Somatic Embryogenesis and Plant Regeneration in Barley (Hordeum vulgare L.)

  • Commercial cultivars and elite germplasms of barely (Hordeum vulgare L) are still recalcitrant to genetic transformation because of the lack of an efficient regeneration system. In this study, we established an efficient plant regeneration procedure from embryogenic calli derived from mature embryos. Callus induction from germinated mature embryos was best as over 95% in CIM medium (CI medium containing $2.5mg/{\ell}$ dicamba) under dark incubation. Development of embryogenic callus was highest as over 50% in CI3D medium (EC medium supplemented with $3mg/{\ell}$ 2,4-D). The highest regeneration of plants from embryogenic callus (40%) was obtained with CIS medium ($SI+1mg/{\ell}IAA\;and\;2mg/{\ell}\;BA$). These plant regeneration conditions could be useful in improving barley transformation efficiency.

아까시나무(Robinia pseudoacacia L.) 유묘의 뿌리혹 형성에 적절한 배양토, 질소, 인 수준 구명에 관한 연구 (Optimum Potting Medium and Nitrogen and Phosphorus Levels in the Soil for Root Nodule Formation in Black Locust (Robinia pseudoacacia L.) Seedlings)

  • 본 연구는 아까시나무 유묘에서 뿌리혹 형성에 가장 적절한 배양토의 종류와 토양의 양료 수준을 구명하기 위해 수행하였다. 상업용 상토, 완숙 혹은 미숙 퇴비, 유기질 비료, 산림용 고형비료, 화학비료 등의 토양 첨가물을 이용하여 질소와 인의 수준을 다양하게 조절한 후 포트에 아까시나무 종묘를 식재하여 3개월 간 온실에서 실험을 실시하여 초기 뿌리혹 형성에 적절한 토양 환경을 조사하였다. 피트모스, 질석, 지올라이트를 함유한 상업용 상토는 높은 보수력으로 인해 초기 활착과 건중량 생산에는 적절하였으나 무기 양료 중 질소(0.052%), 인(91 ppm)의 부족으로 인해 뿌리혹 형성에는 부적합하였다. 퇴비는 미숙 퇴비이건 완숙퇴비이건 구별 없이 뿌리혹 형성에 가장 유리했는데, 질소(0.08-0.21%)와 인(141-1,228 ppm)의 함량이 적절하였지만, 칼륨(0.14-1.58 cmol/kg)의 함량은 부족한 편이었다. 화학(복합)비료는 토양 내 질소(0.14-0.21%), 인(406-618 ppm), 칼륨(1.39-3.13 cmol/kg)의 함량을 적절한 수준으로 유지하여 초기 건중량 생산에는 지장을 주지 않았지만 뿌리혹 형성에는 부적절하였다. 산림용 고형비료는 뿌리혹 형성에 적합한 반면, 유기질 비료는 뿌리혹 형성에 적합하지 않았다. 배양토에는 적절한 수준의 질소(0.05-0.2%)와 인(100-600 ppm)이 함유되어 있을 경우에 초기 뿌리혹 형성이 제대로 이뤄졌다. 아까시나무 유묘의 경우 토양의 질소와 인 중에서 질소보다 인이 초기의 뿌리혹 형성을 더 촉진하는 것으로 판단되었다.

  • As the LCD fabrication factories (Fab) are highly capital-intensive and the markets are very competitive, it is an essential requirement of operational management to achieve full-capacity production while meeting customer demands on time. In a typical LCD Fab, medium-term schedules such as release plans and production plans are critical to achieve the goal of full-capacity production and on-time delivery. Presented in this paper is a framework for weekly planning system generating medium-term schedules using a finite-capacity planning method. Also this paper presents a release planning method applying capacityfiltering algorithm, especially backward capacity-filtering procedure, which is one of the finite-capacity planning methods. In addition, performance analyses using actual data of a TFT-LCD Fab show that the proposed method is superior to existing methods or commercial S/W products generating release plans.

