Development of Hepatocyte Spheroids Immobilization Technique Using Alternative Encapsulation Method

  • Primary hepatocytes of small animals such as rat and rabbit were often used for the study of extracorporeal liver support systems. Freshly isolated rat hepatocytes form spheroids within tow days when cultivated as suspension in spinner vessels. These spheroids showed enhanced liver specific functions and more differentiated morphology compared to hepatocytes cultured as monolayers However, shear stress caused by continuous agitation deteriorated spheroids gradually. In this work we immobilized spheroids to prolong liver specific activities. First, hepatocyte spheroids were suspended in collagen solution containing calcium chloride and then dropped into alginate solution. A thin layer of calcium alginate was formed around the droplet and then was removed after the inner collagen was gelled by treatment of sodium citrate buffer. Spheroids embedded in collagen-gel bead maintained liver specific functions such as albumin secretion rate longer than hepatocyte spheroids exposed to shear stress. Therefore, we suggest that this immobilization technique may offer an effective long-term hepatocyte cultivation and facilitase the development of a bioartificial liver support device.

Insulin 투여가 백서치수 및 치은의 Collagen 생합성에 미치는 영향 (EFFECT OF INSULIN ADMINISTRATION ON THE COLLAGEN BIOSYNTHESIS IN INCISOR PULP AND GINGIVA OF RAT)

  • The effect of insulin on the collagen metabolism in dental pulp and gingiva was studied using Sprague-Dawley rat and ³H-proline was injected intraperitoneally and then sacrificed according to predetermined schedule. Lower incisor pulp and gingiva were removed immediatedly and hydrolysed with acid. After digestion, proline and hydroxyproline were separated and analysed quantitatively employing thin-layer chromatography and radioactivity of each amino acid was counted by liquid scintillation counter. The label incorporated into lower incisor pulp and gingiva and radioactivity of ³H-hydroxyproline were increased in insulin administered group. And disappearance rate of ³H-hydroxyproline was decreased. Those results indicated that amino acid, I.e. proline transport and collagen biosynthesis were increased in dental pulp and gingiva by insulin administration.

Optical Characteristics of Corneal Nanostructure According to the Angle of Collagen-fiber-layer Arrangement

  • Collagen fibers tens of nanometers in size, which constitute most of the corneal volume of the human eye, are layered in a uniform direction, and adjacent fiber layers are arranged at an angle of 90° to each other. According to the results of this study, the transmittance at 45° of interlayer rotation angle is highest, and higher than that of the 90° body structure. The transmittance is examined, concerning the polarization state of the incident light; circularly polarized light case shows higher transmittance than linearly polarized. Through this, a simulation to confirm the deformed structure of collagen fibers, which show higher transmittance than the anatomical structure of the cornea, is attempted.

Inhibitory Effects of Moutan Cortex Radicis Extracts and Paeonol on Rabbit Platelet Aggregation

  • The present study was undertaken to investigate the effects of Moutan Cortex Radicis extracts and paeonol, a major component, on rabbit platelet aggregation and thromboxane (TX) $A_2$ formation. Moutan Cortex Radicis methanol and butanol layers (100 ${\mu}g/mL$) showed the weak inhibitions, and water layer (100 ${\mu}g/mL$) had no effect on the collagen-induced platelet aggregation. Whereas, hexane and EtOAc layers potently inhibited the collagen (3 ${\mu}g/mL$)-induced platelet aggregation with the $IC_{50}$ values of 10.9${\pm}$1.0 and 31.5${\pm}$0.8 ${\mu}g/mL$, respectively. Paeonol isolated from the hexane-acetone layer specifically inhibited the collagen-induced platelet aggregation with the $IC_{50}$ value of 113.1 ${\pm}$ 0.9 ${\mu}M$, whereas it had little effects on the other agonists such as AA-, thrombin-, A23187- and thapsigargin-induced platelet aggregations. Paeonol also potently inhibited the collagen-induced TXB formation in rabbit platelet in a concentration-dependent manner. These results suggest that paeonol may inhibit rabbit platelet aggregation by interfering with an essential step in collagen-induced platelet activation and $TXA_2$ formation. Paeonol may be a promising candidate for an antiplatelet agent.

