며, MVHS(Modified Volumetric Heat Source) 모델을 사용하여 화재에 의한 연소생성물의 농도 및 거동을 예측하였다. 해석 조건은 다양한 화재 발생 위치에 따라서 측정 된 발열량을 이용하였다. 본 연구를 통해 교행로 내에 위치한 방화문에 의해 비사고터널로의 확산을 막을 수 있음을 보였고 특히, 배연시스템의 가동을 통해 유동방향을 변화시킴으로써 사고터널내의 연기 확산을 제어할 수 있음을 확인하였다.

고려인삼(Panax ginseng C.A.Meyer)의 간이법에 의한 홍삼제조 및 사포닌 성분분석 (Saponin Analysis and Red Ginseng Production using the Simplified Method of Korean Ginseng (Panax ginseng C.A.Meyer))

  • 홍삼 제조공정을 단순화하면서 효율적으로 유효성분을 증대시키기 위해서 4년근 수삼을 구입하여 깨끗이 수세한 후 $96-98^{\circ}C$에 3시간 정도 수증기로 증삼한 후 30시간 정도 열풍건조하여 홍삼제조를 위한 간이법을 개발하였다. 제조한 홍삼으로부터 홍삼엑스($60^{\circ}$ brix 이상)를 제조하였으며 홍삼엑스의 추출수율은 60% 이상이었다. 간이법으로 제조한 홍삼을 재료로 하여 농축한 홍삼엑스의 사포닌 함량을 조사하기 위해서 HPLC를 이용하여 총 사포닌(total saponin)을 분석하였다. 그 결과 PD계 사포닌 중 암세포전이억제효과와 평활근이완작용이 탁월한 ginsenoside-Rg3에는 H사에 비하여 2배 이상 검출되었으며, 간상해 억제작용과 혈소판 응집억제작용이 있는 것으로 알려진 ginsenoside-Rh1종의 경우 3배 이상 검출이 되었다.

Destruction and Removal of PCBs in Waste Transformer Oil by a Chemical Dechlorination Process

  • A practical and efficient disposal of PCBs (polychlorinated biphenyls) in waste transformer oil by a chemical dechlorination process has been reported. The transformer oil containing commercial PCB mixtures (Aroclor 1242, 1254 and 1260) was treated by the required amounts of PEG 600 (polyethylene glycol 600), potassium hydroxide (KOH) and aluminum (Al), along with different reaction temperatures and times. The reaction of PEG with PCBs under basic condition produces arylpolyglycols, the products of nucleophilic aromatic substitution. The relative efficiencies of PCB treatment process were assessed in terms of destruction and removal efficiency (DRE, %). Under the experimental conditions of PEG600/KOH/Al/100 oC/2hr, average DRE of PCBs was approximately 78%, showing completely removal of PCBs containing 7-9 chlorines on two rings of biphenyl which appear later than PCB no. 183 (2,2',3,4,4',5',6-heptaCB) in retention time of GC/ECD. However, when increasing the reaction temperature and time to 150 oC and 240 min, average DRE of PCBs including the most toxic PCBs (PCB no. 77, 105, 118, 123 and 169) in PCB family reached 99.99% or better, with the exception of PCB no. 5 and 8 (2,3-diCB and 2,4'-diCB). In studying the reaction of PEG with PCBs, it confirmed that the process led to less chlorinated PCBs through a stepwise process with the successive elimination of chlorines. The process also permits complete recovery of treated transformer oil through simple segregating procedures.

CP-Ti 및 Ti-6Al-4V 합금의 전기화학적 특성에 미치는 제조공정의 영향 (Effect of Manufacturing Process on Electrochemical Properties of CP-Ti and Ti-6Al-4V Alloys)

  • Ti and its alloys show the excellent corrosion resistance to chloride environments, but they show less corrosion resistance in HCl, $H_2SO_4$, NaOH, $H_3PO_4$, and especially HF environments at high temperature and concentration. In this study, we used the commercially pure titanium and Ti-6Al-4V alloy, and evaluated the effect of the manufacturing process on the electrochemical properties. We used commercial products of rolled and forged materials, and made additive manufactured materials by DMT (Directed Metal Tooling) method. We annealed each specimen at $760^{\circ}C$ for one hour and then air cooled. We performed anodic polarization test, AC impedance measurement, and Mott-Schottky plot to evaluate the electrochemical properties. Despite of the difference of its microstructure of CP-Ti and Ti-6Al-4V alloys by the manufacturing process, the anodic polarization behavior was similar in 20% sulfuric acid. However, the addition of 0.1% hydrofluoric acid degraded the electrochemical properties. Among three kinds of the manufacturing process, the electrochemical properties of additive manufactured CP-Ti, and Ti-6Al-4V alloys were the lowest. It is noted that the test materials showed a Warburg impedance in HF acid environments.

