Recycling of Wastepaper(V): -Calcium Hardness Control of Process Water for Zero-Discharge System- (고지재상연구 (제5보) -공정수 폐쇄화를 위한 칼슘경도 조절-)

  • A new technique for recycling of white water was developed in order to reduce the calcium hardness in a closed OCC recycling system. Calcium ions present in the white water were precipitated as calcium carbonate by reacting with sodium carbonate, and the precipitated $CaCO_3$ was removed from the system using a flotation fractionation method, which has been commonly used in deinking process. In the flotation stage, a mixed gas of $CO_2$-air was purged into the flotation cell because the pH of $Na_2CO_3$-treated white water was reduced to neutral by $CO_2$ gas. Since $CaCO_3$ precipitate tends to stick onto fine fiber surface and then selectively removed from the white water, a proper amount of suspended solid in white water acts as an important factor for deciding the removal efficiency. By the application of $Na_2CO_3$ addition-$CO_2$ flotation to the short circulated white water, the calcium hardness was significantly reduced by 87% and more. Removal of calcium ions with fine fibers led to a drainage improvement, reduction of fresh water consumption, and enhanced efficiency of wet-end chemicals.

The effect of Na2MoO4 addition on the formation and corrosion resistance of Cr-electroplated layer of low carbon steel (저탄소강의 크롬도금층 형성 및 내식성에 미치는 Na2MoO4 첨가의 영향)

  • Presence of cracks in electrodeposited hard chromium layer, which provide a path of corrosive media to steel substrate, is a serious issue in metal finishing with chromium electroplating. In this study, we added sodium molybdate in an electrolyte for chromium electroplating bath. 130g/L of sodium molybdate in Sargent bath for chromium electroplating causes a codepostion of molybdenum with chromium in a rage of 0.61 ~ 3.14 wt.%. The co-deposited molybdenum enhances the crystallinity of chromium layer, thus the hardness is slightly decreases by the addition of molybdate in electrolyte. However, due to the co-deposition of molybdenum, a crack-free chromium layer could be electrodeposited. Such crack-free chromium layer shows a significantly improved corrosion resistance.

Factors Affecting Reactivity of Various Phenolic Compounds with the Folin-Ciocalteu Reagent (다양한 페놀성 물질과 Folin-Ciocalteu 시약의 반응성에 미치는 영향 요인 평가)

  • The Folin-Ciocalteu (F-C) reagent has been extensively used for quantifying total phenolic contents in many different types of food materials. Since several different procedures of the assay methods using the F-C reagent have been applied, we investigated changes in reactivity of various phenolic compounds with the F-C reagent under three different assay conditions and factors affecting reactivity. Among 10 standard compounds tested, compounds with high hydroxyl density (number of -OH/molecular weight) showed a largely different response according to addition sequence of the F-C reagent or $Na_2CO_3$. Preincubation in $Na_2CO_3$ significantly reduced the reactivity of the phenolic compounds bearing galloyl moiety (e.g. gallic acid, tannic acid, and epigallocatechin-3-gallate) with the F-C reagent, while monophenol compounds including ferulic acid and sinapinic acid were more stable as compared to diphenols. There was little change in response to the F-C reagent of all phenolic compounds incubated in acidic pH; their reactivity except ferulic acid was reduced significantly when incubated in neutral or alkaline pH. Changes in reactivity of gallic acid incubated in $Na_2CO_3$ or neutral/alkaline pH conditions were the most prominent. $H_2O_2$ generated from phenolic compounds did not affect the reaction with the F-C reagents. The present results suggest that reactivity of different phenolic compounds with F-C reagent was affected considerably by different procedures of the assay, and the total phenolic contents could be fluctuated according to standard compounds and assay scheme.

