Cytokinetic Study of MCF-7 Cells Treated with Commercial and Recombinant Bromelain

  • Background: Breast cancer is a leading cause of death in women. The available chemotherapy drugs have been associated with many side effects. Bromelain has novel medicinal qualities including anti-inflammatory, anti-thrombotic, fibrinolytic and anti-cancer functions. Commercially available bromelain is obtained through tedious methods; therefore, recombinant bromelain may provide a cheaper and simpler choice with similar quality. Materials and Methods: This study aimed to assess the effects of commercial and recombinant bromelain on the cytokinetic behavior of MCF-7 breast cancer cells and their potential as therapeutic alternatives in cancer treatment. Cytotoxic activities of commercial and recombinant bromelain were determined using (sulforhodamine) SRB assay. Next, cell viability assays were conducted to determine effects of commercial and recombinant bromelain on MCF-7 cell cytokinetic behavior. Finally, the established growth kinetic data were used to modify a model that predicts the effects of commercial and recombinant bromelain on MCF-7 cells. Results: Commercial and recombinant bromelain exerted strong effects towards decreasing the cell viability of MCF-7 cells with $IC_{50}$ values of 5.13 ${\mu}g/mL$ and 6.25 ${\mu}g/mL$, respectively, compared to taxol with an $IC_{50}$ value of 0.063 ${\mu}g/mL$. The present results indicate that commercial and recombinant bromelain both have anti-proliferative activity, reduced the number of cell generations from 3.92 to 2.81 for commercial bromelain and to 2.86 for recombinant bromelain, while with taxol reduction was to 3.12. Microscopic observation of bromelain-treated MCF-7 cells demonstrated detachment. Inhibition activity was verified with growth rates decreased dynamically from 0.009 $h^{-1}$ to 0.0059 $h^{-1}$ for commercial bromelain and to 0.0063 $h^{-1}$ for recombinant bromelain. Conclusions: Commercial and recombinant bromelain both affect cytokinetics of MCF-7 cells by decreasing cell viability, demonstrating similar strength to taxol.

Resistance of Cryptosporidium parvum oocysts following commercial bleach treatment

  • We investigate the resistance of Cryptosporidium (C.) parvum oocysts to commercial bleach treatment. The viability and infectivity of C. parvum oocysts suspended in 100, 50, 25, 12.5, 6.3 or 3.2% aqueous commercial bleach for 10, 30, 60, 120 or 180 min at room temperature were assessed by nucleic acid Syto-9 staining, histologic examination of ileum and infectivity to immunosuppressed neonatal C57BL/6N mice. Although the viability was decreased compared with normal oocysts, all oocysts in contact with serially diluted commercial bleach for 180 min were alive by nucleic acid dye Syto-9 staining. And, microscopic examination of ileum sections revealed developmental stages of C. parvum in all mice. The oocyst shedding patterns between mice infected with oocysts contacted with commercial bleach and normal control mice were not significantly different each other. Although commercial bleach is widely used as a bacterial and viral disinfectant, the present findings indicate that it is not an effective disinfectant for C. parvum oocysts under practical conditions. Authors conclude that, therefore, it is undesirable to recommend commercial bleach as a disinfectant for C. parvum oocysts.

  • This study examined how the quality of TikTok service affects user satisfaction and continuance intention. This study classified the quality of Tiktok service as service quality, information quality, and system quality, and examined the effect of each dimension on user satisfaction and continuous intention. In addition, the role of perceived commercial viability was also examined. In this study, we conduced an online survey on 237 Chinese TikTok users and analyzed the data with the SEM and a moderated regression analysis. Results are as follows. First, information quality and system quality are positively associated with user satisfaction. Second, information quality is positively associated with continuance intention. Third, user satisfaction fully mediates system quality and continuance intention, and it partially mediates information quality and continuance intention. Fourth, although user satisfaction is positively associated with continuance intention, commercial viability does not function as a moderating variable.