중형 트럭의 피칭 운동 저감을 위한 현가계의 설계 변수에 관한 해석적 연구 (An Analytical Study of Suspension Design Parameters in order to Reduce the Pitching Motion of Medium Truck)

  • Ride quality of medium truck became a very important factor in the suspension design, to the demand of more comfortable ride of passengers. This study describes how to determine and evaluate design parameters related to the chassis suspension system with time and frequency analysis. The spring stiffness and damping force of the chassis suspension system were obtained by observing the vertical acceleration PSD. The simulation was carried out on various road profiles, which was suggested by ISO. The pitching motion of the medium size truck was observed to improve the ride quality. A computer simulated truck model was constructed using DADS, a commercial dynamic analysis software, in order to simulate the truck motions. From the analyzed process of suspension parameters, it was concluded that the spring and the shock absorbers affect the pitching of the vehicle. In order to validate the computer simulated truck model, a physical prototype was constructed and tested.

  • Phage utilizing medium and BL agar supplemented with antibiotic Tc(tetracycline) were developed as selective media for the isolation and counting of bifidobacteria in dairy products. The former was based on the host specificity of phage. When bifidobacteria and Lactobacillus casei HY 2782 were mixed together in dairy product, L. casei HY 2782 was laysed by J1 phage which has host specificity to L. casei HY 2782 whereas bifidobacteria grew well on the selective medium added wIth J1 phage. The latter was found to inhibit the growth of S. salivarius subsp. thermophilus, L. acidophilus, and L. casei, but commercial bifidobacteria grew well in Tc-containing BL agar.

Trichoderma koningii의 conidiospore로부터의 원형질체 분리에 관하여 (Isolation of protoplast from conidiospore of Trichoderma koningii)

  • Conditions for isolation of protoplasts from conidiospores of Trichoderma koningii ATCC 26113 were tested. Maximum production of conidial protoplasts was obtained by preincubation of conidiospores on liquid minimal medium for 8 1/2 hrs. and by reaction with cell wall lytic enzyme for 3 hrs. Among effective cell wall lytic enzymes (Driselase, p-Glucuronidase, Novozyme and Zymolyase), Driselase was the most effective one on the production of conidial protoplasts. The production of conidial protoplasts was also enhanced by addition of 2-Deoxy-D-Glucose $(25{\mu}g/ml)$ into liquid minimal medium. Over 70% of the initial swollen conidia, preincubated in liquid minimal medium supplemented with 2-Deoxy-D-Glucose $(25{\mu}g/ml)$, were converted to protoplasts by incubation with 2% (w/v) commercial lytic enzyme Driselase at $28^{\circ}C$ for 3 hrs. The reversion frequency of the conidial protoplasts was about 30 times (25-50%) higher than that of mycelial protoplasts (0.6-1.3%).

중형상용차량용 LPG엔진의 개발에 관한 연구 (Development of an LPG Engine for Medium-Duty Commercial Vehicles)

  • This study is aimed to develop a proto-type LPG engine for medium-duty commercial vehicles in order to substitute for conventional diesel engine. Recently, it is recognized that diesel engines are main cause for smoke pollution in urban site. So, it is expected to reduce this environmental emission by developing and substituting LPG engine which has the advantage of practical use in a short development period in aspects of infrastructures. For that, after analysing the specifications and performance characteristics of a base diesel engine, parts of combustion chamber, intake system, fuel supply and ignition systems suitable for LPG combustion were re-designed and manufactured. And and engine controller for fuel supply and ignition distributions was matched by feedback mapping based on the speed-load conditions. The torque and power of LPG engine were increased by 6∼12% on the overall driving conditions compared to the base diesel engine, and fuel consumption rate marked the similar level based on the fuel price. Exhaust emissions such as THC, CO, NOx recorded the same order with conventional LPG engine for passenger car.

