Cognitive Enhancing Activity of Betula platyphylla Sap in Scopolamine Induced Amnesic Mice

  • Cognitive enhancing activity of Betula plantyphylla sap was determined in scopolamine induced amnesic mice using passive avoidance test. Oral acute administration of the sap effectively reversed memory deficit in a dose dependent manner. Then the sap was standardized on the basis of sugar contents using HPLC combined with refractive index detector. Glucose and fructose were the main sugars present in the sap.

  • This study reports on early adolescents' health information seeking behaviors that investigated cognitive, affective, and physical aspects of behaviors as they experienced to find information on their needs of health information seeking. In spite of the current widespread internet health information use by adolescents, little research exists to illuminate how they are engaged in cognitive, affective, and physical information behaviors in information search process. Qualitative data were collected through individual interviews informed by Kuhlthau's information search process. Forty adolescents from S city in South Korea participated in the project. Findings report thoughts, feelings, and actions aspects of information search process. This study expects to extend our knowledge of the adolescents' health information seeking behaviors of Kuhlthau's information search process.

Cognitive Enhancing Activity of the Steamed and Fermented Extracts of Codonopsis lanceolata Radix (양유(洋乳)의 증숙 및 발효 추출물의 인지능 개선 활성)

  • This study was designed to determine and compare the cognitive enhancing effect of various Codonopsis lanceolata extracts by steaming, fermentation and high hydrostatic pressure process. We prepared water extract of C. lanceolata, steamed C. lanceolata, steamed and fermented C. lanceolata and C.lanceolata by high hydrostatic pressure process and fermentation. Cognitive enhancing effect of extracts was evaluated in scopolamine-induced memory impairment mice using by passive avoidance test and Morris water maze tests. MTT assay was conducted to investigate neuroprotective effect on glutamate induced cell death in HT22 cells. Steamed and fermented C. lanceolata water extract decreased escape latency in Morris water maze test and increased the latency time of the passive avoidance test compared to other extracts. Furthermore, the steamed and fermented C. lanceolata water extract showed neuroprotective effect. These results suggest that steaming and fermentation process more improve cognitive enhancing effect of C. lanceolata than other process.

Odorant Stimulation Promotes Survival of Rodent Olfactory Receptor Neurons via PI3K/Akt Activation and Bcl-2 Expression

  • Olfactory stimulation activates multiple signaling cascades in order to mediate activity-driven changes in gene expression that promote neuronal survival. To date, the mechanisms involved in activity-dependent olfactory neuronal survival have yet to be fully elucidated. In the current study, we observed that olfactory sensory stimulation, which caused neuronal activation, promoted activation of the phosphatidylinositol 3'-kinase (PI3K)/Akt pathway and the expression of Bcl-2, which were responsible for olfactory receptor neuron (ORN) survival. We demonstrated that Bcl-2 expression increased after odorant stimulation both in vivo and in vitro. We also showed that odorant stimulation activated Akt, and that Akt activation was completely blocked by incubation with both a PI3K inhibitor (LY294002) and Akt1 small interfering RNA. Moreover, blocking the PI3K/Akt pathway diminished the odorantinduced Bcl-2 expression, as well as the effects on odorant-induced ORN survival. A temporal difference was noted between the activation of Akt1 and the expression of Bcl-2 following odorant stimulation. Blocking the PI3K/Akt pathway did not affect ORN survival in the time range prior to the increase in Bcl-2 expression, implying that these two events, activation of the PI3K pathway and Bcl-2 induction, were tightly connected to promote post-translational ORN survival. Collectively, our results indicated that olfactory activity activated PI3K/Akt, induced Bcl-2, and promoted long term ORN survival as a result.

