Genetic Diversity and Differentiation of Colletotrichum spp. Isolates Associated with Leguminosae Using Multigene Loci, RAPD and ISSR

  • Genetic diversity and differentiation of 50 Colletotrichum spp. isolates from legume crops studied through multigene loci, RAPD and ISSR analysis. DNA sequence comparisons by six genes (ITS, ACT, Tub2, CHS-1, GAPDH, and HIS3) verified species identity of C. truncatum, C. dematium and C. gloeosporiodes and identity C. capsici as a synonym of C. truncatum. Based on the matrix distance analysis of multigene sequences, the Colletotrichum species showed diverse degrees of intera and interspecific divergence (0.0 to 1.4%) and (15.5-19.9), respectively. A multilocus molecular phylogenetic analysis clustered Colletotrichum spp. isolates into 3 well-defined clades, representing three distinct species; C. truncatum, C. dematium and C. gloeosporioides. The ISSR and RAPD and cluster analysis exhibited a high degree of variability among different isolates and permitted the grouping of isolates of Colletotrichum spp. into three distinct clusters. Distinct populations of Colletotrichum spp. isolates were genetically in accordance with host specificity and inconsistent with geographical origins. The large population of C. truncatum showed greater amounts of genetic diversity than smaller populations of C. dematium and C. gloeosporioides species. Results of ISSR and RAPD markers were congruent, but the effective maker ratio and the number of private alleles were greater in ISSR markers.

Genetic Variation of Scytalone Dehydratase Gene in Colletotrichum spp. (Colletotrichum spp. 유래 scytalone dehydratase 유전자의 유전적 다양성 비교)

  • The production of dihydroxynaphthalene (DHN) melanin is known to be essential factor for pathogenicity in Colletotrichum lagenarium. However, the genetic diversity of melanin genes was not much known among Colletotrichum spp. To investigate the variability of melanin gene in Colletotrichum spp. that cause anthracnose on diverse crops including tomato, we cloned and sequenced partial sd, one of DHN melanin genes encoding for scytalone dehydratase, from eight strains of C. coccodes, C. acutatum, C. truncatum C. caricae, and C. musae. The size of PCR-amplified sd ranged 437 bp to 545 bp. The nucleotide sequence identity of sd among the Colletotrichum strains tested varied from 49% to 99%. All of the PCR-amplified sd from eight strains contain an intron and have two exons coding for 122 amino acids. Overall, the size and nucleotide sequence of sd varied among the five Colletotrichum spp. Sequence identity of the predicted scytalone dehydratase protein of 122 amino acids ranged 50 to 99%. Phylogentic analysis based on the sd nucleotide sequences revealed that the five Colletotrichum spp. could be genetically divided.

  • Colletotrichum species are important fungal pathogen that cause great damages on various host plant species worldwide. In Korea, Colletotrichum species cause massive economic losses on apple, peach, grape, and essecially, sweet persimon productions. In the past, Identification of the pathogen and the studies on the genetic relationships among the pathogenic isolates were mainly based on morphology, cultural characteristics, and the difference in pathogenicity. However, in recent years, these traditional methods have been replaced with molecular methods to solve the difficulty of classification on pathogens. Therefore, in this study, RAPD and PCR-RFLP methods were employed for the studies of genetic relationship among the different isolates of Colletotrichum species that cause damages on sweet persimon. As a results of genetic relationship analysis, Colletotrichum species tested were divided into two big groups or five small groups.

  • Paclitaxel (taxol) has long been used as a potent anticancer agent for the treatment of many cancers. Ever since the fungal species Taxomyces andreanae was first shown to produce taxol in 1993, many endophytic fungal species have been recognized as taxol accumulators. In this study, we analyzed the taxol-producing capacity of different Colletotrichum spp. to determine the distribution of a taxol biosynthetic gene within this genus. Distribution of the taxadiene synthase (TS) gene, which cyclizes geranylgeranyl diphosphate to produce taxadiene, was analyzed in 12 Colletotrichum spp., of which 8 were found to contain the unique skeletal core structure of paclitaxel. However, distribution of the gene was not limited to closely related species. The production of taxol by Colletotrichum dematium, which causes pepper anthracnose, depended on the method in which the fungus was stored, with the highest production being in samples stored under mineral oil. Based on its distribution among Colletotrichum spp., the TS gene was either integrated into or deleted from the bacterial genome in a species-specific manner. In addition to their taxol-producing capacity, the simple genome structure and easy gene manipulation of these endophytic fungal species make them valuable resources for identifying genes in the taxol biosynthetic pathway.

  • Colletotrichum species are important fungal pathogens that cause great damages on various host plant species worldwide. In Korea, Colletotrichum species cause massive economic losses on apple, peach, grape, and especially, sweet persimon productions. In the past, identification of the pathogen and the studies on the genetic relationships among the pathogenic isolates were mainly based on morphology, cultural characteristics, and the difference in pathogenicity. However, in recent years, these traditional methods have been replaced with molecular methods including AFLP. AFLP method with the merits of both RAPD and RFLP has been widely used for the genetic relationship studies of various organisms. Therefore, in this study, AFLP method was employed for the studies of genetic relationships among the different isolates of Colletotrichum species collected from various parts of sothern Korea. As a result, two specific band pattern groups were observed among different isolates of Colletotrichum species.

