Effect of Tyrosinase Inhibitors on the Melanogenesis of Gold Fish(Jet Black Color) (Tyrosinase 저해제가 검은툭눈붕어의 멜라닌 생성에 미치는 영향)

  • The in vivo effect of tyrosinase inhibitors in the melanogenesis of gold fish (jet black color) was evaluated by measuring surface color and observing melanin pigment. The fish was firstly cultivated in 0.9% NaCl solution for 1 week to induce melanogenesis, and then, it was transferred to each treatment group containing tyrosinase inhibitor. The fish was grouped into control. food additive group (addition of 5 mM glutathione, 5 mM cysteine, and 1 mM benzoic acid), microbial inhibitor group (addition of culture broth of Aspergillus oryzae in shiitake and glucose medium), and plant extract group (addition of the mixed extracts of green tea, beet, red chicory, and nameko). After 6 days, the fish was anesthetized by electric shock, and color of pectoral region, lateral region, and dorsal fin was measured. Hunter's L and b values of treated group were generally higher than those of control group, indicating that the tyrosinase inhibitors could inhibit the melanogenesis of the fish. Effect of plant extract was apparent, though relatively weak, not because it did not work in vivo, but because a sufficient amount of extract could not be added to fish globes. If a large amount of extract was added, fish gradually died due to a microbial contamination. Microscopic observation of melanin in lateral scale and dorsal fin showed that in the treated groups with tyrosinase inhibitors, the number of melanophore per unit area and the size of one melanophore decreased.

Changes in color stability and antioxidant properties of dietary pigments after thermal processing at high pressures (고온가압 처리에 의한 식용색소의 화학안정성 및 산화방지활성 변화)

  • Various dietary pigments are added to processed foods to improve their sensory and commercial properties. In this study, autoclave sterilization (121℃ for 15 min at 15 psi) was performed on 34 food pigments, and changes in their color stability and antioxidant activity were analyzed. The autoclaving process drastically reduced the peak color intensities of water-soluble paprika and beet red (BR) by ~90%. Turmeric oleoresin (TO), water-soluble β-carotene, and grape skin color were also unstable and showed a remaining color intensity of 45-60%. The colors of all the synthetic pigments tested were stable under this process. The scavenging activities of BR and paprika against ABTS, DPPH, and AAPH radicals decreased significantly, whereas those of TO were enhanced after the autoclaving treatment. The results suggest that the chemical and bioactive properties of certain dietary pigments are affected by the autoclaving process, and this phenomenon should be considered during food processing.

  • The attractiveness of the gingiva is determined by its color, shape, and the shape and location of the boundary between the teeth and the gingival tissue. The standards beauty, balance, and health of the gingiva are all different, but the general public would agree that a coral pink gingiva is more beautiful than black or brown gingiva. Hence, one would be able to smile more confidently in public if he or she receives a gingival pigmentation removal surgery that changes the color of black or brown gums to a beautiful pink color with relative simplicity. The color of one's gingiva varies from pale pink to deep bluish purple, depending on many health components. The most prominent among these include the vascular supply, epithelial thickness, the degree of keratinization, and the presence of pigment in the epithelium. Melanin, carotene, reduced hemoglobulin, and oxyhemoglobulin are the main pigments contributing to the normal color of the oral mucosa. The health of one's gingival tissue are essential for an attractive smile. Excessive melanin deposits in the basal and early basal layers of the epithelium stored in the form of melanosomes frequently cause pigmentation.   • Two urostyloid ciliates, collected from brackish water in Korea, were identified as Diaxonella pseudorubra pseudorubra (Kaltenbach, 1960) Berger, 2006 and Pseudokeronopsis flava (Cohn, 1866) Wirnsberger, Larsen and Uhlig, 1987. The description was based on living, protargol impregnated specimens. These species are described as follows: Diaxonella pseudorubra pseudorubra: body size in vivo $145-230{\times}40-60\;{\mu}m$, elongated ellipsoidal in shape. Cytoplasm reddish and flexible. Adoral zone of membranelles occupied 30-40% of the body; composed of 33-44 membranelles; 1-3 frontoterminal cirri, 1-4 frontal row cirri, 4-6 buccal cirri, 6-10 transverse cirri. Midventral rows composed of 14-24 cirri, four left marginal rows, one right marginal row. Two kinds of cortical granules; the larger one is yellowish and the smaller one is reddish. Pseudokeronopsis flava: body size in vivo $150-210{\times}30-45\;{\mu}m$, elongated ellipsoidal shape. Cytoplasm yellowish and flexible. Adoral zone of membranelles occupied 25-30% of body; composed of 44-58 membranelles in number. Frontal cirri forming bicorona composed of 5-7 cirral pairs, 2-3 frontoterminal cirri, one buccal cirrus, and 2-3 transverse cirri. Midventral rows composed of 18-33 cirri, 34-53 left marginal cirri, and 40-58 right marginal cirri. Two kinds of cortical granules; the larger one is colorless and "blood-cell-shaped," and the smaller one is yellowish. Diaxonella pseudorubra pseudorubra is different from the most similar subspecies, D. pseudorubra pulchra, in cytoplasmic color and number of midventral cirri. Pseudokeronopsis flava is different from its most similar congeners in pigment granular color, number of bicorona, number of midventral cirri, and position of the contractile vacuole.