조선·해양 기자재용 강재의 내식성에 미치는 분체도장 중 산화물 첨가의 영향 (Effect of Oxide Particles Addition to Powder Coating on Corrosion Resistance of Steel Used as Marine Equipments)

  • The demand for powder-coated steel used in the marine industry is increasing owing to their superior corrosion resistance. However, the powder coatings used in commercial products can deteriorate easily by the penetration of brine. In an attempt to suppress brine penetration into the powder coating and significantly increase the corrosion resistance, three types of oxide particles were added to the coating. Electrochemical impedance spectroscopy tests in 3.5% NaCl solution were performed to evaluate the corrosion behaviors of the powder coating with oxide particles. The results showed that the addition of SiO2 particles to a powder coating severely decreased the corrosion resistance due to the easy detachment of agglomerated SiO2 particles with a coarse size from the coating layer. In contrast, the TiO2 and SnO2-added coatings showed better corrosion resistance, and the TiO2-added coating performed best in the test conducted at room temperature. However, conflicting results were obtained from tests conducted at a higher temperature, which may be attributed to the effective suppression of brine penetration by the fine SnO2 particles uniformly distributed in the coating.

Skeletal Muscle Troponin I (TnI) in Animal Fat Tissues to Be Used as Biomarker for the Identification of Fat Adulteration

  • In this study, the existence of skeletal muscle troponin I (smTnI), well-known as a muscle protein in fat tissues, and the utilization of smTnI as a biomarker for the identification of fat adulteration were investigated. A commercial antibody (ab97427) specific to all of animals smTnI was used in this study. Fat and meat samples (cooked and non-cooked) of pork and beef, and chicken considered as representative meats were well minced and extracted by heating and non-heating methods, and the extracts from fat and meat tissues were probed by the antibody used in both enzyme-linked immunosorbent assay (ELISA) and immunoblot. The antibody exhibited a strong reaction to all meat and fat extracts in ELISA test. On the other hand, the results of immunoblot analsis revealed a 23 kDa high intensity band corresponding to the molecular weight of smTnI (23786 Da). These results demonstrate that the existence of smTnI in all animal fat tissues. Since there are monoclonal antibodies specific to each species smTnI, smTnI in fat tissues could be used as a biomarker to identify or determine animal species adulterated in meat products. Therefore, an analytical method to identify fraudulent fat adulteration can be developed with an antibody specific to each species smTnI.

Rapid High Performance Liquid Chromatographic Quantification of Major Isoflavones in Soybeans and Soybean Pastes

  • A simple HPLC quantification method was developed for genistein, genistin, daidzein, and daidzin in soybeans and soybean products. The procedure used a $4.6{\times}100\;mm$ $Chromolith^{(R)}$ RP-18e column with a mobile phase of 1% HOAc in 20% MeOH to 1% HOAc in 80% MeOH for 10 min. The injection volume was $2\;{\mu}L$ at a flow rate of 2 mL/min. Detection was carried out under UV at 254 nm. Under these conditions, the major isoflavones daidzein, daidzin, genistein, and genistin in soybean and soybean pastes were eluted within 7 min with baseline separation. Optimal extraction of the above four major isoflavones was achieved when 40 g of soybean or soybean paste was refluxed in 100 mL of 95% ethanol for 2 hr. Ten different soybean cultivars and nine commercial soybean pastes were analyzed by this method. The total isoflavone content was highest in the cultivar Somyung ($2,497\;{\mu}g/g$ dry weight). The isoflavone content in soybean pastes varied widely from manufacturer to manufacturer (an almost five-fold difference between the highest and lowest values). Such variations were presumably due to differences in fermentation conditions, type of soybeans used, and levels of such additives as starch and salt.

산소/질소 분리를 위한 다층구조 제올라이트 5A를 함유한 탄소분자체 분리막 제조 (Hierarchical 5A Zeolite-Containing Carbon Molecular Sieve Membranes for O2/N2 Separation)

  • 다층 구조를 가진 5A 제올라이트를 탄소 분자체 분리막에 첨가한 복합막을 제조하고 질소/산소의 분리 특성을 평가하였다. 제올라이트의 첨가는 선택도에는 미세한 영향을 주지만 투과도를 크게 증가시키는 방법으로 전체적인 탄소막의 질소/산소의 분리 성능을 상승시켰다. 특히 메조포어를 함유한 다층구조의 제올라이트 첨가제는 분리막의 투과도를 보다 효율적으로 상승시켜 아주 우수한 분리 성능에 도달하였다. 이 연구의 결과는 저렴한 탄소막 전구체와 제올라이트 소재를 활용하고도 고성능의 질소/산소 분리막을 손쉽게 제조할 수 있다는 것을 제시한다.
