더덕의 花粉發芽에 관한 溫度와 培地條件 (Factors Influencing Pollen Germination in vitro of Codonopsis lanceolata)

  • The present study has been undertake to obtain the fundatmental data of optimum germination condition and to establish storage time for artificial pollinationin Codonopsis lancelata pollen. In vitro condition for germination of freshly collected and stored pollen were examined. The optium temperature for germination of fresh pollen was $25^{\circ}C$. The optium sucrose concentration in the medium ranged from 30 to 40 % and optium pH 6.0% for pollen germination. The rate of pollen germination accelerated considerably in the medium with 1% agar. 30% sucrose, and pH 6. C. lanceolata pollen remained viable for 15 days when stored at 5$^{\circ}C$ with silica gel as desiccant.

In Vitro evaluation of lipid accumulation inhibitory effect in 3T3-L1 cell and antioxidant enzyme activity of Codonopsis lanceolata using different solvent fractions

  • This study was conducted to evaluate the effect of anti-obesity and antioxidant enzyme activities in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The cytotoxicity of different solvent fractions of C. lanceolata on 3T3-L1 preadipocytes were evaluated using the MTT assay, the rate of cell survival progressively decreased in a dose-dependent manner. Butyl alcohol fraction at $200{\mu}g/mL$ exhibited a pronounced cytotoxic effect (75.73%) on 3T3-L1 cell comparable to that of the hexane fraction (79.82%), methylene chloride fraction (84.02%), ethyl acetate fraction (87.62%) and DW fraction (86.30%) at the same concentration. The Oil Red O solution was used to determine whether different solvent fractions of C. lanceolata induce adipocyte differentiation in 3T3-L1 preadipocytes. Confluent 3T3-L1 cells were treated with $50{\mu}g/mL$ concentration of solvent fraction extracts from C. lanceolata. Inhibitory degree of lipid accumulation against solvent fraction extracts showed a significant level compared with the control. Both lipid accumulation and adipocyte differentiation showed relatively high effect on methyl chloride fraction. The root extract of C. lanceolata had the highest SOD enzyme activity of 84.5% in ethyl acetate partition layer and while water partition layer of diploid showed the lowest SOD enzyme activity of 57.9%. The activity of CAT, APX and POD showed a significantly higher activity in ethyl acetate partition layer compared with the other fraction. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on anti-obesity and antioxidant capacity.

초고압 발효 더덕 추출물의 인지능력 개선 효과 (Memory Enhancing Effect of Codonopsis lanceolata by High Hydrostatic Pressure Process and Fermentation)

  • Alzheimer's disease (AD), most common form of dementia is characterized that memory deficit and loss of cognitive function. The Codonopsis lanceolata (C.lanceolata) was treated by high hydrostatic pressure process and fermentation. This study was evaluated cognitive enhancing effect C.lanceolata extract by high hydrostatic pressure process and fermentation and compared with common C.lanceolata extract using Morris water maze and passive avoidance test. And their neuroprotective effect on glutamate induced oxidative stress in HT22 cell was investigated by MTT assay. High hydrostatic pressure process and fermented C.lanceolata extract (HFCE) and common C.lanceolata extract (CCE) (100 and 300 mg/kg) were administered to mice. Results showed HFCE enhanced cognitive function than CCE as shown by decrease in escape latency time. HFCE increased the latency time of the passive avoidance test compared to CCE. Furthermore, HFCE showed significant neuroprotective effect against glutamate cytotoxicity in HT22 cells. These results indicate that high hydrostatic pressure process and fermented more improve spatial cognitive ability of C. laanceolata.

저장조건과 포장재에 따른 야산더덕의 향기성분의 변화 (Change of Volatile Flavor Components of Codonopsis lanceolata Cultivated on a Wild Bill and Stored at Various Conditions)

  • We investigated the changes of volatile flavor components from Codonopsis lanceolata, which were packed in woven polypropylene(WP) film or low density polyethylene(LDPE) film and stored for 15 and 30 days at refrigerated($2{\∼}4^{\circ}C$ ) or room($18{\∼}20^{\circ}C$) temperature. A hundred and sixty seven volatile flavor components in the fresh C. lanceolata were identified by GC/MS analysis. When determining the flavor components from C. lanceolata cultivated on a wild hill and stored at 4 conditions for 30days, six volatile components such as 1-hexadecene, 2,6-dimethyl-2-octanol, 2-methyl-2-dodecanol, $\alpha$-guaiene, $\delta$-cadinene and trans-2-hexen-1-ol were detected as common components of all stored samples, and The types of common flavor components of C. lanceolata were different according to storage conditions. The numbers were 16 from refrigerated, 7 from room temperature stored, and 10 components from WP or LDPE packed conditions, respectively. The total peak area ratio of the major 10 compounds were $52.0{\∼}86.8\%$, and the percentage of trans-2-hexen-1-ol, which was the only common compound among the major 10 components, was the highest as $26.4{\∼}68.1\%$ The major flavor profile, describe by highly trained panel, were green, aldehydic, earthy and camphoreous. As the result from sensory evaluation, the freshness of C. lanceolata was maintained better by controlling storage temperature rather than selection of package materials. The best condition for characteristic flavor of C. lanceolata was packing with LDPE and chilling.