Application of 630-nm and 850-nm Light-emitting Diodes and Microcurrent to Accelerate Collagen and Elastin Deposition in Porcine Skin

  • Background and Objectives Skin aging is reportedly associated with regulation in collagen and elastin synthesis. This study investigated the potential of combining light-emitting diode (LED) treatments using a 630-nm and 850-nm LED with simultaneous microcurrent application. Materials and Methods The dorsal skin of female pigs was treated with a home-use device. We examined the treatment effects using photography, thermocamera, microscopic pathology, and histological examination to determine the mechanism of action, efficacy, and safety of the procedure. A histological observation was performed using hematoxylin and eosin, Masson's trichrome, Victoria blue, and immunohistochemical staining. We also used the Sircol soluble collagen and elastin assay kit to measure the amounts of collagen and elastin in the porcine back skin tissue after 2 and 6 weeks. Results Evaluation by visual inspection and devices showed no skin damage or heat-induced injury at the treatment site. Histological staining revealed that accurate treatment of the targeted dermis layer effectively enhanced collagen and elastin deposition. Collagen type I, a protein defined by immunohistochemical staining, was overexpressed in the early stages of weeks 2 and 6. Combined therapy findings showed the superior capability of the 630-nm and 850-nm LED procedures to induce collagen; in contrast, elastin induction was more pronounced after microcurrent treatments. Conclusion The home-use LED device, comprising a combination of 630-nm and 850-nm LEDs and microcurrent, is safe and can be used as an adjunctive treatment for self-administered facial rejuvenation.

Inhibitory Effects of Collagen Coated Coffee Bean Intake on Skin Aging

  • To evaluate the protective effect of collagen peptide-coated coffee extract on skin aging, cell viability was measured with a MTT assay using cultured CCD-986sk fibroblasts, and its effect on wrinkles in the skin of hairless mice induced by UVB-irradiation was examined. In addition, its effect on procollagen synthesis and anti-oxidative, and its inhibitory activity against collagenase, elastase, tyrosinase and MMP-1 were analysed. After the 30-minute topical treatment, the animals were exposed to UVB irradiation (60-100 mJ/cm2) for 4 weeks and its intensity increased during the period. Under the experimental conditions set in this study, the skin thickness of hairless mice significantly decreased (11.8-21.3%) compared to the control group. Based on these results, the prolonged oral intake of a collagen peptide mixture with coffee is expected to significantly increase the synthesis of procollagen in dermal fibroblasts, thereby contributing to the alleviation of wrinkling and lowered elasticity due to structural damage to the dermal layer caused by UV. The oral intake of collagen-coated coffee contributes to increasing collagen biosynthesis in a dose-dependent manner and alleviates the symptoms of thickened keratin caused by UV irradiation. However, it did not inhibit the enzymes involved in skin aging, whitening, wrinkle improvement, and antioxidation. Based on the these results, it can be concluded that the intake of collagen peptide-coated coffee extract can be utilized as an alternative material for the prevention or treatment of diseases associated with photoaging.

Habitats and Air Uptake Based on Analysis of Skin Structure of Two Korean Bullheads, Pseudobagrus brevicorpus and P. koreanus (Pisces; Bagridae}

  • The skin of the Korean bullheads, Pseudobagrus brevicorpus and P. koreanus, is composed of epidermis, dermis and subcutis. The epidermis has three layers, the outermost layer, middle layer and stratum germinativum. The epidermis consists of two types of gland cells, an unicellular mucous cell of sulfomucin and a large club cell having sometimes two nuclei. The epidermis has numerous intraepithelial blood vessels in P. brevicorpus but not at all in P. koreanus. Lymphatic spaces containing lymphocytes are well developed in mainly the stratum germinativum. The dermis lacks scales and consists mostly of bundles of coarse collagen fibers. The collagen bundles are arranged in parallel to each other in the dorsum and lateral region toward the dorsum, but vertically at intervals in the abdomen and lateral region toward the abdomen. Considering this unique skin structure, the two species are likely to exercise cutaneous respiration as a dual respiratory system to overcome hypoxic conditions which frequently occurs in their habitats.

