Weighted Gene Co-expression Network Analysis in Identification of Endometrial Cancer Prognosis Markers

  • Objective: Endometrial cancer (EC) is the most common gynecologic malignancy. Identification of potential biomarkers of EC would be helpful for the detection and monitoring of malignancy, improving clinical outcomes. Methods: The Weighted Gene Co-expression Network Analysis method was used to identify prognostic markers for EC in this study. Moreover, underlying molecular mechanisms were characterized by KEGG pathway enrichment and transcriptional regulation analyses. Results: Seven gene co-expression modules were obtained, but only the turquoise module was positively related with EC stage. Among the genes in the turquoise module, COL5A2 (collagen, type V, alpha 2) could be regulated by PBX (pre-B-cell leukemia homeobox 1)1/2 and HOXB1(homeobox B1) transcription factors to be involved in the focal adhesion pathway; CENP-E (centromere protein E, 312kDa) by E2F4 (E2F transcription factor 4, p107/p130-binding); MYCN (v-myc myelocytomatosis viral related oncogene, neuroblastoma derived [avian]) by PAX5 (paired box 5); and BCL-2 (B-cell CLL/lymphoma 2) and IGFBP-6 (insulin-like growth factor binding protein 6) by GLI1. They were predicted to be associated with EC progression via Hedgehog signaling and other cancer related-pathways. Conclusions: These data on transcriptional regulation may provide a better understanding of molecular mechanisms and clues to potential therapeutic targets in the treatment of EC.

PC-3 세포에서 cobalt chloride에 의해 down-regulation되는 puromycin-sensitive aminopeptidase의 apoptosis에 미치는 효과 (Cobalt Chloride-Induced Down-Regulation of Puromycin-Sensitive Aminopeptidase Involved in Apoptosis of PC-3 Cells)

  • 저산소상태(hypoxia)는 세포유형 및 성장조건에 따라 apoptosis를 유발하거나 또는 apoptosis의 진행을 억제한다. 저산소상태 유발물질인 cobalt chloride ($CoCl_2$) 역시 여러 세포 유형에서 apoptosis를 유발한다고 알려져 있으나 그 기작은 불명확하다. 본 연구에서는 PC-3 세포에서 $CoCl_2$에 의해 down-regulation되는 puromycin-sensitive aminopeptidase (PSA)의 세포 내 기능을 조사하였다. 본 연구 결과, PC-3 세포에 puromycin을 처리한 결과 전체 세포 집단의 약 42%에서 apoptosis가 유도되었다. PSA가 apoptosis에 관여하는지를 확인하고자 PSA siRNA로 내재성 PSA의 발현을 억제시킨 다음 apoptosis 연관 특성을 조사한 결과 PSA의 발현 억제에 의해 효과적으로 apoptosis가 유도되었다. 이러한 결과는 PC-3 세포에서 PSA가 세포생존에 중요한 역할을 할 것임을 보여주는 것이라 사료된다.

  • Biocides, or non-agricultural pesticides, are a broad class of chemical. They are including biological agents used to disinfect/sanitize, pesticides for non-agricultural use, wood preservatives, antifoulants, etc. Since the early 1980s, many adverse effects of biocides to human health and ecosystem have been found in the world. Especially, antifouling biocide like TBT caused serious toxic effects on the marine organisms. Therefore, OECD began to work on biocides in mid 1996 to help Member countries co-operate in the assessment and registration of these products. EU also announced the Biocidal Products Directive (BPD, 98/8/EC) in 1998 to harmonize regulatory approaches to allow EU countries to conduct evaluations of biocides more efficiently. Korea just start to consider of biocides regulation. Some biocides products are regulated, but not all the biocides which are using in Korea. Therefore, we need to make a appropriate regulation for the all biocides categories. In addition, there are necessary to develop risk assessment tools, to survey the use pattern and amount, to research on the ecosystem contamination by the biocides .

  • This paper presents a simple and cost effective circuit topology of single-ended type high frequency quasi-resonant PWM inverter using IGBTs, which can operate under wide soft switching operation range based on ZCS for main power switch as compared with a conventional active voltage-clamped ZVS-PWM high frequency quasi-resonant inverter developed previously. In principle, this new circuit topology can efficiently operate under a constant frequency PWM control-based power regulation scheme. In particular, it is noted that the zero current soft switching (ZCS) commutation can achieve for the main active power switch. On the other hand, the zero voltage soft switching (ZVS) commutation can also achieve for the auxiliary active power switch. The operating principle of this high-frequency Inverter treated here and its power regulation characteristics are illustrated on the basis of the simulation and feasible experimental results.

프로게스테론과 에스트라디올 $17{\beta}$에 의한 Long-Chain Acyl-CoA Synthetase 4의 발현 조절 (Regulation of Long-Chain Acyl-CoA Synthetase 4 Expression with Progesterone and Estradiol-$17{\beta}$)

  • Acyl-CoA synthetase 4(ACSL4) is an arachidonate-preferring enzyme abundant in steroidogenic tissues and postulated to modulate eicosanoid production. The human and mouse ACSL4 gene are mapped on chromosome X. The female mice heterozygous for ACSL4 deficiency became pregnant less frequent1y and produced small litters, with 40% of embryos surviving gestation. In this study, we examined the regulation of ACS4 by estradiol-$17{\beta}$ and progesterone (P4) in the human endometrial cancer cell line HTB-1B. ACSL4 mRNA was increased in a dose-dependent manner. Also, expression of ACSL4 gene was up-regulated in a time-dependent manner in HTB-1B cells. However, combined treatment with progesterone and estradiol-$17{\beta}$ modestly decreased the levels of ACS4L mRNA as compared with the estradiol-$17{\beta}$ and progesterone respectively. Overall, these results suggest that the ACSL4 gene is regulated by progesterone and estradiol-$17{\beta}$ in the HTB-1B cells.