  • The objective of this study was to determine the effect of semen extenders on the motility, viability and fertility in vitro of spermatozoa during storage of fresh boar semen diluted in different commercial extenders used for pig artificial insemination (AI). In this experiment, semen were diluted in Androhep plus, Beltsville Thawing Solution (BTS), Modena, Seminark and Vitasem LD. Five ejaculates were collected from three Duroc boars and sub-samples were diluted ($30{\times}10^6$ spermatozoa/ml) in different extenders. Semen was stored at $170^{\circ}C$ for 10 days. Sperm motility and viability was assessed using Computer-Assisted Semen Analysis (CASA) and flow-cytometry on 1, 3, 5 and 10 day post collection The motility of spermatozoa stored in different extenders was gradually decreased by increasing the duration of storage of semen. However, there was not significant1y different in the sperm motility and viability among other extenders. On the other hand, the in vitro-matured oocytes were fertilized and cultured in vitro to assess the fertility of boar spermatozoa stored for 3 and 10 days in different extenders. The percentage of morula and blastocyst were taken as indicators of fertility in vitro of spermatozoa. Therefore, there were no differences in the rate of embryos developed to the molular and blastocyst stage. There were no differences in the motility and fertility in vitro among 5 kinds of commercial boar semen extenders.

Development of fluoride varnish with sustained fluoride release and biocompatibility (불소 지속방출 및 생체적합성을 가진 불소 바니쉬의 개발)

  • The purpose of this study was to fabricate a biocompatible fluoride varnish with sustained fluoride release, and to compare it with commercial fluoride varnishes. For the experimental fluoride varnish, bis-GMA (BG) or two types of rosin (KR-610: K0, KR-612: K2) were used as bases. Either ethyl acetate or ethanol was added as solvent and 5 wt% NaF was used. 5 mg of experimental F- varnishes and 2 commercial products, Cavity shield (CS) and Flor-opal (FO), were applied on a labial surface of bovine teeth ($10mm{\times}7mm$). The amount of fluoride release was measured at 1 hr, 2 hrs, 3 hrs, 4 hrs, 8 hrs, 12 hrs, 1 day, 3 days, 5 days, 10 days, 15 days, 20 days and 30 days. MTT test was done with diluted F- varnishes using ethanol. Statistical analysis was done with one-way ANOVA and Duncan multiple range test (${\alpha}=0.05$). BG showed the highest fluoride release at 1 hr (P<0.05), while that of K0 was highest at 2 hrs (P<0.05). From 1 day to 5 days, experimental fluoride varnishes showed higher fluoride release than the commercial products (P<0.05), and there were no significant differences after 5 days (P>0.05). For MTT test, K0 and FO showed higher cell viability than other experimental groups (P<0.05), with no significant differences with K2 (P>0.05). Considering the sustained fluoride release and cell viability of the experimental rosin-based fluoride varnishes compared with commercial products, it will be appropriate for clinical application.

  • A breeding plan was carried out on four commercial strains of silkworm (Bombyx mori L.) 101, 102, 103 and 104 to improve some important traits. Genetic gain or response to selection $({\Delta}G)$, heritability of cocoon shell weight (CSW) and specific combining ability effects were estimated to determine the strains that can be improved. Strain 101 had lowest heritabitity, ${\Delta}G$ and viability. Strain 102 was acceptable in selection response but its viability was low. Therefore these two strains were not suitable for more selection. As a result, only lines 103 and 104 were chosen for further improvement. Intra population selection based on independent culling level method practiced from third to sixth generation for both productive and viability traits simultaneously. While CSW and CW had increasingly enhanced during primary generations, they went slightly up after third generation. According to negative genetic correlation, viability decreased during primary generations, but after third generation that paid attention to balanced development of both productive and viability traits, viability increased so that the pupation rate reached to $91\%$ in 103 and $97\%$ in 104 for last generation $(G_8)$.

Survival of Double-Microencapsulated Bifidobacterium breve in Milk in Simulated Gastric and Small Intestinal Conditions

  • Bifidobacteria are probiotic organisms that provide both flavor and health benefits when incorporated as live cultures into commercial dairy products. Because bifidobacteria are very sensitive to environmental conditions (acids, temperature, oxygen, bile salts, the presence of other cultures, etc.), their viability in human gastrointestinal tract is limited. The microencapsulation of bifidobacteria is a process to protect them against harsh environmental conditions, thereby increasing their viability while passing through human gastrointestinal tract. To confirm the survival rate of microencapsulated Bifidobacterium breve CBG-C2 in milk, their survival rate was compared with several kinds of free bifidobacteria and lactic acid bacteria in commercial yogurt products under simulated gastric and small intestinal conditions. Double-microencapsulation of the bacteria was employed to increase the survival rate during digestion. The outer layer was covered with starch and gelatin to endure gastric conditions, and the inner layer was composed of a hard oil for the upper small intestinal regions. Almost all microencapsulted bifidobacteria in the milk survived longer than the free bifidobacteria and lactic acid bacteria in the commericial yogurt products under the simulated gastric conditions. Numbers of surviving free bifidobacteria and lactic acid bacteria in the commercial products were significantly reduced, however, the viability of the microencapsulated bificobacteria in the milk remained quite stable under gastric and small intestine conditions over 3$\sim$6 hrs. Thus double-microencapsualtion of bifidobacteria in milk is a promising method for improving the survival of bifidobacteria during the digestive process.