Extraction Process Optimization of Poncirus trifoliata and Prunus mume for Antibacterial Activity against Vibrio parahaemolyticus (장염비브리오 항균활성을 위한 탱자와 매실의 추출조건 최적화)

  • The present investigation was carried out particularly to optimize the extraction process of Poncirus trifoliata and Prunus mume to develop a functional foodstuff having antibacterial activity against Vibrio parahaemolyticus. The extracts were prepared from the fruits under various conditions, and then optimum extraction conditions were decided in case maximal antibacterial activity was obtained. In extraction process by using hot water, the best antibacterial activity of Poncirus trifoliata was achieved at $80{\sim}100^{\circ}C$ for 30 min, while that of Prunus mume was achieved at $100^{\circ}C$ for 30 min, as showing 14.8 and 16.6 mm of inhibition diameter, respectively. The extraction process with addition of NaOH increased the activity of Prunus mume, but addition of $K_2CO_3$ and $NaHCO_3$ drastically decreased the activity. In the processes at high temperature up to $120^{\circ}C$ by using pressure extractor and with various concentrations of NaOH, the maximum activity was observed in the extract of Prunus mume at $120^{\circ}C$ for 30 min with 0.05 N NaOH. The fermented ethanol extract of the fruits showed less activity than those of the extracts with water and NaOH solution.

Optimization of Culture Conditions for the Bioconversion of Vitamin $D_3\;to\;1{\alpha}$,25-Dihydroxyvitamin $D_3$ Using Pseudonocardia autotrophica ID9302

  • We assessed the ability of a Pseudonocardia sp. from soil samples to bioconvert vitamin $D_3$. The optimal culture conditions for the bioconversion of vitamin $D_3$ to active $1{\alpha}$,25-dihydroxyvitamin $D_3$ were investigated by varying the carbon and nitrogen sources, the metal salt concentrations, the initial pH, and the temperature. Microbial transformations were carried out with the addition of vitamin $D_3$ dissolved in ethanol. They were sampled by extraction with methanol-dichloromethane and the samples were examined by HPLC. Optimum culture conditions were found to be 0.4% yeast extract, 1% glucose, 3% starch, 1% fish meal, 0.2% NaCl, 0.01% $K_2HPO_4$, 0.2% $CaCO_3$, 0.01% NaF, and pH 7.0 at $28^{\circ}C$. The optimal timing of the addition of vitamin $D_3$ for the production of calcitriol by Pseudonocardia autotrophica ID9302 was concurrent with the inoculation of seed culture broth. Maximum calcitriol productivity and the yield of bioconversion reached a value of 10.4mg/L and 10.4% respectively on the 7th day in a 75L fementer jar under the above conditions.

Domestic and International Trends in Technologies for Sodium Reduction (국내외 나트륨 저감 기술 동향)

  • Sodium chloride (NaCl) is a very important as one of major food ingredients in food industries. Recently, as the potential risk of adult diseases such as hypertension by overingestion of sodium, health authorities of many countries are executing policies for the reduction of sodium to suppress the overingestion of sodium by intake of NaCl. As general ways, the replacement of NaCl with either alternative salts, such as solar salts and minerals, for examples calcium, magnesium, potassium, lactic acid, and so on, and the addition of flavor enhancers were used to reduce the contents of sodium in foods. Recently, controls of particle size of sodium chloride or release point are emerging as new salt-manufacturing technologies for the sodium reduction. Upon reducing NaCl in foods it is important to develop practically adaptable technologies on the basis of the consideration of the unique functions of NaCl in foods, in particular effects on rheological characters, function as a humectant, shorten shelf life time, and so on.

Oxidation of Benzyl Ethers in Sodium Hypochlorite Mediated Piperidine-1-oxyl System (촉매량의 Piperidine-1-oxyl과 NaOCl계에서 벤질 에테르 유도체들의 산화 반응)

  • The oxidation of various benzyl ethers and benzyl alkyl ethers to benzoates has been studied in two-phase system of $CH_3CO_2Et$ and aqueous NaOCl (6.6 mol eq.). The oxidant N-oxo-4-methoxy-2,2,6,6-tetramethylpiperidium bromide (N-oxoammonium salt) was prepared in situ and recycled by addition of 4-methoxy-2,2,6,6-tetramethylpiperidine-1-oxyl (0.03 mol eq., 4-methoxy-TEMPO), co-catalyst KBr (0.03mol eq.) and second oxidant NaOCl. Thus the catalytic amount of 4-methoxy-TEMPO was used. An adjustment of the pH value of below 8.0 was also required for this reaction with 2.5 hr of reaction time at 0∼5$^{\circ}C$. Under these conditions benzyl ethers were oxidized to benzoates. The selectivity of oxidation of benzyl alkyl ethers is dependent on the acidity of hydrogen and steric effect of alkyl group.

