공시균인 Saccha. erythraea IFO 13426으로부터 VB-C에 의한 erythromycin 생산 유도능이 시사된 바 있으므로, 공시균으로부터 VB-C와 특이적으로 결합하는 autoregulators 및 receptor gene을 탐색하여, EM의 생산 조절 기구를 규명하고자 하였다. 탐색의 일환으로 기존의 Streptomyce속 receptor gene의 공통배열을 primer로 이용하여 PCR을 수행하였고, 예상 크기인 120bp의 단편을 pUC19 vector에 ligation하여 E. coli DH5$\alpha$에 형질전환한 후, plasmid를 분리하여 BamHI을 처리하여 2% agarose gel에 전기영동한 결과, pUC19 (2.7kbp)외에 receptor gene PCR 산물이 120bp위치에 존재하는 것을 확인하였다. 형질전환된 plasmid로 PCR을 수행하여 염기배열을 결정한 후 해석한 결과 Streptomyces sp. 유래의 receptor gene과 유사함을 확인하였다. 따라서 Saccha. erythraea IFO 13426에는 항생물질인 erythromycin의 생산에 관여한다고 추정되는 autoregulator receptor protein을 코드하는 유전자가 존재할 것으로 예상되어 120 bp의 PCR product를 probe로 이용하여 Southern 및 colony hybridization을 통하여 3.2 kbp의 SacI 단편을 가지는 plasmid(pESG)를 제작하였고, 이를 sequencing한 결과, autoregulator receptor protein 유전자가 KpnI과 SalI을 포함하는 영역에 존재한다는 것을 알 수 있었으며 이를 EsgR이라 명명하였다. 유전자 해석 결과, EsgR은 205개의 아미노산으로 구성되어 있으며, 이는 기존의 autoregulator receptor proteins과 비교시 30%이상의 상동성을 나타내었으며, 기존의 autoregulator receptor prorein들이 하부의 항생물질 생합성 유전자들의 제어를 위해 보유하고 있는 helix-turn-helix DNA binding motif를 EsgR이 보유하고 있는 점에서, EsgR은 Saccha. erythraea가 보유하는 autoregulator receptor protein을 code하는 유전자로 추정되었다.
A rapid and quantitative real-time PCR was developed to target the invasion A (invA) gene of Salmonella spp. We developed quantitative standard curves based on plasmids containing the invA gene. Based on these curves, we detected Salmonella spp. in artificially contaminated buffered peptone water (BPW) and milk samples. We were able to determine the invA gene copy number per ml of food samples, with the minimum detection limit of $4.1{\times}10^{3}$ copies/ml of BPW and $3.3{\times}10^{3}$ copies/ml of milk. When applied directly to detect and quantify Salmonella spp. in BPW and milk, the present real-time PCR assay was as sensitive as the plate count method; however, copy numbers were one to two logs higher than the colony-forming units obtained by the plate count methods. In the present work, the real-time PCR assay was shown to significantly reduce the total time necessary for the detection of Salmonella spp. in foods and to provide an important model for other foodborne pathogens.
돼지고기에 오염된 살모넬라의 수를 직접 측정하는 방법으로 경쟁적 PCR(competitive PCR)을 사용하였다. 세가지 DNA추출방법을 비교한 후 guanidine thiocyanate-phenol-chloroform 방법을 일부 수정하여 인위적으로 살모넬라를 오염시킨 돼지고기로부터 DNA를 직접 추출하는 방법으로 선택하였다. Rahn 등(Mol. Cell. Probes 1992년)이 이미 보고한 284-bp iuvA gene의 특이성을 평가하기 위해 살모넬라 57균주와 살모넬라가 아닌 균주 24개를 사용하였다. 시험해 본 모든 살모넬라 균주는 invA 양성으로 살모넬라가 아닌 모든 균주는 invA 음성으로 판명되었으며 살모넬라가 아닌 균주 중 양성으로 잘 못 판명된 경우는 없었다. 돼지고기 0.1 g을 사용할 경우 검출한계는 1,460 cfu였다. 경쟁적 PCR을 위해 invA gene중 82-bp를 결실시킨 DNA조각을 pGEM-4Z백터에 클로닝을 하였다. 인위적으로 오염된 돼지고기로부터 추출한 살모넬라 염색체 DNA와 이와 같은 primer결합자리를 가진 경쟁 DNA를 동시에 경쟁적 PCR로 증폭하였다. 경쟁적 PCR을 사용하여 결정한 균수와 MPN방법으로 결정한 균수는 거의 유사하였으며 전체 과정을 수행하는 데 약 5시간이 소요되었다.