류마티스 관절염에서 HO-1에 의한 VEGF 발현 유도에 대한 연구 (Effects of Heme Oxygenase-1 on VEGF Expression in Rheumatoid Arthritis)

  • Heme oxygenase-1 (HO-1), an inducible heme-degrading enzyme, is expressed by macrophages and endothelial cells in response to various stresses and mediators of inflammation. HO-1 has been recently implicated in regulation of angiogenesis via expression of VEGF. The purpose of this study was to determine the effects of HO-1 modulation on the collagen-induced arthritis (CIA) model and on angiogenesis via up- regulation of VEGF expression in human synovial fibroblast. DBA/1J mice were treated with an inhibitor of HO-1, tin protoporphyrin IX (SnPP), or with an inducer of HO-1, cobalt protoporphyrin IX (CoPP), from day 1 to day 35 after CIA induction. The clinical evolution of disease was monitored visually. At the end of the experiment, histopathologic changes were examined on the joints. VEGF expression in paws were measured by immunohistochemical stain. mRNA expression of HO-1 and VEGF stimulated with various concentration of $TNF-{\alpha}$, CoPP accessed on human synovial fibroblast by RT-PCR. Effects of pretreatment with SnPP on mRNA expression of HO-1 and VEGF in the presence of CoPP and $TNF-{\alpha}$ in synovial fibroblast was accessed by Real-time RT-PCR. Administration of cobalt protoporphyrin IX significantly induced the inflammatory response, with increased arthritis index and expression of VEGF in the paws of the arthritis models. Treatment with SnPP significantly reduced the severity of CIA through inhibition of joint inflammation and cartilage destruction. The expression of VEGF were also significantly reduced by SnPP treatment in the paw. CoPPIX as inducer of HO-1, increased HO-1 and VEGF expression dose dependently in synovial fibroblast. In contrast, inhibition of HO-1 activity by SnPPIX abrogated CoPPIX-induced HO-1 and VEGF production in synovial fibroblast. Stimulation with $TNF-{\alpha}$ increased HO-1 and VEGF expression itself and showed additive effect on HO-1 and VEGF expression when it treated with CoPP. When SnPP was treated with CoPP and $TNF-{\alpha}$, it abrogated the CoPP induced HO-1 and VEGF expression and also abrogated $TNF-{\alpha}$ induced HO-1 and VEGF expression in synovial fibroblast. The effects of HO-1 induction in rheumatoid arthritis results in aggravation of arthritis via up-regulation of VEGF. I concluded that inhibition of the expression or activity of HO-1 could be a therapeutic target of rheumatoid arthritis.

화석에너지와 CO2배출량 규제 하의 경제와 환경의 효율성 분석 (Analysis of the Economy and Environment Efficiencies under the Regulation of Fossil Fuel and Carbon Dioxide Emission)

  • 본 연구의 목적은 2000-2009년 국가별로 환경과 화석에너지의 제약 하에서 경제와 환경의 효율을 측정하고자 함이다. 본 연구는 국가별 소득수준에 따른 OECD국가, 중고소득국가, 저소득국가 등 세 그룹으로 구분하여, 생산가능집합에서 바람직한 산출물인 GDP와 오염물을 동시에 포함한 '환경지향형, 경제지향형, 환경-경제지향형, Two-stage DEA모형' 등의 네 가지 방법을 제시하여 비교한다. OECD국가는 상대적으로 경제효율성이 높으나 환경효율성은 낮았고 비OECD국가는 환경효율성이 높고 경제효율성이 낮았다. 화석에너지와 온실가스의 저감의 여지는 OECD국가가 더 높았다.

  • 전력계통의 주파수조정예비력(Frequency Regulation Control)은 부하 변동에 대응하여 규정된 주파수를 유지하고 안정된 전력계통 운영이 가능하도록 단시간 내에 응동할 수 있는 예비력이다. 본 논문은 우리나라 전력계통에 적합한 위와 같은 주파수조정예비력을 제시하고 전력산업 구조개편에 따라 분리된 발전사업자들이 전력시장에서 대부분의 주파수조정예비력을 담당하는 화력발전소 예비력 확보 방안을 연구 검토한 것이다.

  • HDAC and HAT (histone acetyltransferase) are involved in co-regulation in chromatin remodeling and the functional regulation of gene transcription. Abnormal recruitment of HDAC is related to carcinogenesis. Thus, the identification of potent histone deacetylase (HDAC) inhibitor has been considered as very intriguing approach for development for cancer chemotherapy. More recently, anti-inflammatory activity of SAHA cytokines was reported via reduction of proinflammatory cytokinres in vitro and in vivo. (omitted)

Emerging Co-signaling Networks in T Cell Immune Regulation

  • Co-signaling molecules are surface glycoproteins that positively or negatively regulate the T cell response to antigen. Co-signaling ligands and receptors crosstalk between the surfaces of antigen-presenting cells (APCs) and T cells, and modulate the ultimate magnitude and quality of T cell receptor (TCR) signaling. In the past 10 years, the field of co-signaling research has been advanced by the understanding of underlying mechanisms of the immune modulation led by newly identified co-signaling molecules and the successful preclinical and clinical trials targeting co-inhibitory molecules called immune checkpoints in the treatment of autoimmune diseases and cancers. In this review, we briefly describe the characteristics of well-known B7 co-signaling family members regarding the expression, functions and therapeutic implications and to introduce newly identified B7 members such as B7-H5, B7-H6, and B7-H7.