Fungicide Selection for Control of Lycium chinense Anthracnose Caused by Colletotrichum spp. (구기자 탄저병 방제를 위한 살균제 선발)

  • Anthracnose caused by Colletotrichum spp. has been known as a significant disease which commonly infects to the fruits of Chinese matrimony (Lycium chinese) in the field conditions. To select effective fungicides for the control of Chinese matrimony anthracnose, the antifungal activity of 15 fungicides were evaluated with 13 different strains of Colletotrichum spp. in the laboratory condition. Six fungicides (Dithianon WG, Tebuconazole SC, Tebuconazole WG, Trifloxystrobin + Tebuconazole SC, Azoxystrobin SC and Polyoxin D zinc salt + Carbendazim WP) out of them, showed effective suppression with the mycelium growth of pathogenic fungus, and were selected to test in vivo of the field condition. Five fungicides, Dithianon WG, Tebuconazole WG, Trifloxystrobin + Tebuconazole SC, Tebuconazole SC and Azoxystrobin SC, were significantly effective to protect anthracnose of Chinese matrimony, the variety "Chungyang Jerae".

Development of Rapid Molecular Detection Marker for Colletotrichum spp. in Leaf and Fruit Tissues of Sweet Persimmon

  • Sweet persimmon (Diospyros kaki Thunb.) is widely cultivated in the southern part of Korea and its cultivation is increasing. However, anthracnose disease caused by Colletotricuhum species is one of the major hinderances to the cultivation and production of sweet persimmon. Therefore, in the current study, PCR was used to specifically detect Colletotrichum spp., based on the sequences of the ITS II regions in the rDNA. Using the sequence data, CO-1 was designated to detect Colletotrichum together the with ITS 4 primer. The result showed that a single segment of ca. 500 bp was observed only in Colletotrichum, but not in any other fungal and bacterial isolates. The annealing temperatures and template DNA quantites were also investigated to identify optimal conditions for detection. Using these species-specific primers, a unique band was obtained at annealing temperatures ranging from $55^{\circ}C\;and\;61^{\circ}C$ and template DNA levels from 10 pg- $10{\mu}g$.

Comparison of Pepper Anthracnose Pathogens from Korea and China

  • Anthracnose of pepper casued by Colletotrichum spphas been a great problems for pepper production in Korea and China. Especially Colletotrichum gloeosporioides was found predominantly over cultivation areas during infection periods and caused severe rots on bath unripe and ripe fruits that resulted in major yield losses. In this study, comparison of Colletrichum spp.isolated from Korea and China in morphology and pathgenicity, and RAPD-PCR analysis were conducted. Based on morphological characteristics, the pathogen isolates, K1 and C1, K2 and C2, and K3 and C3 were identified as Colletotrichum gloeosporioides (G) type, C. gloeosporioides (R) type and C. coccodes, respectively. in pathogenicity test, K1 and C1, and K2 and C2 were found to attack mainly fruits and to be the most virulent among isolates. K3 and C3 were strongly virulent to leaves and seedling. Pathogenicity between Korean and Chinese isolates. K3 and C3 were strongly virulent to leaves and seedling. Pathogenicity between Korean and Chinese isolates did not show any difference. Results of the RAPD-PCR analyses indicate the varying levels of molecular diversity within and between Colletotrichum spp.of Korea and China. The similarities between K1 and C1, K2 and C2, and K3 and C3 were 85.71%, 71.43% and 50.0% respectively.

Anthracnose of Blueberry (Vaccinium spp.) of Caused by Colletotrichum gloeosporioides in Korea (Colletotrichum gloeosporioides에 의한 블루베리 탄저병 발생)

  • Anthracnose of blueberry (Vaccinium spp.) caused by Colletotrichum gloeosporioides was observed in the exhibition orchard of Gyeongsangnam-do Agricultural Research and Extension Services in Korea, 2007. The typical symptoms were initially water-soaked lesions and then rotten on fruits, the infected fruits were dropped but some of them were hunged and mummified. The pathogenic fungus grown on potato dextrose agar showed greyish to dark gray color. Conidia were straight, cylindrical apex obtuse in shape and $10{\sim}20{\times}3{\sim}5\;{\mu}m$ in size. Appressoria were brown in color, clavate or ovate in shape and $6{\sim}18{\times}4{\sim}12\;{\mu}m$ in size. Optimum temperature for mycelial growth was $25{\sim}30^{\circ}C$. On the basis of mycological characteristics and pathogenicity test on host plants, the fungus was identified as Colletotrichum gloeosporioides (Penz.) Penz. & Sacc.. This is the first report on the anthracnose of blueberry (Vaccinium spp.) caused by C. gloeosporioides in Korea.

Anthracnose of Perilla Caused by Colletotrichum spp. and Glomerella cingulata

  • Serve outbreaks of anthracnose were observed on perilla plants grown in greenhouses and open fields in several locations in Korea during the disease survey from 1997 to 2000. A total of 53 isolates of Colletotrichum spp. and Glomerella sp. was obtained from diseased perilla plants and identified based on their morphological and cultural characteristics. Forty isolates were identified as Colletotrichum gloeosporioides, three isolates as C. coccodes, five isolates as C. dematium, and the other five isolates as Glomerella cingulata, the teleomorph of C. gloeosporioides. All isolates of C. gloeosporioides tested by artificial inoculation were strongly virulent on perilla plants, but isolates of the other species were weakly or not virulent. Anthracnose symptoms induced on the perilla plants by artificial inoculation with the isolates of C. gloeosporioides were similar to those observed in the fields. This study revealed that C. gloeosporioides is the main causal fungus of perilla anthracnose.