  • The possibility of employment of ceramics, known to be emitting far infrared waves, to keep the growth and the viability of the antagonistic microorganisms was examined. Among four kinds of commercial ceramics, the ceramic powders composed of soft ferrite as a main component has exhibited to grow Pseudomonas cepacia and Saccharomyces cerevisiae better. The ceramic powder in the growth medium has increased the number of cells of P. cepada about ten times and that of S. cerevisiae two to five times more than that in the control at $27^{\circ}C$. The viability of the microorganism at low temperature which was measured from the regrowth behavior at $27^{\circ}C$ after five days store at $4^{\circ}C$ has shown that the lag time of the two microorganisms reduced about three hours without any defect in the rate of logarithmic growth. These results demonstrated that the ceramic powders was available to the growth and viability at low temperature of antagonistic microorganisms.

Impact of Prebiotic on Viability of Lactiplantibacillus plantarum D-2 by Encapsulation through Spray Drying and Its Commercialization Potential

  • This study investigated the impact of inulin (INL) on viability of L. plantarum D-2 (LPD2) by encapsulation through spray drying (SD) and its commercialization potential to alternative of conventional wall material maltodextrin (MD). LPD2, derived from sea tangle (Saccharina japonica) kimchi, is probiotics exhibiting significant attributes like cholesterol reduction, antioxidant properties, and resilience to acidic and bile environments. To enhance storage viability and stability of LPD2, encapsulation was applied by SD technology. The optimum encapsulation condition with MD was 10% MD concentration (MD10) and inlet temperature (96℃). The optimum concentration ratio of MD and INL was 7:3 (INL3) for alternative of MD with similar encapsulation yield and viability of LPD2. Viability of LPD2 with INL3 exhibited almost 8% higher than that with MD10 after 50 days storage at 25℃. Physicochemical characteristics of the encapsulated LPD2 (ELPD2) with MD10 and INL3 had no significant different between flowability and morphology. But, ELPD2 with INL3 had lower water solubility and higher water absorption resulting in extension of viability of LPD2 compared to that with MD10. The comprehensive study results showed that there was no significant difference in the encapsulation yield and physicochemical properties between ELPD2 with MD10 and INL3, except of water solubility index (WSI) and water absorption index (WAI). INL have the potential to substitute of MD as a commercial wall material with prebiotic functionality to enhance the viability of LPD2 by encapsulation.

  • The objective of this study was to compare the effect of semen extenders on the sperm motility, viability, acrosome integrity and functional integrity of plasma membrane (HOST: hypo-osmotic swelling test) during liquid preservation of Korean Native boar semen. In this experiment, semen was diluted in Androhep plus, Beltsville Thawing Solution (BTS), ModenaTM, Seminark and Vitasem LD. Sperm-rich fractions were collected from three Korean Native boars and sub-samples were diluted ($30{\times}10^6$ spermatozoa/ml) in different semen extenders. Semen samples were stored at $17^{\circ}C$ for 96 hours. On everyday (0, 24, 48, 72, 96 h) after storage, the sperm characteristics relevant for fertility, such as sperm motility, viability, acrosome integrity and HOST positive were evaluated. The motility of spermatozoa stored in different extenders was no significantly different among other extenders (P>0.05). Also, no difference was observed among samples processed with different extenders in the percentage of sperm viability, acrosome integrity and HOST positive. All extenders maintained a high percentage (70%) of sperm motility, viability and acrosome integrity through 96 h of storage. The result of this study show that there was no significant differences among extenders in their capacity to preserve motility, viability and membrane integrity of spermatozoa from normal, fertile Korean Native boars for 96 h of liquid preservation at $17^{\circ}C$.