더덕을 첨가하여 제조한 약주의 이화학적, 관능적 및 항산화 특성 변화 (Changes of Physicochemical, Sensory and Antioxidant Activity Characteristics in Rice Wine, Yakju Added with Different Ratios of Codonopsis lanceolata)

  • 쌀에 더덕을 각각 0, 10, 20, 30% 첨가하여 제조한 더덕 약주의 이화학적, 관능적 및 항산화 특성의 변화를 조사하였다. 더덕첨가량의 증가에 따라 pH는 감소되었고 총산의 함량은 증가된 것으로 나타났다. 총당의 함량은 더덕 첨가량의 증가에 따라 증가하였고 알코올 함량은 감소하였다. 색도 중 L값은 더덕 첨가량의 증가에 따른 변화가 없었으나 a와 b값은 증가한 것으로 나타났다. DPPH 자유라디칼 소거 활성은 더덕 첨가 시 증가된 것으로 나타났다. 관능검사 결과 전체적 기호도에서 20% 더덕을 첨가하여 제조한 약주가 제일 우수한 것으로 나타났다. 이화학적,관능적 및 항산화 특성으로 보아 20% 더덕을 첨가하여 약주를 제조하는 것이 적합하다고 생각된다.

산더덕과 재배더덕에 존재하는 토양미생물 및 향기 유발에 영향을 미치는 미생물 탐색 (Detection of soil microorganisms of an upland or cultivated Codonopsis lanceolata and investigation of them affecting on flavor substances)

  • We investigated microbial populations of an upland and cultivated Codonopsis lanceolata. The microbial populations from both types of soils were also investigated. There were more than 10 microorganisms existed in upland than cultivated one. The total viable cell counts of C. lanceolata from upland and cultivated one, especially in the upper zone, were 9.7x10$\^$6/ CFU/g and 4.2${\times}$10$\^$6/ CFU/g, respectively. As a results, upper parts of C. lanceolata in upland were considered to harbour approximately more than 2.3 fold higher microorganisms than in cultivated one. However, the total viable cell counts between the two soil habitat, that is, 1.2${\times}$10$\^$7/ CFU/g from upland and 1.0x10$\^$7/ CFU/g from cultivated, were not significantly different. We also examined the unique flavor producing microorganisms in the soil extract broth including 25% C. lanceolata extract. One microorganism was detected in upper pars of C. lanceolata and upland soil. No. 6, microorganism causing the characteristic flavor of C. lanceolata was continued as Actinomyces by microscopy.

Immune Cells Activity, Nitrite Scanvenging and ABTS Radical Scavenging Activities of Codonopsis lanceolata Ethanol Extracts from Districts in Korea

  • This study was executed to evaluate the immune activity, nitrite scavenging activity and ABTS radical scavenging activity against extracts of various concentration of ethanol solvent from Codonopsis lanceolata cultured at 6 local regions. The immune responses from both human T and B cell line was significantly enhanced in the cell growth compared to control while the cell growth was influenced at a certain period of culture. The results revealed that the cell growth of both human T and B cell was altered in a time dependent manner. The nitrite scavenging activity of ethanol extracts from various solvent concentration of C. lanceolata were affected by pH. At a pH of 1.2, the nitrite scavenging effect of all of the extracts tested observed higher than that of the other two pH ranges. There was no distinct detection of nitrite scavenging effects of the pH range 6.0. The ABTS radical scavenging activity was progressively increased in a dose-dependent manner, and the activity was the highest in 100% ethanol extract. The result from this investigation suggests that the extract of Codonopsis lanceolata could be an addition to basic medicine for immune modulation and natural food additives.