  • The effect of moistening and air-drying of acid-conditioned dentin before priming on the formation of resin-dentin hybrid zone was investigated, Freshly extracted human molars were used and divided at random into 5 groups, Groups 1 - 3 consisted of specimens conditioned with 10 % phosphoric acid for 20 seconds; Group 1 served as a control in which the conditioned dentin was simply blot-dried with a damp facial tissue; Group 2 was air dried for 30 seconds ; Group 3 was air dried for 30 seconds and immediately remoistened for 10 seconds with air-water syringe. and then the specimen was blot-dried with a damp facial tissue. Groups 4-5 were not acid conditioned ; In group 4, the smear layer on the dentin was blot dried before primer placement; Group 5 was air dried only for 30 seconds, The acetone-based primer and bonding agent of All Bond 2 (Bisco. Inc., USA) and composite resin (Z-100, 3M Dental products, USA) were applied for acid conditioned dentin and non-conditioned dentin. The morphologic ultrastructure of resin-dentin hybrid zone was examined by the use of SEM and TEM. and the existence of inorganic material and analysis of Ca/P weight-percent ratio in the resin-dentin hybrid zone were revealed by the EDAX, The results were as follows : 1. In the moistened specimens from acid-conditioned groups, the resin penetrated about 3-$4{\mu}m$ into dentin and the denatured collagen smear layer was not present at the surface. The resin tag was formed to a thickeness of 3-$4{\mu}m$ at the upper part of dentinal tubule and compactively connected to each other by means of many lateral branching. 2. In the air-dried specimens from acid-conditioned groups, the resin penetrated about 2.0-$2.5\;{\mu}m$ into dentin and an upper thin black layer to a thickness of 30-35nm was identified between adhesive resin and demineralized collagen layer. The resin tag to have a diameter of $2.5{\mu}m$ was formed at the upper part of dentinal tubule. However the funnel shape of the tag was not notable compared to the moistened specimens. 3. In the remoistened specimens from acid conditioned groups, the resin penetrated about 2.0-$2.5{\mu}m$ into dentin and an upper black layer was not present. The resin tag at the upper part of dentinal tubule was formed less than $2{\mu}m$ and was weakly connected to each other by means of few lateral branching. 4. In the non-conditioned groups, the smear layer was formed to a thickness of $0.5{\mu}m$ at dentin surface. However, the resin-dentin hybrid zone was not identified by TEM. The evidence of resin penetration into intertubular and intratubular dentin did not show. 5. All the acid-conditioned groups showed that the detected calcium and phosphorus weight percent ratios at the $2{\mu}m$ upper portion from the resin-dentin interface into the resin were much higher than that at the $2{\mu}m$ lower portion from the resin-dentin interface to dentin. 성견의 발치와 협측골 외측에 적용한 교원질 차단막과 재조합 골형성단백질의 치조제 보존 효과 (Application of rhBMP-2 loaded collagen membrane on the buccal plate for ridge preservation: a pilot study in dogs)

  • Objective: The aim of this study was to determine the effect of multi-layer of a collagen membrane alone or loaded rhBMP-2 on the buccal plate for ridge preservation after tooth extraction. Material and methods: Following bilateral extraction of the maxillary 1st and 3rd premolars in five dogs, rhBMP-2 loaded collagen membrane was applied to the buccal plates at the 1st premolar and collagen membrane only was applied to the buccal plates at the 3rd premolar unilaterally. The collagen membranes applied in the experimental groups were piled into four layers. The corresponding sites of the contralateral side healed naturally. After 3months of healing, the animals were sacrificed. Radiographic and histologic analyses were performed. Results: There was no significant difference in the healing of extraction socket at both 1st and 3rd premolars. In microcomputed tomography, the widths of the residual ridge of the experimental groups were similar with the controls. Histologically, the experimental groups did not exhibit different pattern compared to the controls regardless of the addition of rhBMP-2. Conclusion: Layering of the collagen membrane with or without rhBMP-2 on the buccal plate failed to show the effectiveness in dimensional preservation of the extraction socket.

각막 간질 대체물로 콘드로이틴 설페이트가 결합된 콜라젠 스폰지의 생체 적합성 평가 (The Evaluation of Biocompatigbility of Collagen/Chondroitin Sulfate Sponge as a Scaffold for Corneal Stromal Layer)

  • 조직공학에서 생체재료의 생체적합성과 조직재생능력은 생체재료가 생체에 적합한지를 판단하는 가장 중요한 요소이다. 콜라젠은 인체조직을 이루는 주된 단백질이고, GAGs은 조직의 미세환경을 결정짓는 주요한 요소로 알려져 있다. 본 연구에서는 1형 콜라젠 스폰지를 탈수 열처리 (A군)하고 EDC로 가교결합한 것 (B군)과 CS를 첨가하여 EDC로 가교결합한 군 (C군)의 스폰지 형태의 콜라젠 지지체을 제조하였다. 제작된 콜라젠 스폰지를 3 mm의 디스크 형태로 토끼 각막의 실질부위에 주머니형태로 이식하였다. 8주 동안 각막의 신생혈관 생성, 혼탁, 지지체의 투명도 정도를 확인하고, 2주, 4주, 8주 후에 적출하여 염증과 각막 섬유모세포의 이동을 확인하였다. 모든 군에서 염증, 신생혈관 생성, 혼탁은 일어나지 않았다. 그러나 CS가 첨가된 콜라젠 스폰지에 섬유모세포의 이동이 많았고, 이식물의 투명도가 증가하였다. 1형 콜라젠 스폰지는 각막 간질에의 생체적합성이 뛰어나 각막 간질로의 대체 가능성이 크고, CS가 첨가된 1형 콜라젠 스폰지는 조직공학적 생인공각막의 재구성에 큰 도움을 줄 것으로 기대된다.