  • 디지털 영상물에 대한 저작권을 보호하기 위해 저작권자의 고유한 영상을 기존 디지털 영상에 삽입하는 것을 워터 마킹이라고 부른다. 삽입된 워터 마크가 효과적으로 사용되기 위해서는 영상의 다양한 왜곡이나 변형에도 강건할 수 있게 그 강도가 강해야 하며, 동시에 워터 마크가 삽입된 영상의 왜곡을 식별할 수 없게 그 강도를 약하게 하여야 한다. 비가시성을 유지한 채 가능한 한 강한 워터마크를 삽입하는 것은 워터마크를 적용 하는 데 매우 중요한 문제이다. 대부분의 워터 마크 알고리듬에서 삽입될 워터 마크의 강도는 원영상과 워터마크 영상간의 물리적인 차이에 근거하여 계산된 특정 값으로 고정한 채, 이 값을 모든 영상에 동일하게 적용하고 있다. 인간의 경우 자극의 탐지는 주변 영상의 밝기나 대비 등 여러 요소에 의해 변화되므로 디지털 영상에 삽입된 워터 마크의 탐지도 영상의 특징에 따라서 변화될 수 있다. 본 연구에서는 원영상으로부터 세 수준의 밝기와 세 수준의 대비를 갖는 기본 영상들을 만든 후, 기본 영상에서 탐지될 수 있는 워터 마크의 강도가 변화되는 지를 살펴보았다. 연구 결과 영상의 밝기가 밝아질수록, 대비가 커질수록 지각될 수 있는 워터 마크의 강도는 더 커지는 것으로 나타났다. 또한 회귀분석을 통해 영상의 밝기와 대비에 근거하여 삽입될 워터 마크의 강도를 결정할 수 있는 회귀 식을 만들어 내었다. 이러한 회귀 식은 주어진 영상의 통계 치에 근거하여 워터 마크 강도를 적응적으로 변화시킬 수 있는 알고리듬을 개발하는 데 유용하게 사용될 수 있을 것이다.

Neural correlates of visual mean representation (시각적 평균 표상의 신경기제)

  • Visual scene contains lots of redundant information. To process this redundant information without increasing brain's volume, human visual system may summarize incoming information. If similar but different information are given to visual system, visual system extracts statistical properties of the information. One example of the statistical representation is representation of mean size. The mean representation is accurate and durable. The process of mean representation is suggested to be parallel. However, previous studies on the mean representation mostly used behavioral methods. The purpose of this study was to investigate which neural regions extracted the mean size of a set of circles using fMRI method. According to previous studies, BOLD signal of certain areas that were in charge of cousin stimuli decreased when the same stimuli presented repetitively. We used this paradigm and found that BOLD signal of right occipital area was decreased when same mean site was presented repeatedly. This results suggest that right occipital area is the locus of mean representation of visual stimuli.

  • 본 연구는 한국어 화장품 리뷰 코퍼스의 자질기반 감성 분석을 위하여, 이 도메인에서 실현되는 중요한 다단어 표현(MWE)의 유한상태 그래프 사전과 문법을 구축하는 방법론을 제시하고, 실제 구축된 사전과 문법의 성능을 평가하는 것을 목표로 한다. 본 연구에서는 자연어처리(NLP)에서 중요한 화두로 논의되어 온 MWE의 어휘-통사적 특징을 부분문법 그래프(LGG)로 형식화하였다. 화장품 리뷰 코퍼스에 DECO 한국어 전자사전을 적용하여 어휘 빈도 통계를 획득하고 이에 대한 언어학적 분석을 통해 극성 MWE(Polarity-MWE)와 화제 MWE(Topic MWE)의 전체 네 가지 하위 범주를 분류하였다. 또한 각 모듈간의 상호관계에 대한 어휘-통사적 속성을 반복적으로 적용하는 이중 증식(double-propagation)을 통해 자원을 확장하였다. 이 과정을 통해 구축된 대용량 MWE 유한그래프 사전 DECO-MWE의 성능을 테스트한 결과 각각 0.844(Pol-MWE), 0.742(Top-MWE)의 조화평균을 보였다. 이를 통해 본 연구에서 제안하는 MWE 언어자원 구축 방법론이 다양한 도메인에서 활용될 수 있고 향후 자질기반 감성 분석에 중요한 자원이 될 것임을 확인하였다.