Liming Materials and Desalinization of Marine Originated Tidal Soil (석회(石灰)의 종류(種類)와 해성간척지(海成干拓地) 토양(土壤)의 제염(除鹽))

  • In comparision with calcium sulphate, the effect of calcium-carbonate, -silicate and -hydroxide on desalinization of tidal saline soil was investigated in a continuous leaching column experiments after mixing with an equivalent amount of Ca to sodium plus magnessium in the saline soil. One half of liming materials was mixed to the top one-tenth of column soil and the remainder was spread on the surface. Results obtained are as follows ; 1. Gypsum made easy to percolate and desaline (Na) tidal marine soil but accumulated magnessium in subsoil. 2. $Ca(OH)_2$, $CaCO_3$, and $CaSO_3$ precipitated Mg in the soil which limes were mixed, but they washed down magnessium more severely from the immediate bellow the limed soil and less from the subsequent soil layers. This leaching was more severer at the treatment of $Ca(OH)_2$and lowest at the treatment of $CaSiO_3$. 3. The alkalinity of lime in addition to the dissociation of exchangeable Na raised pH of limed leached tidal soil and slowed down the percolation rate which retarded desalining Na from limed saline soils. This effect was most severe in the $Ca(OH)_2$ treated soil. 4. pH of leached soils was correlated possitively with exchangeable Na and negatively with exchangeable Mg giving follwing relationship pH= 7.77+0.489 Na/Mg r = 0.845.

Inhibitory Effects of Bile Acids on the Cholesterol Biosynthesis in Cultured Hepatocytes (배양 간세포내에서의 콜레스테롤 합성에 대한 담즙산의 저해효과)

  • The present work tested the inhibitory effects of bile acids on the cholesterol biosynthesis and the activity of HMG-CoA reductase in cultured rat hepatocytes. The uptake of bile acids in hepatocytes were increased in according to the different bile acid concentrations and culture times. The rate of cholesterol synthesis in cells were inversely decreased to the bile acid concentrations and culture times. As expected, insulin injection (4 units/100g body weight) showed an enhancing effect of the cholesterol synthesis and the HMG-CoA reductase activity. The addition of bile acids in medium of insulin-treated hepatocytes also showed the suppressing effect. This effect was directly confirmed in isolated hepatic icrosomes by the test of HMG-CoA reductase activity. In the test of $Na^+$,$K^+$-ATPase activity in the isolated hepatocyte membrane, only the cholic acid did not stimulate the enzyme system. The reason of such difference is not obvious, but this result indicates that the cholic acid could be absorbed by simple diffusion.

Effects of Starter Candidates and NaCl on the Production of Volatile Compounds during Soybean Fermentation

  • We inoculated different combinations of three starter candidates, Bacillus licheniformis, Staphylococcus succinus, and Tetragenococcus halophilus, into sterilized soybeans to predict their contributions to volatile compound production through soybean fermentation. Simultaneously, we added NaCl to soybean cultures to evaluate its effect on the volatile compounds profile. Cells in soybean cultures (1.5% NaCl) nearly reached their maximum growth in a day of incubation, while cell growth was delayed by increasing NaCl concentrations in soybean cultures. The dominance of B. licheniformis and S. succinus in the mixed cultures of three starter candidates switched to T. halophilus as the NaCl concentration increased from 1.5% to 14% (w/w). Seventeen volatile compounds were detected from the control and starter candidate-inoculated soybean cultures with and without the addition of NaCl. Principal component analysis of these volatile compounds concluded that B. licheniformis and S. succinus made major contributions to producing a specific volatile compound profile from soybean cultures where both species exhibited good growth. 3-Hydroxybutan-2-one, butane-2,3-diol, and 2,3,5,6-tetramethylpyrazine are specific odor notes for B. licheniformis, and 3-methylbutyl acetate and 2-phenylethanol are specific for S. succinus. Octan-3-one and 3-methylbutan-1-ol were shown to be decisive volatile compounds for determining the involvement of S. succinus in the soybean culture containing 7% NaCl. 3-Methylbutyl acetate and 3-methylbutan-1-ol were also produced by T. halophilus during soybean fermentation at an appropriate level of NaCl. Although S. succinus and T. halophilus exhibited growth on the soybean cultures containing 14% NaCl, species-specific volatile compounds determining the directionality of the volatile compounds profile were not produced.