  • The results of influence on the saccharification and $Sikhea$ by adding mulberry fruit in this study are as follows. pH was decreased with the amount of mulberry fruit increased during the saccharification period. A significant change was not found after 4 hours of saccharification. It is considered that 4-5 hours was proper for saccharification because brix increased by more than 1 brix per hour up to 4 hours while there were little change after 5 hours. L was decreased while a and b values were increased in the Hunter' value of mulberry fruit $Sikhea$. The DPPH free radical scavenging activities and SOD-like activity of mulberry fruit $Sikhea$ were increased as the addition quantity of mulberry fruit increased. Anthocynin color pigment in mulberry fruit has the inhibiting effect on microbes. The preferences of mulberry fruit $Sikhea$ were 10% < 30% < 15% < 20% < 0% < 25%, and less than 25% of mulberry fruit addition seems to be desirable. The addition of mulberry fruit will help decrease the amount of sugar to use in $Sikhea$ because the sugar level of $Sikhea$ increases with the addition of mulberry fruit. It will also improve the preference for Sihkea by giving it better color and sourness.

Enhancement of Skin Color by Postharvest UV Irradiation in 'Fuyu' Persimmon Fruits (수확 후 자외선 조사에 의한 '부유' 단감의 과피 착색 증진)

  • The effects of UV irradiation, as a hormetic stimulus, on the postharvest persimmon fruits (Diospyros kaki cv. Fuyu) were investigated in regards to the change of carotenoid contents and flesh softening, when the UV irradiation was combined with or without the pretreatment of 1-methylcyclopropene (1-MCP) as an ethylene action inhibitor. The major carotenoid pigments in persimmon fruits were ${\beta}$-carotene, lycopene and ${\beta}$-cryptoxanthin. Of them, the lycopene was a pigment, which increased markedly after harvest. UV irradiation increased the contents of ${\beta}$-carotene and lycopene, enhancing the skin color to scarlet. The treatment accelerated however also the softening of fruit flesh. But the softening of UV irradiated fruits could be delayed significantly by pretreatment with 1-MCP without reducing the advantageous effect of UV irradiation on the carotenoid increase.