C9orf72-Associated Arginine-Rich Dipeptide Repeat Proteins Reduce the Number of Golgi Outposts and Dendritic Branches in Drosophila Neurons

  • Altered dendritic morphology is frequently observed in various neurological disorders including amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD), but the cellular and molecular basis underlying these pathogenic dendritic abnormalities remains largely unclear. In this study, we investigated dendritic morphological defects caused by dipeptide repeat protein (DPR) toxicity associated with G4C2 expansion mutation of C9orf72 (the leading genetic cause of ALS and FTD) in Drosophila neurons and characterized the underlying pathogenic mechanisms. Among the five DPRs produced by repeat-associated non-ATG translation of G4C2 repeats, we found that arginine-rich DPRs (PR and GR) led to the most significant reduction in dendritic branches and plasma membrane (PM) supply in Class IV dendritic arborization (C4 da) neurons. Furthermore, expression of PR and GR reduced the number of Golgi outposts (GOPs) in dendrites. In Drosophila brains, expression of PR, but not GR, led to a significant reduction in the mRNA level of CrebA, a transcription factor regulating the formation of GOPs. Overexpressing CrebA in PR-expressing C4 da neurons mitigated PM supply defects and restored the number of GOPs, but the number of dendritic branches remained unchanged, suggesting that other molecules besides CrebA may be involved in dendritic branching. Taken together, our results provide valuable insight into the understanding of dendritic pathology associated with C9-ALS/FTD.

Protective role of caffeic acid in an Aβ25-35-induced Alzheimer's disease model

  • BACKGROUND/OBJECTIVES: Alzheimer's disease (AD) is characterized by deficits in memory and cognitive functions. The accumulation of amyloid beta peptide ($A{\beta}$) and oxidative stress in the brain are the most common causes of AD. MATERIALS/METHODS: Caffeic acid (CA) is an active phenolic compound that has a variety of pharmacological actions. We studied the protective abilities of CA in an $A{\beta}_{25-35}$-injected AD mouse model. CA was administered at an oral dose of 10 or 50 mg/kg/day for 2 weeks. Behavioral tests including T-maze, object recognition, and Morris water maze were carried out to assess cognitive abilities. In addition, lipid peroxidation and nitric oxide (NO) production in the brain were measured to investigate the protective effect of CA in oxidative stress. RESULTS: In the T-maze and object recognition tests, novel route awareness and novel object recognition were improved by oral administration of CA compared with the $A{\beta}_{25-35}$-injected control group. These results indicate that administration of CA improved spatial cognitive and memory functions. The Morris water maze test showed that memory function was enhanced by administration of CA. In addition, CA inhibited lipid peroxidation and NO formation in the liver, kidney, and brain compared with the $A{\beta}_{25-35}$-injected control group. In particular, CA 50 mg/kg/day showed the stronger protective effect from cognitive impairment than CA 10 mg/kg/day. CONCLUSIONS: The present results suggest that CA improves $A{\beta}_{25-35}$-induced memory deficits and cognitive impairment through inhibition of lipid peroxidation and NO production.

Effect of Water and Ethanol Extracts Codonopsis lanceolata on Spatial Learning and Memory in Mice (더덕 물 추출물과 에탄올 추출물의 인지능 개선 활성 비교)

  • Alzheimer's disease (AD), most common form of dementia is characterized that memory deficit and loss of cognitive function. This study was evaluated cognitive enhancing effect of water and ethanol extracts of Codonopsis lanceolata and compared using Morris water maze and passive avoidance test. The water and 70% ethanol extracts (100, 300 and 500 mg/kg) were administered to mice. The neuroprotective effect on glutamate-induced cell death in HT22 cells was additionally investigated using MTT assay. Results showed 70% ethanol extract of Codonopsis lanceolata enhanced cognitive function than water extract, as shown by decrease in escape latency time in Morris water maze test. In passive avoidance test, 70% ethanol extract also increased the latency time compared to the water extract. Furthermore, 70% ethanol extract significantly protected neuronal cell against glutamate cytotoxicity and showed higher than neuroprotective effect of water extract. These results indicate that 70% ethanol extract more improve spatial cognitive ability and protected neuronal cells than water extract.