모넨신, 니저리신, 사이클로스포린 등을 하이드록실레이션 시키는 균주인 S. benihana에 존재하는 여러 가지 CYP를 클로닝하기 위해, 방선균 CYP의 보존된 부분을 통해서 degenerate primer를 제작하였고, colony hybridization을 통해서 스크리닝 한 결과 총 5 종류의 CYP가 검색되었다. 아미노산 서열의 분석 결과 방선균의 CYP 들과 매우 높은 유사성을 가졌으며, 이들 CYP의 앞 뒤 서열의 검색 결과 이 중 4개의 CYP의 downstream에는 FD 유전자가 존재함을 알 수 있었다. CYP503의 경우 다른 나머지 4개의 CYP의 서열과 차이가 많았으며, 2차 대사산물의 변형과 관련되어 있을 것으로 예상되며, ChoP와 유사성을 보이는 나머지 4개의 CYP는 스테로이드 계열 물질의 하이드록실레이션과 밀접한 연관이 있을 것으로 추정된다.
High expression of mitofusin-2 (MFN2), a mitochondrial fusion protein, has been frequently associated with poor prognosis of patients with cervical cancer. Here, we aimed to identify the function of MFN2 in cervical cancer to understand its influence on disease prognosis. To this end, from cervical adenocarcinoma, we performed an MTT assay and quantitative RT-PCR (qRT-PCR) analysis to assess the effects of MFN2 on the proliferation and of HeLa cells. Then, colony-formation ability and tumorigenesis were evaluated using a tumor xenograft mouse model. The migration ability related to MFN2 was also measured using a wound healing assay. Consequently, epithelial-mesenchymal transition (EMT) of MFN2-knockdowned HeLa cells originating from adenocarcinoma. markers related to MFN2 were assessed by qRT-PCR. Clinical data were analyzed using cBioPortal and The Cancer Genome Atlas. We found that MFN2 knockdown reduced the proliferation, colony formation ability, migration, and in vivo tumorigenesis of HeLa cells. Primarily, migration of MFN2-knockdowned HeLa cells decreased through the suppression of EMT. Thus, we concluded that MFN2 facilitates cancer progression and in vivo tumorigenesis in HeLa cells. These findings suggest that MFN2 could be a novel target to regulate the EMT program and tumorigenic potential in HeLa cells and might serve as a therapeutic target for cervical cancer. Taken together, this study is expected to contribute to the treatment of patients with cervical cancer.
Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a multifunctional cytokine that has been implicated in the regulation of pre-implantation embryo development across several species. The aim of this study was to determine the effects of mouse granulocyte-macrophage colony-stimulating factor (mGM-CSF) on development of porcine parthenotes and nuclear transferred embryos, and on their expression of implantation-related genes. In the presence of bovine serum albumin, mGM-CSF did not increase the percentage of oocytes that developed to the blastocyst stage and at day 7 did not increase oocyte cell number. Addition of 10 mM GM-CSF to protein-free culture medium significantly increased the compaction and blastocoel formation of 1- to 2-cell parthenotes and cloned embryos developing in vitro. However, cell number was not increased when they were cultured in the presence of GM-CSF. Semi-quantitative reverse transcripts polymerase chain reaction (RT-PCR) revealed that mGM-CSF enhances mRNA expression of the leukemia inhibitory factor receptor, but does not influence interleukin-6 or sodium/glucose co-transporter protein gene expression in blastocyst stage parthenotes. These results suggest that mGM-CSF may enhance viability of porcine embryos developing in vitro in a defined culture medium.