발효더덕의 화학성분 (Chemical Compositions of Fermented Codonopsis lanceolata)

  • 더덕(Codonopsis lanceolata Bench. et Hook.)은 예로부터 한방에서 염증을 제거하는 작용이 있고, 인후염이나 림프절 결핵에 효과가 있으며 오랫동안 먹어도 독성이 없는 상품(上品)의 한약자원이다. 민간에서는 고혈압 및 당뇨병 치료에도 이용하며, 최근에는 항암효과와 변비 예방 및 치료에 효능이 있는 식이섬유가 다량 함유되어 있는 것이 밝혀져 약용을 겸한 건강식품으로 주목을 받는 산채이다. 본 연구는 등급 외 더덕을 이용하여 발효더덕을 제조한 후 약이성 음식으로의 활용을 위한 가능성을 타진하고자 계획 수행되었다. 따라서 발효더덕의 영양성분 분석을 통한 식품영양학적 접근을 하고자 분석을 하였다. 식품영양학적 접근에서의 일반성분은 건량기준으로 당질(79.3%) 조단백질 (13.0%), 조지방(2.40%) 및 조회분(5.3%)이었고 각 시료 100 g의 함유열량은 390.5 kcal로 분석되었으며, 총 식이섬유소 함량은 건량기준으로 총 당질 중 47.4%를 나타내었다. 또한, 총 18 종의 아미노산으로 구성되었으며 필수아미노산(8,118.18 mg)과 비필수아미노산(10,913.42 mg) 함량을 나타내었으며, 무기질 중 칼륨의 함유량이 가장 높았고 그 다음이 인, 칼슘, 마그네슘 순으로 나타나 알칼리성 재료임을 알 수 있었으며, 지방산 함량의 경우 총 포화지방산(37.86%), 단일불포화지방산(3.84%) 및 다가불포화지방산(35.64%)으로 구성 되어 있어 다른 식물류에 비해 불포화지방산의 함량이 높은 것으로 나타났다. 조사포닌의 경우 60.1 mg/g이 함유되어 있는 것으로 나타났다. 또한, 유리당 함량의 경우 maltose (15.4%), glucose(8.9%), fructose(0.8%)의 함량을 나타내었다.

  • 더덕(Codonopsis lanceolata)은 주로 한국, 중국 등 동아시아 지역에 재배되고 있으며, 더덕의 뿌리는 기침, 기관지염, 천식, 결핵, 소화 불량의 증상을 치료하기 위한 기능성 식품 및 전통 의학으로 사용되어져 왔다. 보고된 바에 의하면 phenylpropanoids, polyacetylenes, saponins, flavonoids와 같은 다양한 식물 천연물 성분들이 항비만, 항염, 항암, 항산화, 항미생물 활성과 같은 약리학적 작용에 관여한다고 보고되어 있다. MS기반 대사체학 분석을 이용한 주산지의 마커 성분을 선정하는 것은 다른 지역에서 재배된 약용 식물의 안전성뿐만 아니라 화학적 조성과 생물학적 효능의 변화와도 관련이 있기 때문에 부작용 없이 더덕의 유익한 효과만을 보장하는데 중요하다. 본 연구에서는 국내산 더덕의 주산지 특성을 구별하기 위해 UPLC-QTOF-MS를 기반으로 하는 대사체 프로파일링과 다변량 통계분석 기법인 PCA 분석을 수행하여 판별모델을 확립하였다. 그 결과 인제(강원도), 횡성(강원도), 무주(전라북도)의 3개 그룹이 PCA와 loading plot 분석결과 tangshenoside I, lancemaside A, lancemaside G는 더덕 주산지를 구별하기 위한 잠재적 대사체 마커들로 제안하였다.

  • For the investigation of resources in Codonopsis species, the studies were carried out to evaluate the pharmaco-constituents from the leaves of Codonopsis lanceolata (Campanulaceae) whose roots have been used to antitussive, expectorant, detoxicate, tonic, edible, etc. as a folk medicine in Korea. From the EtOAc and BuOH fractions of MeOH extract, three flavonoid compounds, $luteolin-7-O-{\beta}-_D-glucopyranoside$$(C_{21}H_{20}O_{1i},\;mp\;254{\sim}255^{\circ}$, compound 1), $luteolin-5-O-{\beta}-_D-glucopyranoside$$(C_{21}H_{20}O_{1i},\;mp\;279{\sim}281^{\circ}$, compound 2) and luteolin $(C_{15}H_{10}O_6,\;mp\;327{\sim}330^{\circ}$, compound 3) were isolated and identified on the basis of their physicochemical properties and spectroscopic evidences(UV, IR, $^1H-NMR$, $^{13}C-NMR$, MS etc.) in comparison with authentics respectively.