  • Paint is used as top coat in Automobile Refinishing that is divided into 1K paint of base type and 2K paint of urethane type. after using 1K paint, it can be reusable. However 2K paint is not available that is mixed a certain amount of base with hardner, so 2K paint has to be used all at a time or discarded the rest. This study covered that colored Urethane paint and colorless urethane paint in top coating process of AR. Top coat urethane paint is divided into colored urethane paint and transparent urethane paint like clear coat. Colored urethane paint is used for paint that contains resin, pigment, solvent and additives. It is color such as black and white. While transparent urethane paint is used for paint that contains resin, solvent and additives. Urethane paint has a paint system that cur after several hours of mixing base and hardner. By a paint technician of a field and empirical data, we can also recognize that according to worker's paint style and experience, although he or she paint same car model and area, paint use of one's is different. There aren't guidelines workers refer to about paint use by car models and work scope because ARP works various car models that are in different color, size and area. After using urethane paint, the rest of paint has come, and there are highs and lows in paint use each time he or she works. Therefore, this study suggest a basic guideline to use paint efficiently and regularly by understanding area, the type of work, and paint use that are appropriate for each car model and regardless of workers and work scopes in AR.

  • The cutaneous erythrophore differentiation from larvae to adult in bombina orientalis-the Korean fire bellied toad-is discussed electron microscopically to reveal the structural changes of carotenoid vesicles and pterinosomes in the process of erythrophore differentiation. Typical structure of adult erythrophores distributed beneath the basement membrane is composed much carotenoid vesicles and a small quantity of pterinosomes. Marginal electron density of these carotenoid vesicles is higher than those of internal regions and pterinosomes have many concentric lamellae. The very first differentiated erythrophores are apeared before and after the foreleg developing stage of metamorphosis and red ventral color is just appeared about $1\\sim2$ weeks after metamorphosis. About 1 year after metamorphosis, external body color of the young toad is similar to adult but the morphology of pigment granules observed in these erythrophores differs from that of adult in structure and electron density. At this stage, the differentiated level of pterinosomes is not remarkable and the accumulation state of carotenoid pigments in erythrophores is also relatively low.

  • A visual field tester using a fixation target with a lack of distinctiveness decreases accuracy and usability in visual field testing. The present study is intended to develop various induction methods of gaze fixation for effective visual field testing. Proposed were four new gaze fixation induction methods (color changing dot; alphanumeric characters; flashing black dot; and bulls eye and cross hair, BECH) by considering visual attention factors such as color, meaning, flashing, and shape and the proposed methods were compared with the existing black dot (BD) method in terms of gaze fixation performance and subjective satisfaction by 32 participants in their 20s to 30s. BECH was found most preferred by increasing gaze fixation performance by 4.8% and subjective satisfaction by 0.4 to 2.0 in a 7-point scale compared to BD. BECH can be applied to tests such as visual field testing and macular pigment optical density testing in which gaze fixation is crucial for accuracy and usability.

Substitution of Pro206 and Ser86 Residues in the Retinal Binding Pocket of Anabaena Sensory Rhodopsin is Not Sufficient for Proton Pumping Function

  • Anabaena sensory rhodopsin is a seven transmembrane protein that uses all-trans/13-cis retinal as a chromophore. About 22 residues in the retinal-binding pocket of microbial rhodopsins are conserved and important to control the quality of absorbing light and the function of ion transport or sensory transduction. The absorption maximum is 550 nm in the presence of all-trans retinal at dark. Here, we mutated Pro206 to Glu or Asp, of which the residue is conserved as Asp among all other microbial rhodopsins, and the absorption maximum and pKa of the proton acceptor group were measured by absorption spectroscopy at various pHs. Anabaena rhodopsin was expressed best in Escherichia coli in the absence of extra leader sequence when exogenous all-trans retinal was added. The wild-type Anabaena rhodopsin showed small absorption maximum changes between pH4 and 11. In addition, Pro206Asp showed 46 nm blue-shift at pH7.0. Pro206Glu or Asp may change the contribution to the electron distribution of the retinal that is involved in the major role of color tuning for this pigment. The critical residue Ser86 (Asp 96 position in bacteriorhodopsin: proton donor) for the pumping activity was replaced with Asp, but it did not change the proton pumping activity of Anabaena rhodopsin.