Overexpression of mitofusin-2 (MFN2), a mitochondrial fusion protein, is frequently associated with poor prognosis in cervical cancer patients. Here, I aimed to investigate the involvement of MFN2 in cervical cancer progression and determine the effect of MFN2 on prognosis in cervical cancer patients. After generating MFN2-knockdown SiHa cells derived from squamous cell carcinoma, I investigated the effect of MFN2 on SiHa cell proliferation using the Cell Counting Kit-8 assay and determined the mRNA levels of proliferation markers. Colony-forming ability and tumorigenesis were evaluated using a colony-formation assay and tumor xenograft mouse models. The migratory and invasive abilities associated with MFN2 were measured using wound-healing and invasion assays. Wnt/β-catenin-mediated epithelial-mesenchymal transition (EMT) markers related to MFN2 were assessed through quantitative RT-PCR. MFN2-knockdown SiHa cells exhibited reduced proliferation, colony formation, migration, invasion, and tumor formation in vivo. The motility of SiHa cells with MFN2 knockdown was reduced through Wnt/β-catenin-mediated EMT inhibition. MFN2 promoted cancer progression and tumorigenesis in SiHa cells. Overall, MFN2 could serve as a therapeutic target and a novel biomarker for cervical cancer.
A multiplex competitive polymerase chain reaction (PCR) method was developed for the rapid identification and quantification of Leuconostoc mesnteroides and Lactobacillus plantarum populations which are the key microorganisms in kimchi fermentation. The strain-specific primers were designed to selectively amplify the target genes encoding 165 rRNA of L. plantarum and dextransucrase of L. mesenteroides. There was a linear relationship between the band intensity of PCR products and the number of colony forming units of each model organism. The PCR quantification method was compared with a traditional plate-counting method f3r the enumeration of the two lactic acid bacteria in a mixed suspension culture and also applied to a real food system, namely, watery kimchi. The population dynamics of the two model organisms in the mixed culture were reliably predictable by the competitive PCR analysis.
2021년 5월 화상병 확산 방지를 위해 화상병 예찰 중에 충북 음성의 화상병이발병한 배 과수원 바로 인근의 밭에서 자라는 팥배나무의 어린 가지가 수침상을 보이면서 마르는 증상을 발견하였다. 채집한 잎과 어린 가지로부터 균 분리를 진행한 결과 화상병원균과 유사한 유백색의 mucoid한 colony를 보이는 2개 균주를 순수 분리하였다. 순수 분리된 병원균을 화상병원균 특이 프라이머 PCR 검정 및 16S rRNA 염기서열을 이용한 계통도 분석을 통해 분리된 균주가 Erwinia amylovora임을 확인하였다. 배 유과와 사과 대목에 병원성 검정을 수행한 결과 접종 6일 후에 접종한 자리에 괴사 증상과 우즈를 확인할 수 있었고, 이들 증상에서 동일한 균을 재분리하였다. 본 연구는 국내에서 최초로 E. amylovora에 의해 발생한 팥배나무에서의 화상병을 보고하고자 한다.
Shiga 독소 생성 대장균(Shiga toxin-producing Escherichia coli; STEC)을 가장 빠르게 검출할 수 있는 초고속 이단계 PCR 방법을 개발하였다. 검색 대상 유전자는 STEC에서 생성되는 Shiga 독소(Shiga toxin; Stx)를 암호화하고 있는 유전자 stx1이며, 1쌍의 stx 유전자 특이 primer를 사용하여 검출을 수행하였다. 초고속 PCR (Ultra-rapid PCR)은 microchip 기반의 6 ${\mu}l$ PCR 용량의 Real-time PCR을 사용하고, PCR 회전의 각 단계 중 혼성과 중합을 한 단계로 하였을 뿐 아니라, 각 단계의 적용시간을 각 1초, 3초(해리, 혼성/중합)가 되게 극단적으로 줄여, 검사소요시간을 최소화하였다. 35회전의 PCR 진단에 사용된 시간은 6분38초였으며, 용융온도분석에서 stx1 특이 유전자가 검출되었음을 확인하는 데까지 총 7분 28초가 소요되었다. 또한 민감도 측정에서 $3{\times}10^0$ CFU/reaction까지 성공적으로 검출 가능함이 확인되었고, 용융온도분석에서 이 증폭산물은 일정한 $81.42{\pm}0.34^{\circ}C$의 용융온도를 갖는 것으로 확인되었다. 이 검사법을 다양한 STEC 균주들에게 적용하여 그 성능을 검증하였으며, 이로써 본 초고속 이단계 PCR 방법은 Shiga 독소 생성 대장균의 초신속 검출에 바로 적용될 수 있을 것으로 기대한다.