다당류를 생산하는 미생물의 분리와 배양특성 (Isolation of the Exopolysaccharide Producing Microorganism and their Cultural Characterisitics)

  • A screening was performed to isolate exoploysaccharide-producing microorganisms, which synthesized specific exopolysaccharide for the substitutive of commercial polysaccharides, from natural sources. Soil bacterium, one of 378 mucoid isolates, was finally selected as potential producer of polysaccharides which made the culture broth very viscous and thus examined in detail for optimal medium composition. Isolated strain was identified as Xanthomonas sp. EPS- 1 from the results of morphological and biochemical characteristics. The composition of optimal medium for exopolysaccharide production was as follows: 50 g sucrose, 1.5 g peptone, 2 g KH$_{2}$PO$_{4}$, 2 g MgSO$_{4}$, -7H$_{2}$O, 3 g NaCl, 0.05 g CaCO$_{3}$, 0.07 g FeSO$_{4}$-7H$_{2}$O and 0.05 g MnSO$_{4}$-7H$_{2}$O in 1 liter of distilled water. From the experiments of temperature and pH dependence, the optimal conditions for exopolysaccharide biosynthesis seemed to be 30$\circ$C and 8.0, respectively. About 14.9 gram of maximum exopolysaccharide per liter was obtained at the initial pH 8.0, 30$\circ$C and 250 rpm in a flask culture. The exopolysaccharide EPS-1 had such potential as an emulsifying agent and a gelling agent in comparision with commercial exopolysaccharide.

상업 및 업무기능 활성화를 통한 지방중소도시 구도심 재생방안 : 당진군을 중심으로 (Old Inner City Regeneration of Local Small-and-Medium Cities for the Invigoration of Commercial and Business Function : case of Dangjin-gun)

  • 본 연구는 지방중소도시 중 성장과 더불어 구도심이 쇠퇴한 대표적인 사례지역인 당진군을 대상으로 중심시가지로서의 구도심을 재생하기 위해 필요한 상업 및 업무기능 활성화 방안을 제시해보았다. 연구는 활성화를 위한 도시재생방향과 중요 도입기능으로 구분하여 컨조인트 분석을 활용하였다. 연구결과에 따른 시사점은 다음과 같다. 첫째, 고밀복합개발은 지양하고 저밀형 상업 업무공간 조성 및 역사문화자원의 활용을 고려할 필요가 있다. 둘째, 지역경제 활성화를 위해서 신산업(하이테크산업) 유치보다는 지역의 특화된 업종 및 시설을 육성하는 방향을 우선적으로 검토할 필요가 있다. 셋째, 구체적이고 실천 가능한 기존 상권 유지 및 저소득층 재정착 대책을 수립할 필요가 있다. 넷째, 유지관리 측면에서 불법행위 규제와 공공 공간의 개선 그리고 이용자의 안전성 확보가 충분히 고려될 필요가 있다. 다섯째, 기능강화 측면에서 대중교통을 통한 접근성 강화 및 지역홍보활동과 이벤트 개최를 중요하게 고려할 필요가 있다. 여섯째, 비즈니스 활동 지원 및 구도심 디자인 가이드라인 마련 등의 지원을 고려할 필요가 있다. 일곱째, 공공은 지역주민의 의견과 전문가의 의견을 잘 조율하여 균형적인 방향으로 사업이 추진될 수 있도록 주된 역할을 담당해야 한다.

일산신도시의 중심상권 활성화를 위한 상권 분석에 관한 연구 (A Study on the Trade Area Analysis of the Ilsan Subway Area for the Regeneration of Main Street)

  • This article assumes that the first newtown has declined in Korea. Urban regeneration projects should be implemented for regeneration for CBD in Newtown as well as small local cities. The purpose of this article is to analyze the character of the subway area and to suggest the key issues for regeneartion in the subway area. Especially focussing on the local important commercial function in 5 subway areas. As a result of analyzing 5 subway areas according to basic statistical data, field interviews, and legal decline diagnostic index, all five commercial districts showed decline. In particular, the decline of Baekseok Station area appears to be serious, and there is a key factor that attracts the decline of the commercial area such as the long - term building or the closure rate of each commercial area. In addition, the vacancy rate is increasing due to large distribution facilities in new cities, the entry of new floating facilities on the outskirts, and unsold new stores, and the recession is accelerating. In this way, the signs of decline in the core of the new city are enriched, but the current Urban Regeneration Act is a legal system with a policy goal of regeneration of small and medium cities in the provinces as a system to promote the function of core bases rather than the overall redevelopment of new cities. Therefore, preemptive response is needed for the regeneration in those areas.
